Investigation of Antimicrobial Activity and Stability of Orixa japonica Thunb. Leaf Extract (상산나무 잎 추출물의 항균활성 및 안정성 조사)

  • The antimicrobial activity of Orixa japonica Thunb. leaf extract towards 13 microorganism strains was evaluated. Both methanol (MEex) and 70% ethanol extracts showed antimicrobial activity towards Streptococcus mutans, Bacillus cereus, Staphylococcus aureus, and Pseudomonas aeruginosa. MEex showed a higher antimicrobial activity than the 70% ethanol extract. In addition, the dichloromethane fraction (DCMfr) of the MEex also had an antimicrobial effect against the microorganisms examined. The minimum inhibitory concentrations (MICs) towards S. mutans, B. cereus, S. aureus, and P. aeruginosa were 49.22, 24.61, 49.22, and 49.22 mg/mL, respectively. In contrast, the MICs of the DCMfr tpwards S. mutans, B. cereus, S. aureus, and P. aeruginosa were 3.31, 0.21, 1.7, and 1.7 mg/mL, respectively. The MEex antimicrobial activity was not affected by a 3 h exposures to pH in the range of 3-11 or by temperatures were maintained between $80^{\circ}C-100^{\circ}C$ for 6 h. However, the MEex antimicrobial activity decreased at a heat treatment of $121^{\circ}C$ 1 h.

  • Green tea, a leaf of the plant Camellia sinensis, is one of the most consumed traditional oriental beverages. Green tea has been considered a medicine and a healthful beverage since ancient times, but recently it has received a great deal of attention because of its antioxidants like polyphenols. Moreover, green tea contains amino acids, carbohydrates, proteins, chlorophyll, volatile compounds, minerals, and phytochemical components that are essential or helpful to human health. Depending on the manufacturing process, green teas are classified into several types. Among these, powdered green tea can be effective in the absorption of ingredients compare with other types of green tea since we take the beverage with powder itself. In this paper, the contents of general ingredients (moisture, proteins, fat, carbohydrates, and ash), minerals (Fe, Mg, Ca, Na, K, and P), hunter color values, and alcohol insoluble substance were determined in total of six powdered green teas commercialized in Korea and Japan.

Antimicrobial Activity of Extracts from Different Parts and Essential Oil from Pinus densiflora on Skin Pathogens (소나무 부위별 추출물 및 essential oil의 피부상재균에 대한 항균 활성)

  • This study was carried out to examine the antimicrobial activity of 70% ethanol and hot water extracts from different parts and essential oil in Pinus densiflora on skin pathogens such as Staphylococcus epiermidis, S. aureus, Propionibacterium acnes, Candida albicans, C. tropicalis, and Trichophyton rubrum. The antimicrobial activities of extracts and essential oil were tested by paper disc assay and minimum inhibitory concentration test. The ethanol extract of pine pollen showed antimicrobial activity against C. tropicalis and T. rubrum. Ethanol extract of pine needle and pine gnarl showed antimicrobial activity against S. aureus, C. tropicalis, and P. acnes. Essential oil from pine needle exhibited antimicrobial activity against C. albicans, C. tropicalis, and P. acnes. The minimum inhibitory concentration of ethanol extracts of pine needle and pine gnarl against P. acnes and C. tropicalis ranged from 0.002% to 0.0063%. The lowest minimum inhibitory concentration (0.025%) against C. albicans and C. tropicalis was obtained from essential oil. These results indicate that ethanol extracts of pine pollen, pine needle, pine gnarl, and essential oil of pine needle could be applicable to control the skin infection pathogens. Especially, ethanol extract of pine gnarl had a broad spectrum of antimicrobial activity and pine extracts and essential oil exhibited higher antimicrobial activity with Candida sp. and P acnes.

