Effect of Hwangryunhaedoktang on Contact Hypersensitivity and Passive Cutaneous Hypersensitivity in Mice

  • During the screening program to discover antiatopic agents from herbal formulas, we investigated the inhibitory effect of Hwangryunhaedoktang (HT) on passive cutaneous anaphylaxis and oxazolone-induced mouse ear dermatitis. HT significantly inhibited PCA reaction in mice at doses of 50 and 200 mg/kg with inhibitory activity of 31 and 53%, respectively. HT at concentration of 0.05 and 0.1% inhibited ear swelling by 23 and 46% at 16 days in oxazolone-induced mouse ear dermatitis. Both HT with anti without human intestinal microflora showed potent inhibitory activity on the ${\beta}$-hexosaminidase release induced by IgE. Based on these findings, HT may be a usable agent for skin disorder contact dermatitis.

Ameliorative effects of Glechoma hederacea var. longituba extract on oxazolone-induced atopic dermatitis-like skin lesions (긴병꽃풀 추출물의 oxazolone-유도 아토피 피부염 개선 효과)

  • Objectives: This study aimed to investigate the anti-atopic properties of Glechoma hederacea var. longituba extracts (GHE) in murine atopic dermatitis model. Methods: BALB/c mouse ear stimulated with oxazolone (OX) for 4 weeks, then 1% GHE was topically applied every two days for 3 weeks to mouse. Ear thickness was measured by a digital thickness gauge. The ears tissues were collected and subjected to hematoxylin-eosin (H&E) and toluidine blue (TB) staining. Results: Treatment with GHE successfully alleviated the symptoms of atopic dermatitis, such as erythema, horny substance, and swelling. The infiltration of lymphocytes and mast cells were significantly reduced following GHE treatment. Conclusion: Taken together, our results showed that GHE possessed the potential to be a novel immunomodulatory drug against atopic dermatitis.

Histopathologic Features in Animal Model of Atopic Dermatitis Induced by Topical Application of Oxazolone

  • Animal models of atopic dermatitis (AD) are widely used to investigate therapeutic effects of candidates for AD. However, the characteristics of each model are not fully understood. This study was designed to compare the animal models of dermatitis induced by dinitrofluorobenzene (DNFB) and oxazolone (Ox). We investigated the effects of DNFB and Ox on skin thicknesses and weights as well as skin lesions associated with AD such as scale, crust and erythematous eruption, and histopathological changes such as hyperkeratosis, dermal and epidermal hyperplasia and immune cell infiltration in inflamed tissues. Multiple application of 0.5% Ox onto the skin increased skin thickness and weight compared to those of DNFB treated mice, as well as those of normal mice. In addition, topical application of DNFB induced marked scale, crust and erythematous eruption, while Ox induced erythematous eruption and mild scale and crust. Histopathological examination revealed that 0.5% Ox induced marked hyperplasia in the dermis and epidermis, large vesicles, spongiotic changes, mild hyperkeratosis and immune cell infiltration in balb/c mice. These data suggest that multiple applications of Ox can induce chronic AD like dermatitis in balb/c mice.

Dioscorea japonica Thunb. Ethanolic Extract Attenuated Oxazolone-Induced Atopic Dermatitis-like Skin Lesions in BALB/c Mice

  • The rhizomes of Dioscorea japonica Thunb. are widely consumed as food and also used to treat diabetes and polyuria in Korea. This study was undertaken to study the anti-atopic dermatitis effects of a 95% ethanolic extract (DJE) of D. japonica in an oxazolone-stimulated murine model of atopic dermatitis (AD). The therapeutic effects of DJE on AD-like skin lesions were assessed on both ears. DJE (1%) or dexamethasone (0.5%; the positive control) were applied to skin lesions for three weeks. Serum levels of IgE and IL-4 were assessed by ELISA (enzyme-linked immunosorbent assay). Histopathological examinations were performed by hematoxylin and eosin (H&E) and toluidine blue staining and revealed DJE significantly reduced dermal thickness and inflammatory cell infiltration when applied to oxazolone-treated ear skin. DJE-treated AD mice also showed lower serum levels of IgE and IL-4 than oxazolone-stimulated controls. Our findings demonstrate DJE might be a useful safe, topical agent for the treatment of atopic diseases.

Inhibitory Effect of Ginsenoside Rg5 and Its Metabolite Ginsenoside Rh3 in an Oxazolone-Induced Mouse Chronic Dermatitis Model

  • The effect of a main constituent ginsenoside Rg5 isolated from red ginseng and its metabolite ginsenoside Rh3 in a chronic dermatitis model was investigated. Ginsenosides Rg5 and Rh3 suppressed swelling of oxazolone-induced mouse ear contact dermatitis. These ginsenosides also reduced mRNA expressions of cyclooxygenase-2, interleukin $(IL)-1{\beta}$, tumor necrosis factor $(TNF)-{\alpha}$ and interferon $(IFN)-{\gamma}$. The inhibition of ginsenoside Rh3 was more potent than that of ginsenoside Rg5. These findings suggest that ginsenoside Rh3 metabolized from ginsenoside Rg5 may improve chronic dermatitis or psoriasis by the regulation of $IL-1{\beta}$ and $TNF-{\alpha}$ produced by macrophage cells and of $IFN-{\gamma}$ produced by Th cells.

