유기인계 농약 중독 환자에서 고용량 Pralidoxime이 치료 결과에 미치는 효과 (Clinical Outcome for High-dose Pralidoxime in Treating Organophosphate Intoxication)

  • Purpose: The optimal dose of oximes for use in the treatment of organophosphorus pesticide poisoning has not been conclusively established. In this retrospective study, we assessed the effectiveness of the use of high-dose pralidoxime infusion in treating organophosphorus pesticide poisoning. Methods: From January 1998 to December 2009, 71 patients visited the hospital Emergency Department (ED) as a result of organophosphate pesticide intoxication. All of these patients received an initial bolus of 2 g of pralidoxime as the first step of treatment. Patients who then received continuous infusion of pralidoxime at a dose of 500 mg/hr were entered into study group 1 (low dose), and those treated by continuous infusion of pralidoxime at a dose of 1000 mg/hr were entered into study group 2 (high-dose). Plasma cholinesterase activities for each patient were evaluated at ED arrival and re-evaluated 24 hours after pralidoxime infusion. The effectiveness of the two treatment modalities was gauged by comparing the required duration of mechanical ventilation, time spent in the intensive care unit (ICU) and total time spent in the hospital. Results: The mean duration of mechanical ventilation was $9.98{\pm}6.47$ days for group 1 and $4.39{\pm}6.44$ days for group 2. The respective mean duration of time spent in ICU and the total number of days in the hospital were $16.38{\pm}18.84$ days and $21.87{\pm}20.16$ days for group 1, and $7.83{\pm}9.99$ days and $11.71{\pm}13.53$ days for group 2. Highdose pralidoxime treatment was associated with shorter required durations for mechanical ventilation, ICU and hospital stay. In addition, plasma cholinesterase reactivation rates were higher for those patients receiving high-dose pralidoxime treatment. Conclusion: The results suggest that high-dose pralidoxime treatment has greater efficacy for patients suffering from organophosphorus pesticide poisoning.

초임계 이산화탄소에 의한 어류조직 중 유기인계 농약의 첨가회수율 (Extraction Efficiencies of Organophosphorus Pesticides Spiked in Fish Tissues by Supercritical Carbon Dioxide)

  • 유기용매에 의한 잔류농약 추출방법을 대체할 신속, 간편, 값싼 방법의 개발을 목표로, 어류조직에 5종의 유기인계 농약 표준용액을 첨가하여, 추출온도와 압력별, 이산화탄소 유속별, 보조용매 종류별, 어류 시료량별, 어류 종류별로 초임계 이산화탄소를 이용하여 첨가회수율을 측정하였다. 조피볼락에 유기인계 농약 표준용액을 첨가한 후 흡수제로 celite를 혼합하여 초임계 이산화탄소로 추출하였을 때 첨가회수율은 추출온도와 추출압력이 낮을수록 증가하였고, 이산화탄소 유속이 높을수록 감소하였다. 보조용매 사용에 따른 회수율의 증가효과는 크지 않았으며, 사용한 보조용매로 methanol, acetone, methylene chloride 중 methylene chloride 10%와 더불어 추출하였을 때 첨가회수율이 가장 높았다. 어류 시료량이 적을수록 첨가회수율이 증가하였다. 조피볼락, 방어, 옥돔, 정어리, 고등어에서의 첨가회수율은 각각 $66.7{\sim}86.3%$, $56.2{\sim}79.2%$, $57.6{\sim}77.8%$, $84.2{\sim}96.3%$, $74.6{\sim}83.6%$로 지방함량이 높은 시료는 지방함량이 낮은 시료보다 첨가회수율이 높았다.

