The Effect of a Natural Zeolite (Clinoptilolite) on the Performance of Broiler Chickens and the Quality of Their Litter

  • The objective of this study was to assess the effect of clinoptilolite (a natural zeolite) on growth and performance of broilers as well as on the quality of their litter. A total of 5,200 one-day old broiler chickens (Cobb 500) were used in two consecutive rounds lasting 42 days each. The broilers were given free and continuous access to a nutritionally non-limiting diet (in meal form) that was either a basal diet (B) or a 'zeolite diet' (Z; the basal diet supplemented with clinoptilolite at a level of 2%). A $2{\times}2$ factorial design consisted of two feeding treatments (B and Z) and two bedding types, sawdust (S) and sawdust with zeolite (Sz, which was the result of adding $2kg\;zeolite/m^2$), was used. In each round the broilers were randomly assigned to one of four (n = 650), treatment groups: two fed on the basal diet (B) and had bedding of either sawdust (group BS) or sawdust and zeolite (group BSz) and two fed on the Z diet and had as bedding either sawdust (ZS) or sawdust and zeolite (ZSz). Average growth rates were significantly (p<0.05) different between broilers of different groups; broilers that were fed on the 'zeolite diet' (Z) and were placed either in a compartment with sawdust bedding or sawdust bedding and zeolite (ZS and ZSz) as well those that were fed on the basal diet in a compartment with sawdust bedding and zeolite (BSz) grew at a faster rate (p<0.05) compared with those of the control group (BS). The incorporation of NZ in broilers diets and in their bedding material decreased the organic content in litter samples throughout the experimental period. The lowest organic content was recorded in group ZSz where NZ was added in both feed and litter. Mean ammonia concentration (ppm) was significantly higher in group ZS in comparison to groups BSz and ZSz (27.00 vs. 20.55 and 21.71 respectively). The results of this study showed that the incorporation of the clinoptilolite both in feed and into the litter had a positive effect on broiler growth and also on the quality of their litter.

Development and Validation of an HPLC Method for the Pharmacokinetic Study of Dipyridamole in Human (디피리다몰 체내동태 연구를 위한 혈청 중 디피리다몰의 HPLC 정량법 개발 및 검증)

  • A rapid, selective and sensitive reversed-phase HPLC method for the determination of dipyridamole in human serum was developed, validated, and applied to the pharmacokinetic study of dipyridamole. Dipyridamole and internal standard, loxapine, were extracted from human serum by liquid-liquid extraction with diethyl ether and analyzed on a Nova Pak $C_{I8}$ column with the mobile phase of 40 mM ammonium acetate:methanol:acetonitrile (35:35:30)(v/v/v, pH 7.8). Detection wavelength of 280 nm and flow rate of 1.0 mL/min were fixed for the study. The assay robustness for the changes of mobile phase pH, organic solvent content, and flow rate was confirmed by $3^3$ factorial design using a fixed dipyridamole concentration (50 ng/mL) with respect to its peak area and retention time. And also, the ruggedness of this method was investigated at three different laboratories using same quality control (QC) samples. This method showed linear response over the concentration range of 2-2000 ng/mL with correlation coefficients greater than 0.999. The lower limit of quantification using 0.5 mL of serum was 2 ng/mL, which was sensitive enough for pharmacokinetic studies of dipyridamole. The overall accuracy of the quality control samples ranged from 103.94 to 105.86% for dipyridamole with overall precision (% C.V.) being 4.60-11.49%. The relative mean recovery of dipyridamole for human serum was 97.64%. Stability studies showed that dipyridamole was stable during storage, or during the assay procedure in human serum. The peak area and retention time of dipyridamole were not significantly affected by the changes of mobile phase pH, organic solvent content, and flow rate under the conditions studied. This method showed good ruggedness (within 15% C.V.) and was successfully used for the analysis of dipyridamole in human serum samples for the pharmacokinetic studies of orally administered Dimor tablet (75 mg as dipyridamole) at three different laboratories, demonstrating the suitability of the method.

Development and Validation of HPLC Method for Pharmacokinetic Study of Promethazine in Human (염산프로메타진 체내동태 연구를 위한 혈청 중 프로메타진의 HPLC 정량법 개발 및 검증)

  • A rapid, selective and sensitive reversed-phase HPLC method for the determination of promethazine in human serum was developed, validated, and applied to the pharmacokinetic study of promethazine. Promethazine and internal standard, chlorpromazine, were extracted from human serum by liquid-liquid extraction with n-hexane containing 0.8% isopropanol and analyzed on a Capcell Pak CN column with the mobile phase of acetonitrile-0.2 M potassium dihydrogen phosphate (42:58, v/v, adjusted to pH 6.0 with 1 M NaOH). Detection wavelength of 251 nm and flow rate of 0.9 mL/min were fixed for the study. The assay robustness for the changes of mobile phase pH, organic solvent content, and flow rate was confirmed by $3^{3}$ factorial design using a fixed promethazine concentration (10 ng/mL) with respect to its peak area and retention time. In addition, the ruggedness of this method was investigated at three different laboratories using same quality control (QC) samples. This method showed linear response over the concentration range of 1-40 ng/mL with correlation coefficients greater than 0.999. The lower limit of quantification using 1 mL of serum was 1 ng/mL, which was sensitive enough for pharmacokinetic studies. The overall accuracy of the quality control samples ranged from 96.15 to 105.40% for promethazine with overall precision (% C.V.) being 6.70-11.22%. The relative mean recovery of promethazine for human serum was 63.54%. Stability (freeze-thaw and short-term) studies showed that promethazine was stable during storage, or during the assay procedure in human serum. However, the storage at $-80^{\circ}C$ for 4 weeks showed that promethazine was not stable. Extracted serum sample and stock solution were not allowed to stand at ambient temperature for 12 hr prior to injection. The peak area and retention time of promethazine were not significantly affected by the changes of mobile phase pH, organic solvent content, and flow rate under the conditions studied. This method showed good ruggedness (within 15% C.V.) and was successfully used for the analysis of promethazine in human serum samples for the pharmacokinetic studies of orally administered Himazin tablet (25 mg as promethazine hydrochloride) at three different laboratories, demonstrating the suitability of the method.

