Rabbit Liver and Lung Microsomal Metabolism of $\beta$-Nicotyrine:Isozyme Specificities toward the Oxidation of $\beta$-Nicotyrine

  • Studies on the biodisposition of beta-nicotyrine by lung and liver microsomes was examined in order to provide a better understanding of its fate in this tissue. beta-nicotyrine (100$\mu$M) was incubated with microsomes (1 mg/ml) prepared from New Zealand White rabbits. The rate of oxidation observed in lung microsomal incubations was 1.7 nmoles $\beta$-nicotyrine oxidized mg$^{-1}$ min$^{-1}$ compared with 2.7 nmoles $\beta$-nicotyrine oxidized mg$^{-1}$ min$^{-1}$ by the liver microsomal preparation. However, when these rates were expressed as a function of cytochrome P-450 content, the specific activity of the metabolic oxidation catalyzed by lung (8.3 nmoles $\beta$-nicotyrine oxidized nmole cytochrome P-450$^{-1}$ min$^{-1}$) was approxiamtely 4 times greater than liver microsomes (2.3 nmoles $\beta$-nicotyrine oxidized nmole cytochrome P-450$^{-1}$ min$^{-1}$). Isozyme studies on the oxidation of $\beta$-nicotyrine employed several methods of altering activities of specific isozymes present in pulmonary microsomes, including the use of the isozyme 2 and 6 specific inhibitor $\alpa$-methyl ABT, metabolic inhibitor(MI) complex formation. The results of this inhibition study would appear to indicate the $\beta$-nicotyrine is metabolized predominantly by pulmonary isozyme 5.

Kinetics and Mechanism of the Oxidation of Carbon Monoxide on CoO-$\alpha-Fe_2O_3$ Catalysts

  • The oxidation of carbon monoxide by gaseous oxygen on 0.53, 1.02, and 1.51 mol $\%$CoO-doped $-Fe_2O_3$ catalysts has been investigated in the temperature range from 340 to 480$^{\circ}C$ under various CO and $O_2$ partial pressures. The oxidation rates have been correlated with 1.5-order kinetics; the 0.5-order with respect to $O_2$ and the first-order with respect to CO. In the above temperature range, the activation energy is 0.34 $\pm$ 0.01 eV${\cdot}$$mol^{-1}$. The electrical conductivity of 0.53, 1.02, and 1.51 mol %CoO-doped $\alpha$-$Fe_2O_3$ has been measured at 350$^{\circ}C$ under various $P_{CO}and $P_{O_2}$. From the conductivity data it was found that $O_2$ was adsorbed on Vo formed by doping with CoO, while CO appeared essentially to be chemisorbed on the lattice oxygen of the catalyst surface. The proposed oxidation mechanism and the dominant defect were supported by the agreement between the kinetic data and conductivities.

Kinetics and Oxygen Vacancy Mechanism of the Oxidation of Carbon Monoxide on Perovskite$Nd_{1-x}Sr_xCoO_{3-y}$ Solutions as a Catalyst

  • The oxidation of carbon monoxide by gaseous oxygen in the presence of a powdered $Nd_{1-x}Sr_xCoO_{3-y}$ solid solution as a catalyst has been investigated in the temperature range from 150$^{\circ}$C to 300$^{\circ}$C under various CO and $O_2$ partial pressures. The site of Sr substitution, nonstoichiometry, structure, and microstructure were studied by means of powder X-ray diffraction and infrared spectroscopy. The electrical conductivity of the solid solution has been measured at 300$^{\circ}$C under various CO and $O_2$ partial pressures. The oxidation rates have been correlated with 1.5-and 1.2-order kinetics with and without a $CO_2$ trap, respectively; first-and 0.7 order with respect to CO and 0.5-order to $O_2$. For the above reaction temperature range, the activation energy is in the range from 0.25 to 0.35 eV/mol. From the infrared spectroscopic, conductivity and kinetic data, CO appears essentially to be adsorbed on the lattice oxygens of the catalyst, while $O_2$ adsorbs as ions on the oxygen vacancies formed by Sr substitution. The oxygen vacancy mechanism of the CO oxidation and the main defect of $Nd_{1-x}Sr_xCoO_{3-y}$ solid solution are supported and suggested from the agreement between IR data, conductivities, and kinetic data.

