Molecular Cloning, Characterization and Expression Analysis of an ILF2 Homologue from Tetraodon nigroviridis

  • Interleukin-2 enhancer binding factor 2 (ILF2) was reported to regulate transcription of interleukin-2 (IL-2), a central cytokine in the regulation of T-cell responses. This property of ILF2 was well characterized in human and mammals, but little is known in bony fish. In this paper, an ILF2 homologue was cloned and well characterized from Tetraodon nigrovirid is for the further investigation of the function of ILF2 in bony fish. The full-length Tetraodon ILF2 cDNA was 1380 bp in size and contained an open reading frame (ORF) of 1164 bp that translates into a 387 amino-acid peptide with a molecular weight of 42.9 kDa, a 5' untranslated region (UTR) of 57 bp, and a 3' UTR of 159 bp containing a poly A tail. The deduced peptide of Tetraodon ILF2 shared an overall identity of 58%~93% with other known ILF2 sequences, and contained two N-glycosylation sites, two N-myristoylation sites, one RGD cell attachment sequence, six protein kinase C phosphorylation sites, one amino-terminal RGG-rich single-stranded RNA-binding domain, and a DZF zinc-finger nucleic acid binding domain, most of which were highly conserved through species compared. Constitutive expression of Tetraodon ILF2 was observed in all tissues examined, including gill, gut, head kidney, spleen, liver, brain and heart. The highest expression was detected in heart, followed by liver, head kidney and brain. Stimulation with LPS did not significantly alter the expression of Tetraodon ILF2. Gene organization analysis showed that the Tetraodon ILF2 gene have fifteen exons, one more than other known ILF2 genes in human and mouse. Genes up- and down-stream from the Tetraodon ILF2 were Rpa12, Peroxin-11b, Smad4, Snapap and Txnip homologue, which were different from that in human and mouse.

구조방정식을 이용한 종합병원 행정직원의 역할스트레스가 소진, 직무만족 및 조직몰입에 미치는 영향분석 (A Study using Structural Equations on how the Role Stress of Administrative Workers in General Hospitals Affects Burnout, Job Satisfaction, and Organizational Commitment Analysis)

  • 본 연구의 목적은 의료기관 행정직 종사자의 역할 스트레스가 소진과 직무만족, 조직몰입에 미치는 영향을 실증적으로 규명하고자 실시되었다. 본 연구는 200병상 이상의 종합병원 중 10곳의 행정직원을 대상으로 2015년 7월 13일부터 8월 14일 까지 한 달 동안 설문조사를 실시하였다. 조사된 설문은 AMOS 18.0을 이용하여 확인적 요인분석, 평균분산추출, 연구모형의 적합도 지수, 연구모형의 경로계수 추정치 분석 등을 실시하였다. 본 연구의 주요결과로는 의료기관 행정직 종사자의 역할갈등과 역할과다는 소진에 정(+)적인 영향을, 소진은 직무만족에 부(-)적인 영향을 미치는 것으로 나타났고, 조직몰입과의 관계는 유의하지 않은 것으로 나타났다. 본 연구결과는 부서간의 연계 업무 등에 있어 역할갈등을 줄이기 위해서는 업무에 대한 명확한 지침과 업무지시의 통일성 등이 필요하고, 정확한 업무 측정과 계량으로 구성원의 역할을 적정화할 때 조직에 대한 애착과 헌신도도 높아지고 궁극적으로 개인의 직무성과도 향상될 수 있음을 시사하고 있다.

민간병원과 공공병원 직원들의 노동조합몰입수준 비교분석 (Comparison of Commitment in Labor Union Employees' in the Private Hospitals and Public Hospitals)

  • The purpose of this study was to provide basic materials needed to enhance quality of organizational life by identifying the improvements of labor union management in the perspective of hospital organization management. Materials were collected from administrators, nurses and medical technicians in target hospitals from March 3 to March 3 to March 30, 2010 through survey questionnaires. The main results of this study were as follows: 1. The commitment level of the subjects according to their satisfaction was higher when the administrators both in Private hospitals and Public hospitals were not satisfied in their works. 2. The commitment level of the subjects according to the subjects' job dedication level was higher those who had higher emotional attachment to their job in all vocations in both Private hospitals and Public hospitals. 3. The commitment level of the subjects according to the subjects' labor union involvement was higher in those who had higher satisfaction in labor union and perceived their colleagues' attitudes more positively in all vocations in both Private Hospitals and Public Hospitals. 4. From the results of multiple regression analysis to identify major influencing factors of labor union commitment level, In Private hospital employees, those who had senior officer posts in labor union and higher normative commitment showed higher commitment union. Therefore hospital managers should have democratic and flexible attitudes toward labor union. Additionally, as colleagues attitudes is important determinant in union commitment, hospital managers should have countermeasures to enhance the work atmosphere of hospital employees.

