Metabolic Analysis of Poly(3-Hydroxybutyrate) Production by Recombinant Escherichia coli

  • Poly(3-hydroxybutyrate) (PHB) production by fermentation was examined under both restricted- and ample-oxygen supply conditions in a single fed-batch fermentation. Recombinant Escherichia coli transformed with the PHB production plasmid pSYLl07 was grown to reach high cell density (227 g/l dry cell weight) with a high PHB content (78% of dry cell weight), using a glucose-based minimal medium. A simple flux model containing 12 fluxes was developed and applied to the fermentation data. A superior closure (95%) of the carbon mass balance was achieved. When the data were put into use, the results demonstrated a surprisingly large excretion of formate and lactate. Even though periods of severe oxygen limitation coincided with rapid acetate and lactate excretion, PHB productivity and carbon utilization efficiency were not significantly impaired. These results are very positive in reducing oxygen demand in an industrial PHA fermentation without sacrificing its PHA productivity, thereby reducing overall production costs.

Optical Emission Characteristics of Atmospheric Pressure Dielectric Barrier Discharge (대기압 유전체배리어방전의 발광특성)

  • Plasma properties of dielectric barrier discharges (DBDs) at atmospheric pressure were measured and characterized using optical emission spectroscopy. Optical emissions were measured from argon, nitrogen, or air discharges generated at 5-9 kV using 20 kHz power supply. Emissions from nitrogen molecules were markedly measured, irrespective of discharge gases. The intensity of emission peaks was increased with applied voltage and electrode gap. The short wavelength peaks (315.9 nm and 337.1 nm) measured at the middle of DBDs were significantly increased with applied voltage. The optical emission from DBDs decreased with the addition of oxygen gas, which was especially significant in argon discharge. Emission from oxygen molecules cannot be measured from air discharge and argon discharge with 4.8% oxygen. The emission intensity at 337.1 nm and 357.7 nm related with nitrogen molecule was sensitively changed with electrode types and discharge voltages. However, the pattern of argon emission spectrum was nearly the same, irrespective of electrode type, oxygen content, and discharge voltage.

Influence of Oxygen Supply Method on the Performance of IGCC Plants (IGCC 플랜트에서 산소공급방식이 성능에 미치는 영향)

  • In this paper, two types of integrated gasification combined cycle (IGCC) plants using either an air separation unit (ASU) or an ion transport membrane (ITM), which provide the oxygen required in the gasification process, were simulated and their thermodynamic performance was compared. Also, the influence of adopting a pre-combustion $CO_2$ capture in the downstream of the gasification process on the performance of the two systems was examined. The system using the ITM exhibits greater net power output than the system using the ASU. However, its net plant efficiency is slightly lower because of the additional fuel consumption required to operate the ITM at an appropriate operating temperature. This efficiency comparison is based on the assumption of a moderately high purity (95%) of the oxygen generated from the ASU. However, if the oxygen purity of the ASU is to be comparable to that of the ITM, which is over 99%, the ASU based IGCC system would exhibit a lower net efficiency than the ITM based system.

Successful treatment of central retinal artery occlusion using hyperbaric oxygen therapy

  • Central retinal artery occlusion (CRAO) is considered an ophthalmologic emergency. The prognosis of this disease is very poor. Currently, there is no generally effective therapy available to treat CRAO. Hyperbaric oxygen therapy (HBOT) can increase the volume of oxygen delivered to the ischemic retinal tissue until spontaneous or assisted reperfusion occurs. We report the case of a patient who experienced sudden visual loss due to CRAO that was treated with HBOT. The patient was an 81-year-old woman who presented with CRAO in her right eye (OD). She exhibited "hand motion" visual acuity before treatment. She underwent three sessions of HBOT at a pressure of 2.8 atmospheres absolute, performed over 3 days. After 4 days in hospital, her visual acuity improved to 0.4 (OD) for far vision and 0.5 (OD) for near vision. Her vision was stable without the supply of oxygen; therefore, she was discharged.

Oxygen Fluctuation Monitored with High Frequency in a Eutrophic Urban Stream (the Anyang Stream) and the Effect of Weather Condition (부영양 도심하천(안양천)에서 고빈도 관측에 의한 산소고갈과 기상조건의 영향 연구)

  • The variation of dissolved oxygen (DO) was monitored with high frequency by an automatic data-logging sensor in a eutrophic urban stream (the Anyang Stream) located in a metropolitan area of Seoul, South Korea. In general, DO showed the diel variation of increase in daytime and decrease at night, implying that primary production is a major mechanism of oxygen supply in this ecosystem. The fluctuation of oxygen was determined by rainfall. DO depletion was most obvious after a rainfall resulting in an anoxic condition for a day, which is thought to be caused by scouring of periphyton and organic ooze at the stream bottom. Seasonally DO was higher in winter and frequently depleted in warm seasons. DO depletion was often at a dangerous level for fish survival. Fish survey showed that little fish was living at the study site and oxygen depletion may be the major stress factor for aquatic animals. From the results it can be suggested that a high frequency monitoring of oxygen should be established for the proper assessment of aquatic habitats and better management strategy.

