Comparison of Nitrogen Metabolism in Yak (Bos grunniens) and Indigenous Cattle (Bos taurus) on the Qinghai-Tibetan Plateau

  • The objective of the present study was to examine whether yaks possess any adaptive mechanisms of nitrogen (N) metabolism to survive in the harsh foraging environment of the Qinghai-Tibetan Plateau. A grazing experiment on native alpine meadows was conducted to determine availability of herbage biomass and body weight (BW) change of yaks over the year, followed by two indoor feeding trials to investigate adaptation mechanisms of N metabolism in yaks fed at similar intake level to grazing conditions. Three castrated males of each of three genotypes; yak (Bos grunniens), indigenous cattle (Bos taurus) and their crossbred - cattleyak (Bos taurus male${\times}$Bos grunniens female), were used in the housed trials. Results showed that: i) Monthly herbage biomass production and daily grazing intakes by yaks over the year ranged from 220 to 4,664 kg DM per ha, and 1.90 to 8.50 kg DM, respectively. For about seven months each year, yaks suffer from malnutrition as a result of inadequate pasture conditions; ii) Urinary N excretion and N retention by yaks were significantly affected by feeding level, and yaks had a lower (p<0.05) average daily urinary N excretion (0.39 g/kg $BW^{0.75}$) and a greater (p<0.05) N retention (-0.09 g/kg $BW^{0.75}$) than indigenous cattle (0.47 and -0.16 g/kg $BW^{0.75}$, respectively). Fasting daily urinary N excretion was greater (p<0.05) for indigenous cattle than yaks (353 vs. 248 mg/kg $BW^{0.75}$). Purine derivative N excretion and purine derivative N index (PNI) increased with increasing feeding level, while the value of PNI was greater (p<0.05) for yaks and cattleyak (0.11 and 0.12, respectively) than for indigenous cattle (0.09) during the feeding trials. These results suggest that yaks could rely, in part, on the recycling of N to adapt to the harsh forage environment on the Qinghai-Tibetan Plateau.), were used in the housed trials. Effect of Omija (Schisandra chinensis Baillon) Addition Ratio on Quality Characteristics of Mulberry Extracts (오미자 첨가율이 오디청의 품질 특성에 미치는 영향)

  • The purpose of this study was to investigate the quality characteristics of mulberry extracts added with Omija (Schisandra chinensis Baillon) in order to determine the best conditions to enhance sour taste of mulberry extracts during manufacturing. The total acidity of Omija was 5.71%, which was 20 times that of mulberry; sour taste of mulberry extracts could be enhanced by adding Omija during the extraction period. Overall, we observed reduction of pH and soluble solid contents as well as increased total acidity and anthocyanin content with a higher Omija adding ratio. In terms of taste acceptability, which is the most important parameter of beverage development, OC-3 (added with 18.75% Omija) and OC-4 (added with 25% Omija) values were $4.20{\pm}0.75$ and $4.05{\pm}0.80$, respectively, for mulberry extracts with Omija. This result was significantly different (P<0.05) from the control group (without Omija addition), which showed an OC-3/OC-4 value of $2.30{\pm}1.24$. The results of this study indicate that Omija addition at a ratio of 18.75~25% and extraction period of 10~12 days are optimum conditions to improve sour taste when developing mulberry extracts.

Study of Effective Cryoprotectants on the Cryopreservation of Porcine Mesenechymal Stem Cells (돼지 중간엽 줄기세포 동결에 있어서 동결보호제에 따른 특성 연구)