Streptozotocin유도 당뇨백서에서 치주조직 손상의 치유에 관한 실험적 연구 (AN EXPERIMENTAL STUDY OF PERIODONTAL INJURY HEALING IN STREPTOZOTOCIN INDUCED DIABETIC RAT)

  • To observe the healing process of the gingival wound in diabetic condition, the author induced diabetes mellitus by intravascular injection of streptozotocin in rat tail vein and made fresh wound in rat mandibular posterior gingiva using scalpel. The healing processes of gingival wound were examined periodically by light microscopy. The results were as follows. 1) The healing was completed at second week in control group, but it delayed until fourth week in experimental group and the organization was persisted for first two weeks. 2) The inflamed gingiva of dibetic rat demonstrated scanty polymorphonuclear lerkocytic infiltration at the early stage of experiment, but it soon became numerous as in cotrol group and the lymphocytic infiltrations were same degree as in control group. 3) The tissue destruction was broader in the experimental group than in the control group. The epithelization was began at the early stage of healing and the epithelial attachment was reformed with the completion of the wound healing. 4) Loss of Sharpey's fibers and destruction of cementum and alveolar bone were observed with the inflammatroy reaction, but these were reformed with the completion of the wound healing.

병원 조직의 리더십유형과 조직몰입, 직무성과의 구조적인 관계 (Structural Relationship between Leadership Type of Hospital Organization and Organizational Commitment and Job Performance)

  • 본 연구는 리더십유형과 조직몰입 및 직무성과의 인과관계를 연구하여 병원조직의 조직성과 향상 및 효과적인 인적자원관리 방법을 제시하는데 그 목적이 있다. 조사기간은 2016년 10월 6일부터 10월 20까지 이며, 병원에 근무하고 있는 직원 375명을 최종 조사대상으로 선정하였다. 분석방법은 측정변수의 타당도를 분석하고자 신뢰도 및 확인적 요인분석을 실시하였으며, 리더십과 조직몰입 및 직무성과의 인과관계를 파악하고자 공분산 구조방정식 분석을 실시하여 결론을 도출하였다. 결과를 보면 리더십, 조직몰입 및 직무성과는 직종(p<0.05), 연령(p<0.05), 근무기간(p<0.05)별로 유의한 차이가 나타났으며, 개별적 배려(p<0.001), 조건적 보상(p<0.001)이 조직몰입과 직무성과에 높은 인과관계를 나타냈다. 앞으로 조직 구성원의 개별적인 특성을 고려한 배려와 능력을 발휘할 수 있는 리더십과 업무달성도를 측정한 후 합리적인 보상체계의 구축이 필요하다. 또한 직종별 뿐만 아니라 성별, 연령, 근무기간 등을 감안하여 조직의 성과를 촉진시킬 수 있는 상황에 맞는 리더십이 필요하다. 그리고 관리자는 구성원들이 조직에 대한 충성심과 애착을 가질 수 있도록 조직문화 형성과 관련교육프로그램 제공 등의 노력이 요구된다.

조직유착모형의 경험적 적합성에 관한 고찰 - 교사들의 경우를 중심으로 한 한 . 미간 비교연구 - (An Empirical Testing of Employee Attchment Model: A Comprison of South Korean and U.S. Teachers)