Metabolism-Dependent Cavalent Binding of $S(-)-^3H-Nicotine$ to Lung Microsomes in Vitro

  • Incubation of $S(-)-^3H$-nicotine with rabbit lung microsomes in the presence of dioxygen and NADPH results in the formation of metabolities that bind covalently to microsomal macro-molecules. The addition of cytochrome P-450 monooxygenase inhibitors, $\alpha$-methylbenzyl ami-nobenzotriazole and aroclor 1260, inhibited both (S)-nicotine metabolism and covalent binding. The relative rates of oxidation of nicotine $\Delta^{1',5'}$ iminium ion to continine indicates that lung $100,000\times{g}$ supematant catalyzed this oxidation approximately 18 times slower than that of liver system based on mg of protein, and increased covalent interactions. Since than that of liver system based on mg of protein, nd increased covalent interactions. Since the activity of lung iminium oxidase appears much lowr than the liver, it is tempting to speculate that localized concentrations of nicotine $\Delta^{1',5'}$ iminium ion in the lung will survive for a longer period of time. These results support that cytochrome P-450 catalyzed oxidation of nicotine leads to the formation of reactive nad electrophilic intemediates capable of chemical interactions with biomacromolecules.

강화도 남단 갯벌의 혐기성 유기물 분해능과 황산염 환원력 및 저서 동물이 이에 미치는 잠재적 영향 (Rates of Anaerobic Carbon Mineralization and Sulfate Reduction in Association with Bioturbation in the Intertidal Mudflat of Ganghwa, Korea)

  • 본 연구에서는 강화도 남단 갯벌에서 혐기성 유기물 분해능과 황산염 환원력을 정량화하고 유기물 분해에 있어 황산염 환원의 상대적 중요성에 미치는 저서 동물의 잠재적 영향에 대해 토의하고자 하였다. 혐기성 유기물 분해능은 $26{\sim}85\;mmol\;C\;m^{-2}\;d^{-1}$비 범위로 조사되었고, 이를 강화도 남단의 갯벌 면적 (약 90 $km^2$)으로 환산하면 하루 동안 약 46 ton의 유기물이 분해되는 것이라 할 수 있다. 황산염 환원력은 $22.6{\sim}533.4\;nmol\;cm^{-3}\;d^{-1}$의 범위로 조사되었으며, 전체 혐기성 유기물 분해의 $31{\sim}129%$를 차지하는 것으로 나타났다. 이는 연구지역에서의 혐기성 유기물 분해가 황산염 환원에 의해 주도되고 있음을 의미한다. 한편, 10월에 혐기성 유기물 분해에서 황산염 환원이 차지하는 비중이 상대적으로 낮게 나타난 반면, 공극수 내 Fe(II)의 농도가 증가한 것으로 나타났다. 이러한 결과들은 대형 저서 동물 활동에 의해 Fe(II)의 재산화가 촉진됨으로써 공극수 내 Fe(III)의 공급이 원활해졌고, 그로 인해 유기물 분해과정에서 철 환원 작용이 황산염 환원 작용을 약화시켰음을 의미한다.

순환여과시스템에서 온도가 질산화 반응에 미치는 영향 (Effect of Temperature on Nitrification in a Recirculating Aquaculture System)

  • The effects of temperature on nitrification of enriched nitrifiers were investigated by using kinetics and thermodynamics method through the batch test. Aquaculture recirculating water, which was sampled at Chung Cheong Buk-Do Inland Fisheries Research Institute, was analized to observe the characteristics of nitrification. Temporal variation of ammonium, nitrite and nitrate concentration was measured at batch experiments. Activation energy was calculated using Arrhenius equation with the oxidation rates of specific ammonium or nitrite ion. These oxidation rates were measured at temperature range of $6-35^{\circ}C$ and ammonium concentration range of 0.2-1.8 mg/L. Two distinct activation energy of Nitrosomonas sp. at temperature $6-15^{\circ}C\;and\;15-35^{\circ}C$ was 93.1 and 25.0 KJ/mol, respectively. Nitrate accumulation was observed at temperature over $15^{\circ}C.$

