Effect of Ultrasound-Induced Hyperthermia on Cellular Uptake of P-gp Substrate and Non-P-gp Substrate in MDR Cells

  • A previous report recently demonstrated that ultrasound-induced hyperthermia (USHT:0.4 watts (W)/$cm^2$ at $41^{\circ}C$) could increase cellular uptake of P-glycoprotein (P-gp) substrates in P-gp expressing cancer cell lines. Since P-gp plays a major role in limiting drug permeability in the multi-drug resistant (MDR) cells, studies were conducted to elucidate the mechanism of USHT on cellular accumulation of P-gp and non-P-gp substrate in MDR cells. To accomplish this aim, we studied the effects of USHT on the accumulation of P-gp substrate, R123 and non-P-gp substrate, antipyrine in MDR cells. We demonstrated that USHT increased permeability of hydrophobic molecules (R123 and $[^{14}C]$-antipyrine). The enhanced permeability is reversible and size-dependent as USHT produces a much larger effect on cellular accumulation of $[^{14}C]$-antipyrine (MW 188) than that of R123 (MW 380.8). These results suggest that USHT could affect MDR cells more sensitive than BBMECs. Also, the present results point to the potential use of USHT to increase cellular uptake of P-gp recognized substrates, mainly anti-cancer agents into cancer cells.

Positional relationship between mandibular third molar and mandibular canal in cone beam computed tomographs

  • Purpose: To provide diagnostic information by evaluation of the positional relationship between the mandibular third molar and the mandibular canal. Materials and Methods: Eighty-nine mandibular third molars were classified as mesioangular, horizontal, vertical, distoangular groups. The distances between the mandibular third molar and the mandibular canal were measured in cone-beam computed tomographs. The height and width ratios of distances from the mandibular third molar and the mandibular canal to the mandibular inferior border and to the lingual cortical plate were calculated. Results: The vertical and buccolingual distances between the mandibular third molar and the mandibular canal were 0.03 mm, 2.96 mm in the mesioangular, 0.37 mm, 3.38 mm in the horizontal, -1.50 mm, 1.38 mm in the vertical, -1.10 mm, 4.20 mm in the distoangular group. There were significant differences in vertical (P < 0.05), but not in buccolingual (P>0.05). The height and width ratios of distances on the mandibular third molar were 47.1 %, 36.1 % in the mesioangular, 47.4%, 34.4% in the horizontal, 37.0%, 46.7% in the vertical, 40.9%, 37.4% in the distoangular group. There were significant differences between the mesioangular and the vertical group, and the horizontal and the vertical group in height ratio (P < 0.05), and also between the mesioangular and the vertical group in width ratio (P < 0.05). The height and width ratios of distances on the mandibular canal showed no significant differences between groups (P > 0.05). Conclusion : The mesioangular group showed the nearest distance between the mandibular third molar and the mandibular canal vertically. The root apex of the mandibular third molar was positioned more buccally in the vertical group than in the mesioangular group.

Properties of an Extracellular 5-Fluorocytosine Deaminase (세포외 5-Fluorocytosine Deaminase의 특성)

  • - Some properties of an extracellular cytosine deaminase excreted from an isolate from soil samples were examined after 20~80%' ammonium sulfate fractionation. The enzyme catalyzed the conversion of cytosine and 5-fluorocytosine into uracil and 5-fluorouracil by substrate specificity, respectively. The optimum temperature and storage time on the stability of the enzvme preparation were below $50^{\circ}C$ keeping above 90% of the residual activity and near 4 days keeping above 80% of the residual one in Tris-HCI buffer. The maximum activity was also obtained at 8.0 in pH and 37'C in temperature. The pHs and temperatures for enzyme activity ranged from 8.0~8.5 and from 37~$45^{\circ}C$. respectively. The presence of $Ag^-,Hg^{2-}, Zn^{2-}, Cu^{2-}, Sn^{2-}, \; or\; Pb^{2-}$ in the reaction mixture resulted in the marked inhibition in enzyme activity, but 1 mM of $K^+, Fe^[3+}, Mg^{2+}, \; or \; Na^+$. slightly increased the activity. The enzyme preparation was heavily affected by most of inhibitors tested such as 1 mM of EIITA, NaCN and pentachlorophenol, and completely inactivated by p-CMB and TCA of 1 mM, or 10 mM.

