Studies on Lao-Chao Culture Filtrate for a Flavoring Agent in a Yogurt-Like Product

  • Lao-chao is a traditional Chinese fermented rice product with a sweet and fruity flavor, containing high levels of glucose, a little alcohol and milk-clotting characteristics. In order to optimize commercial production of lao-chao, Rhizopus javanicus and Saccharomyces cerevisiae were selected as the mold and yeast starter, respectively. A commercial mixed starter (chiu-yao) was used as control. Fermentation of the experimental combination revealed a sharp drop in pH (to 4.5) on the fourth day, remaining constant thereafter. Content of reducing sugars gradually decreased throughout the entire fermentation period. Of the free amino acids, higher quantities of alanine, leucine, proline, glutamic acid, glutamine and $NH_3$ were noted. For sugars, glucose revealed the highest concentration, while organic acid levels, including those for oxalic, lactic, citric and pyroglutamic acid, increased throughout the fermentation period. Twenty-one compounds were identified by gas chromatography from aroma concentrates of the lao-chao culture filtrate, prepared using the headspace method. For the flavor components, higher quantities of ethanol, fusel oil and ester were determined in both culture filtrates. In regard to the evaluation of yogurt-like product, there were significant differences in alcoholic smell, texture and curd firmness.

The Effect of Different Sources of Urease Enzyme on the Nutritive Value of Wheat Straw Treated with Urea as a Source of Ammonia

  • Wheat straw samples (3-4 cm) were sprayed with solutions of urea (U) alone or with a dry addition of garden soil (GS), midden soil (MS), soya bean meal (SM) or jack bean meal (JM) as crude urease sources and with a pure urease (UR) enzyme. Each of the urease sources was included at two levels: 30 and 60 g/kg except pure urease, which was added at a level of 2.5 & 5.0 g/kg treated straw dry matter. Untreated straw without urease source was used as a control. After treatment, samples were sealed in polythene bags and stored for 2, 7, 14, 21 and 35 days at $19{^{\circ}C}$. The urease sources, their levels and treatment time produced significant effects on ammonia production (p<0.01). The addition of urease offered more flexibility in hydrolyzing urea in the shortest possible time. Incorporation of soya bean and jack bean meal was effective in reducing the modified acid detergent fiber (MADF) content of straw and the same time increasing organic matter (OM) digestibility. Overall effect, addition of soya bean to urea at a ratio of 1:1 appeared to be the most satisfactory urease source for the treatment of urea and wheat straw.

Protected (bypass) Protein and Feed Value of Hazelnut Kernel Oil Meal

  • In situ and in vivo digestion trials were conducted to determine the degradation of dry matter (DM), crude protein (CP) and effective protein degtadability (EPD), and digestibility of nutrients of Hazelnut kernel oil meal (HKOM), and effects of HKOM on nitrogen (N) balance. In the in situ study, nylon bag were suspended in the rumen of 3 Karayaka rams to estimate protected protein. Protein sources were analyzed for pepsin soluble protein (PSP) using a Pepsin Digestion Method. In the digestion trials, 4 Karayaka rams (36 mo.) were used in a $4{\times}4$ Latin square to evaluate the digestibility of nutrients and N retention to measure effects of diets containing HKOM, soybean meal (SBM) corn gluten meal (CGM) and urea (U). The degradability of DM and CP, and PSP content of HKOM were lower (p>0.05) than that of SBM, but higher (p<0.001) than that of CGM. EPD of HKOM was higher (p<0.01) than that of SBM or CGM. The apparent digestion coefficients of organic matter and CP for HKOM were lower than for SBM, but higher than for CGM. N retention of HKOM was higher than that of SBM and lower than that of CGM (p>0.05). In conclusion, these data may indicate that the HKOM is a high digestible feed source with a value between SBM and CGM.

