Anti-Atherosclerosis Activity of Rosmarinic Acid in Human Aortic Smooth Muscle Cells (사람 동맥 평활근 세포에 대한 Rosmarinic Acid의 항동맥경화 활성)

  • Rosmarinic acid frequently found as a secondary metabolite in herbs and medicinal plants, has exhibited antimicrobial, antiviral, antioxidative, and anti-inflammatory activities. The proliferation and migration of human aortic smooth muscle cells (HASMC) in response to activation by various stimuli plays a critical role in the initiation and development of atherosclerosis. This study was conducted to examine the effects of Rosmarinic acid on the proliferation and migration of HASMC. Rosmarinic acid suppressed the proliferation of HASMC via induction of the expression of apoptotic proteins including cleaved poly ADP-ribose polymerase (PARP), and caspase-3. Rosmarinic acid decreased anti-apoptotic Bcl-2 and increased pro-apoptotic Bax. Moreover, treatment of rosmarinic acid decreased the G1/S cycle regulation proteins (cyclin D1, cyclin E, CDK2, CDK4 and CDK6) and increased p21, p27 and p53. Rosmarinic acid also blocked HASMC migration via suppression of MMP-9 and MMP-2. Taken together, these results indicate that rosmarinic acid has the potential for use as an anti-atherosclerosis agent.

A study of dietetic on the gastric & duodenal ulcer (위.십이지장궤양의 식요방법(食療方法)에 관한 연구)

  • Objectives : This research examined about dietary therapy on the gastric & duodenal ulcer based on the literary findings, for clinical treatment and prevention of the gastric & duodenal ulcer. Methods : This Research examined on histories, compositions, applications, and effects of dietetic on about 200 cases of dietary therapy of the gastric & duodenal ulcer from recent Chinese literatures Results : 1. Various vegetables, animals and mineral materials including herbs, grains, vegetables, fruits, food and drink were used for the dietary therapy. 2. Methods of the preparation for use as therapeutics were decoction, pulvis, gruel, medicinal wine, cake, tea, paste and gelatin and etc. 3. Frequently used materials were cuttle fish bone, Bletillae rhizoma, oyster shell, egg shell, sugar, aloe, licorice, lily, red jujube, and pig stomach. 4. Four properties of cold, warm, cool and hot were equal in frequencies. Most common tastes were sweet and bitter. Conclusion : Though dietary therapy for the gastric & duodenal ulcer is not based on clinical or experimental data, but through experience. It is mostly based on Yin-Yang and five elements, visceral manifestation, channels and their collateral channels and chinese herbal medicine theories. If we use them properly according to oriental medicine method, it will be effective on treating and preventing the gastric & duodenal ulcer.

Aqueous Extract of Ma huang Decreases Neuropeptide Y Expression in the Hypothalamus of Rats

  • Ma huang, the dried plant stem of Ephedra Intermedia Schrenk et CA, is one of the well known medicinal herbs, and has been used for the diaphoretic, antiasthmatic, and diuretic actions. Ma huang is an ephedrine type alkaloid used for the weight loss and energy expenditure. Medications based on the Ma huang have been found to be effective in the treatment of obesity. Neuropeptide Y (NPY), a 36-amino-acid peptide and concentrated in the hypothalamus, stimulates feeding desire and decrease energy expenditure. In the present study, the effect of Ma huang on the expression of NPY in the rat hypothalamus was investigated using immunohistochemistry. The present results demonstrated that Ma huang treatment suppressed weight gain and NPY expression in the hypothalamus depending upon the dosage used. Based on the results, it can be suggested that Ma huang treatment is effective in curbing the desire for food via modulation of NPY expression under the normal conditions.

Effects of the Geijibokryunghwan on Carrageenan-induced Inflammation and COX-2 in Hepatoma Cells

  • In oriental medicine, Geijibokryunghwan(GBH) was used to improvement various symptoms created by the thrombosis. We investigated the effects of an oriental medicinal prescriptions, Geijibokryunghwan (GBH) consisting of herbs of Cinnamomi Ramufus (Geiji; 桂枝), Poria cocos (Bokrung; 茯?), Moutan Cortex Radicis(Modanpi; 牧丹皮), Paeoniae Radix (Jakyak; 芍藥) and Persicae Semen (Doin; 桃仁) on tumor growth-inhibitory activity and cancer chempreventive activity in assays representing three maior stages of carcinogenesis. Cancer chempreventive agents include nonsteroidal anti-inflammatory drugs (NSAIDS) such as indomethacin, aspirin, piroxicam, and sulindac, all of which inhibit cyclooxygenase (COX). Effects of the GBH extracts on carrageenan-induced edema Inflammation using female (C57BL/6XC3H) Fl (B6C3Fl ) mice and tumorigenesis were examined. Finally, cyclooxygenase metabolites were determined after extracts treatment. These data suggest that GBH extracts merits investigation as a potential cancer chempreventive agent in humans.

