Tumor Cell Clone Expressing the Membrane-bound Form of IL-12p35 Subunit Stimulates Antitumor Immune Responses Dominated by $CD8^+$ T Cells

  • IL-12 is a secretory heterodimeric cytokine composed of p35 and p40 subunits. IL-12 p35 and p40 subunits are sometimes produced as monomers or homodimers. IL-12 is also produced as a membrane-bound form in some cases. In this study, we hypothesized that the membrane-bound form of IL-12 subunits may function as a costimulatory signal for selective activation of TAA-specific CTL through direct priming without involving antigen presenting cells and helper T cells. MethA fibrosarcoma cells were transfected with expression vectors of membrane-bound form of IL-12p35 (mbIL-12p35) or IL-12p40 subunit (mbIL-12p40) and were selected under G418-containing medium. The tumor cell clones were analyzed for the expression of mbIL-12p35 or p40 subunit and for their stimulatory effects on macrophages. The responsible T-cell subpopulation for antitumor activity of mbIL-12p35 expressing tumor clone was also analyzed in T cell subset-depleted mice. Expression of transfected membranebound form of IL-12 subunits was stable during more than 3 months of in vitro culture, and the chimeric molecules were not released into culture supernatants. Neither the mbIL-12p35-expressing tumor clones nor mbIL-12p40-expressing tumor clones activated macrophages to secrete TNF-${\alpha}$. Growth of mbIL-12p35-expressing tumor clones was more accelerated in the $CD8^+$ T cell-depleted mice than in $CD4^+$ T cell-depleted or normal mice. These results suggest that $CD8^+$ T cells could be responsible for the rejection of mbIL-12p35-expressing tumor clone, which may bypass activation of antigen presenting cells and $CD4^+$ helper T cells.

  • We have cloned and characterized an antiapoptotic gene, p35, which blocks apoptosis, from Bombyx mori nuclear polyhedrosis virus (BmNPV) K1 strain. The 897 bp p35 has an open reading frame of 299 amino acids. The BmNPV-K1 p35 showed a high identity to Autographa californica nuclear polyhedrosis virus and BmNPV T3 strain. The BmNPV-K1 p35 was different from the amino acid sequences of BmNPV T3 at 6 positions. The p35 gene of BmNPV-K1 was 99.2% identical at the nucleotide level and 98% identical at the amino acid level to BmNPV T3. The location of p35 gene in the BmNPV-K1 genome was confirmed by Southern blot analysis and its expression patterns at the transcriptional level in the infected cells were con- firmed by Northern hybridization analysis.

  • The surface of $Li[Ni_{0.35}Co_{0.3}Mn_{0.35}]O_2$ cathode was modified by $[Li,La]TiO_3$ coating using pH controlled coating solution. At low pH values (acidic solution), cathode powders, which is oxides, have a positive surface charge, whereas, they have a negative surface charge at high pH values. As a result, their charge could affect the formation of the coating layer on the surface of cathode powder. To determine the optimal pH value, the surface coating of the pristine powder was carried out at various pH values of the coating solution. The surface morphology of coated samples was characterization by SEM and TEM analyses. Impedance analysis and cyclic voltammogram presented that internal resistance of the cell was dependent upon the pH of coating solution.

An Observation on the Viability and pH of Lactobacillus casei isolated from Yakult. (유산균음과에서 분리한 Lactobacillus casei의 저장온도별 균수 및 pH의 경시적 변화 관찰)

  • The authors studied the viability and pH of Lactobacillus casei which was isolated from the Yakult in terms of its storage temperature time(days). The following results have been obtained: (1) The viability test to Lactobacillus casei under storage in 4$\circ$C temperature condition in 35 days, has shown that the original population of $4.0\times 10^8$ organisms was decreased to $0.1\times 10^8$, under storage in 10$\circ$C temperature the organisms to $0.08\times 10^8$, in room temperature the organisms to $0.009\times 10^8$, and in 35$\circ$C temperature the organisms to $0.0009\times 10^8$. The correlation coefficient of storage period (days) and number of bacterial viability were significant as r=0.956(p<0.01) in 4$\circ$C, r=-0.999 (p<0.01) in 100C, r=0.975(p<0.01) in room temperature and r=-0.923(p<0.01) in 35$\circ$C. (2) The pH test to Lactobacillus casei unde.r storage in 4$\circ$C temperature condition for 35 days has shown that the original pH of 3.54 were acidified to 3.35, under storage in 10$\circ$C temperature the pH were acidified to 3.20, in room temperature the pH were acidified to 3.15, and in 35$\circ$C temperature the pH were acidified to 3.0. The correlation coefficient of variation of pH between storage period (days) and temperature were significant as r=-0.972(p<0.01) in 4$\circ$C, r=-0.922 (p<0.01) in 10$\circ$C, r=-0.963 (p<0.01) in room temperature and r=-0.953 in 35$\circ$C.

