Efficacy of formalin bath against gill infections with Pseudodactylogyrus spp. in cultured eel Anguilla japonica (포르말린 약욕이 뱀장어 아가미흡충의 구제에 미치는 효과)

  • Effects of formalin on removal of Pseudodactylogyrus spp. were examined against naturally infected eel, Anguilla japonica (weight 89.9~96g) at a water temperature of $28^{\circ}$. Prior to experiments for removal of the parasite, the hematological toxicity of formalin bath at 0~500 ppm for 30 min~24 h was assessed by hematocrit values (Ht). Based on the results of Ht, appropriate methods of treatment, concentrations and durations, were examined in the main study. There was no significant (P>0.05) change of Ht in 100 to 200 ppm for 24 h. In contrast, Ht increased significantly (P<0.05) at above 300 ppm. This suggests that physiological damage was caused by formalin bath treating with 300 to 500 ppm. Formalin bath with 100 and 200 ppm for 24 h caused significant decreases (P<0.05) in the infection of the parasite. In conclusion, the 100 ppm formalin for 24 hour-bath was found most recommendable for the effective treatment of Pseudodactylogyrus spp. for the gills of the infected eel because of the median lethal concentration ($LC_{50}$) of formalin to eel; cumulative mortalities were found to be 0 and 13.3%, respectively, following 24 h bathing.

A Study on the Toxicity of Pb and Cu Compound in Carassius auratus(goldfish) (납 및 구리화합물이 Carassius auratus(goldfish)에 미치는 독성에 관한 연구)

  • In order to investigate acute toxicity and bioconcentration of heavy metals for a freshwater fish, the fish used in this experiment was goldfish, Carassius auratus. Each ten goldfish was accommonidated in a water and was treated with different concentration of Pb and Cu compound. The 24 $hr-LC_{50}$ was obtained by plotting on the log-normal distribution graph. Furthermore, the combined effect of Pb and Cu was also investigated the fish was treated with Pb or Cu compound only, and Pb and Cu compound together, respectively. These results were summarized as follows: 1. The 24 $hr-LC_{50}'s$ of Pb and Cu were 7.48 mg/l and 0.666 mg/l, respectively. 2. When single or/and combined treatment with Pb(7.0 mg/l) or/and Cu(0.6 mg/l) to Carassius auratus for 24 hours were performed, there was significant difference between the single or/and the combined treatment in their bioaccumulated Cu concentrations. Cu concentrations in goldfish were higher in the combined treatment than in the single treatment. 3. When Carassius auratus was exposed to 0.748 mg/l (1/10 of 24 $hr-LC_{50}$) and 1.496 mg/l of Pb (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 79.14 and 100.11 for Pb, respectively. The BCF of Pb was obtained as a linearity according to the concentration and exposure time as follows log BCF=1.014 log $P\cdot T$+1.011 ($r^2$=0.9041) where, P: pollutant concentration(mg/l) T: exposure time(day) 4. When Carassius auratus was pxposed to 0.0666 mg/l (1/10 of 24 $hr-LC_{50}$) and 0.1332 mg/l of Cu (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 55.42 and 63.24 for Cu respectively. Influence of Palm Kernel Meal Inclusion and Exogenous Enzyme Supplementation on Growth Performance, Energy Utilization, and Nutrient Digestibility in Young Broilers

  • The objective of the present study was to investigate the influence of palm kernel meal (PKM) inclusion and exogenous enzyme supplementation on growth performance, nitrogen-corrected apparent metabolizable energy (AMEn), coefficient of apparent ileal digestibility (CAID) and total tract retention of nutrients in young broilers fed corn-based diets. Four inclusion levels of PKM (no PKM [PKM0], 8% [PKM8], 16% [PKM16], and 24% [PKM24]) and two enzyme additions were evaluated in a $4{\times}2$ factorial arrangement of treatments. A total of 384, one-d-old male broilers (Ross 308) were individually weighed and allocated to 48 cages (eight broilers/cage), and cages were randomly assigned to eight dietary treatments. Results indicated that the inclusion of 8% and 16% PKM increased (p<0.05) the weight gain compared to the PKM0 diet. Birds fed the PKM8 diets had the highest (p<0.05) feed intake. Weight gain and feed intake were severely reduced (p<0.05) by feeding the PKM24 diet. Enzyme supplementation increased weight gain (p<0.05), independent of PKM inclusion level. In PKM0 and PKM8 diets, enzyme addition significantly (p<0.05) lowered feed conversion ratio (FCR); whereas enzyme addition had no effect on FCR of birds fed PKM16 and PKM24 diets. In PKM0 and PKM16 diets, enzyme addition significantly (p<0.05) increased CAID of nitrogen and energy but had no effect in the PKM8 and PKM24 diets. Inclusion of PKM into the basal diet, irrespective of inclusion level, enhanced (p<0.05) starch and fat digestibility. Inclusion of PKM at 16% and 24% resulted in similar CAID of neutral detergent fiber (NDF) but higher (p<0.05) than that of the PKM0 and PKM8 diets. Enzyme addition, regardless of the level of PKM inclusion, significantly (p<0.05) increased CAID of NDF. There was a significant (p<0.05) decrease in AMEn with PKM inclusion of 24%. The present data suggest that inclusion of PKM in broiler diets could be optimized if PKM-containing diets are formulated based on digestible amino acid contents and supplemented with exogenous enzymes. If amino acid digestibility and AME of PKM considered in the formulation, it can be included in broiler diets up to 16% with no deleterious effects on growth performance.

