Dephosphorylation of DBC1 by Protein Phosphatase 4 Is Important for p53-Mediated Cellular Functions

  • Deleted in breast cancer-1 (DBC1) contributes to the regulation of cell survival and apoptosis. Recent studies demonstrated that DBC is phosphorylated at Thr454 by ATM/ATR kinases in response to DNA damage, which is a critical event for p53 activation and apoptosis. However, how DBC1 phosphorylation is regulated has not been studied. Here we show that protein phosphatase 4 (PP4) dephosphorylates DBC1, regulating its role in DNA damage response. PP4R2, a regulatory subunit of PP4, mediates the interaction between DBC1 and PP4C, a catalytic subunit. PP4C efficiently dephosphorylates pThr454 on DBC1 in vitro, and the depletion of PP4C/PP4R2 in cells alters the kinetics of DBC1 phosphorylation and p53 activation, and increases apoptosis in response to DNA damage, which are compatible with the expression of the phosphomimetic DBC-1 mutant (T454E). These suggest that the PP4-mediated dephosphorylation of DBC1 is necessary for efficient damage responses in cells.

결장암세포에서 sanguinarine에 의한 종양억제 유전자 p53 의존적 apoptosis 유도 (Induction of Tumor Suppressor Gene p53-dependent Apoptosis by Sanguinarine in HCT116 Human Colorectal Cancer Cells)

  • 천연 benzophenanthridine alkaloid의 일종인 sanguinarine에 의한 인간 암세포에서의 세포사멸 유도는 암 치료를 위한 잠재적 치료 가능성으로 여겨져 왔으나 기본적인 항암 기전은 여전히 불분명하다. 종양 억제제 p53의 결실 또는 돌연변이는 결장암세포의 항암제 내성에 대한 주요 원인으로 작용하다. 따라서, 본 연구에서는 정상 p53을 가진 HCT116 (p53+/+) 및 p53이 결여된 HCT116 (p53-/-) 결장암세포를 대상으로 sanguinarine에 의해 유도되는 세포사멸에서 p53의 역할을 조사하였다. 본 연구의 결과에 의하면, sanguinarine은 HCT116 (p53-/-) 세포에 비하여 HCT116 (p53+/+) 세포의 생존력을 현저히 감소시켰다. 아울러 sanguinarine은 HCT116 (p53-/-) 세포보다 HCT116 (p53+/+) 세포에서 p53 및 cyclin-dependent kinase 억제제 p21WAF1/CIP1의 발현을 증가시키면서 DNA 손상 및 세포사멸의 유도를 증가시켰다. Sanguinarine은 HCT116 (p53+/+) 세포에서 외인성 및 내인성 세포사멸의 개시에 관여하는 caspase-8 및 caspase-9의 활성을 증가시켰으며, 전형적인 효과기 caspase인 caspase-3을 활성화시켰다. 또한, sanguinarine은 HCT116 (p53+/+) 세포에서 Bax/Bcl-2의 발현 비율을 증가시키고 미토콘드리아 손상을 유발하였지만, HCT116 (p53-/-) 세포에서는 이러한 현상이 관찰되지 않았다. 결론적으로 본 연구의 결과는 sanguinarine은 HCT116 결장암세포에서 p53 의존적으로 외인성 및 내인성 세포사멸의 경로 활성을 통하여 세포사멸을 유도하였음을 의미한다.

