  • This study was designed to investigate the relationship between the nutrient intakes, antioxidants, and inflammatory markers of overweight and obese adults (46 females and 16 males) in Yeongdong area. The study was conducted through questionnaires, anthropometric checkups, 2-day 24 hr recalls and blood biomarker analyses. Body fat was significantly higher in women compared to men. Diastolic blood pressure (DBP) and systolic blood pressure (SBP) were significantly higher in men compared to women. There were no significant differences in height, weight, body mass index, and waist size among the two groups. The intake of nutrients was significantly higher in men compared to women. Ferric reducing ability plasma (FRAP) was significantly higher in men compared to women. Body fat was positively associated with blood IL-6 and IL-8 levels. DBP was positively associated with FRAP. The intake of protein was negatively associated with IL-6 levels. The intake of carbohydrates was negatively associated with total antioxidant capacity (TAC). Prostaglandin E2 (PGE2) levels were negatively associated with TAC. These results suggest that antioxidant and inflammatory markers may be related to the body fat percentage and dietary intake in overweight and obese adults.

  • Oxidative stress has been reported to be one of causes of neuritis. This study examined antioxidative activities of methanol extracts of six amphibian species known to be medicinal animals (Rana catesbeiana, R. coreana, R. rugosa, R. dybowskii, R. nigromaculata, and Hyla japonica) and investigated their effects of inhibiting nitric oxide (NO) production and cytotoxicity on the murine macrophage RAW264.7 cells. As inflammation is closely associated with reactive oxygen species, assays on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, xanthine oxidase inhibitory activity, superoxide anion radical scavenging activity and NO scavenging activity of the extracts of the six species were performed to investigate their antioxidative activity. The results obtained were as follows; All extracts showed antioxidative activity, and the activity of R. dybowskii was the highest in comparison among those. Anti-inflammatory effects of the extracts were also examined, the five extracts except that of R. rugosa did not show cytotoxicity for RAW264.7 cells at the maximal concentration ($1,000{\mu}g\;mL^{-1}$). Selectivity index, meaning NO scavenging activity compared to cytotoxicity, showed the highest level in the extract of R. dybowskii. These results will be very useful basic data for future studies on prevention and treatment of human diseases to understand the biological roles of amphibian extracts throughout the antioxidative or anti-inflammatory pathways.

  • This study compared the composition and texture of muscle and levels of detoxification and antioxidant enzymes in the livers of stocked and cultivated rockfish Sebastes schlegeli released after the primary culture stage in Tongyoung, South Korea. The crude lipid content of muscle was significantly higher ($P$<0.05) in cultivated rockfish than stocked rockfish, while the texture did not differ significantly ($P$>0.05). The condition factor (CF) and hepatosomatic index (HSI) did not differ significantly and the growth of stocked and cultivated rockfish was similar. The levels of the detoxification enzymes cytochrome P450 (CYP) and Ethoxyresorufin-O-deethylase (EROD) were significantly lower in the livers of stocked rockfish, perhaps because of their reduced exposure to xenobiotic compounds. In addition, stocked rockfish had a significantly ($P$<0.05) lower CAT and higher GR than cultivated rockfish, but similar levels of tGPx, SOD, GSH, and GSSG. The present study shows that the growth rates of stocked and cultivated rockfish are similar and that stocked rockfish are exposed to fewer xenobiotic compounds and less oxidative stress.

  • Background: Based on the assertion that apocynin diminishes acute lung injury (ALI) by inhibition of NADPH oxidase, the effect of apocynin was tested in interleukin-$1{\alpha}$ (IL-1)-induced ALI in rats. Methods: IL-1 was insufflated into the trachea of Sprague-Dawley rats to induce ALI, and apocynin (8 mg/kg) was given intravenously for inhibition of NADPH oxidase. In addition, we determined whether apocynin inhibited generation of superoxide anions from isolated human neutrophils. Five hours after IL-1 instillation, lung injury parameters, expression of cytosolic phospholipase A2 (cPLA2) by cells from bronchoalveolar lavage (BAL), an index of oxidative stress in lung tissues (${\gamma}$-glutamyltranspeptidase, activity), and ultrastructure of alveolar type II (AT II) cells were evaluated. Results: Apocynin decreased the generation of free radicals from phorbol myristate (PMA)-activated neutrophils in vitro, but did not ameliorate ALI. IL-1 induced enhancement of the expression of cPLA2 on neutrophils was not altered by apocynin. Conclusion: Apocynin induced suppression of the generation of superoxide anions from neutrophils by inhibition of NADPH oxidase does not attenuate IL-1-induced ALI in rats.