Literature Review of Clinical Studies for Herbal Medicine Treatment on Juvenile Idiopathic Arthritis (JIA) (소아 특발성 관절염의 한약치료에 대한 문헌 고찰)

  • Objectives The purpose of this study is to investigate the clinical studies on the effect of herbal medicine in JIA and to seek better approach of herbal medicine to treat JIA. Methods This study researched randomized controlled trials through various databases in the world about herbal medicine treatments in JIA. Results 11 out of 414 studies were selected and analyzed. All studies were conducted in China. Most of the studies were using herbal medicines, as an adjunctive treatment to the main regimen, and the total effective rate was higher in the treatment group than in the control group. One study was comparing the herbal oriental medicine treatment to the western medicine treatment, and it showed that treatment group was more effective than control group. 5 out of 11 studies reported treatment cases from a single herbal medicine; most of them used Total glucosides of Paeonia (TGP). Clematidis Radix et Rhizoma (威靈仙) were also commonly used, as well as medicinal herbs belong to Eliminating wind-dampness medicine (祛風濕藥) and Activating blood resolving stasis medicine (活血祛瘀藥). Conclusions Based on this study, herbal medicines for JIA treatment will be effective even if used alone or in conjunction with other medicines. To apply clinically, further studies are needed.

A Comparative Anatomical Study of Saururus chinensis (Lour.) Baill. and Houttuynia cordata Thunb. (Saururaceae) (삼백초와 약모밀의 내부형태 비교 연구)

  • Objectives : Saururus chinensis and Houttuynia cordata (Saururaceae) are perennial herbs using for medicinal purposes in Korea. The objectives of this study are to compare anatomical key characters between two medicinal plants and to provide fundamental information for the identification of two herbal medicines by using anatomical features. Methods : Cross-sections of root, rhizome, stem, petiole, and leaf for each species were observed in this study. Materials were analyzed through dehydration, paraffin embedding and micro-sectioning, and double staining with Safranin O and Fast-Green FCF. Observations of permanent preparation were conducted using light microscope. Results : S. chinensis and H. cordata were distinguished with anatomical differentiations; Idioblasts with essential oil were scattered in the parenchyma cell of cortex, pith, and phloem of S. chinensis, on the other hand, in H. cordata, idioblasts were distributed ring-shaped in the cortex of the root. S. chinensis had two cycles of vascular bundles in the stem while H. cordata had one cycle. Hypodermis layer was conspicuous in a stem of H. cordata, crystals were observed the only parenchyma in a stem of S. chinensis, and epidermal oil cells were developed in the epidermis of H. cordata. S. chinensis had air cavity at the cortex and pith of the stem. The shape of cross-section was polygonal in the stem of S. chinensis and was circular in the stem of H. cordata. Conclusions : We investigated anatomical study of Korean S. chinensis and H. cordata. To identify two herbal medicines, we considered main anatomical features and provided identification key here.

Research on Standardization of Sasang Typology Formulae English Translation (사상(四象) 방제명(方劑名)의 영역(英譯) 표준화(標準化)에 대한 연구(硏究))

  • Objectives: This study proposes a new form of English translation in comparison with that shown in English books related to Sasang Constitutional medicine formulae which have been published up to now. Methods: This study formulated a new form of English translation guidelines by making a comparative analysis of the English translation cases listed in "Korean-English Dictionary of Oriental Medicine", "Longevity and Life Preservation in Eastern Medicine", and "Introduction to Sasang Constitutional Medicine". Results: This study as a common rules suggested that names of preparation form should be placed at the very end of the formula name and the first letters of the major words of the formula name should be capitalized. In addition, for the sake of accuracy in English translation of the formula name, this study divided the formula names into 8 types. The 8 sorts of types include: (a) Materia Medica + Preparation Form, (b) Materia Medica + Indication + Preparation Form, (c) Materia Medica Numbers + Preparation Form, (d) Indication + Preparation Form, (e) Concept + Preparation Form, (f) Concept + Materia Medica + Preparation Form, (g) Concept + Indication + Preparation Form, (h) Etcetera. In these types, medicinal herbs should be written in scientific name, and translation of indication can follow Noun+~ing participle form. In case of the terms having a Korean-medicine-based meaning, this study aimed for source-oriented translation. Conclusions: This study provides a general English translation method and examples of the formula names for Sasang constitutional medicine.