Interaction of promyelocytic leukemia/p53 affects signal transducer and activator of transcription-3 activity in response to oncostatin M

  • Promyelocytic leukemia (PML) gene, through alternative splicing of its C-terminal region, generates several PML isoforms that interact with specific partners and perform distinct functions. The PML protein is a tumor suppressor that plays an important role by interacting with various proteins. Herein, we investigated the effect of the PML isoforms on oncostatin M (OSM)-induced signal transducer and activator of transcription-3 (STAT-3) transcriptional activity. PML influenced OSM-induced STAT-3 activity in a cell type-specific manner, which was dependent on the p53 status of the cells but regardless of PML isoform. Interestingly, overexpression of PML exerted opposite effects on OSM-induced STAT-3 activity in p53 wild-type and mutant cells. Specifically, overexpression of PML in the cell lines bearing wild-type p53 (NIH3T3 and U87-MG cells) decreased OSM-induced STAT-3 transcriptional activity, whereas overexpression of PML increased OSM-induced STAT-3 transcriptional activity in mutant p53-bearing cell lines (HEK293T and U251-MG cells). When wild-type p53 cells were co-transfected with PML-IV and R273H-p53 mutant, OSM-mediated STAT-3 transcriptional activity was significantly enhanced, compared to that of cells which were transfected with PML-IV alone; however, when cells bearing mutant p53 were co-transfected with PML-IV and wild-type p53, OSM-induced STAT-3 transcriptional activity was significantly decreased, compared to that of transfected cells with PML-IV alone. In conclusion, PML acts together with wild-type or mutant p53 and influences OSM-mediated STAT-3 activity in a negative or positive manner, resulting in the aberrant activation of STAT-3 in cancer cells bearing mutant p53 probably might occur through the interaction of mutant p53 with PML.

Poly(ADP-ribosyl)ation of p53 Contributes to TPEN-Induced Neuronal Apoptosis

  • Depletion of intracellular zinc by N,N,N,N-tetrakis(2-pyridylmethyl) ethylenediamine (TPEN) induces p53-mediated protein synthesis-dependent apoptosis of mouse cortical neurons. Here, we examined the requirement for poly(ADP-ribose) polymerase (PARP)-1 as an upstream regulator of p53 in zinc depletion-induced neuronal apoptosis. First, we found that chemical inhibition or genetic deletion of PARP-1 markedly attenuated TPEN-induced apoptosis of cultured mouse cortical neurons. Poly(ADP-ribosyl)ation of p53 occurred starting 1 h after TPEN treatment. Suggesting the critical role of PARP-1, the TPEN-induced increase of stability and activity of p53 as well as poly(ADP-ribosyl)ation of p53 was almost completely blocked by PARP inhibition. Consistent with this, the induction of downstream pro-apoptotic proteins PUMA and NOXA was noticeably reduced by chemical inhibitors or genetic deletion of PARP-1. TPEN-induced cytochrome C release into the cytosol and caspase-3 activation were also blocked by inhibition of PARP-1. Taken together, these findings indicate that PARP-1 is essential for TPEN-induced neuronal apoptosis.

Quercetin-induced apoptosis ameliorates vascular smooth muscle cell senescence through AMP-activated protein kinase signaling pathway

  • Aging is one of the risk factors for the development of cardiovascular diseases. During the progression of cellular senescence, cells enter a state of irreversible growth arrest and display resistance to apoptosis. As a flavonoid, quercetin induces apoptosis in various cells. Accordingly, we investigated the relationship between quercetin-induced apoptosis and the inhibition of cellular senescence, and determined the mechanism of oxidative stress-induced vascular smooth muscle cell (VSMC) senescence. In cultured VSMCs, hydrogen peroxide (H2O2) dose-dependently induced senescence, which was associated with increased numbers of senescence-associated β-galactosidase-positive cells, decreased expression of SMP30, and activation of p53-p21 and p16 pathways. Along with senescence, expression of the anti-apoptotic protein Bcl-2 was observed to increase and the levels of proteins related to the apoptosis pathway were observed to decrease. Quercetin induced apoptosis through the activation of AMP-activated protein kinase. This action led to the alleviation of oxidative stress-induced VSMC senescence. Furthermore, the inhibition of AMPK activation with compound C and siRNA inhibited apoptosis and aggravated VSMC senescence by reversing p53-p21 and p16 pathways. These results suggest that senescent VSMCs are resistant to apoptosis and quercetin-induced apoptosis attenuated the oxidative stress-induced senescence through activation of AMPK. Therefore, induction of apoptosis by polyphenols such as quercetin may be worthy of attention for its anti-aging effects.