Study on Effects of Media, EGF, ${\beta}-ME$ and Hormones on IVM of Porcine Oocytes (배양액 종류, EGF, ${\beta}-ME$ 및 호르몬이 돼지 난자의 체외 성숙율에 미치는 영향에 관한 연구)

  • 본 연구는 안정된 돼지 체외 성숙 난자를 얻을 목적으로 배양액의 종류 및 배양액에 EGF, ${\beta}-ME$, 호르몬 첨가가 돼지 난포란의 체외 성숙에 미치는 영향을 조사하였다. 난포란을 TCM-199, NCSU-23 및 PZM-3으로 48시간 배양했을 때 체외 성숙율은 각각 $22.1{\pm}0.70%,\;30.6{\pm}0.70%$$30.4{\pm}2.82%$였다. TCM-199로 48시간 배양했을 때 체외 성숙율은 NCSU-23 및 PZM-3 보다 약간 낮은 체외 발생율을 나타냈다. 난포란을 25 ng/ml의 EGF를 첨가한 TCM-199, NCSU-23 및 PZM-3로 48시간 배양했을 때 체외 성숙율은 각각 $46.3{\pm}2.8%,\;76.6{\pm}3.1%$$72.2{\pm}2.6%$로 나타났다. 난포란의 배양 시 배양액에 25 및 50 ng/ml의 EGF를 첨가 후 48시간 배양했을 때 첨가하지 않은 군에 비해 높은 체외 성숙율을 나타냈다(p<0.05). 난포란을 NCSU-23 및 PZM-3 배양액에 $25{\mu}M/ml$${\beta}-ME$를 첨가한 후 48시간 배양했을 때 체외성숙율은 각각 $43.9{\pm}1.41%,\;41.7{\pm}l.41%,\;44.4{\pm}0.70%,\;40.6{\pm}0.70%$로 나타났다. 난포란을 $25{\mu}M/ml$${\beta}-ME$를 첨가한 NCSU-23로 48시간 배양했을 때 첨가하지 않은 군에 비해 높은 체외 성숙율을 나타냈다(p<0.05). 난포란의 배양 시 NCSU-23에 PMSG, hCG, PMSG+hCG, hCG+${\beta}$-estradiol, PMSG+${\beta}$-estradiol을 첨가 후 배양하였을 때 체외 성숙율은 각각 75.6%, 77.8%, 80.0%, 86.4% 및 84.8%로서 무첨가 군(64.4%)에 비해 높게 나타났다(p<0.05).

A Study on the Effects of Sirtuin 1 on Dendritic Outgrowth and Spine Formation and Mechanism in Neuronal Cells (신경세포에서 sirtuin 1이 수상돌기 성장과 가시형성에 미치는 영향 및 기전에 관한 연구)

  • Increasing evidence suggests that depression is associated with impairments in neural plasticity. Sirtuin 1 plays an important role in neural plasticity, and the activation of mechanistic target of rapamycin complex 1 (mTORC1) signaling is known to improve neural plasticity. In this study, we aimed to determine whether sirtuin 1 affects dendrite outgrowth and spine formation through mTORC1 signaling. Resveratrol (sirtuin 1 activator; 1 and 10 μM) and sirtinol (sirtuin 1 inhibitor; 1 and 10 μM) were treated in primary cortical culture with and without dexamethasone (500 μM). Levels of sirtuin 1, phospho-extracellular signal regulated protein kinase 1/2 (ERK1/2), phospho-mTORC1, and phospho-p70 ribosomal protein S6 kinase (p70S6K) were evaluated using Western blot analysis. Dendritic outgrowth and spine density were assessed using immunostaining. Resveratrol significantly increased levels of sirtuin 1 expression and phosphorylation of ERK1/2 (a downstream target of sirtuin 1), mTORC1, and p70S6K (a downstream target of mTORC1) in a concentration-dependent manner under dexamethasone conditions. Resveratrol also significantly increased dendritic outgrowth and spine density. Conversely, sirtinol significantly decreased levels of sirtuin 1 expression and phosphorylation of ERK1/2, mTORC1, and p70S6K in a concentration-dependent manner under normal conditions. Moreover, sirtinol significantly decreased dendritic outgrowth and spine density. Consistent with the results of sirtinol, sirtuin 1 knockdown using sirtuin 1 siRNA transfection significantly decreased dendritic outgrowth and spine density as well as phosphorylation levels of ERK1/2 and mTORC1. These data suggest that sirtuin 1 enhances dendritic outgrowth and spine density by activating mTORC1 signaling.

