Osmotic Stress-Inducible Expression of a Lipid Transfer Protein Gene in Poplar

  • We have cloned an LTP gene (PoLTP1) from poplar (Populus alba ${\times}$ P. tremula var. glandulosa) suspension cells and examined changes in its expression levels in response to various stresses and ABA treatment. The full-length PoLTP1 cDNA clone encodes a polypeptide of 116 amino acids with typical characteristics of LTPs, notably a conserved arrangement of cysteine residues. Southern blot analysis indicate that two or three copies of the PoLTP1 are present in the genome of the investigated hybrid poplar. In addition, northern analysis of samples from soil-grown plants indicate that PoLTP1 is tissue-specifically expressed in the leaves and flowers. The gene is significantly up-regulated by treatment with mannitol, NaCl and ABA, but not by either cold or wounding. These results indicate that PoLTP1 is involved in osmotic stress responses in poplar plants and suspension cells.

The effect on photosynthesis and osmotic regulation in Beta vulgaris L. var. Flavescens DC. by salt stress

  • This study was to investigate the effect of salt stress on physiological characteristics such as plant growth, photosynthesis, solutes related to osmoregulation of Beta vulgaris. A significant increase of dry weight was observed in 50 mM and 100 mM NaCl. The contents of Chl a, b and carotenoid were lower in NaCl treatments than the control. On 14 day after NaCl treatment, photosynthetic rate (PN), the transpiration rate (E) and stomatal conductance of CO2 (gs) were reduced by NaCl treatment. On 28 day after NaCl treatment, the significant reduction in gs and E was shown in NaCl 200 mM. However, PN and water use efficiency (WUE) in all NaCl treatments showed higher value than that of control. Total ion contents (TIC) and osmolality were higher than the control. On 14 day after treatment, the contents of proline (Pro) increased significantly in 200 mM and 300 mM NaCl concentration compared with control, whereas on 28 day in all treatments it was lower than that of the control. The contents of glycine betaine (GB) increased with the increase of NaCl concentration. The contents of Na+, Cl-, GB, osmolality and TIC increased with the increase of NaCl concentrations. These results suggested that under severe NaCl stress conditions, NaCl treatment did not induce photochemical inhibition on fluorescence in the leaves of B. vulgaris, but the reduction of chlorophyll contents was related in a decrease in leaf production. Furthermore, increased GB as well as Na+ and Cl- contents resulted in a increase of osmolality, which can help to overcome NaCl stress.

Relationship between Drought-Tolerance and Physiological Parameters in Korean Barley Genotypes (보리 품종의 한발저항성과 생리적 지표와의 상관)

  • Thirty-six barley varieties including Korean modern and local varieties were tested for drought-tolerance in the field of plastic rain shelter, Drought treatment was initiated at initial tillering stage (March 27, 2002) by withholding irrigation and lasted until harvest. Soil water potential maintained at around -0.05㎫ in the control plot and varied from -0.05㎫ (at the initial stage of drought treatment) to -0.29㎫ in the drought treatment plot. At forty days after drought treatment, relative water content (RWC), osmotic potential (OP), osmotic adjustment (OA), and $^{13}\textrm{C}$ discrimination ($\Delta$) were measured and then plants were sampled for leaf area index (LAI) and dry weight (DW). Barley was harvested at maturity for determining DW, grain yield, 1000 grains weight and number of spikelet. The tested varieties revealed wide spectrum of drought tolerance. Dongbori-1, Chalbori, Changyeongjaerae, Samdobori and Weolseong 87-31 showed strong drought-tolerance while Songhagbori and Suwonmaeg360 showed weak drought-tolerance. The drought injury indexes (drought/control ratio) of DW and yield revealed significant positive correlation with leaf RWC in drought treatment plot and $\Delta$ in the control plot, but obvious negative correlation with leaf OP and OA under drought condition. In addition, all the drought indexes of OP, $\Delta$ and RWC showed obvious positive correlation with the drought injury indexes of DW, 1000 grain weight and yield. Thus, OP and RWC under drought condition and $\Delta$ under well-watered condition would be used as the evaluation criteria for drought- tolerance of barley genotypes. However, further investigation is needed for the relationship between $\Delta$ and drought-tolerance as the other reports were not consistent with our result.

