Anti-inflammatory and Anti-oxidant Activities of Aster Scaber Ethanol Extract

  • In mountainous regions, wild herbs which can also be edible in nature for humans and animals possess a wide array of biologically diversified properties. It is because of the fact that due to the cold weather of mountains; they are enriched in certain kinds of phytochemicals such as anti-oxidants, anti-inflammatory and many more. One such kind of an herb is Aster scaber (AS) in Korean. It is a widely cultivated culinary herb in Korean peninsula and used as a side dish in Korean culinary cuisine. In view of its extensive use in cuisine, we geared to unravel the anti-oxidant and anti-inflammatory effects of AS in murine alveolar macrophage cell line (MH-S). 2,2'-Azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) assays revealed a dose dependent (7.8~1,000 ㎍/mL) inhibition of oxidation by AS 70% ethanol (ASE) extract as compared to Trolox and Ascorbic acid respectively. Nitric oxide assay (NO) showed a dose dependent decrease (5~40 ㎍/mL) in MH-S cells with ASE when stimulated with Coal Fly Ash (CFA). Moreover, this dose for NO reduction was also found to be least cytotoxic for cells as determined by cellular viability (MTT) assay. The gene expression of pro-inflammatory mediators (iNOS and COX-2) and cytokines (IL-6 and IL-1β) and were also dose dependently inhibited by ASE in MH-S cells through RT-PCR. Therefore, in light of these findings, AS exhibited a strong anti-oxidant and anti-inflammatory agent. These results also justify the extensive use of this mountainous herb in culinary practices for beneficial effects on human health.

Fabrication of Porous Cu Layers on Cu Pillars through Formation of Brass Layers and Selective Zn Etching, and Cu-to-Cu Flip-chip Bonding (황동층의 형성과 선택적 아연 에칭을 통한 구리 필라 상 다공성 구리층의 제조와 구리-구리 플립칩 접합)

  • The feasibility of an efficient process proposed for Cu-Cu flip-chip bonding was evaluated by forming a porous Cu layer on Cu pillar and conducting thermo-compression sinter-bonding after the infiltration of a reducing agent. The porous Cu layers on Cu pillars were manufactured through a three-step process of Zn plating-heat treatment-Zn selective etching. The average thickness of the formed porous Cu layer was approximately 2.3 ㎛. The flip-chip bonding was accomplished after infiltrating reducing solvent into porous Cu layer and pre-heating, and the layers were finally conducted into sintered joints through thermo-compression. With reduction behavior of Cu oxides and suppression of additional oxidation by the solvent, the porous Cu layer densified to thickness of approximately 1.1 ㎛ during the thermo-compression, and the Cu-Cu flip-chip bonding was eventually completed. As a result, a shear strength of approximately 11.2 MPa could be achieved after the bonding for 5 min under a pressure of 10 MPa at 300 ℃ in air. Because that was a result of partial bonding by only about 50% of the pillars, it was anticipated that a shear strength of 20 MPa or more could easily be obtained if all the pillars were induced to bond through process optimization.

Nitrogen Transport In Groundwater-Surface Water Hyporheic Zone at Brackish Lake (기수호의 지하수-지표수 혼합대 내 질소 거동 분석)

  • Sediment, aquifer materials, surface water, and groundwater from brackish Songji lake affected by salinity of seawater, were collected and a pilot scale column experiment was conducted to simulate the nitrogen transport through the hyporheic zone. Upstream experiments of groundwater displayed that groundwater containing a small amount of salt percolated into aquifers and sediments, maintaining low dissolved oxygen concentrations. In addition, partial denitrification occurred in the aquifer due to salinity and low dissolved oxygen, resulting in the accumulation of NO2-. In sediments,nitrogenous compounds were reduced due to adsorption by long residence times or microbial-mediated oxidation/reduction reactions. Downstream experiments of surface water displayed that surface water from the brackish lake, containing high concentrations of dissolved oxygen and salts, infiltrated into the sediments and aquifer, supplying high dissolved oxygen concentrations. This resulted in biological nitrification in the sediments and aquifer, which reduced nitrogen-based pollutants despite the high salt concentration in the surface water. Whereas partial denitrification at low dissolved oxygen concentrations in the upwelling mixing zone was observed by salinity and accumulated NO2-, nitrification at high dissolved oxygen concentrations in the downwelling mixing zone was not significantly affected by salinity. These results confirm that salinity in the brackish water lake has some influence on the nitrogen behavior of the hyporheic mixing zone, although nitrogen behavior is a complex combination of factors such as DO, pH, substrate concentration, and organic matter concentration.