Isolation of an Agarase-producing Persicobacter sp. DH-3 and Characterization of its β-agarase (Agarase를 생산하는 Persicobacter sp. DH-3의 분리 및 β-agarase의 특성)

  • The purpose of this study was to isolate a new marine agarase-producing bacterium. Agarase can hydrolyze agar and agarose to produce agarooligosaccharides or neoagarooligosaccharides, which possess many physiological functions. Strain DH-3 was isolated from seawater collected from the coast of Yeosu at Jeollanam province, Korea. A 16S rDNA sequence analysis showed this strain to be Persicobacter sp. DH-3. Extracellular agarase was prepared from culture media of Persicobacter sp. DH-3 and used for characterization. Relative activities at 20, 30, 40, 50, 60, and $70^{\circ}C$ were 50, 55, 70, 100, 90, and 50%, respectively. Relative activities at pH 5, 6, 7, and 8 were 75, 100, 90, and 75%, respectively. The enzyme showed maximum activity at $50^{\circ}C$ in a 20 mM Tris-HCl buffer at pH 6. This enzyme could be useful, as agar is in liquid state at $50^{\circ}C$. Agarase activities were maintained at 80% or more for 2 hr at 20, 30, and $40^{\circ}C$. Thin layer chromatography analysis suggested that Persicobacter sp. DH-3 produced extracellular ${\beta}$-agarases as it hydrolyzed agarose to produce neoagarohexaose and neoagarotetraose. In addition, zymogram analysis confirmed that Persicobacter sp. DH-3 produces at least three agar-degrading enzymes with molecular weights of 45, 70, and 140 kDa. Therefore, it is expected that agarases from Persicobacter sp. DH-3 could be used to produce functional neoagarooligosaccharides.

Isolation and Characterization of Peroxidase from Jerusalem Artichoke Tubers (돼지감자 Peroxidase의 분리와 특성)

  • Peroxidase from Jerusalem artichoke tubers, which might be related to browning reaction, was purified by ammonium sulfate precipitation, DEAE-cellulose and Sephacryl S-200 chromatography. The optimum pH of the purified peroxidase was 5.0 and relatively stable at pH $5.0{\sim}6.0$ using substrate of p-phenylenediamine and $H_2O_2$. D-values for thermal inactivation at 60, 70 and $80^{\circ}C$ were 86, 45 and 33 sec, respectively. Activation energy was 4,111 J/mole. The enzyme showed the most sensitive specificity of substrate for p-phenylenediamine. The compounds such as 1mM potassium cyanide, 10mM sodium diethyldithiocarbamate, L-ascorbic acid, sodium hydrosulfite and L-cysteine inhibited completely while 1mM of $Ca^{2+}\;and\;Cu^{2+}$ activated the purified peroxidase.