Salt treatment for recovery of the mud loach, Misgurnus mizolepis from transport stress

  • Due to the shortage of the fingerling/juvenile mud loach, Misgurnus mizolepis in Korea, these fish have been imported from China. However, the mortality rate during and after their transportation is very high. In this study, we examined various physiological and histological parameters to evaluate the effect of salt treatment on the survival and recovery of mud loaches in holding farms during the quarantine process. Glucose, osmolality, $Na^+$, $Cl^-$, and histological changes were assessed for three different salinities. Non-treated fish (control 0.0%) exhibited lower levels of osmolality, and $Na^+$ and $Cl^-$ concentrations compared with those kept in solar salt solution (0.5% and 1.0%). Glucose levels in control fish were higher than those in fish exposed to 0.5% and 1.0% solar salt solution. Histologically, control fish showed thinner epidermis of skin, branchial hyperplasia and lamellar fusion with an abundance of eosinophilic granule cell-like cells. After solar salt solution treatment, damaged gill structures in the fish almost recovered within 5 days. The present study demonstrates that mud loaches transported from China suffer from skin and gill damage and physiological dysfunction which may increase the mortality and morbidity. Moreover, saline treatment might alleviate the stress responses and ionic/osmotic imbalances, and help heal gill damage.

Effect of electrochemical treatment on consolidation of soft clay

  • In this study, a method of electrochemical consolidation is applied. This method utilizes electro-osmosis, which is an effective ground improvement technique for soft clays, and soil treatment using lime, which is the oldest traditional soil stabilizer. The mechanism of lime treatment for soil involves cation exchange, which leads to the flocculation and agglomeration. Five representative laboratory tests-an electro-osmotic test and four electrochemical tests with various proportions of lime-were performed on dredged marine clay. The objectives of this study are to investigate the effect of electrochemical treatment and to determine the optimum dose for optimal consolidation performance of dredged marine clay. The results show that a better consolidation effect was achieved in terms of current, temperature, and vane shear strength by using electrochemical treatment. The best results were observed for the electrochemical test using 4% lime content.

Effects of Various Pretreatments on Quality Attributes of Frozen and Thawed Peaches (전처리 방법에 따른 냉동 복숭아의 품질 특성)

  • The aim of this study was to investigate the effect of pretreatments on quality of frozen peach. Pretreatments including steam blanching, water blanching, high pressure, and osmotic dehydration were applied to two varieties (Daeokgye and Hwangdo). Pretreated peaches were frozen and thawed at $-20^{\circ}C$ and $5^{\circ}C$, respectively. Steam blanching and osmotic dehydration with ascorbic acid reduced ${\Delta}E$ values without change of pH and acidity. Osmotic dehydration with sucrose decreased drip loss and increased brix. Freezing/thawing resulted in an increase of maximum force, while maximum force decreased with increasing time of steam and water blanching. Furthermore, osmotic dehydration with calcium chloride increased maximum force. High pressure decreased maximum force of Daeokgye and increased that of Hwangdo compared with non-treatment. Total polyphenol content, DPPH radical scavenging activity, and ABTS radical scavenging activity were improved by pretreatment including steam blanching, blanching, and osmotic dehydration with ascorbic acid.

Optimization of Pre-treatment Process for Manufacturing Apple Jangachi (사과장아찌 제조를 위한 전처리 공정의 최적화)

  • In this study, optimized pre-treatment conditions were investigated to improve the quality of gochujang apple jangachi. The moisture content was decreased by 14% in 60% sugar solution and 17% in 30% salt solution during 12 hours. On the one hand, in the case of dry sugar and salt method, moisture content was decreased rapidly by 20% and 24%, respectively during 12 hours. Compared with the sugar and salt solution method, the dry sugar and salt method was more effective in reducing moisture content during 12 hours. In the case of osmotic dehydration of the apple parts in the dry sugar method, the moisture content of the flesh, flesh including peel, and peel of apple decreased by 23%, 20%, and 16%, respectively during 6 hours. However, in the dry salt method, the moisture content of the flesh, flesh including peel, and peel of apple decreased by 19%, 12% and 13%, respectively during 2 hours. During this time, the pH tended to decrease, regardless of the presence of sugar and salt. Total acidity was slightly increased in the case of salt. In hot air drying after osmotic dehydration, the moisture content of flesh including peel of apple decreased from 64% and 67% to 31% and 27%, respectively at 90 minutes, and from 64% and 67% to 11% and 18% at 150 minutes, respectively. The moisture content of the peel decreased from 51% to 10% at 120 minutes. As a result of the sensory evaluation, the overall acceptance of the flesh and flesh including peel was highly evaluated, resultantly, the products were considered to be suitable for the production of gochujang apple jangachi.

