Neem (Azadirachta indica) Seed Cake in Animal Feeding-Scope and Limitations - Review -

  • The different products of neem (Azadirachta indica) are utilized for variety of purposes in industry, health and animal agriculture in the Indian subcontinent. The cake from seeds after oil extraction is a good source of nutrients (CP: 35-38%; EE: 4.5-5.5%; CF: 12-15%; Ca: 0.75%; P: 0.45% on DM), and in particular, the one out of its kernel is proteinaceous and is relatively balanced in its amino acid and mineral profile. But the cake is toxic and bitter to taste owing to triterpenoids (nimbin, salannin, azadirachtin), which restricts its safe inclusion in livestock diet. Several feeding trials with raw cake have revealed poor palatability and adverse performance among different categories of livestock and poultry. Internal organ changes included histological alteration in intestine, liver, kidney and distruption of spermatogenesis and ovarian activity. Ruminants appears to tolerate reasonably higher levels of the cake and to a limited low levels of dietary inclusion also proved to be tolerable in monogastric farm animals. Debitterization through solvent (hexane, ether) extraction, water washing, alkali (NaOH, 1.5, 2.5 or 3%, wt/wt) soaking and urea (1.5 or 3%, wt/wt) - ammoniation have been tried with appreciable success in improving the palatability and nutritive value of the cake. For enhanced utilization, decortication of neem seeds is to be done effectively at industrial level with maximum oil recovery. The resultant proteinaceous kernel by-product could be a cheaper unconventional protein supplement after suitable processing.

Relationship between Intersequence Pauses, Laying Persistency and Concentration of Prolactin during the Productive Period in White Leghorn Hens

  • Prolactin is considered to influence the taking of pauses in between ovulatory sequences in White Leghorn hens. Therefore modulating concentrations of prolactin using bromocriptine - a dopamine agonist during early life (17 to 36 weeks of age) could overcome the inhibitory effects of high concentration of prolactin on ovarian activity. The effect of modulation of prolactin concentration on egg production, sequence length and inter sequence pauses were studied by analyzing the oviposition records from 19 to 72 weeks were studied and compared with untreated controls. Bromocriptine administered subcutaneously (100 $\mu$g kg$^{-1}$ body weight or orally through feed (640 $\mu$g day$^{-1}$ bird$^{-1}$) resulted in a steady and sustained decrease in prolactin levels (p<0.01) during and after the withdrawal of treatment up to one reproductive cycle (72 weeks of age). The treated birds had comparatively longer sequences (p<0.01) and fewer pauses (p<0.01). Egg production increased (p<0.01) by fourteen per cent through subcutaneous administration and eleven per cent through oral feeding, over the control birds. It is concluded that the physiological pauses that occur during ovulatory sequences can be disrupted effectively using bromocriptine. Prolactin levels are modulated which may interfere with the follicular recruitment and subsequent oviposition thereby improve egg laying potential of the bird.

Anti-mutagenic Activity of Salvia merjamie Extract Against Gemcitabine

  • Gemcitabine is an anti-cancer drug with clinically uses in the treatment of various neoplasms, including breast, ovarian, non-small cell lung, pancreaticand cervical cancers, T-cell malignancies, germ cell tumours, and hepatocellular carcinomas. However, it has also been reported to have many adverse effects. Naturally occurring anti-mutagenic effects, especially those of plant origin, have recently become a subject of intensive research. The present study was therefore designed to investigate the anti-mutagenic effects of Salvia merjamie (Family: Lamiaceae) plant extracts against the mutagenic effects of gemcitabine. The anti-mutagenic properties of Salvia merjamie were tested in Inbred SWR/J male and female mice bone marrow cells. The mice were treated in four groups; a control group treated with 30 mg/kg body weight gemcitabine and three treatment groups, each with 30 mg/kg body weight gemcitabine together with, respectively, 50, 100 and 150 mg/kg body weight Salvia merjamie extract. Chromosomal aberration and mitotic index assays were performed with the results demonstrating that Salvia merjamie extract protects bone marrow cells in mice against gemcitabine induced mutagenicity. This information can be used for the development of a potential therapeutic anti-mutagenic agents.