Histopathological Study of Acute Toxicity of Ammonia on Common Carp Cyprinus carpio (잉어, Cyprinus carpio에 미치는 암모니아 급성독성의 조직병리학적 연구)

  • The carp (Cyprinus carpio) used in the experiment were hatched in the spring this year and reared to 5.96g($4.84{\sim}6.55g$) in mean weight in a nursery pond at Daeyon fish farm, Pusan, Korea. The sample fish were exposed to different conditions of total ammonia (TA-N) concentrations 10, 20 and 30ppm and pH 6.5, 7.0, 7.5 and 8.0 at water temperatures 20, 25 and $30^{\circ}C$ for 24, 48 and 72 hours. After the procedure, the gill, liver and kidney of the fish were examined histopathologically. In this experiment, with the rise of water temperature, increase of pH and ammonia concentration, and the extension of exposure time the three organs showed the tendency of apparent abnormal changes such as hypertrophy and necrosis in their tissues. At $20^{\circ}C$ of water temperature gill tissue did not show any abnormality regardless of the change of pH at 10 ppm of ammonia concentration for 24 hours of exposure, but beyond the conditions given above, there occurred hypertrophy and the epithelium of gill lamellae was detached. The detach of gill lamellae epithelium initiated from the proximal part of the gill lamellae then gradually spread toward the uppermost tip. The heavier vacuolation of the liver was observed with the rise of water temperature and pH, and such morbid state in the liver was considered to be the result of edema in the liver tissue. The kidney showed no damage to the renal tubule epithelium at pH 6.5, but it was damaged at pH 8.0 when exposed to 30 ppm ammonia at $20^{\circ}C$ for 24 hours.

Factors Influencing Biohydrogenation and Conjugated Linoleic Acid Production by Mixed Rumen Fungi

  • The objective of this study was to evaluate the effect of soluble carbohydrates (glucose, cellobiose), pH (6.0, 6.5, 7.0), and rumen microbial growth factors (VFA, vitamins) on biohydrogenation of linoleic acid (LA) by mixed rumen fungi. Addition of glucose or cellobiose to culture media slowed the rate of biohydrogenation; only 35-40% of LA was converted to conjugated linoleic acid (CLA) or vaccenic acid (VA) within 24 h of incubation, whereas in the control treatment, 100% of LA was converted within 24 h. Addition of VFA or vitamins did not affect biohydrogenation activity or CLA production. Culturing rumen fungi at pH 6.0 slowed biohydrogenation compared with pH 6.5 or 7.0. CLA production was reduced by pH 6.0 compared with control (pH 6.5), but was higher with pH 7.0. Biohydrogenation of LA to VA was complete within 72 h at pH 6.0, 24 h at pH 6.5, and 48 h at pH 7.0. It is concluded that optimum conditions for biohydrogenation of LA and for CLA production by rumen fungi were provided without addition of soluble carbohydrates, VFA or vitamins to the culture medium; optimum pH was 6.5 for biohydrogenation and 7.0 for CLA production.

  • An analysis of the bonding situation in CaM₂P₂ (M=Fe, Ni) with ThCr₂Si₂ structure is made in terms of DOS and COOP plots. The main contributions to covalent bonding are due to M-P and P-P interactions in both compounds. Particularly, the interlayer P-P bonding by variation in the transition metal is examined in more detail. It turns out that the shorter P-P bonds in CaNi₂P₂ form as a result of the decreasing electron delocalization into ${{\sigma}_p}^*$ of P₂ due to the weaker bonding interaction between the metal d and ${{\sigma}_p}^*$ as the metal d band is falling from Fe to Ni.

Twenty-Four Hour pH Study and Manometry in Gastric Esophageal Substitutes in Children

  • Purpose: Studies on the physiology of the transposed stomach as an esophageal substitute in the form of a gastric pull-up or a gastric tube in children are limited. We conducted a study of motility and the pH of gastric esophageal substitutes using manometry and 24-hour pH measurements in 10 such patients. Methods: Manometry and 24 hour pH studies were performed on 10 children aged 24 to 55 months who had undergone gastric esophageal replacement. Results: Six gastric tubes (4, isoperistaltic; 2, reverse gastric tubes) and 4 gastric pull-ups were studied. Two gastric tubes and 4 gastric pull-ups were transhiatal. Four gastric tubes were retrosternal. The mean of the lowest pH at the midpoint of the substitute was 4.0 (range, 2.8-5.0) and in the stomach remaining below the diaphragm was 3.3 (range, 1.9-4.2). In both types of substitute, the difference between the peak and the nadir pH recorded in the intra-thoracic and the sub-diaphragmatic portion of the stomach was statistically significant (p<0.05), with the pH in the portion below the diaphragm being lower. The lowest pH values in the substitute and in the remnant stomach were noted mainly in the evening hours whereas the highest pH was noted mainly in the morning hours. All the cases showed a simultaneous rise in the intra-cavitatory pressure along the substitute while swallowing. Conclusion: The study suggested a normal gastric circadian rhythm in the gastric esophageal substitute. Mass contractions occurred in response to swallowing. The substitute may be able to effectively clear contents.