Effect of Osmotic Priming Treatment on Seed Viability and Seedling Growth in Watermelon (수박종자의 Osmotic Priming 처리가 종자활력 및 유묘생장에 미치는 영향)

  • In nine commercial watermelon cultivars, the effects of osmotic seed priming were evaluated for seed viability and seedling growth at different germination temperatures and field conditions. Generally, primed seeds showed improved germination rate and reduced time for emergence at different germination temperatures, as compared to untreated seeds. In particular, priming effect on seed germination was significantly high at $15^{\circ}C$, which is an environment normally resulting in poor germination. Depending on the watermelon cultivar, germination rate under low temperature ($15^{\circ}C$) was improved by up to 15~66% in primed seeds, as compared to untreated seeds. However, priming effect on germination was gradually reduced as the temperature reached to its optimum level for seed germination. Seed priming tended to improve the viability, fresh weight, and dry weight of watermelon seedlings, but its effect on seedling stage was not large, as compared to untreated seeds. Primed watermelon seeds showed improved emergence rate and facilitated germination in the field, but their seedling growth after 30 days from germination was not significantly affected. Our results indicated that seed priming can greatly improve the seed germination at poor temperature conditions in the watermelon.

Hypoosmotic shock adaptation by prolactin involves upregulation of arginine vasotocin and osmotic stress transcription factor 1 mRNA in the cinnamon clownfish Amphiprion melanopus

  • We cloned cDNA-encoding arginine vasotocin (AVT) from the brain of the cinnamon clownfish Amphiprion melanopus, and that was predicted to encode a protein of 153 amino acids. We examined changes in the expression of AVT mRNA in the brain and arginine vasotocin receptor (AVTR) mRNA and osmotic stress transcription factor 1 (OSTF1) mRNA in the gills of the cinnamon clownfish using quantitative real-time PCR in an osmotically changing environment (seawater (35 psu) ${\rightarrow}$ brackish water (BW, 17.5 psu) and BW with prolactin [PRL]). The expression of AVT, AVTR, and OSTF1 mRNA in the brain and gills increased after transfer to BW, and the expression was repressed by PRL treatment. AVT-immunoreactive cells were almost consistently observed in the telencephalon. The plasma $Na^+$ and $Cl^-$ levels decreased in BW, but the level of this parameter increased in BW with PRL treatme during salinity change. These results suggest that AVT, AVTR, and OSTF1 play important roles in hormonal regulation in osmoregulation organs, and that PRL improves the hyperosmoregulatory ability of cinnamon clownfish in BW environment.

A Case of Osmotic Demyelination Syndrome in a Patient with Severe Hyponatremia Complicated by Rhabdomyolysis (횡문근융해증이 합병된 중증 저나트륨혈증 환자에서 발생한 삼투성 탈수초 증후군 1예)

  • Hyponatremia, the most common electrolyte disorder, has been rarely reported as causing rhabdomyolysis. Osmotic demyelination syndrome (ODS), a demyelinating disease of the central pons and/or other areas of the brain, is infrequently reported as associated with rapid correction of hyponatremia. This paper reports a case of ODS after correction of severe hyponatremia complicated by rhabdomyolysis. A 47-year-old female with a history of chronic alcoholism presented herself at the hospital with altered consciousness after three days of nausea and vomiting. She was on a thiazide diuretic for essential hypertension. Her blood tests upon her hospital admission showed hyponatremia ($Na^+$ 98 mEq/L), hypokalemia ($K^+$ 3.0 mEq/L), and elevation of her serum creatine phosphokinase (3,370 IU/L) with an increase in her serum myoglobin level 11,267 ng/mL). She was treated with intravenous fluid therapy that included isotonic and hypertonic salines along with potassium chloride. She became more alert, and her neurological condition gradually improved after the first five days of her therapy. On the ninth day after her admission, she developed progressive quadiaresis associated with dysarthria, dysphagia, and dystonia despite the resolution of her hyponatremia. Magnetic resonance imaging of her brain on 16th day revealed symmetrical areas of signal hyperintensity in her central pons, basal ganglia, and precentral gyrus in T2-weighted images, which are consistent with ODS. Her neurological symptoms steadily improved after six weeks with only supportive treatment and rehabilitation.
