Identification of a p-Cresol Degradation Pathway by a GFP-Based Transposon in Pseudomonas and Its Dominant Expression in Colonies

  • In this study, the chromosome-encoded pcuRCAXB genes that are required for p-cresol degradation have been identified by using a newly constructed green fluorescent protein (GFP)-based promoter probe transposon in the long-chain alkylphenol degrader Pseudomonas alkylphenolia. The deduced amino acid sequences of the genes showed the highest identities at the levels of 65-93% compared with those in the databases. The transposon was identified to be inserted in the pcuA gene, with the promoterless gfp gene being under the control of the pcu catabolic gene promoter. The expression of GFP was positively induced by p-cresol and was about 10 times higher by cells grown on agar than those in liquid culture. In addition, p-hydroxybenzoic acid was detected during p-cresol degradation. These results indicate that P. alkylphenolia additionally possesses a protocatechuate ortho-cleavage route for p-cresol degradation that is dominantly expressed in colonies.

Effects of phytotoxins on Change of pH in the Continuous Cropping Soil (연작지(連作地) 토양(土壤)의 독소물질(毒素物質)이 토양(土壤) pH 변화(變化)에 미치는 영향(影響))

  • The experiment was conducted to investigate the origin of pH changes in a continuous cropping soil. The results obtained were as follows : 1. The concentration of phytotoxin increased and at the same time the soil pH decreased with the prolonged practice of continuous cropping. 2. The decrease of soil pH by the phytotoxin materials was higher in the distilled water and tap water than in the soil. 3. The effect of phytotoxin on plant germination was higher in benzoic acid than in p-hydroxybenzoic acid. 4. The treatment of acetone to the continuous cropping soil recovered the initial soil pH and sesame growth was normal in the soil.

Antimicrobial Activity of Thinned Strawberry Fruits at Different Maturation Stages

  • Among the phenolic compounds that is generally present in strawberry fruits, five simple phenolics, three flavonoids, and a stilbene were tested for their antimicrobial activity against seven fungi and one oomycete. trans-Cinnamic acid showed strong antimicrobial activity, and the antimicrobial effect of the simple phenolics decreased with an increase in the number of hydroxyl groups. Phytophthora capsici was the most susceptible to the phenolic compounds tested in this study. trans-Cinnamic acid, p-hydroxybenzoic acid, and kaempferol were mainly detected in 'Seolhyang' strawberry fruits, and the total phenolic contents of the fruits decreased during their development. Extracts of the green (1-10% red color) and red (above 90% red color) strawberry fruits reduced the mycelial growth and zoospore germination rate of P. capsici, and the extract of red strawberry fruit showed strong antimicrobial activity against the zoospore germination of P. capsici. These results indicate that strawberry fruits contain antimicrobial phenolic compounds and that strawberry fruit extract can be used as a natural fungistat.

Effect of Phenolic Compounds Identified from Crop Residues (Wheat.Rye) on the Germination and Growth of Various Weeds (맥류작물(麥類作物)(밀.호밀) 잔여물(殘餘物)로부터 동정(同定)된 Phenolic Compounds 가 잡초(雜草)의 발아(發芽) 및 생육(生育)에 미치는 영향(影響))

  • The phenolic compounds identified from rye and wheat residues seemed to be p-coumaric, p-hydroxybenzoic, vanillic, ferulic, salicylic and syringic acids. Total phenol content of rye and wheat straw determined at heading stage was 0.1803% and 0.1702%, respectively. Total phenol content in straw was higher than that of root at all growth stages. The germination and growth of plants, such as Oryza sativa, Echinochloa creugalli, Cyperus serotinus, Portulaca oleacea, Amaranthus retroflexus, Digitaria saguinalis, and C. album were inhibited by treatments of authentic phenolic compounds as the concentrations increased. However, at the early stage, the germination and shoot growth of P. distinctus were markedly stimulated by them, and then the further growth of shoot and root was markedly inhibited by the prolonged treatment of phenolic compounds. The aqueous extracts from rye and wheat straw completely inhibited the germination of A. retroflexus and C. album at a high concentrations. The content of starch and protein in bulbs of P. distinctus was lower in the p-hydroxybenzoic acid treated plot, at $10^{-2}M$, than the untreated control.

  • Corneal permeability of various n-alkyl p-hydroxybenzoates (parabens) was studied in vitro using excised rabbit corneas, and the effect of lipophilicity of parabens on the corneal permeability was also investigated. Permeability coefficients were obtained from the least-square linear regression after the steady state had been reached. Lipophilicity of parabens was calculated by distribution coefficients determined in octanol-S | $12{\phi}{\parallel}$) rensen's buffer solution (pH 5.0). The relationship between lipophilicity and corneal permeability of parabens was not linear, but the optimum lipophilicity for the maximum permeation was found. The influence of tween 80 on corneal permeability of methyl and butylparaben was not significant.

Antioxidant Compounds Isolated from the Roots of Phlomis umbrosa Turcz.