The Hepatotprotective and Antioxidative Effects of Onion (Allium cepa) Extracts in Rat Hepatocyte Primary Culture (양파(Allium cepa) 추출물의 간보호 및 항산화 효과)

  • The objectives of present study were to investigate the hepatoprotective and antioxidative effects of onion extracts. Primary cultures of rat hepatocytes were incubated with 1.5 mM tert-butyl hydroperoxide(t-BHP), potent oxidizing agent for liver injury for 1 hr in the presence or absence of various concentrations (0, 0.01, 0.05, 0.1 or 0.3 mg/ml) of onion extract. Cytotoxicity and cell viability were determined by measuring glutamic oxaloacetic transaminase(GOT) activity, lactate dehydrogenase(LDH) activity and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) value. Lipid peroxidation was evaluated using thiobarbituric acid reactive substances(TBARS) assay. Effects on antioxidant system were determined by measuring catalase, glutathione peroxidase(GSH-Px), glutathione reductase(GSH-Rd) activities as well as DNA strand breaking assay. Incubation with t-BHP alone increased GOT and LDH activities and TBARS concentration but decreased MTT reduction. Onion extracts at the concentration of 0.05 mg/ml began to decrease GOT and LDH activities induced by 1.5 mM t-BHP. Decreased MTT reduction began to be increased by onion extract at the concentration of 0.01 mg/ml. Onion extracts at the concentration of 0.01 mg/ml began to decrease TBARS concentration induced by t-BHP. Taken together, onion extracts prevented t-BHP-induced hepatocyte injury and lipid peroxidation. Catalase, GSH-Px and GSH-Rd activities of hepatocytes were significantly decreased by 1.5 mM t-BHP for 1 hr incubation. Onion extracts, on the other hand, at the concentration of 0.1 mg/ml began to prevent t-BHP-induced decrease in catalase, GSH-Px and GSH-Rd activities. Onion extracts prevented hydroxyl radical-induced single-strand breakage in dose-dependent manner when plasmid DNA was incubated with various concentrations of onion extracts in the presence of Fenton regents producing hydroxyl radical. These results demonstrate that onion extracts suppressed t-BHP-induced cytoctoxicity, decreased viability and lipid peroxidation and increased GSH-Px, GSH-Rd and catalase activities. Thus hepatoprotective and antioxidant effects of onion extract seem to be due to, at least in part, the increase in antioxidant enzyme activities as well as prevention from hydroxyl radical-induced oxidation, followed by inhibition in lipid peroxidation.

Catalase, glutathione peroxidase(GSH-Px) and glutathione reductase(GSH-Rd) activities of hepatocytes were significantly decreased by t-BHP. Onion extracts at the concentration of 0.1 mg/ml prevented t-BHP-induced decrease in catalase, GSH-Px and GSH-Rd activities. Study on Reduction of Microbial Contamination on Daruma by Combination Treatment of Strong Acidic Hypochlorous Water and Ultrasonic Waves (강산성차아염소산수와 초음파를 병용처리한 조미오징어 반가공품의 미생물 오염도 저감화에 관한 연구)

