In the Xanthan gum fermentation by Xanthomonas campestris there are problems of the large energy consumption by long fermentation time, the mass transfer of oxygen and nutrients by high viscous fermentation broth. In this study, the media optimization and the fed batch fermentation were carried out to decrease fermentation time and increase Xanthan gum yield. The $O_2$ uptake rate (OUR) and $CO_2$ evolution rate(CER) which were obtained from the analysis of fermentation exit gas using a gas chromatograph were investigated. As a result, the fermentation time decreased at optimal assimilable nitrogen concentration but increased at poor or rich assimilable nitrogen concentration, the Xanthan gum biosynthesis was stimulated under the limited condition of assimilable nitrogen source and the optimum fermentation medium was obtained as follow; Glucose=30g / l, Peptone=8.0g / l, $K_2HPO_4=2.0g/l$, $MgS0_47H_2O=10g/l$, Sodium acetate=20g/l, Sodium pyruvate=0.5g/1. As the agitation speed and nitrogen concentration increased, the $O_2$ uptake rate and $CO_2$ evolution rate increased. The OUR and CER were 37.3mmol $O_2/\;l$ hr and 20.2 mmol $CO_2/\;L$ hr at peptone 11g / l and agitation speed 990RPM, respectively. In fed batch fermentation, the final concentration of Xanthan gum was enhanced up to 29g / l.
Efficient simultaneous reduction conditions for $NO_x$ and $NH_3$-slip was investigated in SCR (Selective Catalytic Reduction) process with load variation by applying dual catalysts (SCR catalyst, $NH_3$ decomposition catalyst) system. $N_2O$ formation characteristics were analyzed to look into possible undesirable reaction pathways. In the experiments of catalyst characteristics, various operational variables were tested for the combined catalytic system, such as $NH_3/NO_x$ ratio, temperature, oxygen concentration and $H_2O$. The reaction characteristics of $NO_x$, $NH_3$ and $N_2O$ were analyzed and optimal conditions could be evaluated for the combustion facility with varied load. In terms of $NO_x/NH_3$ simultaneous reduction and $N_2O$ formation suppression, optimal condition was considered NSR 1.2 and temperature $300^{\circ}C$. At this operational condition, $NO_x$ conversion was 98%, $NH_3$ reduction efficiency was 95%, generated $N_2O$ concentration 9.5 ppm with inlet $NO_x$ concentration of 100 ppm. In $NH_3-SCR$ process with $NH_3$ decomposition catalyst, $NO_x$ and $NH_3$ can be considered to be reduced simultaneously at limited conditions. The results of this study may be utilized as basic data at facilities requiring simultaneous $NO_x$ and $NH_3$ reduction for facilities with load variation.
Sulfur-Iodine (SI) cycle, which thermochemically splits water to hydrogen and oxygen through three stages of Bunsen reaction, HI decomposition, and $H_2SO_4$ decomposition, seems a promising process to produce hydrogen massively. Among them, the decomposition of $H_2SO_4$ ($H_2SO_4=H_2O+SO_2+1/2O_2$) requires high temperature heat over $800^{\circ}C$ such as the heat from concentrated solar energy or a very high temperature gas-cooled nuclear reactor. Because of harsh reaction conditions of high temperature and pressure with extremely corrosive reactants and products, there have been scarce and limited number of data reported on the pressurized $H_2SO_4$ decomposition. This work focuses whether the $H_2SO_4$ decomposition can occur at high pressure in a noble-metal reactor, which possibly resists corrosive acidic chemicals and possesses catalytic activity for the reaction. Decomposition reactions were conducted in a Pt-lined tubular reactor without any other catalytic species at conditions of $800^{\circ}C$ to $900^{\circ}C$ and 0 bar (ambient pressure) to 10 bar with 95 wt% $H_2SO_4$. The Pt-lined reactor was found to endure the corrosive pressurized condition, and its inner surface successfully carried out a catalytic role in decomposing $H_2SO_4$ to $SO_2$ and $O_2$. This preliminary result has proposed the availability of noble metal-lined reactors for the high temperature, high pressure sulfuric acid decomposition.
