담배 연기 중 산소 자유 라디칼 측정에 의한 품질 평가 (Evaluation of Cigarette Quality by Measurement of Oxygen Free Radicals in Smoke)

  • 지금까지 연기중의 Nicotine, CO등 몇몇 화합물의 함량 측정과 관능 검사등을 통하여 담배의 품질을 평가해 왔다. 그러나 담배 연기중의 수 많은 화학 성분들을 고려할 때 전연기 성분이 생체에 미치는 영향을 측정함으로써 그 품질을 평가할수 있는 새로운 방범이 요구된다. 담배연기에 의한 생체 손상 쿵 가장 일차적이고 영향이 큰 것으로 알려진 활성 산소종(H2O2, O2-, ·OH)은 그 반응성이 크고 life time이 짧기 때문에 측정이 어렵다. 저자들은 이들을 분해하는 효소를 이용하여 연기중에서 생성되는 이들 산소 자유 라디칼을 측정하였으며 그 결과 담배의 종류에 따라 이들의 생성이 현저한 차이를 보여 일반적으로 고급 담배로 알려진 것들은 낮았고 저급 담배일 수록 높았다. 따라서 이들 산소 자유 라디칼의 측정은 흡연과 건강이라는 측면에서 매우 유용한 담배의 품질 평가 방법으로 이용될수 있을 것으로 생각된다.

Indium Tin Oxide(ITO) Thin Film Deposition on Polyethylene Terephthalate(PET) Using Ion Beam Assisted Deposition(IBAD)

  • Tin-doped indium oxide(ITO) thin films were deposited on polyethylene terephthalate(PET) at room temperature by oxygen ion beam assisted evaporator system and the effects of oxygen gas flow rate on the properties of room temperature ITO thin films were investigated. Plasma characteristics of the ion gun such as oxygen ions and atomic oxygen radicals as a function of oxygen flow rate were investigated using optical emission spectroscopy(OES). Faraday cup also used to measure oxygen ion density. The increase of oxygen flow rate to the ion gun generally increase the optical transmittance of the deposited ITO up to 6sccm of $O_2$ and the further increase of oxygen flow rate appears to saturate the optical transmittance. In the case of electrical property, the resistivity showed a minimum at 6 sccm of $O_2$ with the increase of oxygen flow rate. Therefore, the improved ITO properties at 6 sccm of $O_2$ appear to be more related to the incorporation of low energy oxygen radicals to deposited ITO film rather than the irradiation of high energy oxygen ions to the substrate. At an optimal deposition condition, ITO thin films deposited on PET substrates showed the resistivity of $6.6{\times}10^{-4}$ ${\Omega}$ cm and optical transmittance of above 90%.

일산화탄소 중독시 고압산소투여가 8-hydroxydeoxyguanosine 생성에 미치는 영향 (Effects of Hyperoxia on 8-Hydroxydeoxyguanosine Formation in Carbon Monoxide Exposed Rats)

