Microbiological Evaluation of Antibiotic Resistance and Pathgoenicity in Autothermal Thermophilic Aerobic Digestion Treated Swine Manure

  • In both untreated and conventionally stabilized swine manures antibiotic resistant (AR) microorganisms, Staphylococcus-like and Salmonella-like microorganisms were detected. Also pathogens with MAR phynotype were detected. Presence of such microorganisms suggest high level of pathogen-related health risk to farmers who may be in direct contact with the manure and its conventionally stabilized product In contrast the autothermal thermophilic aerobic digestion (ATAD) treatment have efficiently reduced AR and pathogenicity from the swine manure. When soil was fertilized using swine manure and its stabilized products, despite no detection of MAR-exhibiting pathogen-like microorganisms in fertilized soil, potential pathogen-related health risk could not be ruled out from the fertilized soil since the organic fertilization led to increase in AR and pathogenicity in the soil microbial communities. As conclusion, this microbiological study demonstrated that an ATAD process is applicable in control of pathogen-related health risk in livestock manure.

Effects of Functional Ingredients Supplementation as a Bulking Agent in Composting of Swine Manure (돈분의 퇴비화에 있어 악취 제거 기능성 물질의 첨가 효과)

  • This study was conducted to evaluate the effects of functional ingredients with supplementation as a bulking agent in composting of swine manure. Treatments were T1 & T5 ; Swine mature+Vermiculite, T2 & T6 ; Swine manure+Perlite, T3 & T7 ; Swine manure+Vermiculite+Perlite, T4 & T8 ; Swine manure+Bark. T1, T2, T3 and T4 were supplemented with functional ingredients on d 0 of composting, T5, T6, T7 and T8 were supplemented with functional ingredients on d 3 of composting. Functional ingredients were Fermkito and Yucca. During the composting period, changes of temperature were showed traditionally composting trend. Volatile fatty acids were decreased regardless of treatments in functional ingredients supplementation. Organic matter, T-N and C/N ratio were showed higher bark treatments than other treatments. pH and EC were net differences among the treatments. P$_2$O$_{5}$ and $K_2$O were showed level of 1-3%. In heavy metal, Cr were showed higher in vermiculite treatment than other treatments. In conclusion, vermiculite and perlite treatments were greater composting effects than bark.

The Nutrients and Microbial Properties of Animal Manure and Spent Mushroom Compost Tea and the Effect of Growth of Lettuce (Lactuca sativa L.) (가축분뇨와 폐버섯 퇴비차의 양분 및 미생물적 특성과 상추의 생육에 미치는 영향)

  • In this study, experiments were conducted to determine the effect of different compost teas on plant growth reponses and yield of leaf lettuce. Compost tea is a liquid extract of compost obtained by mixing compost and water for a defined period of time. The pig manure and spent mushroom compost were made by steeping compost in water. Compost tea was aerated from 24 hours and molasses and kelp were added as supplements. The four types of compost were tested growth of lettuce. EC of animal manure compost tea was higher than that of spent mushroom compost tea. Mineral nutrients were significantly higher in animal manure compost tea compared with spent mushroom compost tea. Compost tea contains nutrient and a ranges of different organisms. The beneficial fungi and actinomycetes were prominent in a spent mushroom compost tea. Compost tea from animal manure had the higher numbers of total bacteria. The actinomycetes densities were high in spent mushroom compost tea. But actinomycetes were not founded in animal manure compost tea. The growth characteristics of lettuce in animal manure compost tea were higher than those of spent mushroom compost tea. And also SPAD value in leaf was high in plot treated with animal manure compost tea. The fresh yield of lettuce in animal compost tea was higher by 181% that of control plot. The effect of compost tea on growth of lettuce was largely attributable to mineral nutrient.

Changes of Soil Properties and Temperature by Green Manure under Rice-based Cropping System

  • The cultivation of green manure crop is considered as a good management practice by increasing soil organic matter and fertility levels. This experiment was conducted to improve the soil environment under rice-based cropping system at paddy soil (fine loamy, mixed, nonacid, mesic, family of Aeric Fluventic Haplaquepts) in National Institute of Crop Science (NICS), Korea in 2006 to 2007. The variation of soil temperature in green manure plots was lower than without green manure (control) during spring season (April to May). The temperature variation of no tillage plot (broadcast before rice harvest) was the lowest among treatments. After green manure cropping, the soil bulk density and porosity ratio were improved at the top soil. The production of green manure was the highest athairy vetch and barley mixture plot by partial tillage. However, mixture treatment had no improvement on soil organic matter. After rice cropping with green manure application, soil quality was improved such as soil physical properties except mixture treatment. Therefore, we suggest that soil quality should be improved by green manure cultivation under rice-based cropping system.