Pendimethalin의 제초활성(除草活性)에 미치는 광(光), 유기인계화합물(有機燐系化合物) 및 식물생장조절제(植物生長調節劑)의 영향(影響) (Effect of Light, Organophosphorus Compounds and Plant Growth Regulators on Phytotoxicity of Pendimethalin)

  • Pendimethalin의 약해(藥害) 발현(發現)에 미치는 광(光), 유기인계(有機燐系) 화합물(化合物) 및 식물생장(植物生長) 조절제(調節劑)의 처리(處理) 영향(影響)을 조사(調査)하였다. 벼 발아후(發芽後) 생육(生育)에 나타난 pendimethalin의 약해(藥害)는 광조건하(光條件下)에서 크게 경감(輕減)되었는데, 이는 광(光)에 의한 pendimethalin의 분해(分解)에 의한 것이었다. 유기인계(有機燐系) 화합물(化合物) 처리(處理)에 의한 pendimethalin 약해경감효과(藥害輕減效果)는 사용약제(使用藥劑)의 종류(種類)에 따라 다르며, 실험약제(實驗藥劑) 중 edifenphos의 효과(效果)가 가장 좋았다. 그러나 이러한 약해경감(藥害輕減) 효과(效果)는 edifenphos를 pendimethalin과 동시(同時) 또는 후(後)에 처리(處理)할 때만 나타나며, pendimethalin 처리전(處理前)에 edifenphos가 처리(處理)되면 약해경감(藥害輕減) 효과(效果)는 없었다. 식물생장(植物生長) 조절제(調節劑) IAA, $GA_3$ 및 kinetin 등의 처리(處理)에 의한 pendimethalin 약해(藥害)의 경감효과(輕減效果)는 이들을 단독(單獨)으로 처리(處理)하면 얻을 수 없었지만, IAA 3ppmw+$GA_3$ 3ppmw 및 5ppmw와 kinetin 1ppmw+$GA_3$ 10ppmw의 조합처리(組合處理)로 약해경감(藥害輕減) 효과(效果)를 얻었다.

금 나노 입자 응집 원리를 이용한 유기인계와 카바메이트계 비색-신속 농약검출법 개발 (Development of a Colorimetric Rapid Detection Method for Organophosphorus and Carbamate Pesticides using Gold Nanoparticle Aggregation Principle)

  • 본 연구에서는 시판되고 있는 혈청 내 AChE가 acetylthiocholine과 반응하여 GNP에 aggregation 일으키는 원리를 이용하여 신선채소 농산물 중에 저농도 농약을 신속하고 간편하게 분석할 수 있는 비색-신속 농약 검출법을 개발하는 연구를 수행하였다. 먼저 비색-신속 농약 검출법의 최적화를 위해 GNP 입자의 크기에 따른 응집정도를 확인하여 15~20 nm 직경의 GNP를 선정하였고, 혈청의 희석배수와 acetylthiocholine의 농도를 확인하여 GNP 응집 차이가 가장 큰 혈청 1000배 희석과 acetylthiocholine 1 mM을 최적화 조건으로 선정하였다. 비색-신속 농약 검출법의 평가를 위해 최적화된 비색농약분석법을 이용하여 유기인계 농약은 dimethyl amine으로 카바메이트계 농약은 carbofuran으로 민감도를 분석한 결과 모두 7.5 ng/mL까지 검출이 가능한 것으로 확인되었으며 이는 기존의 비색-신속 농약 검출법과 비교했을 때 높은 민감도와 특이성을 나타내었다. 농약 이외에 화학물질인 곰팡이독소 등에 대한 반응성은 확인되지 않아 높은 특이성을 나타내었다. 또한 상추, 깻잎, 양상추에 대한 시료 전처리법을 확립하고 임의로 오염시킨 3종(상추, 깻잎, 양상추)의 농산물에 대해서 회수율을 확인한 결과유기인계와 카바메이트계 농약을 83.85~133.16% 정도의 회수율이 확인되었다. 이상의 결과 볼 때 본 연구에서 개발한 비색-신속 농약 검출법을 이용한다면 시판 농산물의 잔류농약을 신속하고 민감도 높게 검출할 수 있을 것으로 판단된다.