  • Luteolin and chicoric acid are the most abundant phytochemicals in dandelion (Taraxacum officinale) leaf. In this study, four kinds of extraction methods [hot water, ambient temperature (AT) water, ethanol, and methanol] were applied to analyze the contents of both phytochemicals and verify their anti-inflammatory and antioxidative activities. The methanol extract showed the most potent nitric oxide (NO) inhibitory effect. The luteolin and chicoric acid concentrations were 3.42 and $12.86\;{\mu}g/g$ dandelion leaf in the methanol extract. The NO-suppressive effect of luteolin and chicoric acid was identified in a dose-dependent manner with $IC_{50}$ values of $21.2\;{\mu}M$ and $283.6\;{\mu}M$, respectively, in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells without cytotoxicity. Malondialdehyde (MDA) concentration, as an index for free radical injury on cell membrane, was also dose-dependently inhibited by the two compounds. The suppressive effect was further examined using mRNA and protein expression levels, which were attributable to the inhibition of inducible nitric oxide synthase (iNOS). These results suggest that two phytochemicals in dandelion leaf, luteolin and chicoric acid, may play an important role in the amelioration of LPS-induced oxidative stress and inflammation.

  • In a previous study, a mulberry fruit extract(MFE) supplement exhibited anti-inflammatory activity and improved serum lipid profiles in arthritic rats. The objective of this study was to determine whether dietary MFE could ameliorate inflammatory parameters and serum lipid levels in humans. Twenty-six middle-aged subjects(mean body mass index=27 $kg/m^2$) consumed MFE(100 $m{\ell}/day)$ after lunch for 4 wks. Anthropometric measurements, serum oxidative stress markers and serum lipid profile analyses were performed at baseline and then at 4 wk following the study. There were no significant differences in anthropometric measurements, including BMI, WHR, and body fat composition. After the 4 wk-intervention, serum levels of C-reactive protein(CRP), ferric-reducing ability of plasma(FRAP), serum triglyceride(TG) and LDL-cholesterol had significantly decreased(p<0.05), whereas serum levels of HDL-cholesterol significantly(p<0.05) increased. These findings suggest the consumption of mulberry extract may be protective against inflammation and the atherosclerotic state in elderly obese men at high risk for cardiovascular disease(CVD).

  • Selaginella tamariscina is a traditional herb used in medicine. Phytochemical amentoflavone, a biflavonoid class of flavonoids, was isolated from the plant of Selaginella tamariscina. In this study, the antibacterial effects and combination effects of amentoflavone and conventional antibiotics such as ampicillin, cefotaxime, and chloramphenicol were investigated. These results showed that amentoflavone had a considerable antibacterial effect and synergistic interaction with antibiotics against various bacterial strains (fractional inhibitory concentration index ${\leq}$ 0.5), except for Streptococcus mutans. To study the mechanism(s) involved in the synergistic activities between amentoflavone and antibiotics, we detected hydroxyl radical formation using 3'-(p-hydroxyphenyl) fluorescein and measured the $NAD^+/NADH$ ratio by $NAD^+$ cycling assay. The results indicated that the formation of hydroxyl radical would be a cause of the synergistic effect and that this oxidative stress originated from a transient NADH depletion. This study suggests that amentoflavone synergizes with antibiotics and has potential as a therapeutic agent for antimicrobial chemotherapy.

  • Oxygen radical absorbance capacity(ORAC) is known to be a sensitive and simple method to determine total antioxidant capacity(TAC) in biological samples. While TAC has received great attention with its relation to pathogenesis in the progression of several diseases, little is known about association of ORAC with metabolic risk factors. The aim of this study is to evaluate the relationship between ORAC and serum lipid profiles, fasting glucose and anthropometric measures. One hundred seventeen volunteers participated in the study. Perchloric acid treated serum was used to determine $ORAC_{pca}$. Mean$ORAC_{pca}$ of subjects whose serum total cholesterol(TC) concentrations were $\geq$ 200 mg/dl was significantly(P < 0.05) higher than that of subjects whose TC concentrations were < 200mg/dl. There were significantly positive correlations between $ORAC_{pca}$ and serum concentrations of TC(P < 0.05) and low density lipoprotein (LDL) cholesterol(P < 0.01). The positive relation between cholesterol concentrations and $ORAC_{pca}$ in serum may suggest an elevated TAC against oxidative stress associated with the cardiovascular risk factors. • The purpose of this study was to investigate the effects of oral supplementation of coenzyme Q10 (CoQ10) for 4 weeks on the plasma free oxygen radical and total antioxidant capacity at resting and after one bout exercise in healthy old men. Thirty volunteers with an average (+/-SD) age of 62.59+/-5.3 years participated in this study and were divided with three groups; CoQ10 (200 mg daily) group, vitamin C & E (800mg, 400 IU daily) group, and placebo group. A cycle exercise (60% HRR) test was performed at the end of study. Blood samples were taken for the analyses at rest and pre-, post-, 30min after cycle exercise, before and after the 4 weeks of supplementation. After supplementation, there were no significant differences in the plasma free oxygen radical levels and total antioxidant capacity at resting. Plasma free oxygen radical level and total antioxidant capacity in three groups were significantly elevated after exercise, however, it did not vary significantly between groups. CoQ10 supplementation showed significant difference in total antioxidant capacity during recovery phase compared with placebo group. Our results demonstrated that supplementation of CoQ10 in healthy old men improve blood total antioxidant capacity after one bout exercise, despite no alteration of plasma free oxygen radical levels.