  • This study was performed to develop treatment method for reducing microbial contamination on Daruma (a semi-processed product of seasoned and dried squid) by combination of strong acidic hypochlorous water (SAHW) and ultrasonic waves (UW). The available chlorine concentration, oxidation reduction potential (ORP) and pH of SAHW were $69.67{\pm}0.58ppm$, $1071.33{\pm}4.16mV$ and 2.79, respectively. The 1.49 log CFU/g of viable cell count and 1.32 log CFU/g of Staphylococcus aureus was reduced, and Escherichia coli was reduced below detection limit when the Daruma was treated with 20 times (w/v) of sodium hypochlorite solution (SHS) for 120 min. The 3.62 log CFU/g of viable cell count and 3.22 log CFU/g of Staphylococcus aureus was reduced, and Escherichia coli was reduced below detection limit when the Daruma was treated with 20 times (w/v) of SAHW for 120 min. The antibacterial effects of SAHW were stronger than those of SHS at same available chroline concentration. SAHW treatment after washing strongly alkalic electrolyzed water (SAEW) showed better bactericidal effects than SAHW treatment only. The 4.0 log CFU/g of viable cell count was reduced, S. aureus was reduced below regulation limit (Log 2.0 CFU/g), and E. coli was reduced below detection limit when the Daruma was treated with 20 times (w/v) of SAHW for 90 min after washing with 20 times (w/v) of SAEW for 60 min. The viable cell number was reduced below detection limit and S. aureus was reduced below regulation limit when the Daruma was treated with 20 times (w/v) of SAHW for 60 min in ultrasonic washer. E. coli was reduced below detection limit when the Daruma was treated with 20 times (w/v) of SAHW for 10 min in ultrasonic washer. These results suggest that combination of SAHW and UW may be a good technique to reduce the microbial contamination in daruma.

  • The 10mm diameter aggregates made of clay, carbon and $Fe_2O_3$ were prepared to investigate the mechanism of black core formation. The specific gravity, absorption rate, percent of black core area, fracture strength, total Fe analysis, and XRF were measured at various compositions, sintering temperatures, sintering times, sintering atmospheres, and sintering methods. Small addition of $Fe_2O_3$ did not affect physical properties of the aggregates; however, the percent of black core area increased with increasing carbon contents and increasing sintering temperature. Specific gravity of the aggregates decreased and the water absorption ratio increased with increasing percent of black core area. The aggregates sintered at oxidation atmosphere showed clear border between shell and black core area. Hence, the aggregates sintered at reduction atmosphere showed only black core area in the cross-section of the aggregates. The specific gravity of the aggregates sintered at reduction atmosphere increased with increasing carbon contents and that was the lowest of all comparing other aggregates sintered at different atmospheres. Adsorption rate increased with increasing carbon contents at all atmospheres. The fast sintered aggregates showed lower specific gravity, higher absorption rate, and more black core area than the normally sintered aggregates. It was turned out that the aggregates having more black core area showed higher fracture strength than that of aggregates with no black core area. From the total Fe analysis, the concentration of Fe and FeO was higher at black core area than at shell. Because the concentration of $Fe_2O_3$ in the shell was higher than other area, the color of the shell appeared red. It was also turned out from the XRF analysis that carbon was exist only at black core area.

Inhibitory effects of persimmon (Diospyros kaki Thumb.) against diet-induced hypertriglyceridemia/hypercholesterolemia in rats (떫은감 (Diospyros kaki Thumb.)이 흰쥐의 식이성 이상지질혈증에 미치는 영향)

  • Purpose: This study aimed to investigate the potential of freeze-dried persimmon powder (Diospyros kaki Thumb.) to protect against dyslipidemia induced by a high-fat/cholesterol diet (HFD) in a rat model. Methods: Fifty Wistar rats were randomly divided into five groups: normal control (NC), high-fat/cholesterol control (HC), tannin in HFD (HT, 1% of diet), immature persimmon in HFD (HI, 7% of diet), and mature persimmon in HFD (HM, 7% of diet). Tannin was used as a positive control. Biochemical, molecular, and histopathological changes were observed in the blood and liver. Results: We confirmed that a high fat/cholesterol diet successfully induced dyslipidemia, which was characterized by significantly altered lipid profiles in the plasma and liver. However, oxidized low-density lipoprotein levels, histopathological damage in the liver, and hepatic triglyceride levels were significantly reduced in all HT, HI, and HM groups compared to those in the HF group. In contrast, plasma apolipoprotein B level was significantly reduced only in the HT and HM groups, whereas reduction of the LDL-C level was detected only in the HI group. Although HF-induced sterol regulatory element-binding protein (SREBP) gene expression was significantly reduced in all treated groups, downstream gene expression levels varied among the different groups; significant reduction of fatty acid synthase (FAS) and 3-hydroxy-3-methylglutaryl-CoA (HMGCR) gene expression was detected only in the HI group, whereas cholesterol $7{\alpha}$-hydroxylase (CYP7A1) gene expression was significantly elevated only in the HM group. Conclusion: Taken together, the data suggest that protection of LDL oxidation and hepatic lipogenesis might be, at least partly, attributed to tannin in persimmons. However, the identified mechanisms varied up to the maturation stage of persimmon. In the case of immature persimmon, modulation of FAS and HMGCR gene expression was prominent, whereas in the case of mature persimmon, modulation of CYP7A1 gene expression was prominent.