Effect of broccoli sprouts germination by soaking water condition (침지조건이 브로콜리 발아에 미치는 영향)

  • In order to optimization germination condition of broccoli, we carried out germination ratio experiment under soaking time, soaking temperature, oxygen concentration. The germination ratio results 98.5% following as soaking water having 10 ppm DO(demanded oxygen) during 4 hr at $20{\sim}30^{\circ}C$. The result of provide the dissolved oxygen water, when the concentration of oxygen water were 7, 12 ppm, germination ratio were observed about 76%, 92%. Also, we showed the growth rate of 2 times in 12 ppm compared with 7 ppm. Consequently, germination ratio of broccoli increased cultivation condition at soaking water having high concentration oxygen more than supply to oxygen water of high concentration.

Effect of Highly Concentrated Oxygen Administration on Addition Task Performance and Physiological Signals (고농도 산소가 덧셈과제 수행능력과 생리신호에 미치는 영향)

  • This study investigated the effect of 40% oxygen administration on the addition task performance in three levels of difficulties and physiological signals. Ten male and female college students were selected as the subjects for this study. The experiment consisted of two runs: one was a addition task, with normal air (21% oxygen) administered and the other was with hyperoxic air (40% oxygen) administered. The experimental sequence in each run consisted of Rest1 (3 min), Task1 (1 min, one digit addition task), Task2 (1 min, two digit addition task), Task3 (1 min, three digit addition task), and Rest2 (4 min). Blood oxygen saturation and heart rate were measured throughout the five phases. The accuracy rates of the addition task were enhanced with 40% oxygen administration compared to 21% oxygen. Difference in the accuracy rates grew higher with the rise of difficulty. When 40% concentration oxygen is supplied, blood oxygen saturation increased and heart rate was decreased comparing to 21%. This study showed that the supply of high concentration oxygen increases blood oxygen saturation, which in turns accelerates brain activation resulting from cognitive process and enhances arithmetic abilities. Particularly when difficulty is high, demand for oxygen increases and, as a result, the effect of high concentration oxygen becomes more significant.

Degradation of Phenol in Water Using Circulation Dielectric Barrier Plasma Reactors (순환식 유전체 장벽 플라즈마 반응기를 이용한 수중 페놀 처리)

  • Objectives: The purpose of this study was evaluating the applicability of the circulation dielectric barrier plasma process (DBD) for efficiently treating non-biodegradable wastewater, such as phenol. Methods: The DBD plasma reactor system in this study consisted of a plasma reactor (discharge, ground electrode and quartz dielectric tube, external tube), high voltage source, air supply and reservoir. Effects of the operating parameters on the degradation of phenol and $UV_{254}$ absorbance such as first voltage (60-180 V), oxygen supply rate (0.5-3 l/min), liquid circulation rate (1.5-7 l/min), pH (3.02-11.06) and initial phenol concentration (12.5-100 mg/l) were investigated. Results: Experimental results showed that optimum first voltage, oxygen supply rate, and liquid circulation rate on phenol degradation were 160 V, 1 l/min, and 4.5 l/min, respectively. The removal efficiency of phenol increased with the increase in the initial pH of the phenol solution. To obtain a removal efficiency of phenol and COD of phenol of over 97% (initial phenol concentration, 50.0 mg/l), 15 min and 180 minutes was needed, respectively. Conclusions: It was considered that the absorbance of $UV_{254}$ for phenol degradation can be used as an indirect indicator of change in non-biodegradable organic compounds. Mineralization of the phenol solution may take a relatively longer time than that required for phenol degradation.

Oxygen Mass Balance Analysis in an Intermittently Aerated Wetland Receiving Stormwater from Livestock Farms (축산유역 강우유출수 처리를 위한 간헐 포기식 인공습지에서 산소수지분석)

  • In order to assess the role of aeration in stormwater wetlands, oxygen supply and consumption in a wetland treating runoff from livestock farms were estimated and analyzed. Furthermore, oxygen mass balance was conducted during day time and night time. Internal production by algal photosynthesis dominated the oxygen production particularly in the shallow marsh due to the large amount of algae. Consequently, algal respiration was also the major oxygen depletion element with nitrification and biodegradation estimated as 5.35% and 6.43% of the total oxygen consumption. This excessive portion of oxygen consumption by algae was associated to the highly turbid water caused by the resuspension of sediment particles in the aeration pond, which also affected the subsequent wetland. Moreover, an abundance of oxygen was estimated during the day indicating that oxygen produced by algal activity is sufficient to meet the oxygen demand in the wetland. Thus, supplemental aeration was deemed not necessary at daytime. In contrast, oxygen was greatly depleted at night when algal photosynthesis stopped which induced denitrification. Therefore, it was suggested that supplemental aeration may be operated continuously instead of intermittently to avoid oxygen deficit in the wetland at night or it may be stopped entirely to further enhance denitrification.
