Effect of Ovariectomy and Dietary Calcium Levels on Bone Metabolism in Rats Fed High Calcium Diet during the Growth Period (성장기 동안 고칼슘 식이를 섭취한 흰쥐의 골격대사에 난소절제 및 칼슘 섭취 수준이 미치는 영향)

  • This study explored the effects of dietary calcium levels and/or ovariectomy on bone formation, bone composition and calcium metabolism using female Sprague-Dawley weanling rats(mean body weight$\pm$SEM : 232.3$\pm$6.7g) as a model. Rats received high(1.5%) calcium diets for eight weeks during the growth period and were randomly assigned to ovariectomy and sham groups. The two groups were than each randomly divided into three sub-groups and fed 0.1%, 0.5% and 1.5% calcium diets for eight weeks after operation. The results indicate that body weight gain was higher in ovariectomy groups than sham groups regardless of dietary calcium levels and food intakes. Serum Ca concentration was decreased in low Ca groups after operation and serum P concentration increased in ovariectomy groups. Serum Ca concentration was decreased in low Ca groups after operation and serum P concentration increased in ovariectomy groups. Serum alkaline phosphatase activity was increased in ovariectomy groups and was not influenced by dietary calcium levels after operation. Urinary hydroxyproline decreased in high Ca intake groups regardless of whether rats had received an ovariectomy or not. The weight, length and breaking force of the femur were not significantly different in all groups. Ash, calcium, phosphate and magnesium contents in the femur and lumbar were not significantly different regardless of ovariectomy operation and dietary calcium levels. But high/normal calcium intake after ovariectomy and sham operation increased the weight and calcium content in bone. Therefore, high calcium intake influenced the formation of peak bone mass during the growth period and calcium levels and calcium levels and calcium levels continued to influence bone growth and composition after ovariectomy.

  • Purpose of study: To compare on bone formation between ovariectomized rats and normal rats after graft of alloplastic bone material. Material and methods: Twenty female rats were used in this study. They were divided into two groups by each ten rats. One group was ovariectomized and grafted alloplastic bone material. Other group was non-ovariectomized and grafted alloplastic bone material. After a healing time of 4 weeks, the animals were sacrificed and decalcified preparations were routinely processed for histologic evaluations. Results: Ovariectomy group was significantly lesser bone formation compared with non-ovariectomy group. Conclusions: Ovariectomy acts as a negative factor in new bone formation.

Induction of FSTL1 Gene in Rat Myometrium by Exogenous Estrogen

  • The aim of this study was carried to the induction of follistatin-like 1 (FSTL1) in rat myometrium tissue by exogenous estrogen. PCR was performed by using estrogen treatment after ovariectomy of myometrium mRNA and only ovariectomy of myometrium mRNA and on ovariectomy of myometrium mRNA in rat normal uterus tissue. The PCR products were visualized on agarose gel (1.2%) electrophoresis. FSTL1 mRNA expression level was significantly higher (**P<0.001 or *P<0.05) in estrogen treatment after ovariectomy than in only ovariectomy, and was significantly higher (**P<0.001) in no ovariectomy than in estrogen with treatment after ovariectomy. In conclusion, Although the mechanisms of FSTL1 gene were uncertain, It also might be inducted by exogenous estrogen. And also it is thought to be related to the estorgen mechanism.

  • An elective laparoscopic ovariectomy on a healthy dog revealed a cystic structure in the left ovary. The surgical procedure was successful. Histopathological examination showed the presence of a teratoma adjacent to the ovary. To the best of the authors' knowledge, this is the first reported case of an ovarian teratoma removed by laparoscopic ovariectomy in a dog by using a multiport laparoscopic ovariectomy technique.