Separation of Ni and Fe from $H_2SO_4$ leaching solution of scrapped Fe-Ni alloy (Fe-Ni 합금(合金) 스크랩의 황산(黃酸) 침출액(浸出液)으로부터 Ni와 Fe의 분리(分離))

  • Cementation and solvent extraction processes were studied to separate nickel and iron ions from the $H_2SO_4$ leaching solution with 47 g/L $Fe(Fe^{2+}/Fe^{3+}=1.03),$, 23.5 g/L Ni and 0.90M $H_2SO_4$ which leached from Fe-Ni alloy. Iron powder was used as a reducing agent for the cementation of Ni ion from the leaching solution. The reduction percentage of Ni ion was $17{\sim}20%$ by adding 4 times stoichiometric amount of iron powder at $60{\sim}80$. This may result from the fact that the cementation of Ni ion occurred after the reduction of $Fe^{3+}$ to $Fe^{2+}$ and the neutralization of $H_2SO_4$ with iron powder. The cementation process was proved to be unfeasible for the separation/recovery of Ni ion from the leaching solution including $Fe^{3+}$ as a major component. $Fe^{2+}$ present in the leaching solution was converted to $Fe^{3+}$ for solvent extraction of Fe ion using D2EHPA in kerosene as a extractant. The oxidation of $Fe^{2+}$ to $Fe^{3+}$ was completed by the addition of 1.2 times stoichiometric amount of 35% $H_2SO_4$. 99.6% $Fe^{3+}$ was extracted from the leaching solution (23.5 g/L $Fe^{3+}$) by 4 stages cross-current extraction using 20 vol.% D2EHPA in kerosene. $NiSO_4$ solution with 98.5% purity was recovered from the $H_2SO_4$ leaching solution of scrapped Fe-Ni alloy.

The influence of herbicides on soil microflora -Influence of butachlor- (제초제(除草劑)가 토양미생물(土壤微生物) Flora에 미치는 영향 - Butachlor 제(第)의 영향 -)

  • The purpose of this investigation is to find out the effects on the changes of microflora and the effects on the inorganic nutrients of the fertilized soil and the non-fertilized soil under submerged condition with the treatment of butachlor at the levels of 250, 500, and 1000ppm respectively. The survey was made within the period of 72 days with 7-day intervals under the incubated condition. The result came out to be the following: 1. The Fluctuated changes of soil microflora A. Bacteria: The plots treated by 500ppm of butachlor in case of non-fertilized soil showed the decreasing tendency of bacteria until 21st day. But there were no effects on the other plots. The plots treated by 250ppm and 500ppm in case of fertilized soil showed decreasing tendency on the 7th day, and 1000ppm until 21st days. B. Actinomycetes: There was no effects on the actinotnycetes in case of the non-fertilized soil but the fertilized soil showed some decrease. In both cases, actinomycetes have generally shown a little increase according to the time passage. C. Fungi: In both cases of the fertilized soil and the non-fertilized soil, the plots treated by the media showed some decreasing tendency in comparison with the control plots. There was no change to the number of fungi according to the time passage. 2. The fluctuated changes of nutrient A. $NH_4-N$: The whole plots showed decrease of $NH_4-N$ by adding butachlor. The higher the intencity of butachiar showed the more decreasing tendency of $NH_4-N$. In case of the non-fertilized soil, the highest increase of $NH_4-N$ appeared from the 7th day to 14th day, but showed degrease thereafter. The increasing tendency was seen in case of the fertilized soil plots. B. $NO_3-N$: Decrease of $NO_3-N$ was shown in the whole plots by the treatment of media, and on the 44th day of cultivation almost none of $NO_3-N$ was detected. C. $NO_2-N$: Whole plots showed the number of $NO_2-N$ highest on the 35th day, and there were nothing measurable on the 44th day. D. Eh: On the fertilized soil, the condition of reduction went on strongly but on the non-fertilized soil, the condition of reduction kept on till 42nd day and oxidation appeared thereafter.
