Variations in Geochemical characteristics of the Acid Mine Drainages due to Mineral-Water Interactions in Donghae Mine Area in Taebaek, Korea (태백 동해광인일대의 물-광물의 반응에 의한 산성광산배수의 지구화학적 특성 변화)

  • There are several abandoned coal mines around Donghae mine area in the Taebaek coal field. Two major creeks, Soro and Sanae, are contaminated with the colored precipitates formed from the coal mine drainages. Bed rocks of the study area consist of limestone, shale, and sandstone. Limestone consisted mainly of calcite and dolomite, and shale of quartz, pyropyllite and chlorite, and sandstone of quatz and illite. Coal coal spoil dumps composed mainly of pyrite and chlorite. The oxidative dissolution of sulfide minerals leads to acid mine drainage and adds the metal ions in the stream water. The ion concentrations of Fe, Ca, Mg, Al, Si, SO$_{4}$in the stream polluted by AMD are generally higher than those in the unpolluted stream water. High concentrations of Ca and Mg, Al and Si can be resulted from dissolution of carbonate minerals such as calcite, dolomite and aluminosilicates such as chlorite, pyrophyllite. Although the Fe, Al, Si, SO$_{4}$ contents are considerbly high in the acid water released from the mine adits, they become decreased downstream due to dilution of unpolluted water and precipitation of oxide/hydroxide and sulfate minerals on the bottom of stream.

Changes in the Functionality of Cheonggukjang During Fermentation Supplemented with Angelica gigas, Rehmanniae Radix, and Red ginseng (당귀.지황.홍삼 첨가에 따른 발효 청국장의 기능성 변화 연구)

  • Cheonggukjang is one of the traditional fermented soy-based foods in Korean diets. Studies in cell cultures, humans have revealed anti-hypertension, anti-stress, anticancer, antioxidant, immune enhancing effects. Angelica gigas, Rehmanniae radix, and Red ginseng are popular medicinal plants and widely used for oriental medicine. In this study a strategy had been developed to mobilize beneficial phenolics from Angelica gigas, Rehmanniae radix, and Red ginseng combined with fermented soy by Cheonggukjang fermentation for antioxidant and Type II diabetes management. The quality and functional characteristics of Chenggukjang fermented with Angelica gigas, Rehmanniae radix and Red ginseng. Cheonggukjang (CKJ), Angelica gigas Cheonggukjang (CKJ-DD), Rehmanniae radix Cheonggukjang (CKJ-RG), Angelica gigas and Rehmanniae radix Cheonggukjang (CKJ-DD+RG) and Red ginseng Cheonggukjang (CKJ-RED) were evaluated. The mobilized phenolic profile was evaluated for antioxidant activity and the potential to inhibit ${\alpha}$-amylase linked to hyperglycaemia. This research has important implications for the development of functional soy-based-fermented foods enriched with Angelica gigas, Rehmanniae radix and Red ginseng phenolics for oxidative stress - induced diabetic complications. Furthermore, Hunter's color values of 5 types cheonggukjang, lightness (L-values), redness (a-values) and yellowness (b-values) were evaluated. Free amino acid content of CKJ-RED (0.993 mg/gd. w.) showed higher than that of CKJ (0.205 mg/g-d.w.).

The Effects of Smoking, Drinking and Exercise on Autioxidant Vitamin Intakes and Plasma Antioxidant Status in Elderly People Living in Ulsan (울산지역 노인의 생활습관에 따른 항산화 영양소 섭취실태 및 혈액내 항산화 영양 상태 비교)

  • Body antioxidant status is an important factor in the prevention of many chronic diseases caused by oxidative stress, especially in the elderly and is affected by health-related habits, such as smoking, drinking and regular physical activity. The aim of this study was to investigate the relationship between these health-related habits and plasma antioxidant status in the elderly. Plasma antioxidant status was examined by determining plasma levels of antioxidant vitamins (vitamin C, A, E, $\beta$ -carotene), total antioxidant status (TAS) and thiobarbituric acid-reactive substance (TBARS) . The subjects included 225 elderly persons aged over 60 yews (63 males, 162 females) living in the Ulsan area. They were interviewed to collect data on their general characteristics and health behaviors such as smoking, exercise and alcohol consumption by means of questionnaires. Their dietary intakes were obtained by means of semi-quantitative food frequency questionnaires (FFQ) The study population was divided into two or three groups according to their smoking, drinking, and exercise status. The ratio of smoker, drinker and exerciser was 16.7%, 31.0% and 44.2% respectively. The dietary antioxidant vitamin intakes were not significantly different among groups of smoking and drinking status, but tended to be higher in non-smokers and non-drinkers than in smokers and drinkers. Plasma vitamin C and $\beta$ -carotene levels were significantly higher in non-smokers, but Plasma vitamin A and TBARS levels were significantly lower in non-smokers than in smokers. Plasma TAS was not significantly different among the smoking groups, but showed a tendency to decrease with an increase in the number of packyear. Plasma vitamin C and $\beta$ -carotene levels of the non-drinkers were higher than those of drinkers and past-drinkers, but plasma vitamin A, C, E, TAS and TBARS showed no difference among the groups of drinker. All vitamin intakes of the exercisers were slightly higher than those of the non-exercisers, but vitamin C intake was significantly higher in female exercisers than in non-exercisers. Plasma $\beta$ -carotene levels were significantly higher in male exercisers and plasma vitamin A, C, E, TAS and TBARS levels tended to be higher in exercisers than in non-exercisers. These results suggested that change to non-smoker, modulation of alcohol consumption and regular exercise could enhance antioxidant defences against reactive oxygen species and might increase the likelihood of a healthier life span.