  • Hyperbaric oxygen (HBO) therapy for carbon monoxide (CO) poisoning eventually inducing the hypoxia-reoxygenation condition, may produce oxygen free radicals, which forms 8-hydroxydeoxyguanosine (8-OH-dG) by attacking C-8 position of deoxyguanosine (dG) in DNA. Effects of oxygen partial pressure or duration of HBO therapy with or without CO poisoning on the tissue 8-OH-dG formation were investigated. Male Sprague-Dawley rats were grouped and exposed to air (control group), 4000 ppm of CO for 10 to 30 minutes (CO only group), air for 30 minutes after 30 minute exposure to 4000 ppm of CO(CO-air exposure group), HBO after 30 minute exposure to 4000 ppm of CO(CO-HBO group), or HBO therapy fo. $10{\sim}120$ minutes(HBO only group). The 8-OH-4G concentrations in the brain and the lung tissues were measured with high performance liquid chromatography and electrochemical detector (ECD). Average concentrations of the 8-OH-dG of each group were statistically compared. In the brain tissues, 8-OH-dG concentrations of the CO only group, the CO-air exposure group, and the CO-HBO group did not significantly differ from those of the control group. Similar insignificance was also found between the CO-HBO group and the HBO only groups. No appreciable dose-response relationship was observed between the 8-OH-dG concentration and the oxygen partial pressure or the duration of HBO. However, the 8-OH-dG concentrations of the 30 minute CO only group were higher than those of the CO-air exposure group (p-value<0.05). In the lung tissues, there were no significant differences between the 8-OH-dG concentrations of the control group and those of the CO only group, the CO-air exposure group, and the CO-HBO group. However, mean 8-OH-dG concentration of the CO-air exposure group was significantly higher than that of the CO only group under the same CO exposure condition(p-value<0.05). With the duration of CO exposure, the 8-OH-dG concentrations of the lung tissues decreased significantly (p-value<0.05). The concentrations of 8-OH-dG in the lung tissues proportionally increased with the duration of HBO, but no such relation was observed with the oxygen partial pressure. These results suggest that the brain may be more resistant to oxygen free radicals as compared with the lungs, and that oxygen toxicity following HBO may be affected by factors other than oxygen free radicals.

동물실험을 통한 솔잎(松葉) 유효성분의 항노화효과 구명 및 구조 해명 I. 간장의 활성산소 및 제거효소에 미치는 솔잎 추출물의 영향 (Investigation of Anti-aging Effect and Determination of Chemical Structures of Pine Needle Extract (PNE) through the Animal Experiments I. Effects of PNE on Oxygen Radicals and Their Scavenger Enzymes in Liver of SD Rats)

  • These studies were designed to investigate the effects of pine (Pinus densiflora Sieb et Zucc.) needle extract (PNE) on oxygen radicals and their scavenger enzymes in liver membranes of Sprague-Dawley (SD) rats as a study on investigation of anti-aging effect and determination of chemical structures of PNE through the animal experiments. Male SD rats were fed basic diets (control group) and experimental diets (0.5% and 1.0%-PNE group) for 6 weeks. There were no significant differences in hydroxyl radical (·OH) formations of liver mitochondria and microsomes in 0.5%-PNE group, while ·OH formations were significantly decreased (10% and 18%, respectively) in liver mitochondria and microsomes of 1.0%-PNE group compared with control group. Microsomal hydrogen peroxides and cytosolic superoxide radicals were remarkably decreased (20% and 20∼25%, respectively) in 0.5% and 1.0%-PNE groups compared with control group. Mn-SOD activities in mitochondria were significantly increased about 10% in 1.0%-PNE group, while Mn-SOD activities in mocrosomes were remarkably increased (16∼20%) in 0.5% and 1.0%-PNE groups compared with control group. There were no significant differences in Cu, Zn-SOD activities of liver cytosol in 0.5% and 1.0%-PNE groups, while glutathione peroxidase (GSHPx) and catalase (CAT) activities were significantly decreased (28∼30% and 15∼30%, respectively) in liver cytosols of 0.5% and 1.0%-PNE groups compared with control group. These results suggest that these PNE may play a effective role in a attenuating a oxygen radical formations and increasing a scavenger enzyme activities.