Ar+O2 혼합가스 취입에 의한 용철의 탈탄 반응속도 (Decarbonization Kinetics of Molten Iron by Ar+O2 Gas Bubbling)

  • Molten iron with 2 mass % carbon content was decarbonized at 1823 K~1923 K by bubbling $Ar+O_2$ gas through a submerged nozzle. The reaction rate was significantly influenced by the oxygen partial pressure and the gas flow rate. Little evolution of CO gas was observed in the initial 5 seconds of the oxidation; however, this was followed by a period of high evolution rate of CO gas. The partial pressure of CO gas decreased with further progress of the decarbonization. The overall reaction is decomposed to two elementary reactions: the decarbonization and the dissolution rate of oxygen. The assumptions were made that these reactions are at equilibrium and that the reaction rates are controlled by mass transfer rates within and around the gas bubble. The time variations of carbon and oxygen contents in the melt and the CO partial pressure in the off-gas under various bubbling conditions were well explained by the mathematical model. Based on the present model, it was explained that the decarbonization rate of molten iron was controlled by gas-phase mass transfer at the first stage of reaction, but the rate controlling step was transferred to liquid-phase mass transfer from one third of reaction time.

Fe-2.3%Cr-1.6%W 합금의 고온산화 부식속도와 스케일 분석 (High-temperature Oxidation Kinetics and Scales Formed on Fe-2.3%Cr-1.6%W Alloy)

  • The T23 steel, whose composition was Fe-2.3%Cr-1.6%W, was arc-melted, and oxidized between $600^{\circ}C$ and $900^{\circ}C$ in air for up to 7 months. The amount of precipitates in the arc-melted microstructure was as large as 11.4 vol.%. The precipitates increased the oxidation rate of the arc-melted T23 steel. Owing to the low amount of Cr in the T23 steel, breakaway oxidation occurred after a few hours during oxidation above $700^{\circ}C$ in both arc-melted and as-received T23 steels. The scales that formed on arc-melted and as-received T23 steels were similar to each other. They consisted primarily of the outer $Fe_2O_3$ layer and the inner ($Fe_2O_3$, $FeCr_2O_4$)-mixed layer. The precipitates increased the microhardness and the oxidation rates.

금강밀과 dark northern spring밀의 기울과 배아에서 추출한 기름의 광산화 과정 중 천연산화방지성분의 변화 (Changes in Natural Antioxidants in Oils Extracted from the Bran and Germ of Keumkang and Dark Northern Spring Wheats During Photo-oxidation)

  • 금강밀과 Dark Northern Spring(DNS)밀의 제분 중 발생하는 기울과 배아로부터 추출한 기름(WBG oil)을 $5^{\circ}C$, 1700 Lux로 12일 동안 광산화시킬때 토코페롤, 카로티노이드, 인지질 함량 변화를 측정하고 이들과 WBG oil의 광산화 관련성을 평가하였다. 토코페롤, 카로티노이드, 인지질은 WBG oil의 광산화 중 분해되었으며 DNS밀 WBG oil이 금강밀 WBG oil에서보다 토코페롤의 분해가 빨랐으나, 카로티노이드와 인지질 분해 속도는 느렸다. WBG oil의 광산화 정도와 산화방지성분의 상관관계는 매우 높았으며 인지질이 WBG oil의 광산화에 가장 큰 영향을 나타내었다. 금강밀 WBG oil에 비해 리놀레산 함량이 낮고 올레산 함량이 높았던 DNS밀 WBG oil은 지방산 조성은 물론 인지질의 높은 함량과 낮은 분해 속도에 기인하여 높은 광산화안정성을 나타내었다. 본 결과는 빛의 존재하에서 DNS밀 WBG oil보다는 금강밀 WBG oil이 토코페롤 source로 사용될 수 있는 가능성을 보여주었다.
