Characterization of Agarase from an Isolated Marine Bacterium, Simiduia sp. SH-1 (해양성 Simiduia sp. SH-1 균주의 분리 및 한천분해효소의 특성조사)

  • Agarase from a novel agar-degrading bacterium isolated from seawater in Namhae at Gyeongsangnamdo province of Korea was characterized. The SH-1 strain was selected from thousands of colonies on Marine agar 2216 media. Almost full 16S rRNA gene sequence of the agarolytic SH-1 strain showed 99% similarity with that of bacteria of Simiduia genus and named as Simiduia sp. SH-1. Agarase production was growth related, and activity was declined from stationary phase. Secreted agarase was prepared from culture media and characterized. It showed maximum activity of 698.6 units/L at pH 7.0 and 30℃ in 20 mM Tris-HCl buffer. Agarase activity decreased as the temperature increased from an optimum of 30℃, with 90% and 75% activity at 40℃ and 50℃, respectively. Agarase was not heat resistant. Slightly lower agarase activity was observed at pH 6.0 than at pH 7.0, without statistical difference, and 80% and 75% activity were observed at pH 5.0 and 8.0, respectively. Neoagarotetraose and neoagarobiose were the main final products of agarose, indicating that it is β-agarase. Simiduia sp. SH-1 and its β-agarase would be useful for the industrial production of neoagarotetraose and neoagarobiose, which have a whitening effect on skin, delaying starch degradation, and inhibiting bacterial growth.

An Analysis of the Effect of the Pilot Project for the Management of the Elderly's Food Service in Cheongju City: Focused on improving hygiene control (청주시 노인급식 관리지원 시범사업의 효과분석 : 위생관리개선을 중심으로)

  • In this study, the hygiene control status of meals for the elderly was investigated for the elderly care facilities, communal living families, and day care centers, which are members of the Cheongju Social Welfare Food Management Support Center, and the improvement of food hygiene management was compared and analyzed to find out the effectiveness of the pilot project after conducting management support such as hygiene education for the elderly and providing related supplies and information. The study found that after the first support, the average score of overall feeding hygiene control increased significantly from 63.13 points (out of 100) to 75.10 points (p<0.001) and from 75.10 points (out of 100) to 80.89 points (p<0.001). After primary support, 16 out of 29 items showed significant differences in average scores in the checklist, and 7 out of 29 after secondary visit support (p<0.05, p<0.01, p<0.001). Based on the effectiveness of the above-mentioned pilot project to support senior citizens' meal service management, more welfare facilities for senior citizens should be continuously supported by the Foodservice Management Support Center.

CHANGES IN LIVE-WEIGHT GAIN, BLOOD CONSTITUENTS AND WORM EGG COUNTS IN THAI NATIVE AND CROSS-BRED GOATS RAISED IN VILLAGE ENVIRONMENTS IN SOUTHERN THAILAND

  • The aim of the present study was to investigate the effect of internal parasites on growth rates of Thai Native (TN) and crossbred (75% TN $\times$ 25% Anglo-Nubian, AN and 50% TN $\times$ 50% AN) goats (undrenched, drenched every 3 weeks or at 9 weeks) in village environments in southern Thailand in a humid tropical climate. There was no significant (p > 0.05) difference in growth rate ($g/kg^{0.75}/d$) between the genotypes during unsupplemented grazing (0-64 days of the experimental period). However, during supplementary feeding (64-127 days) and throughout the period (0-127 days) TN goats had significantly (p < 0.01) lower growth rates compared with 75% TN $\times$ 25% AN and 50% TN $\times$ 50% AN goats. There was no (p > 0.05) significant difference in growth rates between 75% TN $\times$ 25% AN and 50% TN $\times$ 50 % AN goats. The growth rates of goats drenched every 3 weeks were significantly (p < 0.01) higher than those undrenched or drenched at 9 weeks. The results of this study also indicate that drenching alone did not result in increased weight gain except when the nutritional status was also improved. Parasitic infection affected some blood constituents, such as pack cell volume, haemoglobin, total protein and albumin. This resulted in lower growth rates for control groups and goats drenched at 9 weeks compared to those of goats drenched every 3 weeks.