Membrane-bound p35 Subunit of IL-12 on Tumor Cells is Functionally Equivalent to Membrane-bound Heterodimeric Single Chain IL-12 for Induction of Anti-tumor Immunity

  • In this study, we compared two different tumor cell vaccines for their induction of anti-tumor immunity; one was a tumor cell clone expressing a membrane-bound form of IL-12 p35 subunit (mbIL-12 p35 tumor clone), and the other was a tumor clone expressing heterodimeric IL-12 as a single chain (mb-scIL-12 tumor clone). The stimulatory effect of mb-scIL-12 on the proliferation of ConA-activated splenocytes was higher than that of mbIL-12 p35 in vitro. However, the stimulatory effect of mbIL-12 p35 was equivalent to that of recombinant soluble IL-12 (3 ng/ml). Interestingly, both tumor clones (mbIL-12 p35 and mb-scIL-12) showed similar tumorigenicity and induction of systemic anti-tumor immunity in vivo, suggesting that tumor cell expression of the membrane-bound p35 subunit is sufficient to induce anti-tumor immunity in our tumor vaccine model.

  • We have studied the properties of $GaAs_{0.35}P_{0.65}$ epitaxial films on the GaP according to doping of $NH_3$ gas using VPE method by CVD. The efficiency of $GaAs_{0.35}P_{0.65}$ epitaxial films found to be greatly enhanced by the according of nitrogen doping. The diodes were fabricated by means of Zn diffusion into vapor grown $GaAs_{0.35}P_{0.65}$ epitaxial films doped with N and Te. The effects of nitrogen doping on carrier density of epitaxial films, PL wavelength and the power out, forward voltage of diodes are discussed. In the end, The effect of electrical and optical properties is influenced by the deep level and deep level density of nitrogen doping.

Effect of 35% Hydrogen Peroxide with Dicalcium Phosphate Dihydrate on the Tooth Whitening and Microhardness (35% 과산화수소에 제2인산칼슘를 함유한 치아미백제가 치아의 색과 경도에 미치는 영향)

  • The purpose of this study was to evaluate tooth whitening and microhardness after treatments with tooth bleaching agents containing dicalcium phosphate dihydrate (DCPD) and 35% hydrogen peroxide (HP) which were used in-office bleaching. Thirty enamel specimens were obtained from human premolars and randomly divided into 3 groups(n=10). Tooth bleaching agents were prepared with DCPD (0 g for controls, 0.1 g and 1 g for experimental groups) and HP solution (35% HP). All groups were applied to enamel surfaces for 60 min for 1 day. The pH of each tooth bleaching agent was measured. Tooth color, microhardness of enamel surfaces were also measured. The tooth bleaching agents containing DCPD showed a significant increase in pH compared to the ones without DCPD(p<0.05). Paired t-tests showed significant difference in color values of enamel before and after bleaching in all the groups(p<0.05). As a result, changes in color, containing DCPD group does not contain a statistically significant difference between groups was observed.(p>0.05). In all groups, tooth hardness after bleaching showed a significant decrease in microhardness (p<0.05). However, the DCPD concentration increased in the bleaching, microhardness values slightly decreased. Based on the above results, tooth bleaching agents containing DCPD and 35%HP were equally effective. Due to increases in pH and effective reduction of tooth surface decalcification, the surface characteristics are exposed to a reduced degree of negative effects, resulting in fewer constituent enamel alterations. Thus, commercial availability of the constituents of tooth whitening materials can be achieved.