Studies on the Biological and Chemical Properties of Musty Ginseng Root and its Causal Mechanism (적변삼의 생물.화학적 특성과 그 발생원인에 관하여)

  • Rusty root of ginseng has been known as one of the limiting factors in ginseng production in Korea. An attempt was, therefore, made to elucidate biological and chemical natures of the rusty root, and the redox Potential of the ginseng cultivated soils were measured and compared with diseased and non-diseased soils. Reddish discoloration was most frequently observed on the epidermis of ginseng root and the pigments were accumulated in all epidermal cells of the diseased lesions. The lower the redox potential of the ginseng cultivated soil was, the more severe the rusty root was observed. Fe content in the diseased epidermis was 3 times higher than that of healthy one. Organic acids such as oxalic, malonic, succinic, and citric acids were also higher in the mss root than in the healthy one. Thin layer chromatogram of phenolic acid fractions obtained from the epidermal cells of the rusty root of ginseng exhibited 3 to 4 unidentified substances not found in the healthy root. Also lignification of the epidermal cells and the activity of phenylalanine ammonia lyase were greater in the rusty root than the healthy root. Colony formation and conidia production of F. solani, And mycelial growth and sclerotium formation of Sclerotinia sp. isolated from ginseng root were suppressed in a nutritionally minimal medium supplemented with water extract of rusty ginseng root epidermis. It is, therefore, suggested that rusty root of ginseng is caused by unfavorable rhizosphere environmental stress or stresses resulting abnormal metabolism in the root as a selfdefence mechanism of non-specific resistance responses.

Functional Properties of Bifidobacterium longum and Their Incorporation into Cheese Making Process (비피도박테리움 롱검의 기능성과 치즈 제조에 활용)

  • Members of the genus Bifidobacterium are prevalent in the human colon and represent up to 90% of all bacteria in fecal samples of breast-fed infants, and 3~5% of adult fecal microbiota. Bifidobacteria produce organic acids, thus reducing the colon pH to a level inhibitory for pathogenic bacteria. They can also detoxify a number of toxic compounds and adhere to the colon mucosa, thus preventing the adherence of pathogens and induction of colon cancer. Recently, we identified a novel Bifidobacterium longum subsp. longum strain, KACC 91563, in a fecal sample of a Korean neonate, and demonstrated its functional properties. We showed that B. longum KACC 91563 alleviates food allergy through mast cell suppression and produces antioxidative and antihypertensive peptides by casein hydrolysis. Dairy products are considered as an ideal food system for the delivery of probiotic cultures to the human gastrointestinal tract. Cheese affords protection to probiotic microbes during gastric transit due to its relatively high pH, more solid consistency, higher fat content, and higher buffering capacity. Incorporation of B. longum KACC 91563 into cheese making is currently under study.

A Study on Composition of Thinners Used in Korea (우리나라에서 사용되는 일부 신나의 구성성분에 관한 연구)

  • In this study, 108 thinners were analyzed to identify their composition. The purposes of this study were to provide the data for MSDS and worker exposure levels. Thinners were collected from manufacturing industries, distributors and users. Wide ranges of thinner components were found ; Toluene, xylene, and ethyl benzene were most often found. Next, MIBK, cellosolve acetate, butyl cellosolve, and butyl acetate were found in 20-40 thinners. Others, such as acetone, n-hexane, cyclohexane, heptane, methylcyciohexane, octane and nonane were also found. There were about 5-6 components in each thinner. In the view point of Industrial Hygiene, Benzene was the most important component of thinners, which procures leukemia. Benzene was found from 8 kinds of thinners out of the total 108 surveyed. Thus, Content of benzene in thinners must be evaluated when industrial hygiene surveys are performed. Aromatic hydrocarbons were identified from 71 kinds of thinners out of total 108 and their contents were 10-87%. Alkanes were not contained in automobile painting, spraying, degreasing, urethane and epoxy thinners but lacquer, enamel and coating thinners.

  • The pickling brine generated from the salting process of kimchi production is difficult to treat biologically due to very high content of salt. When pickling brine is treated and discharged, it cannot satisfy the criteria for effluent water quality in clean areas, while resources such as the salt to be recycled and the industrial water are wasted. However, sterilization by ozone, UV and photocatalyst is expensive installation costs and operating costs when considering the small kimchi manufacturers. Therefore there is a need to develop economical process. The study was conducted on the sterilization efficiency of the pickling brine using electrochemical processing. The electrochemical treatment of organic matters has advantages over conventional methods such as active carbon absorption process, chemical oxidation, and biological treatment because the response speed is faster and it does not require expensive, harmful oxidizing agents. This study were performed to examine the possibility of electrochemical treatment for the efficient processing of pickling brine and evaluated the performance of residual chlorine for the microbial sterilization.

