Effects of Phenolic Compounds and Hosts on the vir Gene Expression of Various Ti Plasmids

  • The vir genes expression of Ti plasmid is induced by a family of related phenolic compounds. We investigated the effects of various phenolic compounds, Ti plasmids and hosts on the expression of the vir genes in the same type of octopine Ti plasmids, pTiKU12, pTiAch5 and pTiA6. The vir gene induction of pTiKU12 was remarkably stimulated by p-coumaric acid in relation to acetosyringone, but those of pTiAch5 and pTiA6 were more stimulated by acetosyringone than by p-coumaric acid. The effect of phenolic compound on the vir gene induction was different according to the kind of Ti plasmids. Also, the vir gene expression of A. tumefaciens KU913, which has pTiKU12 was about 6.2 times as much as that of A. tumefaciens KU915, which has pTiKU12 in KU12 host, in the presence of ferulic acid. But no difference was shown in the presence of p-coumaric acid. The vir gene induction abilities of phenolic compounds are different according to the kinds of phenolic compounds, Ti plasmids and hosts.

Effects of Aqueous Extracts from Naturalized and Korean Wild Plants on Seed Germination and Seedling Growth of Zoysiagrass (자생식물과 귀화식물의 수용 추출액이 Zoysiagrass의 발아와 유근생장에 미치는 영향)

  • The soil pH favored by Korean wild plants is 5.33~7.20, while naturalized invader species prefer soils of pH 3.95~6.48. The germination rate of Zoysia japonica was inhibited sharply, up to 60%, in extracts of naturalized plants of concentrations over 50%. Erigeron canadensis extract most strongly inhibited germination, while the Korean wild species, Cassia mimosoides var. nomame increased germination rates in concentrations of 30% and 50%. The seedling growth of Zoysia japonica in extracts of Korean wild species and naturalized species did not show differences, but was inhibited strongly, up to 60%, in the extract of Cassia mimosoides var. nomame concentration of over 15%. Analysis of the extract from Cassia mimosoides var. nomame identified procatechuic acid, $\rho$-hydroquinone, $\rho$-coumaric acid and ferulic acid at 254nm; and vanillic acid, hydroquinone, benzoic acid and cinnamic acid were discovered at 284nm. The seed germination and seedling growth of Zoysia japonica were investigated under different concentrations of phenolic compounds. Ferulic acid and vanillic acid were associated with an increased germination rate, while $\rho$-coumaric acid appeared to inhibit seedling growth.

Synthesis and Evaluation of Coumaroyl Dipeptide Amide as Potential Whitening Agents

  • Coumaroyl dipeptide amide, Coumaric acid-LG-$NH_2$, was prepared successfully using the solid-phase method, and its efficacy as a skin whitening agent was studied. Coumaric acid-LG-$NH_2$ was prepared with Rink-amide resin, and 96.354% of purity was obtained. Using MTT assay and LDH release assay, we found that it exhibited very low cytotoxicity. And, we found that Coumaric acid-LG-$NH_2$ inhibited tyrosinase activity dose-dependently and showed superior tyrosinase inhibitory activity to well-known whitening agent, arbutin. $IC_{50}$ value of Coumaric acid-LG-$NH_2$ was 182.4 ${\mu}M$, and $IC_{50}$ value of arbutin was 384.6 ${\mu}M$. Also, in measurement of melanin contents using B16F1 melanoma cell lines, Coumaric acid-LG-$NH_2$ reduced melanin production induced by ${\alpha}$-MSH statistically significant, and showed superior melanin inhibitory activity to p-coumaric acid or arbutin. In addition, Coumaric acid-LG-$NH_2$ reduced MC1R mRNA expression level. Thus, we concluded that MC1R pathway is the significant pathway of Coumaric acid-LG-$NH_2$, and Coumaric acid-LG-$NH_2$ has great potential to be used as novel whitening agents.

Protective Effect of Sasa Quelpaertensis and p-Coumaric Acid on Ethanol-induced Hepatotoxicity in Mice

  • Excessive alcohol use causes oxidative stress in the liver, and antioxidant therapy has been an attractive approach for the treatment of ethanol-induced liver damage. The present study examined the hepatoprotective effect of Sasa quelpaertensis Nakai (Korean name, Jeju-Joritdae) in C57BL/6 mice intoxicated with ethanol. Mice were intraperitoneally administered with ethanol alone, or together with test materials three times at 12-h intervals. At 3 h after the last dosing, hepatotoxicity was assessed based on serum activities of aspartate aminotransferase and alanine aminotransferase, and hepatic contents of thiobarbituric acid-reactive substances and glutathione. Sasa quelpaertensis extract mitigated the acute ethanol hepatotoxicity as effectively as silymarin. Its n-butanol fraction was more active than methylene chloride or aqueous fraction. p-Coumaric acid, a major constituent of S. quelpaertensis, was found to effectively prevent the ethanol-induced hepatotoxicity. These data suggest that S. quelpaertensis and p-coumaric acid could be useful for the prevention of liver disease caused by alcohol abuse.