BACKGROUND: Biochar is a solid material converted from agricultural biomass such as crop residues and pruning branch through pyrolysis under limited oxygen supply. Biochar consists of non-degradable carbon (C) double bonds and aromatic ring that are not readily broken down by microbial degradation in the soils. Due to the recalcitrancy of C in biochar, biochar application to the soils is of help in enhancing soil carbon sequestration in arable lands that might be a strategy of agricultural sector to mitigate climate change. METHODS AND RESULTS: Data were collected from studies on the effect of biochar application on soil C content conducted in East Asian countries including China, Japan and Korea under different experimental conditions (incubation, column, pot, and field). The magnitude of soil C storage was positively correlated (p < 0.001) with biochar application rate under field conditions, reflecting accumulation of recalcitrant black C in the biochar. However, The changes in soil C contents per C input from biochar (% per t/ha) were 6.80 in field condition, and 12.58 in laboratory condition. The magnitude of increment of soil C was lower in field than in laboratory conditions due to potential loss of C through weathering of biochar under field conditions. Biochar production condition also affected soil C increment; more C increment was found with biochar produced at a high temperature (over 450℃). CONCLUSION: This review suggests that biochar application is a potential measures of C sequestration in agricultural soils. However, as the increment of soil C biochar was affected by biochar types, further studies are necessary to find better biochar types for enhanced soil C storage.
스마트 워치, 스마트 밴드 같은 웨어러블 디바이스가 개발됨에 따라, 웨어러블 디바이스 사용자의 건강상태를 측정할 수 있는 생체신호 모니터링 기술이 각광 받고 있다[1]. 하지만 한 개의 웨어러블 디바이스에서 여러 가지 생체신호를 측정하기 위해 웨어러블 디바이스에 여러 가지 생체신호 센서를 내장하게 되면 디바이스 자체 크기와 같은 제한적인 자원 측면에서 문제가 발생한다. 또한 새로운 생체신호 센서를 추가 할 때 문제가 발생한다. 본 논문에서는 웨어러블 디바이스 사용자의 요청이 있을 때 가속도 센서, ECG 센서, PPG 센서를 Cradle형태의 모듈에서 동작시킨 후 사용한 센서에서 측정되는 생체 신호를 실시간으로 수집하여 다른 웨어러블 디바이스로 전송할 수 있는 플러그인(Plug-in) 모듈인 Bio-Cradle을 제안한다. Bio-Cradle은 다른 웨어러블 디바이스에 플러그인하면, 디바이스의 종류에 무관하게 동기화 된 다수 생체신호를 전송할 수 있다. 즉 Bio-Cradle을 사용함으로써, 다수 생체신호의 동시 측정이 가능해지고, 웨어러블 디바이스 내부에 생체신호 센서를 내장 할 필요가 없어진다.
An adaptive measure of photosynthetic cells to a condition identified with a reduction of cellular energy charge, caused by water deficit-induced impairment of photosynthetic ATP production, was investigated using hydroponically cultured rice seedlings. Water stress treatment of the seedlings resulted in a marked decrease in cellular ATP level, a significant increase in the content of NAD(H) and concurrent decrease in that of NADP(H) in shoots, which accompanied a decrease in the activity of NAD kinase (EC 2.7.1.23) that specifically converts NAD(H) to NADP(H). The decline in the enzyme activity was particularly evident in the $Ca^{2+}/calmodulin-dependent$ kinase, the major form of NAD kinase in plants, whereas the level of active calmodulin remained unchanged during water deficit. The ratio of $NADP^+$ to NADPH was maintained nearly constant and no increases were seen in the level of $H_2O_2$ and the activities of $superoxide/H_2O_2-detoxifying$ enzymes in shoots stress-treated for two days. Based on these results, it may be suggested that rice plants take a strategy to cope with an adverse situation of limited photophosphorylation created by water deficit in that cells facilitate ATP production through glycolysis and oxidative phosphorylation; in doing so, rice cells suppress NAD kinase activity, consequently up-sizing the NAD(H) pool at the expense of the NADP(H) pool. Several parameters associated with the stress symptoms are also of implicative that there is no overproduction of superoxide radical or the related active oxygen at least in rice seedlings.
A new biological nutrient removal system combining $A^2/O$ process with fixed film was developed in this work and the characteristics of denitrification were especially investigated in the combined fixed film reactor(CFFR). Media was added in the anaerobic, anoxic and aerobic reactors, respectively. Tests were made to establish the effluent level of $NO_x-N$, COD, DO and nitrite effects on $NO_x-N$ removal in the CFFR by decreasing hydraulic retention time (HRT) from 10.0 to 3.5 hours and by increasing internal recycle ratio form 0% to 200%. The influent was synthesized to levels similar to the average influent of municipal wastewater treatment plants in Korea. SARAN media with a porosity of 96.3% was packed 40% / 130% / 25% based on its reactor volume, respectively. It was found that COD rarely limited dentrification in the anoxic reactor because of high $C/NO_x/-N$ ratio in the anoxic reactor, while DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent inhibited denitrification in the anoxic reactor. It was proved that the critical points of DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent were 0.15mg/L and 10%, respectively. As the internal recycle ratio increased, DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent increased. Especially, at the condition of internal recycle ratio, 200%, DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent exceeded the critical points of 0.15mg/L and 10%, respectively. Then, denitrification efficiency considerably decreased. Consequently, it was represented that the control of DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent can assure effective denitrification.