Comparison of Dietary Intakes by 24-hr Dietary Recall, Dietary Record and Food Frequency Questionnaire among Elderly People (회상법, 기록법 및 식품섭취빈도조사법을 이용한 노인의 영양소 섭취 수준의 비교)

  • Nutrient intakes estimated using a 24-hr recall, a dirt record and a food frequency questionnaire(FFQ) were compared in a group of ninety-four elderly people(21 males, 73 females) in Cheongju, a city in Chung-Buk province. Mean intakes for energy, protein, Ca, p, Na, K thiamin and niacin obtained from the diet record were higher than those from the 24-hr recall. Mean intakes for energy, protein, Ca, P, Na, K thiamin, niacin and vitamin C from the FFQ were higher than those obtained from the 24-hr recall or the diet record. Correlation coefficients between the nutrient intake values from the 24-hr recall and those from the diet record ranged from 0.84 to 0.95 and were significantly correlated(p < 0.001). About 80% of the subjects in the lowest quintile by the 24-hr recall were also in the lowest two quintiles by the diet record. While the percentage fallen into the opposite category ranged from 0% to 15%. For most nutrients, at least 65% of the subjects when classified by the 24-hr recall fell into the same quintile when classified by the diet record, and the mean kappa value was 0.7. About 52% of the subjects in the lowest category by the 24-hr recall fell into the lowest two categories by FFQ. The mean percentage of the subjects in the lowest quintile by the diet record or in the lowest two quintiles by the FFQ was 51%. For most nutrients, at least 24% of each of the subjects when classified by both the 24-hr recall and the diet record fell into the same category when classified by the FFQ. The kappa values between the 24-hr recall or the diet recall and the FFQ were 0.17. These data indicate that in elderly subjects the 24-hr recall can provide very similar information to that obtained from the diet record but the FFQ can not provide good information if the right FFQ method is not used for only elderly subjects.

24-Hr Ambulatory Double-probe pH Monitoring in LPR (역류성 후두염의 증상을 가진 환자에서의 24시간 이중 탐침 식도 산도 측정)

  • The term laryngopharyngeal reflux (LPR) refers to the backflow of food or stomach acid back up into the larynx (the voice box) or the pharynx (the throat). Esophagopharyngeal reflux is suggested as an etiologic factor in laryngeal disease. To examine a possible esophageal basis for laryngopharyngeal symptoms, we studied 48 patients with persistent laryngopharyngeal symptoms, and 12 relative control subjects. Patients were evaluated for cervical symptoms by questionnaire and underwent gastrofiberoscopy, fiberoptic laryngoscopy, esophageal manometry and 24-hour ambulatory double-probe pH monitoring. We found LPR in fourteen out of 48 patients with cervical symptoms (29%). The LPR group consisted of nine men and five women. The symptoms that LPR patients complained were throat lump sensation, hoareness, sore throat, throat clearing, chronic coughing and dysphagia in order of frequency, and they were not different significantly from non-LPR patients. The laryngoscopic findings in LPR patients were posterior erythema, laryngeal edema and diffuse erythema, and there was also no significant difference between LPR group and non-LPR group. There was statistically significant correlation between LPR and gastroesophageal reflux (GER). We concluded that there is no pathognomonic symptoms or laryngoscopic findings in diagnosis of LPR, and 24-hour ambulatory double-probe pH monitoring is an essential diagnostic tool in LPR.

  • For the development of a glucanolytic yeast strain. the seceretion of endo-1.4-$\beta$-D-glucanase (CMCase) of Bacillus subtilis was performed in yeast using glucoamylase gene (STA1) of Saccharomyces diastaticus. A 1.7 kb-DNA fragment of STA1 gene containing authentic promoter, signal sequence, threonine serine-rich (TS) region and N-terminal region (98 amino acids) of mature glucoamylase was ligated to YEp 24. E. coli-yeast shuttle vector. And then. CMCase structural gene of B. subtilis was fused in frame with the 1.7 kb-DNA fragment of STA1 gene, resulting in recombinant plasmid pYES('24. Yeast transformant harboring pYESC24 had no CMCase activity. So. we deleted TS region and N-terminal region of mature glucoamylase existing between signal sequence and CMCase structural gene in pYESC24. consequently constructed recombinant plasmid pYESC11. The yeast transformed with the newly constructed recombinant plasmid pYESC11 efficiently secreted CMCase to extracellular medium. After 4 days culture. total CMCase activity of this transformant was 44.7 units/ml and over 93% of total CMCase activity was detected in culture supernatant.