  • 본 논문은 피고용자들의 조직에 대한 헌신도와 근속의도에 초점을 두는 피고용자 조직유착(employee organizational attachment)의 인과모형을 비교, 검토한다. 이 논문은 교사들의 조직유착 행위에 대한 두 개의 연구, 즉 미국 시카고 지역 공립학교 교사들에 대한 연구와 한국의 서울시 중등학교 교사들에 대한 연구를 바탕으로 하고 있다. 이 논문의 이론적틀은 Price-Mueller의 조직유착모형이다. 이 모형은 크게 다섯 개 군의 변수들을 포함하고 있다: 1) 일(노동)에 대한 반응변수 - 업무만족도, 헌신도, 근속의도, 2) 심리학적 긴장변수 - 역할애매성, 역할갈등, 업무과부하, 학생들의 자질, 3) 사회적 구조변수 - 자율성, 단순성, 분배정의 승진의 정당성, 4) 경제적 구조변수 - 보수, 직업안정성, 승진기회, 외부 노동시장 기회, 그리고 5) 개인적 성향변수 - 직업헌신도, 노동가치, 기대만족, 긍정적 인성. 자료는 질문지 조사방법으로 수집되었으며, 서울시내 중등교사들을 대상으로 한 표본 조사 결과 649개의 유효사례가 최종분석에 사용되었다. 조직유착의 인과적 모형을 검증하기위해 LISREL을 이용한 공분산구조분석이 사용되었다. 결과에 따르면 근속의도에 대한 외생변수들의 영향을 매개하는 데, 내생변수인 업무만족도와 조직헌신도의 역할이 미국에 비해 크게 작을 뿐만 아니라, 이 모형은 근속의도 변수의 변량을 크게 설명하지도 못한다. 한국과 미국의 이러한 차이는 한국의 교사들이 가지고 있는 고용관계와 노동시장의 구조적 특성들, 즉 강제적 이동과 외부노동시장의 폐쇄성 때문에 나타나는 것으로 보인다. 미국에서 발달한 이러한 조직유착모형은 적용범위(scope condition)를 명시함으로써 예외적인 경험적 자료에 따른 잘못된 무효화를 방지할 수 있을 것이다. 그리고 앞으로 연구는 한국 교사들의 경우와 같은 구조적 제약에서 피고용자들의 조직유착을 가져오는 요인들에 대한 개념적 및 경험적 탐구에 더 많은 관심을 두어야 한다.

  • The extracellular matrix(ECM) is a complex network of different combination of collagens, glycosaminoglycans, laminin, fibronectin, and many other glycoproteins including proteolytic enzymes. The composition and organization of the ECM contributes to the uniques physical or biomechanical properties of a tissue. Fibronectins(FN) are dimeric glycoproteins located on cell surfaces, in the matrix of connective tissue, and in blood. Fibronectins mediate cell attachment to collagen substratum and have been implicated in a variety of important biological processes, including embryogenesis and cell differentiation. The purpose of this study was to determine the effects of surgical induction of anterior disk displacement(ADD) on distribution of fibronectin in the rabbit temporomandibular joint(TMJ) tissues included the articular cartilage, disc, retrodiscal tissue, articular eminence using an immunohistochemical technique. The left TMJ was exposed surgically, and all discal attachments were severed except for the posterior attachment. The disk was then repositioned anteriorly and sutured to the zygomatic arch. The right TMJ served as a shamoperated control. Normal joints were used as a nonoperated control. Fourty-five rabbits were used for experiments in total. For fibronectin immunohistochemical study, eighteen rabbits (one normal group and 5 experimental groups, each group consists of 3 rabbits) were used. The experimental rabbits were sacrified after operation period of 2, 3, 4, 6 and 8 weeks on fibronectin. The obtained results were as follows ; 1. Fibronectin immunoreaction on all TMJ tissues(mandibular condyle, articular disc, retrodiscal tissue, articular eminence) in the normal rabbit was observed. Especially the reverse cell layer and proliferation zone of articular cartilage of condyle show strong positive reaction. 2. Depletion of fibronectin in the all TMJ tissues except hypertrophic zone of articular cartilage occurred at 2 weeks following induction of ADD. 3. The restoration of immunoreaction at 4 weeks was observed and a progressive increasing reaction at 6 weeks, 8 weeks also was found. Our study generally showed degenerative changes in TMJ tissues after ADD although TMJ tissues adapted or degenerated to abnormal loads and stress distribution according to the remodeling capacity of TMJ tissues.

중소건설기업의 직무배태성이 이직의도에 미치는 영향 : 조직몰입을 매개로 (The Effect of Job Embeddedness in Small and Medium Sized Construction Companies on Turnover Intention through Organizational Commitment)

