Mass Transfer Characteristics in the Osmotic Dehydration Process of Carrots (당근의 삼투건조시 물질이동 특성)

  • Diffusion coefficients of moisture and solid, reaction rate constants of carotene destruction, and the fitness of drying models for moisture transfer were determined to study the characteristics of mass transfer during osmotic dehydration. Moisture loss and solid gain were increased with increase of temperature and concentration; temperature had higher osmotic effect than concentration. Diffusion coefficient showed similar trend with osmotic effect. Diffusion coefficients of solids were larger than those of moisture because the movement of solid was faster than that of moisture at the high temperature. Reaction rate constants were affected to the greater extent by concentration changes than by temperature changes. Arrhenius equation was applied to determine the effect of temperature on diffusion coefficients and reaction rate constants. Moisture diffusion required high activation energy in $20^{\circ}Brix$, while relatively low in $60^{\circ}Brix$. To predict the diffusion coefficients and reaction rate constants, a model was established by using the optimum functions of temperature and concentration. The model had high $R^2$ value when applied to diffusion coefficients, but low when applied to reaction rate constants. Quadratic drying model was most fittable to express moisture transfer during drying. In conclusion, moisture content of carrots could be predictable during the osmotic dehydration process, and thereby mass transfer characteristics could be determined by predicted moisture content and diffusion coefficient.

  • 기존의 일반적인 에탄올 제조법으로는 1) ethylene의 가수분해, 2)전분이나 당분의 발효, 3)acetylene으로부터 얻은 acetaldehyde의 환원반응이 있다. 기존의 화석연로에 의존하지 않고 에탄올을 생성하는 방법은 발효에 의한 방법이다. 발효에 의해 생성되는 에탄올의 농도는 batch식 발효에서 13-14%, 고정화 발효방법에서 5-10%정도이다. 이를 고순도의 에탄올로 농축하기 위한 방법으로는 증류법, 투과증발법, 역삼투법등이 있으며 증류를 이용하여 농축하는 경우에는 상당히 큰 열에너지를 필요로 한다.

  • Pfu DNA polymerase from Pyrococcus furiosus was expressed in the E. coli periplasm, and the fully active polymerase was partially purified by applying osmotic shock, ammonium sulfate precipitation, and heat treatment. This method represents a new way of expressing and purifying functional Pfu DNA polymerase without the use of chromatography.

  • A theoretical study for the flux enhancement by pulsation of transmembrane pressure is presented for osmotic pressure controlled ultrafiltration under laminar flow regime. The transient velocity profile is solved analytically using Green's function method. Time dependent convective diffusive equation is solved to quantify the membrane surface concentration and the permeate flux, numerically. The effects of the amplitude and frequency of pulsation on flux, surface concentration and observed retention are studied.

Comparison of Germination Characteristics, and of Logistic and Weibull Functions to Predict Cumulative Germination of Grasses Under Osmotic Water Stress (수분장애시 목초 발아특성 및 누적 발아율 곡선 예측을 위한 Sigmoid 함수들 간의 비교)

  • The germination of seeds is developmentally complex process requiring water uptake, which is regulated by both genotypic and environmental factors. The present study was undertaken to determine the difference in germination characteristics, and to compare the ability of the logistic and Weibull functions to describe the cumulative germination curve when two levels of osmotic potential(0, -5 bar) were put to seeds of alfalfa, tall fescue, orchardgrass, and Kentucky bluegrass. The effects of grass type, osmotic potential, and their interaction on the total germination and coefficient of germination velocity were significant(P<0.01). The Weibull equation for predicting percent cumulative germination curve of alfalfa had significantly lower residuals than the logistic equation regardless of osmotic potential(P<0.01), indicating that the Weibull equation was more efficient than the logistic equation to fit the data of the percent cumulative germination of alfalfa. The rate parameter from the logistic equation was decreased under water stress, whereas the scale and shape parameters were increased. There were significant differences in days to 20% germination estimated from the logistic and Weibull equations.

  • This study was conducted to develop intermediate material for new processed food from sweet pumpkin. Osmotic dehydration was carried out as pretreatment before drying. After the sweet pumpkins were pretreated under optimized osmotic dehydration conditions, they were dried by three drying methods (hot air drying, vacuum drying and cold air drying). The moisture contents of dried sweet pumpkin products by osmosis treatment (sucrose) and hot air drying, vacuum drying, and cold air drying were 14.48, 14.09 and 13.87%, respectively. Cold air drying preserved more vitamin C content and showed lower color difference than hot air drying and vacuum drying. As a result of microscopic analysis, cold air dried sweet pumpkin was observed regular tissue, while hot air and vacuum dried sweet pumpkins were observed a cell collapse following the loss of water.