  • $\beta$-Amylase was purified from sweet potato by acetone fractlonatlon, Sephadex A-50 ion exchange chromatography and Sepgadex G-200 gel chromatographyl The higher enzyme concentration was, the higher heat stability of enzyme became. After 1 hour 30 minute. At 6$0^{\circ}C$ in pH 5, enzyme under concentration of 30$\mu$l/ml lost its activity completely and over the concentration of 100$\mu$g/ml remained 25% of activity. The enzyme was stabilized at range of pH 4~10 and pH stability was increased by glycerol. Five moles of NaCl inhibited completely of the enzyme activity. SDS of 0.05% inhibited the enzyme completely after 12 hours at 37$^{\circ}C$ in pH5. One mole guanidine-HCl and 8M urea inhibited the entire enzyme after 13 hours at 37$^{\circ}C$ in pH 5.

  • Nine novel compounds, namely, 2-(p-aminobenzenesulfonamideo)-6-sulfamylbenzothiozle, 2-benzamido-6-sulfamylbenzothiazole, 2-(p-toluenesulfonamide)-6-sulfamylbenzothiazole, 2-(3,5-dinitrobenzamido)-6-sulfamylbenzothiazole, 2-benzamido-6-nitrobenzothiazole, 2-(p-toluene-sulfonamido)-6-nitrobenzothiazole, 2-(3,5-dinitrobenzamido)-6-nitrobenzothiazole, 2-(p-toluenesulfonamido)-6-chlorobenzothiazole, 2-(3,5-dinitrobenzamido)-6-chlorobenzothiazole were synthesized and evaluated for their in vitro antimicrobial activity against Staphylococcus aureus wood 46, $\betha$-Streptococcus S86, Escherichia coli and antitubercular activity against Mycobacterium tuberculosis H$_{37}$ /Rv. 2-(p-Toluenesulfonamido)-6-chlorobenzothiazole showed antimicrobial activity at 25 $\mu$g/ml against $\betha$-Streptococcus S/86, 2-(3,5-Dinitrobenzamido)-6-sulfamylbenzothiazole was active at 5 $\mu$g/ml and 2-(3,5-dinitrobenzamido)-6-nitrobenzothiazole was consderably active at 1$\mu$g/ml against Mycobacterium tuberculosis H$_{37}$Rv.

  • To elucidate the composition of phytosterols in the fruit body of Pholiota spp. 6 species(P. adiposa, P. aurivella, P. highlandensis, P. nameko, P. squarrosa and Pholiota sp.) were analyzed with Gas chromatography(GC). Pholiota spp. were contained campesterol, stigmasterol, and $\beta$-sitosterol as the major phytosterols.

Pasteurella multocida isolation from pigs with respiratory disease in Korea

  • A total of 131 Pasteurella (P.) multocida strains were isolated from the lungs of 1,064 pigs with respiratory clinical signs nationwide during 2009-2010 and 2013-2014. The strains of P. multocida comprised 77.1% serotype A and 22.9% serotype D. Analysis of a recent P. multocida outbreak in Korean pigs showed that the isolation rate of serotype D decreased annually. The incidence of antimicrobial resistance, as measured using minimal inhibitory concentration values, has decreased recently. Overall, further studies to characterize P. multocida isolated from pigs in Korea are needed to prevent P. multocida infection in the Korean swine industry

New Record of Ptecticus sinchangensis Ôuchi, 1938 (Diptera, Stratiomyidae, Sarginae) in Korea

  • In Korea, species of the genus Ptecticus Loew, 1855 (Family Stratiomyidae) have been known as three species, P. aurifer (Walker, 1854), P. matsumurae Lindner, 1936 and P. japonicus (Thunberg, 1789) (= P. tenebrifer (Walker, 1849)). Additionally, an unrecorded species, P. sinchangensis ${\hat{O}}uchi$, 1938, was found in Korea. We therefore report the species for the first time in Korea with morphological diagnosis and key for the identification of the Korean species of the genus. In total, the members of the genus Ptecticus are officially recognized as four species in Korea.