The Effects of Proprioceptive Neuromuscular Facilitation and Static Stretching Performed at Various Intensities on Hamstring Flexibility (다양한 강도에서 수행된 고유감각신경근육촉진 스트레칭과 정적 스트레칭이 뒤넙다리근의 유연성에 미치는 효과)

  • Background: To prevent or reduce the risk of strain injury, various approaches, including stretching techniques are currently being used. The effect of proprioceptive neuromuscular facilitation (PNF) and static stretching on flexibility has been demonstrated; however, it is not clear which one is superior. Objects: This study aimed to evaluate the differences between the effects of PNF and static stretching performed at various intensities on muscle flexibility. Methods: The maximum voluntary isometric contraction (MVIC) of the hamstrings using the PNF stretching technique was performed in the P100 group, while 70% of the MVIC was performed in the P70 group. The MVIC value obtained during the PNF stretching in both groups was used as a reference for setting the intensity of static stretching. Static stretching was performed at 130% (S130), 100% (S100), and 70% of the MVIC (S70). The active knee extension (AKE) values, defined as the knee flexion angle were measured before stretching (baseline), immediately after stretching (post), and at 3 minutes, 6 minutes, and 15 minutes. Results: PNF stretching produce a greater improvement in flexibility compared with static stretching. Specifically, the ΔAKE was significantly higher in the S100 and S70 groups than in the P100 group at Post. In the comparison of ΔAKE over time in each group, the ΔAKE at Post showed a significant decrease compared to the value at Baseline in the S130 group; however, no significant difference was observed at 6 minutes while a significant increase was noted at 15 minutes. Conclusion: This study found that PNF stretching is more effective than static stretching with respect to increasing and maintaining the flexibility of muscles. In addition, the increase in flexibility at maximal intensity was similar to that observed at submaximal intensity during both PNF and static stretching.

Growth signaling and longevity in mouse models

  • Reduction of insulin/insulin-like growth factor 1 (IGF1) signaling (IIS) extends the lifespan of various species. So far, several longevity mouse models have been developed containing mutations related to growth signaling deficiency by targeting growth hormone (GH), IGF1, IGF1 receptor, insulin receptor, and insulin receptor substrate. In addition, p70 ribosomal protein S6 kinase 1 (S6K1) knockout leads to lifespan extension. S6K1 encodes an important kinase in the regulation of cell growth. S6K1 is regulated by mechanistic target of rapamycin (mTOR) complex 1. The v-myc myelocytomatosis viral oncogene homolog (MYC)-deficient mice also exhibits a longevity phenotype. The gene expression profiles of these mice models have been measured to identify their longevity mechanisms. Here, we summarize our knowledge of long-lived mouse models related to growth and discuss phenotypic characteristics, including organ-specific gene expression patterns.

Antimutagenic and Antibacterial Activities of Korean and American Propolis (한국산과 미국산 프로폴리스의 항돌연변이 및 항균효과)

  • The antimutagenic activities of ethanol extracts of Korean and American propolis were tested using Salmonella Typhimurium TA98 with two indirect mutagens of 3-amino-1,4-dimethyl-5H-pyrido [4,3-b]indole (Trp-P-1) and 2-aminoanthracene (2-AA) with S9 mix. Additionally, their antimicrobial activities against acne-related pathogenic strains of Propionibacterium acnes, Staphylococcus Epidermidis, Staphylococcus aureus and Pseudomonas aeruginosa were evaluated using both paper disk method and agar dilution method. Ethanol extracts of Korean and American propolis showed strong inhibitory effects, in a dose dependant manner, against the mutagenicities induced by Trp-P-1 and 2-AA. The antimutagenic effect of ethanol extracts of Korean propolis showed significantly higher protective activity than that of American propolis against the Trp-P-1 induced mutagenicity of S. Typhimurium TA98 at the lower concentration ($1-10\;{\mu}g$), but significantly lower protective activity at the higher concentration ($50-200\;{\mu}g$). The antimutagenic effect of ethanol extract of Korean propolis showed significantly higher protective activity than that of American propolis against the 2-AA induced mutagenicity at the concentration of $1\;{\mu}g$, but significantly lower protective activity than that of the American at the higher concentration ($50-200\;{\mu}g$). Both extracts showed strong antimicrobial activities against all the acne-related pathogens tested, with minimal inhibitory concentration (MIC) values in the range $1,500-5,000\;{\mu}g/mL$.