  • 본 연구는 서울, 경기, 인천지역 소재 중소건설기업의 종사자를 대상으로 하여 중소건설기업 구성원의 직무배태성이 조직몰입을 매개로 이직의도에 미치는 영향을 규명하는데 목적이 있다. 표집방법은 유의 표집법을 활용하였으며 최종분석에 이용된 표본 수는 316명으로 입력된 자료 처리는 SPSS 20.0 프로그램을 이용하여 통계분석을 실시하였다. 분석결과, 첫째, 중소건설기업 구성원의 직무배태성의 하위요인 적합성, 희생과 이직의도 간의 상관관계에서 유의한 부(-)의 영향을 미치는 것으로 나타났으며, 연계는 유의하지 않은 것으로 나타났다. 둘째, 중소건설기업 구성원의 직무 배태성의 하위요인 적합성, 희생, 연계 순서로 조직몰입에 유의한 정(+)의 영향을 미치는 것으로 나타났다. 셋째, 조직몰입은 이직의도에 유의한 부(-)의 영향을 미치는 것으로 나타났다. 넷째, 조직몰입은 직무배태성과 이직의도 간의 관계를 매개하는 것으로 나타났다. 따라서 중소건설기업 구성원의 직무배태성이 높을수록 이직의도에 영향을 미치는 요인임을 알 수 있었다. 이는 조직 구성원 개인이 추구하는 가치의 적합성, 업무열정, 조직에 대한 애착 등을 향상시킴으로써 이직의도에 긍정적인 영향을 미칠 수 있음을 시사한다.

Erythropoietin-producing Human Hepatocellular Carcinoma Receptor B1 Polymorphisms are Associated with HBV-infected Chronic Liver Disease and Hepatocellular Carcinoma in a Korean Population

  • Erythropoietin-producing human hepatocellular carcinoma receptor B1 (EPHB1) is a member of the Eph family of receptor tyrosine kinases that mediate vascular system development. Eph receptor overexpression has been observed in various cancers and is related to the malignant transformation, metastasis, and differentiation of cancers, including hepatocellular carcinoma (HCC). Eph receptors regulate cell migration and attachment to the extracellular matrix by modulating integrin activity. EphrinB1, the ligand of EPHB1, has been shown to regulate HCC carcinogenesis. Here, we sought to determine whether EPHB1 polymorphisms are associated with hepatitis B virus (HBV)-infected liver diseases, including chronic liver disease (CLD) and HCC. We genotyped 26 EPHB1 single nucleotide polymorphisms (SNPs) in 399 Korean CLD, HCC, and LD (CLD+HCC) cases and seroconverted controls (HBV clearance, CLE) using the GoldenGate assay. Two SNPs (rs6793828 and rs11717042) and 1 haplotype that were composed of these SNPs were associated with an increased risk for CLD, HCC, and LD (CLD+HCC) compared with CLE. Haplotypes that could be associated with HBV-infected liver diseases by affecting downstream signaling were located in the Eph tyrosine kinase domain of EPHB1. Therefore, we suggest that EPHB1 SNPs, haplotypes, and diplotypes may be genetic markers for the progression of HBV-associated acute hepatitis to CLD and HCC.

전기방사로 제조된 다층 Poly Methyl Methacrylate (PMMA)/Poly Vinyl Alcohol (PVA) Bone Plate의 생체적합성 평가 (Biocompatibility of Multilayer Poly Methyl Methacrylate (PMMA)/Poly Vinyl Alcohol (PVA) Bone Plate by Electrospinning Method)

  • Multilayer Poly methyl methacrylate (PMMA)/ Poly vinyl alcohol (PVA) bone plates were fabricated using electrospinning and in vitro investigations were carried out for pre-clinical biocompatibility studies. The initial cellular cytotoxicity of the methacrylate (PMMA)/ Poly vinyl alcohol (PVA) bone plates was measured by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay using fibroblast-like L-929 cells. Cellular adhesion and differentiation studies were carried out using osteoblast-like MG-63 cells. As simulated body fluid (SBF) contains the same ionic concentration of body fluid and any bioactive material tends to deposit bone-like apatite on the samples surfaces into the SBF, in vitro bioactivity of the multilayer bone plates were investigated using SBF. We also studied the internal organization and tensile strength of the multilayer PMMA/PVA bone plates using micro-computed topography (${\mu}$-CT) and universal testing instrument (UTI, Korea) respectively. The cellular cytotoxicity study with MTT confirmed that the cellular viability was 78 to 90% which indicates good cyto-compatibility. Scanning electron microscopic findings revealed a good attachment and adhesion phenomenon of MG-63 cells onto the surfaces of the samples. Cellular differentiation studies also showed that osteogenic differentiation was switched on in a timely manner and affirmed along with that of the control group. Bone-like apatite formation on the surfaces was confirmed within 14 days of SBF incubation. Initial organizations of the multilayer PMMA/PVA bone plates were characterized as dense and uniform. The tensile strength of the post-pressing electronspun mat was higher than that of the pre-electronspun mat. These results suggest that a multilayer PMMA/PVA bone plate system is biocompatible, bioactive and a very good alternative bone plate system.