A COMPARATIVE STUDY OF PRESERVING ABILITY OF HUMAN PERIODONTAL LIGAMENT CELLS STORED IN DIFFERENT TEMPERATURED STORAGE MEDI (저장용액의 온도에 따른 치주인대세포의 생존율)

  • To compare the survival rate of periodontal ligament cells preserved in storage media with good availability at the time of an avulsion injury, periodontal ligament cells were incubated in ${\alpha}-MEM$ culture medium containing 10% FBS in condition of $37^{\circ}C$, 5% $CO_2$. These cells were then cultured in HBSS, ${\alpha}-MEM$, milk(S co., P. co.) and tap water at the temperature of 4, 25, $37^{\circ}C$ each in 60 min. The groups were measured by MTT assay. The results were as follows : 1. Among the storage media at $4^{\circ}C$, ${\alpha}-MEM$ and P-milk had the highest preserving ability of periodontal ligament cells, while that of HBSS S-milk and tap was low in order. 2. Among the storage media at $25^{\circ}C$, ${\alpha}-MEM$ had the highest preserving ability of periodontal ligament cells, while that of P-milk, HBSS, S-milk, tap water was low in order. 3. Among the storage media at $37^{\circ}C$, the preserving ability of periodontal ligament cells was very high in ${\alpha}-MEM$, P-milk, HBSS and S-milk, it's lowest in tap water. 4. The preserving ability of periodontal ligament cells in ${\alpha}-MEM$ was high at $4^{\circ}C$ and it's low in order of $25^{\circ}C$, $37^{\circ}C$, but in HBSS was high at $4^{\circ}C$ and it's low at $25^{\circ}C$, $37^{\circ}C$ 5. The preserving ability of periodontal ligament cells in S-milk and P-milk was high at $4^{\circ}C$, $25^{\circ}C$ and it s low at $37^{\circ}C$. In conclusion, HBSS is the storage medium of choice in an avulsion, but in this study it is preferable to choose milk at $4^{\circ}C$ for tooth since it is easy to get and affect cell viability.

Predictors of Weight Control Behavior According to College Students' BMI, Perception of Body Shape, Obesity Stress, and Self-Esteem (대학생의 체질량지수, 체형인식, 비만스트레스, 자아존중감에 따른 체중조절행동 예측요인)

  • This study was conducted to identify factors influencing the weight control behavior of college students. The subjects included college students in the C area, and data were collected through a self reported structured questionnaire from Dec 01 to 20 of 2015. Overall, 289 subjects were analyzed. Analyses consisted of descriptive statistics, t-tests, ANOVA, Pearson's correlation analysis, and stepwise multiple regression analysis. The findings showed a prominent distortion of college students' perceptions of their bodies based on evaluation by others and obesity stress, which had special impacts on their weight control behavior. Weight control behavior, body self-awareness (r=.31, p<0.001), assessment of body by others (r=.30, p<0.001), and obesity stress (r=.43, p<0.001) were closely related to their weight control behavior. Stepwise multiple regression revealed that their weight control behavior was closely influenced by obesity stress (${\beta}=.37$, p<0.001) and assessment of body by others (${\beta}=.15$, p=0.009). These variables accounted for 20% of the weight control behavior (F=37.30, p<0.001). Overall, assessment of body by others and obesity stress were found to be predictors of their weight control behavior. These findings indicate that it is necessary to develop and implement information programs and design appropriate prevention programs that can induce healthy weight control behaviors. Such programs should include health education, as well as intervention programs to identify such predictors and help college students judge and perceive their body shapes objectively so that they control their weight in a proper manner.