  • Pine(Pinus densiflora Sieb et Zucc.) is one of rhe popular plant drugs which has been used as a medicine in Asia. To investigate the effect of pine needle extract (PNE) on oxygen radicals and their scavenger enzymes in brain membranes of Sprague- Dawley (SD), make SD rats were fed basic diets(control group), and experimental diets (PNE group) with 0.5 and 1.0% of PNE 6 weeks. Mitochondrial hydroxyl radical levels in brain of 0.5%-PNE and 1.0%-PNE groups were significantly inhibited to 30% and 25%, respectively, and microsomal hydrogen peroxide levels in brain of 0.5%-PNE and 1.0%-PNE groups were significantly inhibited to 15% compared with control group. Cytosolic superoxide rdical levels in 1.0%-PNE group were significantly inhibited to 20% compared with control group. Lipid peroxide(LPO) levels in brain mitochondria of 0.5%-PNE and 1.0%-PNE groups were significantly lower(25% and 35%) than that in control group. Mn-superoxide disumtase (SOD) activities in brain of 0.5%-PNE and 1.0%-PNE groups were significantly higher(18% and 12%) than those in control groups, but Cu,Zn-SOD activities in brain of 0.5%-PNE were significantly activated to 15% compared with control group. Glutathione peroxidase(GSHPx) activities in brain of 1.5%-PNE and 1.0% PNE groups were significantly higher(14% and 12%) than those in control group. These results suggest that more beneficial effects such as inhibition of oxygen radicals and lipid peroxide(LPO). and oncreases of scavenger enzymes in brain membranes of SD rats may be effectively modulated by administration of pine needle extract (PNE)

Effects of Flavonoids of Ginseng Leaves on Erythrocyte Membranes against Single Oxygen Caused Damage

  • It has been well known that extended exposure to reactive oxygens causes severe damage to susceptible biomolecules. In this study, the effects of flavonoids including trifling and kaempferol from Ginseng leaves on single oxygen induced photohemolysis of erythrocytes and free radical scavenging activities were investigated . Each flavonoid aglycone (5-50UM) such as kaempferol, quercetin or baicalein exhibited a high protective effect against the photohemolysis. They protected the cells by scavenging 102 and free radicals. Although the free radical scavenging activities of the flavonoid glycosides were not much lower than those of their corresponding aglycones, their insolubility into lipid bilayers of membrane made them less effective in preventing the photohemolysis induced by 1O2. The 102 and free radical scavenging activities of flavonoids were estimated by the decomposition of the flavonoid by 1O2 and the bleaching of free radicals by the flavonoid, respectively. The solubilization of the flavonoid into micelle or erythrocytes was deduced from spectrophotometric and microscopic observations. The cooperation of L-ascorbic acid and a flavonoid, and a possible involvement of lipoxygenase or cyclooxygenase in the photohemolysis mechanism were discussed. 가감온비탕합오산(加減溫脾湯合五散) 완제(完製)(HWW$^{(R)}$)의 항산화 효과에 대한 연구 (Antioxidative Activities of Wen-pi-tang-Hab-Wu-ling-san (WHW$^{(R)}$) in vitro)

  • Objectives: The objective of this study was to investigate the antioxidant effects of manufactured Wen-pi-tang-Hab-Wu-ling-san (WHW$^{(R)}$) in vitro. Methods: WHW$^{(R)}$ was prepared by the pilot manufacture of WHW water extract from a GMP system appointed company. Antioxidative activities were determined by in vitro tests as follows: the scavenging activities of oxygen free radicals including 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide anion, hydrogen peroxide and nitric oxide radicals, as well as ferrous ion chelating capacity and Trolox equivalent antioxidant capacity (TEAC). Results: WHW$^{(R)}$ significantly scavenged oxygen free radicals such as DPPH (IC$_{50}$=115.28 $\pm$ 0.25 $\mu$g/$m\ell$), superoxide anion (IC$_{50}$=8.56 $\pm$ 0.08 $\mu$g/$m\ell$), hydrogen peroxide (IC$_{50}$=240.36 $\pm$ 3.41 $\mu$g/$m\ell$) and nitric oxide (IC$_{50}$=162.28 $\pm$ 0.21 $\mu$g/$m\ell$) radicals. WHW$^{(R)}$ also showed ferrous ion chelating activity (IC$_{50}$=543.19 $\pm$ 4.85 $\mu$g/$m\ell$) and Trolox equivalent effects (IC$_{50}$=45.311 $\mu$g/$m\ell$) in TEAC and ORAC assay, respectively. Conclusion: This study demonstrates that WHW$^{(R)}$ has strong antioxidative properties through free radical scavenging activity. These data suggest that WHW$^{(R)}$ be used as an antioxidant agent.