Effects of dietary supplementation of herbal active ingredients promoting insulin-like growth factor-1 secretion on production performance, egg quality, blood hematology, and excreta gas emission in laying hens

  • Objective: The purpose of this study was to evaluate the effects of supplementing herbal active ingredients (YGF251) which can promote the secretion of insulin-like growth factor-1 (IGF-1) in the diet on production performance, egg quality, blood hematology, and excreta gas emission in laying hens. Methods: A total of 288 ISA Brown (41-week-old) laying hens with an initial body weight of 1.83±0.68 kg were randomly assigned to 1 of 4 dietary treatments in a randomized block design based on body weight. Each treatment had 12 replicate cages having 6 adjacent cages per replicate (hens are kept in cages alone). The experimental period was 35 days. Dietary treatments were based on the corn-soybean meal-wheat-based basal diet and supplemented with 0.00%, 0.05%, 0.10%, or 0.15% YGF251. Results: There was a linear increased egg weight in weeks 1 to 5 (p<0.05), egg mass in week 1 (p<0.05) and weeks 1 to 5 (p<0.05), egg strength on day 7 (p<0.05), 21 (p<0.01), and 35 (p<0.01), eggshell thickness on day 21 (p<0.05) and 35 (p<0.01), haugh unit on day 21 (p<0.01) and 35 (p<0.05), serum IGF-1 concentration on day 21 (p<0.05) and 35 (p<0.01), and serum total protein concentration on day 35 (p<0.05) were observed with the supplementing YGF251 increased in the diet, while feed conversion ratio in weeks 1 to 5 (p<0.05) and excreta ammonia emission (p<0.01) decreased linearly with the dose of YGF251 increased. Conclusion: Dietary supplementation of YGF251 positively affected the production performance and egg quality of laying hens through increasing serum IGF-1 concentration in a dose-dependent manner. Moreover, YGF251 supplementation improved barn environment by reducing excreta noxious gas emission.

IL-35 Over-expression is Associated with Genesis of Gastric Cancer

  • Overexpression of interleukin (IL)-35 has been found in a variety of malignancies, but the expression status in gastric cancer has yet to be elucidated clearly. In the present study, positive expression of EBI3 and p35 was 63.3% and 70.0% of cases, respectively. EBI3 expression was strongly related with larger tumor size and invasion depth (P<0.05). Similarly, expression of p35 was also correlated with larger tumor size (P<0.05). These results indicate that IL-35 might be involved in growth of gastric cancer. Interestingly, EBI3 and p35 expressions were positive correlated with Ki-67 expression. Moreover, EBI3 immunoreactivity was associated with Bcl-2 staining. Our data suggest IL-35 is correlated with genesis of gastric cancer by regulating growth and apoptosis.

The effect of tooth bleaching agent contained 35% hydrogen peroxide on the color, microhardness and surface roughness of tooth-colored restorative materials (35% 과산화수소를 함유한 치아미백제가 심미수복재의 색, 미세경도 및 표면 거칠기에 미치는 영향)

  • Objectives : The purpose of this study was to evaluate the effects of tooth bleaching agent contained 35% hydrogen peroxide on the color, microhardness and surface roughness of tooth-colored restorative materials. Methods : Four types of tooth-colored restorative materials, including a composite resin(Filtek Z350 ; Z350), a flowable composite resin(Filtek P60 : P60), a compomer(Dyract$^{(R)}$ AP ; DY), and a glass-ionomer cement(KetacTM Molar Easymix ; KM) were used in the study. The specimens($8mm{\times}5mm$) were made by using a customized acrylic mold. Each material was divided into two groups equally(n=40) : experimental group(35% HP) and control group(distilled water). 35% HP group was treated 30 mim/5 days for 15 days. Each 30 minute treatment session consisted of two 15 minute cycles of gel application with 20 second light exposure. The authors measured the color, microhardness, and roughness of the specimens before and after bleaching. The data were analyzed with ANOVA and T-test. Results : 35% HP group showed an apparent color change(${\Delta}E^*$) than control group. In particular, DY and KM showed a noticeable color change and statistically significant differences(p<0.05). 35% HP group showed a reduction in microhardness. Z350 and P60 does not have a statistically significant difference(p>0.05), DY and KM showed a statistically significant difference(p<0.05). Percentage microhardness loss(PML) of control group was 0.6 to 5.5% in the group, 35% HP group was 6.6 to 34.6%. Roughness was increased in 35% HP group after bleaching. Especially DY and KM were significantly increased(p<0.05). Conclusions : Bleaching agents may affect the surface of existing restorations; therefore, they should not be used indiscriminately when tooth-colored restorations are present.