폐암 세포에서 Gemcitabine에 의한 세포 사멸과 p53의 역할 (Gemcitabine-induced Cell Death in Lung Cancer Cells : the Role of p53)

  • 연구배경: Gemcitabine은 폐암에서 임상적 유용성이 큰 새로운 항암제이다. 저자들은 폐암세포에서 Gemcita bine에 의한 세포 사멸과 p53의 역할을 규명하고자 하였다. 방 법 : 폐암 세포주로 A549와 H358 세포주를 이용하였고 세포 독성 검사는 MTT assay를 이용하였으며 Gemcitabine 농도는 10nM, 100nM, 1uM, 10uM, 100uM을 사용하였다. 세포 주기 검사는 FACScan을 이용하여 분석하였고 p53 활성화 여부는 western blot을 사용하였다. p53 단백질 분해를 촉진시키는 안정적 세포주 A549-E6과 H358-E6을 제조하고 대조 세포주 A549-neo와 H358-neo 세포주와 비교하여 p53의 기능적 knock-out 실험을 시행하였다. p53의 기능적 knock-out은 p53 유도 약제인 doxorubicine 1 M을 사용하여 western blot으로 확인하였다. 결 과 : A549와 H358 세포주에서 Gemcitabine은 농도에 비례한 세포 독성을 보였고 S phase arrest와 p53의 활성화를 유도하였다. 안정적 세포주 A549-E6과 H358-E6은 MTT assay에서 대조 세포주 A549-noo와 H358-noo에 비해 각각 20-30%, 30-40%의 세포 독성 차단효과를 보였다. 결 론 : Gemcitabine은 S phase arrest를 유발시키고 p53 단백질의 활성화를 유도하며 p53의 기능소실이 Gemcitabine에 대한 저항인자로 작용하고 있음을 확인할 수 있었다. 향후 Gemcitabine에 의해 p53의 활성화가 발생하는 신호경로와 p53 활성화에 의한 아포프토시스의 신호경로에 대한 연구가 필요할 것으로 사료된다.

백화사설초 메탄올 추출물에 의한 HL-60 세포고사과정에 있어서의 transcriptional factors 활성변화 연구 (Study on Transcriptional Factors Activation Change of HL-60 cell Apoptosis by Hedyotis Diffusa's Methanol Extract)

  • Objective : Hedyotis diffusa has been used as an anticancer agent for several decades in oriental medicine. We test whether the methanol extract of the herb affects transcriptional activation factors including $NF-{\kappa}B$ and AP-1. Methods : 1. HL-60 cells were treated with various concentrations(from 200 to $50{\mu}g/ml$) of methanol extract and $H_2O$ extract($200{\mu}g/ml$)of hedyotis diffusa, After 48h later, the cells were tested for viability by MTT assay. 2. The HL-60 cells were treated with $200{\mu}g/ml$ of methanol extract for the indicated periods. First. Nuclear extracts were isolated and incubated with oligonucleotide probe of $NF-{\kappa}B$ and AP-1. Second. Nuclear extracts were isolated and reacted with p50, p65. c-rel pan-Jun, c-Jun, JunB. JunD antibody on ice for 30min. Finally The cell lysates were prepared and analyzed by western blotting using anti-Fas, anti-FasL and anti-p53 antibody. Results : 1. The methanol extract decreases the viability of human lymphoid origin leukemia HL-60 cells in a dose-dependent manner. 2. $NF-{\kappa}B$ is rapidly activated by the addition of the methanol extract, reaches a peak at 30min and gradually returns to resting level. We confirm that $NF-{\kappa}B$ is a heterodimer mainly composed of p65 subunit with c-Rel. 3. Transcriptional activation of AP-1 is detected at 30min and reaches a maximum at 1hr after stimulation of the cells with the methanol extract. AP-1 is mainly composed with Jur-D and partially Jug-B proteins. 4. the methanol extract of Hedyotis diffusa induces the expression of Fas, Fas ligand and p53 proteins of HL-60 cells in a time dependent fashion. Conclusions : These results suggest that the methanol extract of Hedyotis diffusa exerts anticancer effects to induce the death of human leukomic HL-60 cells via activation of trascriptional factors such as $NF-{\kappa}B$ and AP-1, increase in expression of Fas mediated signalling proteins, and induction of tumor suppressor gene. p53.