  • Two triterpenoids, arjunolic acid (1), belleric acid (2), five phenylethanoids, martynoside (3), orobanchoside (4), 3,4-dihydroxyphenethylalcohol-6-O-caffeoyl-${\beta}$-$\text\tiny{D}$-glucoside (5), leucosceptoside B (6), lunariifolioside (7), four phenolic acids, ferulic acid (8), syringic acid (9), vanillic acid (10), 4-hydroxybenzoic acid (11), and one lignan, (+)-syringaresinol-${\beta}$-$\text\tiny{D}$-glucoside (12), were isolated from the roots of P. umbrosa. All isolated compounds were explored for their antioxidant potential in the DPPH and ABTS assays. In DPPH assay, compound 5 showed high antioxidant capacity. Compounds 3, 4, 6, and 7 displayed considerable antioxidant activities. In addition, compounds 5-7 exhibited potential antioxidant capacities in the ABTS assay.

Allelopathic Potentials of Larix leptolepis (S. et Z.) Gorden on Germination and Seeding Growth of Selected Species (植物의 發芽와 生長에 미치는 일본잎갈나무의 알레로패티 效果)

  • A number of field surveys have been performed in larch (Larix leptolepsis) forest floor in which any seeding of this species never find ther. In laboratory work, aqueous extracts from leaves, roots and stems of larch were provided and used to bioassay. Among them, germination and seedling growth of selected species were the most inhibitory in leaves extracts. Though germiation percentage of a species showed high compared with control, its dry weight after a given period grown were clearly low. The results due to be considered by allelopathic effect. And syringic acid and p-hydroxybenzoic acid were identified by HPLC from larch leaves. It is assumed that these phenolic acids affect on other species as allelochemicals of larch.

A Study on the Absorption Mechanism of Drugs through Biomembranes

  • The effect of lipophilicity on the mechanisms of drug absorption through biomembranes was investigated empolying HPLC system and the fluorescence technique. Human erythrocyte ghost membranes were used as a model biomembrane. A series of four parabens (methyl, ethyl, rpopyl, and butyl) and p-hydroxybenzoic acid were used as the model drugs for lipophilicities and their partition coefficients were measured in Sorensen's phosphate buffer solution (pH 5)/octanol system. Absorption amount of parabens through erythrocyte ghost membranes increased with an increase of lipophilicity resulted from the addition of methylene group to the n-alkyl chain of parabens. And the effect of parabens on the fluidity of ghost membrane also increased with an increase of their lipophilicities.

  • Laccases are multicopper-containing enzymes which catalyze the oxidation of phenolic and nonphenolic compounds with the concomitant reduction of molecular oxygen. They often occur as isoenzymes, either constitutive or inducible, that oligomerize to multilateral complexes, what allow for penetration to the woody cell wall structure. White rot basidiomycete fungi may produce a number of laccase isoenzymes, some constitutively and others after induction. Fungal laccase is commonly induced by many ions, such as $Cu^{2+}$, $Cd^{2+}$ $Ca^{2+}$, $Li^+$, $Mn^{2+}$, $Ag^+$, $Hg^{2+}$, Mn and $Fe^{3+}$, phenolic compounds, some organic compounds, such as ethanol, isopropanol, cAMP, caffeine, p-anisidine, viscosinamide and paraquat, and nitrogens and even heat shock. A combination of Cu and pHB (p-hydroxybenzoic acid) made it possible to extend the inducible laccase activities over 30-fold. But the most effective inducer of laccase in the basidiomycete and other higher fungi is 2,5-xylidine, over 160-fold stimulation of laccase activity. The laccases are frequently encoded by gene families, as e.g. in Pycnoporus cinnabarinus, from which the lcc3-1 or the allelic form lac1 and lac3-2 have been cloned and sequenced. In the case of inducible forms the post-inductional laccase formation depends upon the synthesis of mRNA and the induction is due to the synthesis of a new protein.

  • Based on the Benzoflex (Vesicol Chemical Co.) as PVC plasticizer substituents for Di-n-octyl phthalate (DOP), a series of new aromatic carboxylic acid ester compounds were designed as plasticizers, synthesized, and screened for the endocrine disrupting activity. 2-Hydroxybenzoic acid (1) and 2-methoxybenzoic acid (2) as the commercially available starting materials were reacted with diethylene glycol (3) in the presence of p-toluenesulfonic acid using Dean-Stark column to give diethylene glycol di-(2-hydroxy)benzoate (4, KH01) and diethylene glycol di-(2-methoxy)benzoate (5, KH02), respectively. And diethylene glycol di-(3-pyridinyl) ester (7, KH03) and dipropylene glycol di-(3-pyridinyl) ester (9, KH04) were obtained in high yields by treatment of nicotinoyl chloride (6) with diethylene glycol (3) and dipropylene glycol (8) in the presence of triethylamine as a base. To determine the estrogenic disrupting effect of new synthetic phthalate analogues, E-screen assay method was used. Of these compounds, 4 (KH01) was found to be compound without endocrine disrupting effect.