  • Six phenolic acids were isolated from Jindalrae flowers (Rhododendron mucronulatum Turcz.), an edible plant in Korea. These compounds were identified as chlorogenic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid, caffeic acid, ferulic acid, and p-coumaric acid on the basis of IR, UV, $^{1}H$ and $^{13}C$ NMR, FAB-MS, ES-MS and/or El-MS data. Chlorogenic acid (0.2 g) present in both ethyl acetate and ethyl ether fractions comprised up to 38.5% of the total phenolic acid amount (0.52 g) finally recovered by means of polyamide C-200 column chromatography, preparative TLC, recrystallization, and Sephadex LH-20 column chromatography The antioxidant activities were measured in an ethanol solution of linoleic acid in the presence of ferric thiocyanate. Studies on Identification of the Anthocyanins in Elderberries (Sambucus) (Elderberry(Sambucus) Anthocyanin에 관한 연구(硏究))

  • Studies have been carried out on the structure and the content of the anthocyanins in elderberries which were just recently transplanted in Korea. The anthocyanin pigments of elderberries were extracted with 1 % methanolic HCl and purified with Amberlite IRC-50 cation exchange column. The individual pigments were isolated by paper chromatography. Five pigments, identified by various chemical and physical methods were cyanidin-3-monoglucoside(4.3%), cyanidin-3,5-diglucoside(28.3%), cyanidin-3,5-diglucoside with p-coumaric arid(12.9%), cyanidin-3-xyloglucoside-5-glucoside(38.3%), cyanidin-3-xyloglucoside-5-glucoside with p-coumaric acid(16.2%). The content of total anthocyanin in elderberry was 3.13 mg/g fresh weight.

Quantitative Analysis of Phenolic Compounds in Different parts of Panax ginseng C.A. Meyer and Its Inhibitory Effect on Melanin Biosynthesis (인삼의 부위별 페놀성 성분 함량 및 멜라닌 생성억제효과)

  • Comparison of four phenolic compounds among three different parts of Panax ginseng C.A. Meyer was performed in this study. Contents of esculetin, p-coumaric acd, quercetin, maltol in the ethyl acetate fraction were determinated by HPLC analysis. The main root has a highest concentration of p-coumaric acid and the leave has a highest concentration of quercetin and esculetin. This ethyl acetate fraction of the leaves exhibited higher depigmenting activity and lower cell toxicity than the other parts of ginseng. In addition, it showed that quercetin and esculetin among phenolic compounds highly inhibited melanin biosynthesis in ginseng. It implies that leaves of ginseng may be used as skin whitening herb. Quercetin and esculetin proved to be active compounds in the leaves of ginseng.

  • This study was designed to investigate the possible utilization of Ligustrum japonicum leaves as a source of functional ingredients. Contents of total phenolic compounds and condensed tannin were $0.89{\sim}1.53%$ and $0.10{\sim}0.13%$, respectively. The major flavonoid compounds in the leaves of L. japonicum were luteolin, apigenin and their glycosides. Tyrosol, t-cinnamic acid, p-hydroxybenzoic acid, vanillic acid, shikimic acid and protocatecuic acid were detected in free phenolic acid, while tyrosol, t-cinnamic acid, ferulic acid, esculetin, caffeic acid, p-coumaric acid and hydroxytyrosol were detected in esterified phenolic acid. The insoluble phenolic acid contained tyrosol, t-cinnamic and p-caoumaric acid.

Effect of p-Coumaric Acid, Benzoic Acid, and Salicylic Acid on the Activity of Glutathione Reductase and Catalase in in vitro Grown Tobacco Plants (기내배양 담배 식물에서 p-coumaric acid, benzoic acid 및 salicylic acid에 의해 유도되는 생장 및 glutathione reductase와 catalase의 활성)

  • Effects of p-coumaric acid (p-CA), benzoic acid (BA), and salicylic acid (SA) on the activities of glutathione reductase and catalase were studied in in vitro grown tobacco plants. After culturing the tobacco plants in MS medium containing $10^{-5}$ mM of p-CA, BA, and SA, the increase in the activities of two enzymes, glutathione reductase and catalase, were compared from day 20 to day 50 day, with an interval of 10 days. The growth of the tobacco plants treated with p-CA, BA, and SA was the highest on day 50. Analysis of the effect of the three substances on the activity of glutathione reductase showed that BA and p-CA decreased the activity of the enzyme compared with a control, and SA increased the activity of the enzyme. All of them showed the highest activity on day 40. SA increased the activity of catalase, but BA and p-CA reduced the activity of this enzyme. In all the experimental groups, the activity was the highest on day 40. In conclusion, p-CA and BA appear to promote the growth of tobacco plants. The growth was the best on day 50, but the activity of the antioxidative enzyme was inhibited. On the contrary, SA seemed to inhibit the growth of the tobacco plants but to promote the activity of glutathione reductase and catalase. The growth of the plants treated with SA was best on day 40.