Ultrasonographic Diagnosis for the Treatment of Genital Disease and Early Pregnancy Diagnosis in Korean Native Cattle (한우에서 생식기질환의 치료 및 조기임신진단을 위한 초음파영상진단)

  • Ultrasonographic diagnosis of genital disease and early pregnancy diagnosis was performed in Korean native cattle. The size of ovarian follicle in preovulation, luteal stage and follicular cyst was 18.9, 9.2 and 27.6 mm, respectively, and the thickness of follicular wall was 2.3, 1.8 and 2.8 mm, respectively. The size of corpus luteums in formation stage, activity stage, regression stage, cystic corpora lutea and luteal cyst was 6.2, 11.3, 8.6, 26.7 and 25.9 mm, respectively. The thickness of luteal wall in cystic corpora lutea and luteal cyst was 8.4 and 4.9 mm, respectively. The size of embryo or fetus on day 25, 27, 30, 35, 40, 45 and 50 was 0.8, 0.9, 1.3, 1.5, 2.2, 2.8 and 3.8 cm, respectively. The size of amniotic vesicle on day 25, 27 and 30 was 1.2, 2.1 and 3,0 cm, respectively. The diameter of pregnant uterus on day 25 and 27 was 7.0 and 7.8 cm, respectively. It was concluded that the ultrasonographci values determined in this study can be used as references for the treatment of genital disease and early pregnancy diagnosis in Korean native cattle.

Effect of tebufenozide on embryonic and postembryonic development of sycamore lace bug, Corythucha ciliata(Hemiptera: Tingidae) (버즘나무방패벌레(Corythucha ciliata)의 배자발육과 후배자발육에 미치는 tebufenozide의 영향)

  • These studies were carried out to investigate the effects of nonsteroidal ecdysone agonist, tebufenozide on embryonic and postembryonic development of sycamore lace bug, Corythucha ciliata. Tebufenozide exhibited ovicidal activity($LC_{50}=4.0$ ppm). Susceptibility of each instar nymphs to tebufenozide was nearly same($LC_{50}=2.3{\sim}6.0$ ppm). The tebufenozide treatment in the final instar affected the emergence, longevity and fecundity of surving adults significantly. Tebufenozide applied at high concentration (10 ppm) to younger adults delayed prioviposition period, and decreased adult longevity, the number of eggs laid per female and hatchability. The treatment also inhibited the ovarian development of the female adults.

  • Regulation of cell fate and lung cell differentiation is associated with Aminoacyl-tRNA synthetases (ARS)-interacting multifunctional protein 2 (AIMP2), which acts as a non-enzymatic component required for the multi-tRNA synthetase complex. In response to DNA damage, a component of AIMP2 separates from the multi-tRNA synthetase complex, binds to p53, and prevents its degradation by MDM2, inducing apoptosis. Additionally, AIMP2 reduces proliferation in TGF-β and Wnt pathways, while enhancing apoptotic signaling induced by tumor necrosis factor-α. Given the crucial role of these pathways in tumorigenesis, AIMP2 is expected to function as a broad-spectrum tumor suppressor. The full-length AIMP2 transcript consists of four exons, with a small section of the pre-mRNA undergoing alternative splicing to produce a variant (AIMP2-DX2) lacking the second exon. AIMP2-DX2 binds to FBP, TRAF2, and p53 similarly to AIMP2, but competes with AIMP2 for binding to these target proteins, thereby impairing its tumor-suppressive activity. AIMP2-DX2 is specifically expressed in a diverse range of cancer cells, including breast cancer, liver cancer, bone cancer, and stomach cancer. There is growing interest in AIMP2-DX2 as a promising biomarker for prognosis and diagnosis, with AIMP2-DX2 inhibition attracting significant interest as a potentially effective therapeutic approach for the treatment of lung, ovarian, prostate, and nasopharyngeal cancers.

  • Luteinizing hormone as other glycoprotein hormones is characterized by a heterodimeric structure composed a common $\alpha$-subunit noncovalently linked to a specific $\beta$-subunit. The correct conformation of the heterodimer is important for efficient secretion, hormonal-specific post-translational modifications, receptor binding and signal transduction. To determine whether $\alpha$- and $\beta$- subunits can be synthesized as a single polypeptide chain (tethered-bLH and -bFSH) and also display biological activities, the tetheredbLH and -bFSH molecules were constructed and transfected into chinese hamster ovary (CHO-K1) cells. LH and FSH activities were assayed by using the human embryonic kidney (HEK) 293 cells expressing rat LH and FSH receptor genes. The tethered-bLH and - bFSH proteins were efficiently secreted and showed a similar activity to the dimeric bovine LH and FSH $\alpha$/$\beta$ wild type and native purified from bovine pituitary. The tethered-molecules can be permit development of potent new analogues that stimulate ovarian development. Taken together, a single-chain analog can also be constructed to include additional hormone-specific bioactive generating potentially efficacious compounds. These data indicate the potentiality of the single chain approach to further investigate structurefunction relationships of LH and FSH.