Histological and Immunohistological Studies of the Thyroid Glands of the Osteoporotic Rats Induced by Ovariectomy (난소절제로 유발된 골다공증 시 흰쥐 갑상샘에 대한 조직학적 및 면역조직화학적 연구)

  • In order to investigate the histological and immunohistochemical changes of thyroid glands, and serum osteocalcini estradiol, pmgesterone, follicular-stimulating hormone (FSH), and calcium levels in the osteoporotic rats induced by ovariectomy, normal(sham-operated group) and experimental groups(ovariectomized group) were divided. The experimental groups were subdivide6 to 5, 10, 15, and 20 weeks after ovariectomized groups. The histological structure of the thyroid gland were observed in normal structure throughout all experimental periods but the calcitonin-immunoreartive cells in the experimental groups were significantly decreased compared with normal groups and degranulations of calcitonin-immunoreactive cells were observed. Theses numbers were decreased with time after ovariectomy. However, the serum osteocalcin and FSH levels of experimental groups were significantly increased compared with normal groups and these levels were increased along with time after ovariectomy, but serum estradiol, progestemn and calcium levels of experimental group were significantly decreased compared with normal groups and these levels were decreased with time after ovariectomy.

  • This study explored the effect of calcium levels and/or ovariectomy on bone metabolism using female Sprague-Dawley weanling rats as a model . Rats received a low (0.1%) calcium diet for 8 weeks. The rats were then divided into three subgroups that were fed 0.1% ,0.5% and 1.5% calcium diets for 8 weeks after operation. The results of this experiment indicate that body weight gin was higher in ovariectomy groups than in sham groups regardless of calcium level and food intake. Serum Ca and P concentrations were of normal level regardless of calcium level and ovariectomy. Estrogen concentration was low in the ovariectomized group. Serum alkaline phophatase activity and urinary hydroxyproline have been used as markers of bone formation and resorption. These values were increased in ovariectomized groups. The weight, length and breaking force of femur were not significantly different between the groups. Ash, Ca, P and total lipid contents in femur and lumbar were decreased in the groups fed low calcium . Mg content was decreased in the ovariectomy and total protein content was not affected by calcium level and ovariectomy. The effect groups of ovarectomy on calcium contents of bone was more prominent in lumbar than in femur. In conclusion, though low calcium intakes during growth period may retard the attainment of peak bone mass, calcium supplementation after this period increased bone growth and mineral contents, but not significant effect in three calcium levels. Furthermore, calcium intake was shown to have a greater influence on the mineral contents of femur than of lumbar, and removal of endogenous estrogen production by ovariectomy was shown to be more deleterious on the ash and calcium contents of the lumbar than of femur.

Osteoporotic Changes after Ovariectomy in Rats (랫드에서 난소적출이 골다공증 발생에 미치는 영향)

  • Osteoporosis is a bone disease associated with reduced bone mineral density resulting in debilitating bone fractures. The present study was carried out to determine the influence of ovariectomy(OVX) on serum level of sex steroid and bone metabolism, as well as bone mechanical property, in OVX subjects in comparison with controls. Body weight and food intake was significantly increased in OVX subjects than in controls. The serum level of estradiol(E2) was significantly lower in OVX subjects than in controls. The serum level of calcium and alkalin phosphatae were significantly increased in OVX subjects than in controls. The bone strength, based on the three point bending test, was not significantly different. Seven weeks after ovariectomy, the cavities in the bone reached 223% of the normal. In conclusion, seven weeks after ovariectomy osteoporo sis was evidently appeared.

Effect of Unilateral Ovariectomy on Survival Rate of Embryo and Ovarian Steroid Hormone Levels in the Superovulated Angora Rabbits (Angora 토끼에 있어서 과배란처리후 편측난소척출 수정란의 생존율 및 난소 Steroid 홀몬수준에 미치는 영향)