Anaerobic co-digestion of food waste leachate with microalgae for improvement of methane production (메탄생산 향상을 위한 음폐수와 미세조류의 혐기성 통합소화)

  • Food waste leachate (FWL) is a serious pollutant waste coming from the food waste recycling facilities in Korea. FWL has a high organic matter content and high COD to nitrogen (COD/N) ratio, which can disturb efficient methane production in the anaerobic digestion of FWL. In the present study a microalga, Clorella vulgaris (C.V), was used as co-substrate for the FWL anaerobic digestion in order to supply nutrients, decrease the COD/N ratio and increase its methane yield. Different co-digestion mixtures (COD/N ratios) were studied by using biochemical methane potential test and modified Gompertz equation for kinetic study. Mixed substrate of FWL and C. vulgaris in the co-digestion clearly showed more the biomethane yield than the sole substrates. The maximum methane production, 827.7 mL-$CH_4$/g-VS added, was obtained for COD/N ratio of 24/1, whereas the highest improvement of methane yield was found for COD/N ratio of 15/1.

Physicochemical Properties of Landfill Mined Wastes from Old Landfill Site (불량 매립지에서 굴착된 폐기물의 물리화학적 특성평가)

  • This study was carried out to evaluate the Physicochemical characteristics of mined waste(separated waste and soil) and to predict environmental effect of an old landfill site located at north of Seoul. Municipal solid waster(MSW) had been disposed of at the old landfill site used in this study for about 2 years(1990-1992). The old landfill site selected for this study had accepted mainly municipal solid waste. The landfill-mined waste contained separated waste (40.9%) and soil(59.1%) by wet weight basis. The separated waste consisted of combustible(91.0%) and non-combustible(9.0%). The combustible waste was mainly non-biodegradable plastics. The low heating value of the separated combustible waste, which is calculated by Dulong's equation, was as high as 3,470kcal/kg. According to the Korean Extraction Procedure, separated waste and soil were proved to be not hazardous. The total content of heavy metal in the separated waste and soil met standard of California State, USA. Therefore the separated waste may be relandfilled at a sanitary landfill site and/or burned up at an incinerator, and the separated old soil may be used ad landfill cover-soil at a sanitary landfill site. Water quality of two streams was grade IV, of which water could be used as industrial and agricultural water. The streams near the landfill site might not be contaminated by leachate from the old landfill site. It was estimated that organic matter in the old landfill site would not be actively biodegraded within a short period of time.

Ferroelectric Properties of Bi3.25La0.75Ti3O12 Thin Films with Eu Contents for Non-volatile Memory Device Application (비휘발성 메모리 소자응용을 위한 Eu 첨가량에 따른 BET 박막의 강유전 특성)

  • The effect of Eu contents on the ferroelectric properties of $Bi_{4-x}Eu_xTi_3 O_{12}$ (BET) thin films has been investigated. Bismuth Europium titanate thin films with a Eu contents were prepared on the $Pt/Ti/SiO_2/Si$ substrate by metal-organic decomposition technique. The structure and the morphology of the films were analyzed using X-ray diffraction (XRD) and field emission scanning microscopy (FE-SEM), respectively. From the XRD analysis, it was found that BET thin films have polycrystalline structure, and the layered-perovskite phase is obtained when the Eu contents exceeds 0.2 (x > 0.2). Also, the ferroelectric characteristics of the BET thin films were found to be dependent on the Eu content. Particularly, the BET films doped with x = 0.75 show better ferroelectric properties (remanent polarization 2Pr = 60.99 C/$cm^2$ and only a little polarization fatigue up to $3.5{\times}10^9$ bipolar switching cycling) than those doped with other Eu contents.