  • The objective of this study was to investigate the effective cryoprotectants for the cryopreservation of porcine mesenechymal stem cells (pMSCs). In order to understand the effectiveness of various cryoprotectants on pMSCs, we studied the most commonly used cryoprotectants; dimethyl sulfoxide (DMSO), ethylene glycol (EG), DMSO and EG. pMSCs were isolated from bone marrow matrix of piglet (2 month) and characterized by alkaline phopshatase (AP) activity, colony forming, and differentiation to adipocyte. In slow cooling cryopreservation, the pMSCs were exposed to cell medium containing Dulbecco's Modified Eagle's Medium (DMEM) supplemented with 10% DMSO, 1.5M EG and 5% DMSO/0.75M EG, respectively, and freezed to $-1^{\circ}C$/min from $25^{\circ}C$ up to $-80^{\circ}C$ in a cryo-container. The proportion of viable cells and the growing rates in fresh pMSCs were significantly (P<0.05) higher than those of other groups, but did not differ between the cryopreserved groups. The expression of Sox-2 and Nanog gene was increased by extending culture time in cryopreserved groups. The expression of Bax gene in cryopreserved groups was similar with fresh pMSCs. Moreover, the gene expression of adipocyte-specific marker as well as chondrogenic/osteogenic factors in cryopreserved groups was similarly to fresh pMSCs. Taken together, our results suggested that all these cryoprotectants of 10% DMSO, 1.5M EG and 5% DMSO/0.75M EG could be used for cryopreservation of the pMSCs.

Growth and Laying Performance of Japanese Quail Fed Graded Levels of Hazelnut Kernel Oil Meal Incorporated into Diets

  • Two experiments were conducted to evaluate the effect of substituting different levels of hazelnut kernel oil meal (HKOM) for dehulled soyabean meal (DSBM) in diets for Japanese quail. Five diets in which HKOM was replaced with 0, 25, 50, 75 and 100% of DSBM protein in a growing diet for a 5 week period using 450 Japanese quails of initial age of 1 week and in a layer diet for a 112 days period using 180 Japanese laying quails of initial age of 7 week were examined in experiments 1 and 2, respectively. Hence, treatment groups were: control (0 HKOM), 0.25 HKOM, 0.50 HKOM, 0.75 HKOM and 1HKOM. In the experiment 1, 1 HKOM decreased BWG compared with the 0 HKOM and 0.25 HKOM, while it increased FCR compared with the 0HKOM at day 21 (p<0.05). Compared with the other groups, 0.75 HKOM and 1 HKOM decreased BWG (p<0.01) at day 28. While the FCR of 1 HKOM was higher (p<0.01) than that of 0 HKOM, 0.25 HKOM and 0.50 HKOM, respectively, that of 0.75 HKOM was higher than that of 0 HKOM and 0.25 HKOM diets at day 28. However, at day 42 of age, BWG, FCR and the feed intake (FI) were not affected (p>0.05) by inclusion of HKOM. Mortality rate, carcase yield and liver, heart and gizzard weight (% of body weight) were not affected (p>0.05) by inclusion of HKOM to the diet. In the experiment 2, egg production was decreased (p<0.05) by the 1 HKOM diet compared with the 0.50 HKOM diet. While FCR increased by the 1 HKOM diet compared with the 0.25 HKOM and 0.50 HKOM diets (p<0.05). Feed intake for quails fed with the 0 HKOM diet was higher (p<0.05) than for quails fed with 0.50 HKOM diet. The egg yolk weight for 0 HKOM diet group was lower (p<0.05) than for quails in 1 HKOM diet group. In conclusion, the results indicate that DSBM can be replaced by HKOM in diets for growing and laying Japanese quails. However 50% HKOM has higher laying performance than 100% HKOM in the laying period.

Purification and Charaterization of Antimicrobial Peptide from Roots of Pokeweed (미국자리공(Phytolacca americana L.) 뿌리의 항균 펩타이드 정제 및 특성연구)

  • An antimicrobial peptide was purified from the roots of Phytolacca americana L. and was designated as PAMP-r. Purification was carried out by DEAE-cellulose anion exchange, sephadex G-75 gel filtration, Mono S cation exchange, and Resource RPC reverse phage chromatography. The molecular weight of PAMP-r was estimated to be about 4,900 Da by 15% SDS-PAGE under reducing condition. PAMP-r exhibited a broad spectrum of antimicrobial activity. PAMP-r was stable against heat and pH treatment; its activity was not diminished by the heat treatment up to $80^{\circ}C$ for 30 min, and it showed a pH stability in the range between pH 3.0 to pH 8.0.