  • Organophosphorous compounds can be reduced by zinc metal in acidic solution after alkaline hydrolysis. Although phosphates and phosphonates did not evolve any gas, dithioates did evolve hydrogen sulfide and phosphine, thionates and thiolates did evolve only hydrogen sulfide. The evolved gases were qualitatively detected by means of lead acetate and silver nitrate or mercuric bromide papers and determined by spectrophotometrically. The reduction mechanism and analytical method of dithioates were proposed.

  • We studied about analyzing carbamate pesticides, organochlorine, organophosphorus pesticides simultaneously using by GC/MSD. For this, instead of HPLC which is generally used in analyzing carbamate pesticides, GC was used and all hydrogen of -NH group in carbamate pesticides were substituted for trifluoroacetyl group which is stable at heat And eight pesticide resdidues were not detected in nine samples circulated in the city.

단세포전기영동법(single Cell Gel Electrophoresis Assay)을 이용한 농약 살포자의 DNA손상 평가 (Evaluation of DNA damage in Pesticide Sprayers using Single Cell Gel Electrophoresis)

  • Single cell gel electrophoresis (SCGE) assay, also called comet assay, is a rapid and sensitive method to detect DNA damage in single cell level. To evaluate the DNA damage of lymphocytes of pesticides sprayers, SCGE assay was carried out for 50 pesticides sprayer and 58 control subjects. They were interviewed with structured questionnaire to get the information about the kinds and amount of pesticide. Insecticides and fungicides were predominant among pesticides. Major components of pesticides were organophosphorus, organosulfate, cartap, carbamates, and triazole. Sprayed pesticides were classified into two groups. Group I included organophosphorus, organoarsenic, organotin, tetrazine, triazole and gramoxone, which were known to cause DNA damages. Group II pesticide were carbamates, surfactants, organosulfates, etc., which were not found as DNA damaging agents in scientific documents. Olive tail moments of 100 lymphocytes were measured by KOMET 3.1 program for each person. The means of tail moments were compared between farmers exposed to pesticides and control subjects. Farmers showed higher tail moments than control subjects (2.07$\pm$1.40 vs 1.53$\pm$0.77, p<0.05). The means of tail moments also were compared among group I sprayers (n=36), group II sprayers (n=24) and, control subject, and the means or tail moments were 3.4s$\pm$3.2o, 2.66$\pm$2.20 and 1.53$\pm$0.77 respectively. The difference between means of group I sprayers and controls was statistically significant (p<0.05). In conclusion, this study showed higher DNA damage in farmers exposed to pesticides than control subjects, and comet assay could be useful as a biological monitoring method of genotoxic pesticides for farmers.

Application of Solid Phase Microextraction to the Analysis of Pesticides in Vegetables

  • Solid phase micro-extraction (SPME), a solvent-free, rapid and inexpensive method for the extraction of organic compounds from aqueous sample matrices, was evaluated for determination of the 120 pesticides in vegetables such as crown daisy, perilla leaf, leafy lettuce and to mato. The analysis conditions were chosen for the SPME method: 15 min of immersion of the PDMS fiber in 10 ml of the solution with stirring at 1,000 rpm. The recovery tests were carried out in triplicate. The range of recoveries was 0-142% for organochlorine pesticides and $4.9\sim200\%$ for organophosphorus pesticides. The recoveries were very low in the pesticide groups with low solubility in water. The recoveries became lower in proportion to the interference materials in vegetables. The recovery in tomato was relatively higher than that in perilla Ie af and crown daisy. The recovery values obtained by SPE and SPME were compared. In leaf y lettuce, recovery obtained by SPE method ranged from $58.1\%\;to\;136.1\%$ and recovery by SPME ranged from $9.6\%\;to\;176.3\%$ In organophosphorus pesticides. The recovery in SPME method was satisfactory with $136\%$ for ethoprophos, $119\%$ for methidathion and $113\%$ for diazinon. Meanwhile, recovery of EPN, phenthoate and 2,4-DDT revealed relatively low value of $38\%,\;41\%\;and\;3.4\%,$ respectively. However, most of pesticides applied to SPME method sho wed constant recovery and precision. From these results, it can be concluded that solid phase micro-extraction might be an appropriate method for the screening test of pesticides in vegetables.