  • The seedling of barley and weed would be generally inhibited by rice straw residues in the field. This experiment was carried out to investigate the allelopathic effects of rice straw and environmental conditions of germination such as osmotic potential, pH on the seedling of barley and water foxtail. The pH did not affect the germination of barley and water foxtail. Osmotic potential to -2.9bar, -0.6bar did not affect the germination of barley and water foxtail, respectively. Rice straw mixed with soil at the concentration of 4%(w/w) reduced the dry weight of shoot and root, and volume of barley root, as compared to the untreated control. Dry weight of water foxtail by rice straw mulching was decreased by 87.1%, 86.2%, respectively.

  • In order to develope a protoplast fusion of the genus Cellulomonas having high assimilibility of cellulose, the optimum conditions for the protoplast formation of Cellulomonas flavigena NCIB 12901 was investigated and observed by means of Scanning Electron Microscope. The results suggested that the susceptibility of the cell wall by lysozyme treatment on protoplast formation was considerably depend on the cultural periods of the cells. Cells of C. flavigena at mid exponential phase could more efficiently convert to protoplast cells than those at late exponential phase did. The rate of the protoplast formation was 95%, even though the rate was over 99.9% on counting by indirect method after osmotic shock treatment, when cells of the organism at mid exponential phase were treated with lysozyme (400$\mu\textrm{g}$/$m{\ell}$) for 6 hours and observed by SEM. In the evaluation of protoplast formation of the genus Cellulomonas, direct method of the observation under Scanning Electron Microscope was much more reliable than the counting method of protplasts after osmotic shock treatment. Because defferences between the number of spheroplast and protoplast were not able to be figured out on counting the number of protoplast after osmotic shock treatment.

  • Fructans are polyfructose molecules that function as nonstructural storage carbohydrates in several plants. In addition, it has been suggested that, due to their solubility, they can play an important role in helping plants survive periods of osmotic stress. In order to study the effect of levan synthesis on plant growth, the coding region of the levansucrase gene, which was isolated from Zymomonas mobilis, was introduced into tobacco plants using Agrobacterium tumefaciens-mediated transformation. The presence of the levansucrase gene in transgenic plants was verified by genomic DNA gel blot analysis. RNA gel blot and immunoblot analyses showed an accumulation of the corresponding transcript and protein product of the bacterial levansucrase gene in transgenic plants. Furthermore, a thin layer chromatography analysis revealed that fructans were synthesized and deposited in transgenic tobacco plants. When $T_1$ seeds were germinated and grown under polyethylene glycol-mediated drought stress or cold stress, the transgenic seedlings displayed a substantially higher level of growth than that of untransformed plants. These results suggest that fructans may playa significant role in the tolerance of plants under osmotic stress.

On the Red Cell Fragility in the Normal and Anemic Professional Blood Donors (건강인(健康人) 및 직업적(職業的) 매혈(賣血)로 인(因)한 실혈성(失血性) 빈혈자(貧血者)의 적혈구(赤血球) 취약성(脆弱性)에 관(關)하여)

  • The osmotic and mechanical red cell fragility of the professional blood donors, who were found to be anemic as the result of frequent and repeated blood loss the past 5-6 years, were compared with that of the normal person while incubating the blood at $4-6^{\circ}C$ for 28 days. The fragility was expressed as % hemolysis occured during the incubation, and the following results were obtained: 1. The osmotic fragility in the normal persons (i.e, ; control group) progressively increased as the incubation became longer, and % hemolysis in 0.42% NaCl solution at 0, 10, 15, 21 and 28 incubation day was 31.90, 50.20, 41.68, 43.50 and 55.40 respectively. The mechanical fragility. in the normal red cells ranged between the minimum of 0.00% to the maximum of 5.80% both in 0.90 and 0.66% of NaCl solutions. 2. The hemolysis curve obtained in the red cell osmotic fragility from three cases of the anemic persons (i.e,; experimental group) showed a significant left side shift comparing with the normal in general which indicates that the fragility was more increased in the experimental group. The mechanical fragility in the experimental group ranged between the minimum 0.00% to the maximum 19.00% both in 0.90 and 0.66% of NaCl solutions. 3. The red cells of the chronic anemic person due to the frequent blood loss as the professional blood donor exhibit significantly marked increase both in osmotic and mechanical fragility comparing with the normal, and the tendency was more prominent as the incubation period became longer.