Assessment of genotoxicity of Ssanghwa-tang, an herbal formula, by using bacterial reverse mutation, chromosome aberration, and in vivo micronucleus tests

  • Objectives: Ssanghwa-tang (SHT) is a traditional herbal formula comprising nine medicinal herbs, and it is used for reducing fatigue in Korea. SHT exerts various effects such as anti-inflammatory, antioxidant, and anti-aging activities, and protection against acute hepatotoxicity. However, the genotoxicity of SHT has not yet been established. Methods: Ten components were identified in SHT water extract by using high-performance liquid chromatography analysis. We assessed the genotoxicity of SHT by using bacterial reverse mutation (Ames test), chromosome aberration, and in vivo micronucleus tests. Results: The contents of paeoniflorin, glycyrrhizin, and liquiritin apioside in SHT were 15.57, 6.94, and 3.48 mg/g extract, respectively. SHT did not increase the revertant colonies of Salmonella typhimurium and Escherichia coli strains in the presence or absence of metabolic activity. Although SHT did not induce structurally abnormal chromosomes in Chinese hamster lung (CHL) cells in the presence of metabolic activity, the number of structurally aberrated chromosomes increased dose-dependently in the absence of metabolic activity. In the in vivo micronucleus test, SHT did not affect the formation of micronuclei compared with the vehicle control. Conclusions: Genotoxicity of SHT was not observed in the Ames test and in vivo micronucleus test. However, based on the results of chromosome aberration test, it can be presumed that SHT has the potential to induce genotoxicity because it induced structurally abnormal chromosomes in the absence of metabolic activity.

Deodorizing Effect of Several Antibacterial Medicinal Herbs on Oral Malodor (항균작용을 가진 수종 한약재의 구취억제 효과)

  • Objective: We investigated the oral malodor inhibitory effect of Scutellariae Radix (SR), Phellodendri Cortex (PC), Moutan Cortex (MTC) and Magnoliae Cortex (MGC) for the development of a gargle solution. Methods: 1. Against P. gingivalis and Pr. intermedia, the minimal bactericidal concentration (MBC) and the change of viable cells that were exposed to 1% each herbal extract were observed. 2. Deodorizing activity of 2% herbal extract and Garglin $Mint^{(R)}$ against methyl mercaptan were evaluated by gas chromatography (GC). 3. We used the salivary sediment system (SSS) as the malodor model. 4. The clinical examination was repeated 3 times by 2 subjects by $Halimeter^{(R)}$. Baseline VSC of each subject was measured. Then, the control subject gargled with cysteine for 30 sec. After 4 min, subjects would gargle for 30 seconds with herbal extracts (2%) and Garglin $Mint^{(R)}$. Subsequently, the concentration of VSC was measured at 0, 4, 8, 12, 16, 20, 40 and 60 minutes. Results: 1. Against P. gingivalis, MBC of SR, PC and MTC was 0.1%, and MBC of MGC was 1%. Removal time of P. gingivalis was as follows; 5 hr in MGC, 24 hr in SR and PC, and 48 hr in MTC. Against Pr. intermedia, MBC of SR and PC was 0.5%, and MBC of MTC, MGC was 1%. Removal time of Pr. intermedia was as follows; 5 hr in MTC and 24 hr in SR, PC and MGC. 2. Deodorizing effect of herbal extracts against methyl mercaptan was as follows; MGC and MTC had 100%, SR had 82.22%, PC had 66.60%, Garglin $Mint^{(R)}$ had 40.54%. 3. In the experiment using SSS, PC and MTC had statistically significant malodor-inhibitory effects (p<.05). 4. In the clinical examination, PC and MGC had statistically significant inhibitory effects at every elapsed time compared to the control subject. MTC had that until 40 min. SR had that at 0, 4, 8, 20, and 60 min. Conclusions: SR, PC, MTC and MGC have an antibacterial effect and the chemical removable activity of the oral malodor caused by VSC. These four herbs could have potential as effective anti-malodor agents.

Effect of Aerated Compost Tea on the Growth Promotion of Lettuce, Soybean, and Sweet Corn in Organic Cultivation

  • This study investigated the chemical characteristics and microbial population during incubation of four kinds of aerated compost teas based on oriental medicinal herbs compost, vermicompost, rice straw compost, and mixtures of three composts (MOVR). It aimed to determine the effects of the aerated compost tea (ACT) based on MOVR on the growth promotion of red leaf lettuce, soybean and sweet corn. Findings showed that the pH level and EC of the compost tea slightly increased based on the incubation time except for rice straw compost tea. All compost teas except for oriental medicinal herbs and rice straw compost tea contained more ${NO^-}_3-N$ than ${NH^+}_4-N$. Plate counts of bacteria and fungi were significantly higher than the initial compost in ACT. Microbial communities of all ACT were predominantly bacteria. The dominant bacterial genera were analyzed as Bacillus (63.0%), Ochrobactrum (13.0%), Spingomonas (6.0%) and uncultured bacterium (4.0%) by 16S rDNA analysis. The effect of four concentrations, 0.1%, 0.2%, 0.4% and 0.8% MOVR on the growth of red leaf lettuce, soybean and sweet corn was also studied in the greenhouse. The red leaf lettuce with 0.4% MOVR had the most effective concentration on growth parameters in foliage part. However, 0.8% MOVR significantly promoted the growth of root and shoot of both soybean and sweet corn. The soybean treated with higher MOVR concentration was more effective in increasing the root nodule formation by 7.25 times than in the lower MOVR concentrations Results indicated that ACT could be used as liquid nutrient fertilizer with active microorganisms for culture of variable crops under organic farming condition.