  • The ubiquitin-proteasome mediated protein degradation pathway plays an important role in regulating both cell proliferation and cell death. Proteasome inhibitors are well known to induce apoptosis in various human cancer cell lines. We investigated the effect of combined treatment with proteasome inhibitor and TRAIL, and a possible mechanism of the enhancing apoptosis by the both treatment, on TRAIL-resistant non-small cell lung cancer. A549 cells were exposed to the N-Acetyl-Leu-Leu-Norleu-al (ALLN) as a proteasome inhibitor and then treated with recombinant TRAIL protein. In A549 cells under proteasome inhibition conditions by pretreatment with ALLN, TRAIL treatment significantly decreased cell viability compared to that ALLN and TRAIL alone treatment. Also, the both treatment induced cell damage through DNA fragmentation and p53 expression. In addition, the combined treatment of both markedly increased caspase-8 activation, especially the exposure for 2 h, and Bax expression and induced the dissipation of mitochondrial transmembrane potential in A549 cells. Taken together, these findings showed that proteasome inhibition by ALLN enhanced TRAIL-induced apoptosis via DNA degradation by activated P53 and mitochondrial transmembrane potential loss by caspase-8 activation and bax expression. Therefore, our results suggest that proteasome inhibitor may be used a very effectively chemotherapeutic agent for the tumor treatment, especially TRAIL-resistant tumor cell.

A novel p53-activating radioresponse regulator

  • In order to screen ionizing radiation induced early-response genes, we employed subtractive hybridization method and isolated a metabolism associated gene. The gene expression was very sensitive to ionizing radiation as revealed by a rapid induction of its messenger RNA. We characterized the function of this gene in radiation response. This gene activated p53 and enhanced cell killing effect of ionizing radiation. This effect was attributable to p53 phosphorylation and transcriptional activation.

노인에서 일어서기 동작 시 발의 위치에 따른 근활성도의 변화 (Analysis of Muscle Activity on Foot Position during a Sit-to-stand activity in the Elderly)

  • Purpose: The purpose of this study was to investigate changes in muscle activation associated with foot position during a sit-to-stand exercise among normally healthy elderly subjects. Methods: Eight subjects (male=3; female=5; mean age=$70.13{\pm}{\pm}2.53$years) were recruited.The activation of six muscles (neck extensor; lumbar extensor; hamstring; rectus femoris; gastrocnemius; tibialis anterior) was measured by surface EMG (TeleMyo 2400T G2, Noraxon Inc., USA) during a sit-to-stand protocol under three different foot positions (ankle dorsiflexion of 0, 15, or 30 degrees). Results: Muscle activation of the neck extensor and hamstring was decreased according to the change in foot position (p<0.05), but activation of the rectus femoris was increased (p<0.05). Muscle activation of the neck extensor was significantly different between 0 and 15 degrees (p<0.05). Muscle activation of the hamstring was significantly different between 0 and 15 degrees and between 0 and 30 degrees (p<0.05). Muscle activation of the rectus femoris was significantly different between 0 and 30 degrees and between 15 and 30 degrees (p<0.05). However, activation of lumbar extensor, gastrocnemius and tibialis anterior muscles did not significantly differ between foot positions. Conclusion: These findings suggest that muscle activation during a sit-to-stand movement differs depending on foot position. We believe that these differences should be considered when educating the elderly regarding proper body movements.