  • This study was conducted to investigate the effect of unilateral ovariectomy on the embryo survival and changes of plasma progesterone and estradiol concentration. Mature Angora rabbits were divided into 2 groups (Superovulation and unilateral ovariectomy after superovulation). Unilateral ovariectomied rabilts were subdivided into two groups according to the time of ovariectomy (24 hours and 96 hours after mating). The results obtained were summarized as follows; 1. After unilateral ovariectomy, survival rate and collecting rate of embryos recovered from contralateral and ipsilateral oviduct and uterus were not different, and were lower than intact rabbits. 2. Plasma progesterone concentration at 93, 99, 102 and 114 hours after HCG injection in superovulated rabbits were 12.9 $\pm$ 0.5, 34.8 $\pm$ 5.1, 12.2 $\pm$ 2.7 and 43.4 $\pm$ 5.8ng/ml, respectively. Mean progesterone concentrations were significantly higher at 99 and 114 hours than at 93 and 102 hours (p <0.05). But plasma estradiol concentration was not different. 3. Plasma progesterone concentration in unilateral ovariectomized rabbits was somewhat decreased after unilateral ovariectomy. Plasma estradiol concentration was not different.

Ovariectomy, but not orchiectomy, exacerbates metabolic syndrome after maternal high-fructose intake in adult offspring

  • High fructose diet is associated with the global metabolic syndrome (MtS) pandemic. MtS develops in early life, depending on prenatal and postnatal nutritional status. We hypothesized that ovariectomy increases the chances of developing MtS in adult offspring following high fructose intake by the mother. Pregnant C57BL/6J mouse dams drank water with or without 20% fructose during pregnancy and lactation. After weaning, the pups were fed regular chow. The offspring were evaluated until they were 7 months of age after the mice in each group, both sexes, were gonadectomized at 4 weeks of age. The offspring (both sexes) of the dams who had high fructose intake developed MtS. In the offspring of dams who drank tap water, orchiectomy increased the body weight gain and body fat accumulation, while ovariectomy increased the body fat accumulation as compared to the sham controls. In the offspring of dams with high fructose intake, orchiectomy decreased the body weight gain, body fat accumulation, visceral adiposity, and glucose intolerance, while ovariectomy exacerbated all of them as compared to the sham operations. These data indicate that ovariectomy encourages the development of MtS in adult offspring after maternal high fructose intake, while orchiectomy prevents the development of MtS. The sex difference indicates that male and female sex hormones play contradictory roles in the development of MtS.

Effect of Estrogen and Dietary Protein Level on Ca and Skeletal Metabolism in Ovariectomized Rats (난소절제쥐에서 Estrogen을 투여하였을 때 식이 단백질 수준이 Ca 및 골격 대사에 미치는 영향)

  • To investigate the effect of estrogen and dietary protein level on Ca metabolism, female rats were undergone ovariectomy or sham-operation. Ovariectomized rate were divided into either estrogen-or vehicle-treated groups. Each treatment group was again divided into 40%-casein(H) or 10%-casein(L) diet groups. All experimental diets contained 0.2% Ca, 0.4% P and fed to rats for 8 weeks. Apparant Ca absorption and Ca balance were not affected by dietary protein level and ovariectomy, however they were increased by estrogen injection and this effect was even higher in low protein groups. Urinary Ca excretion were higher in high protein groups. GFR was not affected by dietary protein level, ovariectomy, or by estrogen injection. Urinary protein excretion was higher in high protein groups, which implies that the kidney funtion was deteriorated by high protein diet, and this may account partly for the higher urinary Ca in high protein groups. Ovariectomy or estrogen treatment had no effect on urinary protein excretion. Urinary hydroxyproline was higher in ovariectomized rats and increased in high protein grous. Elevated value of ovarictomized rats was lowered by estrogen injection, especially in low protein group. Alkaline phosphatase tended to increase in ovariectomized groups and lowered with estrogen treatment, but this difference was not statistically significant. Serum PTH was not affected by ovariectomy and dietary protein level. Therefore the increased hydroxproline excretion does not seem to be attributed to PTH. Dietary protein level, ovariectomy and estrogen treatment did not affect the weights and components of femur, scapular, and 4th vertebra. Ash/wt ratio of femur was, however, lower in ovariectomized rats and increased with estrogen treatment. Therefore, among the bones studied, femur seemed to be the most vulnerable. The results of this study shows that estrogen treatment may alleviate or reduce bone loss in postmenopausal women somewhat, especially for those people with low protein diet.