  • Effects of Allolobophora caliginosatrapezoides (Ac) polysaccharide fractions on the inflammation and hypersensitivity were studied in vivo. It showed that Ac polysaccharide fractions have the significant inhibitory activities of inflammation and hypersensitivity; They inhibited significantly the carrageenin-induced paw edema and acetic acid-induced writhing syndrome. They also inhibited significantly the Arthus reaction and delayed hypersensitivity in the sheep red blood cell-sensitized mice in accordance with the inhibition of haemaglutinin titer, haemolysin titer, plaque-forming cells and rosette-forming cells. They also improved markedly the oxazolone-induced dermatitis in rats dose-dependently. As the above results, it exhibited that Ac polysaccharide fraction inhibited not only humoral immune response, but also cell-mediated immune response. It seemed that methanol and ether extracts have also another physiological active agents.

Inhibitory Effect of Protopanxatriol Ginsenosides in an Oxazolone-induced Mouse Psoriatic Model

  • When the inhibitory effect of ginsenoside (G) Re isolated from ginseng and its metabolites G-Rg1, G-F1, G-Rh1 and protopanaxatriol in mouse ear skin psoriasis stimulated by oxazolone was investigated, G-Re and its metabolites suppressed mouse ear swelling stimulated by oxazolone. Among these agents tested, G-Rh1 most potently suppressed ear swelling as well as mRNA expression of COX-2 and proinflammatory cytokines $IL-1{\beta},\;TNF-{\alpha}$ and $interferon-{\gamma}$. These findings suggest that G-Rh1 may improve chronic dermatitis and psoriasis.

Effects of Samhwangsasim-tang Gamibang on Atopic Dermatitis in Mice (삼황사심탕가미방(三黃瀉心湯加味方)이 생쥐에 유발된 아토피 피부염에 미치는 영향)

  • Objectives : Atopic Dermatitis (AD) is chronic skin disease characterized by allergic hypersensitivity reactions. Samhwangsasim-tang Gamibang(SHSST) can treat skin disease by cooling down blood heat, clearing away congenital fever and detoxicating. This study was conducted to investigate the effects of SHSST on AD induced by oxazolone in mice. Methods : We investigated the effects of SHSST on skin tickness, skin lesions, erythema and melanin index, water content and histopathological changes in vivo. The effects on body weights and spleen body weight ratio were also investigated in mice with AD. Results : In our results, oral administration of SHSST ameliorated skin lesion such as flare, erythema, exudate and petechia and lowered skin thickness induced by oxazolone. In addition, SHSST lowered erythema index and elevated water content compared to AD control respectively. In the histopathological observation, oral administration of SHSST prevented epidermal hyperplasia and spongiotic changes which is a hallmark of skin inflammation respectively. Finally, SHSST did not affect spleen/body weight ratio. Conclusions : These data imply that SHSST can ameliorate skin lesion of AD such as flare, erythema, exudate and petechia, and be used to treat AD patients with relative safety.

Antiallergic and Antipsoriatic Effects of Korean Red Ginseng

  • Antiallergic and antipsoriatic effects of korean Red Ginseng (KRG, steamed root of panax ginseng C.A. Meyer, Family Araliaceae) were measured. Orally administered KRG water extract potently inhibited passive cutaneous anaphylaxis (PCA). KRG water extract also showed the potent inhibition in oxazolone-induced mouse dermatitis, and suppressed mouse ear swelling by $39\%$ at 16 days at a dose of $0.1\%$. KRG water extract reduced the levels of mRNA of cyclooxygenase (COX)-2, $IL-1\beta$, $TNF-\alpha$ and $INF-\gamma$ increased in oxazolone-applied mouse ears, however, did not inhibit that of IL-4. KRG water extract also inhibited iNOS and COX-2 mRNA expression level of RAW264.7 cell induced by lipopolysaccharide. Based on these findings, we suggest that KRG can improve atopic and contact dermatitis by the regulation of $ IL-1\beta$ and $TNF-\alpha$ produced by macrophage cells and $interferon-\gamma$ produced by Th1 cells.

Changes in the Anti allergic Effect of Citrus unshii Pericarpium According to Storage Period (진피(陳皮)의 저장기간에 따른 항알러지효과 비교)

  • Objectives : The present study was undertaken to determine whether the ethanol extact of Citri Pericarpium (CP, Pericarp of Citrus unshiu Markovich, Rutaceae) is effective against atopic dermatitis according to it's storage period. Methods : To evaluate antiallergic effect of CP and OCP (Old Citri Pericarpium) evaluated in vivo their inhibitory effects against passive cutaenous anaphylaxis (PCA) reaction induced by IgE-antigen complex and scratching behaviors by compound 48/80. The Anti-atopic effects were measured by contact dermatitis, prurient animal model and PCA reaction. Contact dermatitis in mice as a model of the Type IV reaction caused by Oxazolone. Results : The results showed that anti-pruritus effects, analgesic effects of CP was depends on its hesperetin contents. And It also showed that keep longer in storage appeared to be higher in hesperetin contents. Both CP and OCP(Old CP) have a dose-dependent analgesic action in acetic acid induced writhing syndrome. OCP Potently inhibited PCA reaction in mice, although OCP weakly inhibited in long term contact dermatitis model in mice. Conclusions : These results suggest that the Proportional to the storage period, Citri Pericarpium possesses analgesic effects and anti-allergic effects.