The Manufacturing Characteristics of Organic Liquid Fertilizer with Poultry Manure, Soybean Meal, and Rice Bran (계분, 대두박, 쌀겨를 이용한 유기 액비의 제조 특성)

  • This study was carried out to evaluate the manufacturing characteristics of organic liquid fertilizer with poultry manure, soybean meal, and rice bran at plastic house in Chungbuk Agricultural Research and Extension Service. Treatment was given 3 treatments; poultry manure+soybean meal (PM+SM), poultry manure+rice bran (PM+RB), and soybean meal+rice bran (SM+RB). The obtained results from this study were summarized as follows; The pH in liquid fertilizer was consistently increased in PM+SM treatment, and was increased after decreased at early season in PM+RB and SM+RB treatments. The electriacl conductivity(EC) in liquid fertilizer was rapidly increased from $2^{nd}$ weeks to $4^{th}$ weeks after fermentation in PM+SM and PM+RB treatments, and was rapidly increased from $4^{th}$ weeks to 6th weeks after fermentation in SM+RB treatment. The amount of $H_2S$ gas occurrence was the highest as $1,200\;mg{\cdot}kg^{-1}$ in early season, and was the lowest as $50\;mg{\cdot}kg^{-1}$ at $12^{th}$ weeks after fermentation of organic liquid fertilizer. The temperature of organic liquid fertilizer was stabilizing in $4^{th}$ weeks after fermentation. The yield of well of nitrogen, phosphate, and potassium was increased with increasing fermentation periods. It was not change from $4^{th}$ weeks after fermentation in content of calcium, magnesium and sodium in organic liquid fertilizer.

The Effect of Continuous Use of Cow Manure on Productivity of Organic Rice and Feed Value, Stock Carrying Capacity of Organic Rice Straw in Central Region of Korea (한국 중부지역에서 우분의 연용이 유기 벼의 생산성 및 유기 볏짚의 사료가치, 가축사육능력에 미치는 영향)

  • This experiments was conducted to investigate the influence on organic rice cultivation by continuous use of cow manure in the central region of Korea and investigated productivity of organic rice and feed value, stock carrying capacity of organic rice straw. In the results, application of chemical fertilizer showed higher one of 1st and 2nd year in fresh matter yield (15.77, 30.30ton/ha), dry matter yield (9.90, 13.89 ton/ha), dry matter yield of rice straw (4.88, 7.15ton/ha), dry matter weight of kernel per plant (29.81, 39.99g) and number of kernel per plant (1212.58, 1701units), but there were not significantly differences with each experimental plots in 3rd year. And harvest index showed higher one in application of chemical fertilizer (0.52, 0.48, 0.43) until 3 years. But their quantitative difference between application of chemical fertilizer and cow manure was gradually decreased each year. And the average value of $K_{CP}$ and $K_{TDN}$ showed higher one in application of chemical fertilizer (2.01, 2.94, 1.95 head/ha/yr) until 3 years. However, quantitative difference between chemical fertilizer and cow manure was gradually decreased each year. Especially, application of cow manure showed higher value of 3rd year in $K_{ME}$, but there was not significantly difference with each experimental plots. According to the results, continuous use of cow manure will be reduced their quantitative differences between chemical fertilizer as the year progress in the central region of Korea in terms of productivity of organic rice and feed value, stock carrying capacity of organic rice straw.

Biological Characteristics of Organic Soil applying Rye (Secale cereal L.) as Green Manure for the Long Term (장기간 호밀을 풋거름작물로 시용한 유기농 토양의 생물학적 특징)

  • In this study, microorganism community characteristics of organic managed soil which applied rye (Secale cereal L.) as green manure for 25 years, were determined. The chemical properties of organic soil showed high level of organic matter and available $P_2O_5$, while the level of exchangeable cation was low. The analysis of dehydrogenase activity and carbon source utilization indicated that the values in on organic soil were significantly higher than those of the control. It suggested that the microorganism community of organic soil had high microorganism activity, compared to the control. In addition, when the 16S rRNA gene-targeted NGS (Next generation sequencing) analysis was conducted to estimate the class of bacterial community, the class level of bacterial taxon composition on organic soil showed higher portion of Sphingobacteriia, Acidobacteriia, Gammaproteobacteria, Solibacteres and Planctomycetia. By base on the results of various reports in which organic managed soil had high portion of Acidobacteriia and Planctomycetia, the characteristic of taxon composition in organic soil, which showed the high percentages of Ktedonobacteria, Sphingobacteriia, Acidobacteriia and Gammaproteobacteria, was resulted from the application of rye as a green manure for the long term. However, further researches were needed because the crop effect was not considered in this study.