  • Onion (Allium cepa L.) production in Korea has increased gradually over the past 15 years, placing second in food consumption survey with 20.6 g daily intake in 2006. Onions, used as an ingredient in many dishes and accepted by almost all traditions and cultures, have been reported to have a range of health benefits which include anticarcinogenic, antiasthmatic, antibiotic, and antioxidative effects. These effects may be attributable to a powerful flavonoid pigment-containing compounds, such as quercetin and alk(en)yl cysteine sulphoxides (ACSOs). Although antiplatelet and antithrombotic activities of onion have been confirmed by many of in vitro or animal studies, only a few human intervention studies have been examined. The majority of human studies identified that onion improves some cardiovascular markers such as lipid profile and platelet coagulant. With regard to antioxidative effects, somewhat positive effects are confirmed through strengthening the resistance of oxidative DNA damage in lymphocyte and urine, while most studies failed to find inhibitory effects on LDL oxidation. The discrepancies among studies might be ascribed to producing area, processing methods of onion, dosage, subject characteristics, study duration, and measurement methods. In this review, we focused on the preventive effect of cardiovascular disease through onion consumption in human intervention studies.

Chemical Oxidation Treatment of Hydrocarbon-Contaminated Eine Soil by ${H_2}{O_2}$/$Fe^0 System (${H_2}{O_2}$/$Fe^0시스템을 이용한 유류오염 미세토양의 화학적 산화처리)

  • In this Study, application of ${H_2}{O_2}$/$Fe^0 oxidation System (Fenton-like oxidation) for the oxidative treatment of high-level soil contamination with hydrocarbon was suggested. The characteristics of Fenton-like oxidation of diesel-contaminated fine soil was experimentally probed in a batch system varying initial pH, zero valent iron and hydrogen peroxide levels, and initial diesel concentration. Contaminant degradation was identified by total petroleum hydrocarbon(TPH) concentration with gas chromatography. The batch experiments showed that the optimal ${H_2}{O_2}$and $Fe^0 dosage, 10% ${H_2}{O_2}$+ 20% $Fe^0 removed 65% of initial TPH concentration (10,000mg/kg) at a retention time of 24h. And the TPH removal in the ${H_2}{O_2}$/$Fe^0 system effectively proceeded only within a limited pH range of 3-4. The zero valent iron-catalyzed Fenton-like oxidation of diesel-contaminated soil was more competitive to the $FeSO_4-catalyzed system (Fenton oxidation) in removal efficiency and cost especially for the treatment of high level contamination.

Anti-oxidative Capacity of Mulberry Leaf and its Tea (뽕잎 및 뽕잎차의 항산화능)

  • The importance of genetic resources conservation and utilization for economy and the health and well-being of societies in general is recognized all over the world. Mulberry genetic resources contain many natural components and are considerable resource for functional sericulture. We carried out antioxidative capacity analysis of mulberry leaves that were collected from Korea and some of foreign countries and preserved in the greenhouse. The mean antioxidant capacity of 34 accessions was 892.30nmol(ascorbic acid equivalent). Also we evaluated fruity characteristics and selected 4 accessions as breeding resources for the production of mulberry fruit. To investigate the effect of tea on antioxidative capacity, five kinds of tea(coffee mix, green tea added brown rice, mulberry leaf tea, Polygonatum odoratum tea and black tea added lemon) were selected and analyzed. Their's antioxidative capacity were 2,531.01nmol, 1,867.42nmol, 1,053.72nmol, 292.71nmol and 188.91nmol, respectively. The antioxidative capacity of drinking water soaked with mulberry leaf showed 891.96nmol.