Anti Proliferative Properties of Melissa officinalis in Different Human Cancer Cells

  • Background: Medicinal plants, especially examples rich in polyphenolic compounds, have been suggested to be chemopreventive on account of their antioxidative properties. Melissa officinalis L. (MO), an aromatic and medicinal plant, is well known in thios context. However, toxicity against cancer cells has not been fully studied. Here, we investigated the selective anticancer effects of an MO extract (MOE) in different human cancer cells. Materials and Methods: a hydro-alcoholic extract of MO was prepared and total phenolic content (TPC) and total flavonoid content (TFC) were determined by colorimetric assays. Antioxidant activity was determined by DPPH radical scavenging activity. MTT assays were used to evaluate cytotoxicity of different doses of MOE (0, 5, 20, 100, 250, 500, $1000{\mu}g/ml$) towards A549 (lung non small cell cancer cells), MCF-7 (breast adenocarcinoma), SKOV3 (ovarian cancer cells), and PC-3 (prostate adenocarcinoma) cells. Results: Significant (P<0.01) or very significant (P<0.0001) differences were observed in comparison to negative controls at all tested doses ($5-1000{\mu}g/ml$). In all cancer cells, MOE reduced the cell viability to values below 33%, even at the lowest doses. In all cases, $IC_{50}$ values were below $5{\mu}g/ml$. The mean growth inhibition was 73.1%, 86.7%, 79.9% and 77.8% in SKOV3, MCF-7 and PC-3 and A549 cells, respectively. Conclusions: Our results indicate that a hydro-alcoholic extract of MO possess a high potency to inhibit proliferation of different tumor cells in a dose independent manner, suggesting that an optimal biological dose is more important than a maximally tolerated one. Moreover, the antiprolifreative effect of MO seems to be tumor type specific, as hormone dependant cancers were more sensitive to antitumoral effects of MOE.

Non-specific Defensive Factors of the Pacific Oyster Crassostrea gigas against Infection with Marteilioides chungmuensis: A Flow-Cytometric Study

  • In order to assess changes in the activity of immunecompetency present in Crassostrea gigas infected with Marteilioides chungmuensis (Protozoa), the total hemocyte counts (THC), hemocyte populations, hemocyte viability, and phagocytosis rate were measured in oysters using flow cytometry. THC were increased significantly in oysters infected with M. chungmuensis relative to the healthy appearing oysters (HAO) (P<0.05). Among the total hemocyte composition, granulocyte levels were significantly increased in infected oysters as compared with HAO (P<0.05). In addition, the hyalinocyte was reduced significantly (P<0.05). The hemocyte viability did not differ between infected oysters and HAO. However, the phagocytosis rate was significantly higher in infected oysters relative to HAO (P<0.05). The measurement of alterations in the activity of immunecompetency in oysters, which was conducted via flow cytometry in this study, might be a useful biomarker of the defense system for evaluating the effects of ovarian parasites of C. gigas.

Effects of gender and gonadectomy on growth and plasma cholesterol levels in pigs

  • We conducted two studies to determine the effect of gender, gonadectomy (GDX) on growth and plasma cholesterol levels in pigs. In experiment 1, five sham-operated and five GDX female Landrace pigs (26kg) were allowed to have free access to water and feed up to market weight (approximately 100kg). Body weight and feed consumption were recorded biweekly, and daily body weight gain, daily feed intake and feed efficiency (gain/feed) were calculated during the feeding period. In experiment 2, 10 male (26kg) and 10 female (26kg) Landrace pigs were used; five male and five female pigs were assigned to sham-operated or GDX. Pigs were allowed to have free access to water and a diet without added cholesterol (Table 1) until they were 6 months old (male 104 and female 98kg) and thereafter they were fed a hypercholesterolemic diet (Table 1) containing 0.5% cholesterol and 0.1% cholate for 10 days. GDX of female pigs increased average daily gain (P<0.05), compared with their sham-operated counterparts during the growing-finishing period, but had no effect (P>0.05) on feed efficiency. Plasma cholesterol levels in pigs fed a hypercholesterolemic diet for 10 days were much higher (P<0.05) in females than in males (161 vs 104mg/100mL plasma), and were increased by GDX only in male pigs. HDL-cholesterol/LDL+VLDL-cholesterol ratio appeared to be higher in males than in females, and was not influenced by GDX in either sex. Results suggested that the lower growth rate of female pigs than their male counterparts is attributable to the ovarian activity, and the lower plasma cholesterol level in male than in female pigs fed a hypercholesterolemic diet is due to the testicular activity.