  • Organic and chemical fertilizer amendments are an important agricultural practice for increasing crop yields. In order to maintain the soil sustainability, it is important to monitor the effects of fertilizer applications on the shift of soil microorganism, which control the cycling of many nutrients in the soils. Here, culture-dependent and culture-independent approaches were used to analyze the soil microorganism and community structure under six fertilization treatments, including green manure, rice straw compost, rapeseed cake, pig mature compost, NPK +pig mature compost, NPK and control. Both organic and chemical fertilizers caused a shift of the cultural microorganism CFUs after treatments. Bacterial CFUs of the organic fertilization treatments were significantly higher than that of chemical fertilization treatments. The DGGE profiles of the bacterial communities of the samples showed that the green manure treatment was a distinct difference in bacterial community, with a greater complexity of the band pattern than other treatments. Cluster analyses based on the DGGE profile showed that rice straw compost and pig mature compost had a similar banding pattern and clustered together firstly. Rapeseed cake, NPK, NPK +pig manure compost and control clustered together in other sub-cluster and clearly distinguished from green manure.

The Effects of Organic Manure and Chemical Fertilizer Application Levels on the Growth and Nutrient Concentrations of Yellow Poplar (Liriodendron tulipifera Lin.) Seedlings (유기질 및 화학비료 처리수준이 어린 백합나무 생장 및 양분농도에 미치는 영향)

  • Soil nutrient management is important to maintain the constant productivity of seedling production in the nursery for successful forest restoration. This study investigated the effects of organic manure and chemical fertilizer application levels on the growth, soil properties, and nutrient concentrations of yellow poplar seedlings. One-year-old yellow poplar seedlings were treated with the combination of 3 level organic manures(0, 5 Mg/ha, 10 Mg/ha; mixture of poultry manure, cattle manure, swine manure, and sawdust) and 3 level nitrogen-phosphorus-potassium(NPK) chemical fertilizers(0, 1x(urea, $30g/m^2$; fused superphosphate, $70g/m^2$; potassium chloride, $15g/m^2$), 2x). Organic manure significantly increased the soil pH and the concentrations of nitrogen, available phosphorous, exchangeable potassium, calcium, and magnesium. In contrast, the NPK chemical fertilizer decreased the soil pH and exchangeable calcium concentration, did not affect the soil concentrations of nitrogen and magnesium, and increased the concentrations of available phosphorous and exchangeable potassium. Both organic manure and NPK chemical fertilizer treatments increased the seedling height, root collar diameter, and dry weight by 39% and 25%, respectively. The treatment with manure 5 Mg/ha and NPK 2x chemical fertilizer mostly increased seedling dry weight by 2.6 times more than that of the control. Compared to the effects of the fertilization treatments on the soil properties, the effects on nutrient concentrations in the leaves were relatively small. These findings indicate that organic manure that was derived from livestock byproducts and sawdust can be utilized with chemical fertilizer to improve seedling production as well as conserving soil quality.

Prediction of Chemical Organic Composition of Manure by Near Infrared Reflectance Spectroscopy

  • The organic materials included in excreta of livestock are important resources for organic manure and for improving soil quality, although there is still far from effective using. One reason for this is still unclearly standard of quality for evaluation of manure made from excreta of livestock. Therefore, the objective of this study is to develop rapid and accurate analytical method for analyzing organic compositions of manure made from excreta of livestock, and to establish quality evaluation method based on the compositions predicted by near infrared reflectance spectroscopy (NIRS). Sixteen samples of manure, each eight samples prepared from two treatments, were used in this study. The manure samples were prepared by mixing 560 kg feces of swine,60 kg sawdust with moisture content was adjusted to be 65%. The mixture was then keep under two kinds of shelter, black and clear sheets, as a treatment on the effect of sunlight. Samples were taken in every week (form week-0 to 7) during the process of manure making. Samples were analyzed to determine neutral detergent fiber (NDF), acid detergent fiber (ADF) and acid detergent lignin (ADL) by detergent methods, and organic cell wall (OCW) and fibrous content of low digestibility in OCW (Ob) by enzymatic methods. Biological oxygen demand (BOD) was analyzed by coulometric respirometer method. These compositions were carbohydrateds and lignin that were hardly digested. Spectra of samples were scanned by NIR instrument model 6500 (Pacific Scientific) and read over the range of wavelength between 400 and 2500nm. Calibration equations were developed using eight manure samples collected from black sheet shelter, while prediction was conducted to the other eight samples from clear sheet shelter. Accuracy of NTRS prediction was evaluated by correlation coefficients (r), standard error of prediction (SEP) and ration of standard deviation of reference data in prediction sample set to SEP (RPD). The r, SEP and RPD value of forage were 0.99, 0.69 and 7.6 for ADL, 0.96, 1.03 and 4.1 for NDF, 0.98, 0.60 and 4.9 for ADF, 0.92, 1.24 and 2.6 for Ob, and 0.91, 1.02 and 7.3 for BOD, respectively. The results indicated that NIRS could be used to measure the organic composition of forage used in manure samples.