Isolation and characterization of a monodehydroascorbate reductase gene in poplar (Populus alba × P. glandulosa) (현사시나무 monodehydroascorbate reductase (MDHAR) 유전자의 분리 및 발현특성)

  • Monodehydroascorbate reductase (MDHAR) is an important enzyme that plays a role in the detoxification of reactive oxygen species (ROS) by maintaining reduced pool of ascorbate through recycling the oxidized form of ascorbate. In this study, we isolated a PagMDHAR1 gene from Populus alba ${\times}$ P. glandulosa, and investigated its expression characteristics. The PagMDHAR1 cDNA encodes a putative 434 amino acids containing FAD- and NAD(P)H-binding domains. Southern blot analysis indicated that a single nuclear gene encodes this enzyme. Northern hybridization analysis revealed that PagMDHAR1 is highly expressed in both suspension cells and flower tissues, while its expression levels were enhanced by drought, salt, cold, wounding and ABA. Therefore, PagMDHAR1 might be expressed in response to abiotic stress through the ABA-mediated signaling pathway in this poplar species, suggesting that the PagMDHAR1 plays an important role in the defense mechanisms against oxidative stress.

Nutritional Components of Zespri Green Kiwi Fruit (Actinidia delicosa) and Neuronal Cell Protective Effects of the n-hexane fraction (제스프리 그린 키위의 주요 영양성분 및 헥산 분획물의 신경세포 보호효과)

  • The physiological characteristics of kiwi (Actinidia delicosa) fruit were analyzed, which inclued its nutritional composition, in vitro-antioxidant activities, and neuronal cell protective effects. The most abundant components of mineral, amino acid, and fatty acid were found to be potassium (K), glutamic acid, and a-linolenic acid, respectively. The major free sugars were fructose, glucose, and sucrose. In addition, ${\beta}$-carotene and vitamin C contents were $1.35{\mu}g/100mL$ and 29.21 mg/100 g, respectively. The 2,2'-azinobis-(3-ethylbenothiazline-6-sulfonic acid) (ABTS) radical-scavenging activity of the n-hexane fraction obtained from the kiwi extract was 10.52% at a concentration of $1000{\mu}g/mL$. The malondialdehyde (MDA) inhibition of the n-hexane fraction was found to increased in a dose-dependent manner. The intracellular reactive oxygen species (ROS) accumulation after hydrogen peroxide ($H_2O_2$) treatment of PC12 cells was significantly reduced in the presence of the n-hexane fraction compared to PC12 cells treated with $H_2O_2$ only. Moreover, in the a MTT assay, the n-hexane fraction showed in vitro-protective effects against $H_2O_2$-induced neurotoxicity.

Studies on the physicochemical and biochemical characteristics in sesame seed juice under different roasting conditions (참깨의 볶음 조건이 참깨 착즙액의 이화학적 및 생화학적 특성에 미치는 영향)

  • In this study, we investigated the effect of roasting temperature on nutrient content, digestive enzyme activities, and antioxidative properties of sesame seed juice. The sesame seeds were either roasted at 160, 200, and $240^{\circ}C$ or not roasted, and the juice was extracted using a low-speed juice extractor. Owing to the short duration of roasting, benzo[a]pyrene were not detected and trans fatty acids were negligible detected in all sesame seed juices. The sesame seed juice contained abundant nutrients such as minerals, vitamins, and fatty acids. The contents of minerals, vitamin B1 and B3, and sesamol increased with increase in roasting temperature; however, the levels of fatty acids, vitamin B2, sesamin, and sesamolin decreased. In addition, the antioxidant content and antioxidative activities of sesame seed juice increased with increase in roasting temperature. Therefore, these results suggest that roasted sesame seed juice possesses high antioxidative activities, which may be beneficial for preventing oxidative damage in the body.

Analysis of Nutritional Components and Cell-based Antioxidant Activity on Brassica juncea Cultivated in Jeongseon, South Korea (정선 갓의 영양성분 및 세포기반 항산화 활성 분석)

  • The purpose of this study was to demonstrate the quality characteristics of Brassica juncea cultivated in Jeongseon (BJJ), South Korea. We analyzed the nutritional components and antioxidant activity of BJJ. As a result of the free sugar analysis, the contents of glucose and fructose in BJJ were $0.29{\pm}0.02g/100g$ and $0.10{\pm}0.00g/100g$, respectively. The major fatty acids were palmitic acid, octadecenoic acid and stearic acid. The palmitic acid was the highest at 31.22% of all fatty acids. The major minerals were identified as Ca, P, K, Mg and Na. The contents of vitamin $B_1$, vitamin $B_2$, vitamin $B_6$, vitamin C and vitamin E in BJJ were $0.02{\pm}0.00mg/100g$, $0.087{\pm}0.01mg/100g$, $0.02{\pm}0.00mg/100g$, $0.56{\pm}0.06mg/100g$ and $0.20{\pm}0.03mg\;{\alpha}-TE/100g$, respectively. As a result of the free amino acid analysis, total amino acid contents in BJJ were $2,801.21{\pm}115.38mg/100g$. L-proline content was the highest ($744.30{\pm}119.06mg/100g$) in BJJ. BJJ extract inhibits reactive oxygen species production in 3T3-L1 adipocytes. Also, BJJ extract exhibits a protective effect on oxidative stress in $H_2O_2$-induced human dermal fibroblast. These results indicate that BJJ comprises various valuable nutrients which can be used as functional food ingredients.