Bifidobacteria are probiotic organisms that provide both flavor and health benefits when incorporated as live cultures into commercial dairy products. Because bifidobacteria are very sensitive to environmental conditions (acids, temperature, oxygen, bile salts, the presence of other cultures, etc.), their viability in human gastrointestinal tract is limited. The microencapsulation of bifidobacteria is a process to protect them against harsh environmental conditions, thereby increasing their viability while passing through human gastrointestinal tract. To confirm the survival rate of microencapsulated Bifidobacterium breve CBG-C2 in milk, their survival rate was compared with several kinds of free bifidobacteria and lactic acid bacteria in commercial yogurt products under simulated gastric and small intestinal conditions. Double-microencapsulation of the bacteria was employed to increase the survival rate during digestion. The outer layer was covered with starch and gelatin to endure gastric conditions, and the inner layer was composed of a hard oil for the upper small intestinal regions. Almost all microencapsulted bifidobacteria in the milk survived longer than the free bifidobacteria and lactic acid bacteria in the commericial yogurt products under the simulated gastric conditions. Numbers of surviving free bifidobacteria and lactic acid bacteria in the commercial products were significantly reduced, however, the viability of the microencapsulated bificobacteria in the milk remained quite stable under gastric and small intestine conditions over 3$\sim$6 hrs. Thus double-microencapsualtion of bifidobacteria in milk is a promising method for improving the survival of bifidobacteria during the digestive process.

BACKGROUND: In this review paper, the effects of surface cover (SCV), vegetative filter strip (VFS), and vegetated ridge (VRD) on the pollutant runoff from steep-sloping uplands were analyzed to compare the pollutant reduction efficiency in runoff ($PRE_{runoff}$) of the practices and to investigate how slope and rainfall parameters affect the $PRE_{runoff}$. METHODS AND RESULTS: The $PRE_{runoff}$ of SCV, VFS, and VRD for pollutants including suspended solids and biological oxygen demand was compared by analysis of variance. The effect of slope and rainfall parameters on the $PRE_{runoff}$ was explored by either mean comparison or regression analysis. It was found that the $PRE_{runoff}$ differs with the practices due to different pollutant reduction mechanisms of the practices. Though the $PRE_{runoff}$ was likely to be affected by site condition such as slope and rainfall (amount and intensity), more comprehensive understanding was not possible due to the limited data set. CONCLUSION: The $PRE_{runoff}$ of SCV, VFS, and VRD differed due to the distinctive mechanisms of pollutant removal of the practices. It is necessary to accumulate experimental data across a variety of gradient of slope and rainfall for comprehensive understanding of the effects of the practices on pollutant runoff from steep-sloping uplands.
The objective of this study was to construct and assess the applicability of the EFDC model for Saemangeum Reservoir as a 3D hydrodynamic and water quality modeling tool that is necessary for the effective management of water quality and establishment of conservation measures. The model grids for both reservoir system only and reservoir-ocean system were created using the most recent survey data to compare the effects of different downstream boundary conditions. The model was applied for the simulations of temperature, salinity, water quality variables including chemical oxygen demand (COD), chlorophyll-a (Chl-a), phosphorus and nitrogen species and algal biomass, and validated using the field data obtained in 2008. Although the model reasonably represented the temporal and spatial variations of the state variables in the reservoir with limited boundary forcing data, the salinity level was underestimated in the middle and upstream of the reservoir when the flow data were used at downstream boundaries; Sinsi and Garyuk Gates. In turn, the error caused to increase the bias of water quality simulations, and inaccurate simulation of density flow regime of river inflow during flood events. It is likely because of the loss of momentum of sea water intrusion at downstream boundaries. In contrast to flow boundary conditions, the mixing between sea water and freshwater was well reproduced when open water boundary condition was applied. Thus, it is required to improve the downstream boundary conditions that can accommodate the real operations of the sluice gates.