Fermentation Properties of Yulmoo Mulkimchi According to the Ratio of Water to Yulmoo (물비율을 달리한 열무 물김치의 발효특성)

  • In the fermentation of Yulmoo Mulkimchi, various ratios of Yulmoo to water (l/l.14, l/l.5, 1/2, l/2.75, 1/4) were prepared and fermented at 4$^{\circ}C$, 15$^{\circ}C$, 25$^{\circ}C$ for up to 10 days. According to the fermentation time, the pH, acidity, total vitamin C content and microbial growth in Mulkimchi samples were determined together with sensory evaluation. Fermentation temperature on water addition ratio didn't show any difference in pH and microbial growth of Mulkimchi. However, low ratio of water resulted in high acidity and vitamin C content in Mulkimchi. In terms of acid odor and acid taste, the least water addition (l/l.4) sample was significantly strong than those of other samples. The ratio of Yulmoo to water, l/2 showed the highest overall sensorial acceptability and followed by l/l.5, l/l.4, l/2.75 and 1/4 samples. It was found that the content of vitamin C and acid taste of Mulkimchi have correlation with its acceptability.

Electromyographic Activation of Lumbar Multifidus Muscle Induced by Horizontal Shoulder Extension Exercise: Comparison of Four Intensity Levels of the Exercise (등척성 견관절 수평 신전 운동의 강도에 따른 다열근의 활성도 비교)

  • Purpose : To identify whether isometric shoulder horizontal extension (ISHE) exercise could sufficiently activate the lumbar multifidus muscle, and to determine appropriate exercise intensity to increase muscle strength. Methods : Twenty healthy volunteers (10 males and 10 females) participated in this study. ISHE exercises on the dominant side were performed with $90^{\circ}$ shoulder abduction held in $90^{\circ}$ elbow flexion so that their arms were in the horizontal plane. Electromyographic (EMG) measurements of multifidus activation were performed in standing and supine positions, and were taken under four strength conditions: 75%, 50% and 25% of maximum shoulder horizontal extension strength, and maximum strength. Results : The EMG activations of both lumbar multifidus increased significantly with ISHE exercises of larger % strength (p<.05). In their multifidus EMG data measured in standing and supine positions, men differed significantly in their 75%, 50% and 25% strength for both sides (p<.05), and women differed significantly in their 75%, 50% and 25% strength on their 5th lumbar vertebrae's left side, but 75% only for their 5th lumbar vertebrae's right side (p<.05). The EMG data of 5th lumbar vertebrae's left and 5th lumbar vertebrae's right sides appeared to differ significantly at all strength levels for men while standing, but only at the 25% level for women in the supine position (p<.05). These findings indicate that ISHE exercises can be considered a beneficial method to enhance the multifidus strength. Conclusion : This study provides useful information for further study in this field.

Fabrication of $0.25 \mu\textrm{m}$ P-HEMT for X-band Low Noise Amplifier (X-밴드 저잡음 증폭기용 $0.25 \mu\textrm{m}$ T-형 게이트 P-HEMT 제작)

  • We have enhanced the yield of 0.25 ${\mu}{\textrm}{m}$ T-gate $Al_{0.25}$G $a_{0.75}$As/I $n_{0.2}$G $a_{0.8}$As P-HEMT using three-layer E-beam lithography process and selective etching process. The three-layer resist structure (PMMA/copolymer/ PMMA=2000 $\AA$/3000 $\AA$/2000 $\AA$) and three developers (Benzene:IPA=1:1,Methanol:IPA =1:1,MIBK:IPA=1:3) were used for fabrication of a wide-head T-gate by the conventional double E-beam exposure technology. Also 1 wt% citric acid: $H_2O$$_2$:N $H_{4}$OH(200m1:4ml:2.2ml) solution were used for uniform gate recess. The etching selectivity of GaAs over $Al_{0.25}$G $a_{0.75}$As is measured to be 80. So these P-HEMT processes can be used in X-band MMIC LNA fabrication.ion.ion.ion.

  • This was the report of Hydroxyproline detection in silk fibroin and the obtained results were as followings. 1. Hydroxyproline was analized 25∼75 p.p.m. in specific species silk fibroin. 2. The amino acid was included more in crossed species silk fibroin than in the original species fibroin. 3. The number of amino acids in silk fibroin increased from 18 to 19 kinds.