Quality Characteristics and Antioxidant Activities of Commercial Jeupjang (일부 시판 즙장의 품질 특성 및 항산화 활성)

  • Jeupjang has long been considered only a local delicacy. So, it has gradually been forgotten because its name and definition have not been established. This study was performed in order to provide some basic data by comparing the general components, color, free amino acids, free sugar, organic acids and antioxidant activities between commercial Jeupjang(GG, GD, JM, NJ, NS). Moisture contents, crude protein, crude fat and crude ash contents of Jeupjang were 48.07~61.77%, 4.08~10.53%, 3.92~4.64%, and 3.63~9.95%. pH, titrable acidity and salt in Jeupjang were 4.32~5.50, 9.37~14.82 mL(1N-NaOHmL), and 3.27~8.90%. Amino-type nitrogen content is 42.05~ 138.83mg%. Lightness(L), redness(a) and yellowness(b) values in color of Jeupjang were10.04~21.89, 18.92~28.80, and 17.16~37.56. The total content of free amino acids in commercial Jeupjang ranged from 35.22mg% to 306.02mg% in which it showed wide variation among samples. The major free amino acids were glutamic acid, aspartic acid, alanine and proline. The major free sugars were fructose(0.31~1.60%), glucose(6.20~ 13.61%) and maltose(7.34~10.01%). The major organic acids were succinic acid(29.08~ 103.62 mg%) and citric acid(3.08~108.69 mg%). Total polyphenol contents were from 1.16 mg/g to 1.78 mg/g, DPPH radical scavenging activity is from 0.57 AEAC to 1.53 AEAC and ABTS radical scavenging activity is from 1.58 AEAC to 2.85 AEAC. This result might cause the differences of soybean composition, processing methods and brewing periods. For the succession of traditional fermented food such as Jeupjang, quality standards are required. To this end, the standardization of manufacturing processes must be followed.

Distribution and Growth Pattern of Sternaspis scutata (Polychaeta : Sternaspidae) in Chinhae Bay, Korea (진해 만산 오뚜기갯지렁이 Sternaspis scutata (다모강 :오뚜기갯지렁이과)의 분포 및 성장)

  • A population study on the sternaspid polychaete Sternaspis scutata (Ranzani) was conducted in Chinhae Bay, Korea, from lune 1987 to May 1990. Sternaspis scutata was chiefly distributed in the mouth area of the bay, and its distributional delimitation remained unchanged all the year round. Abundance of this species appeared to be related to the organic carbon content of the sediment. It was relatively high in the area where the organic content of the sediment was between 2.0 and $4.0\%(mean\;3.8{\pm}1.11)$ with mean grain size of $8.75\phi$. The relationship between the length of the ventral plate and the body wet weight is Wt=13.2752 $L^{2.7984}(R^2=0.962)$. Therefore, the length of ventral plate can be used as an indicator of body growth of this species. The growth curves fitted to yon Bertalanffy equations for ventral plates and weights are $L_{t}=5.235(1-e^{-0.3625(t-0.4047)})$ and $W_{t}=1364.1(1-e^{-0.3625(t-0.4047)})^{2.7984}$, respectively. Size frequency histograms showed that the settlement occurred during all the year round with its peak in summer.

Effect of Water Treatment Sludge Application on the Growth of Korean Local Corn(Zeas Mays L.) (옥수수에 대한 정수 슬러지(Alum sludge)의 시용효과와 Aluminum이 옥수수의 유식물 생장에 미치는 영향)

  • Effect of water treatment sludge(alum sludge) application on the growth of Korea local corn(Zeas Mays L.) was evaluated and phytotoxicity of alum sludge on the growth of corn roots and plant height was tested. Alum sludge itself and composted alum sludge compost were applicated by 1500kg/ha in the plot respectively. For the fertility test, yield, chlorophyll content, plant height of corn treated by alum sludge and composted alum sludge were better than that of control. No clear toxixity of alum sludge was found. At the food chemical analyses of seed corn, no obvious component variation were evaluated either. Test of pH effect on phytotoxicity of alum sludge on corns showed that plant height and root growth of corns were inhibited at low pH 4 solution. Phytotoxicity test of alum sludge extract which was extracted at various pH solutions, showed that growth and cations uptake inhibition were found the more serious the lower the pH. Plant height and root growth injury were serious at higher than 100ppm Al content solution.

Leaching behavior of the herbicide bentazon in soil column (Soil column중 제초제 bentazon의 용탈 행적)

  • In order to elucidate the leaching behaviour of the herbicide bentazon in soil, soil columns(5cm ID ${\times}$ 34 cm L) were packed with three different soils up to 30 cm height, followed by the treatment of [$^{14}C$]bentazon, and rice plants(Oryza sativa L.) were grown for 9 weeks on these columns, with the columns without growing rice plants as the control for comparison. The amounts of $^{14}C$ activities percolated were about 92% of the originally applied $^{14}C$ irrespective of the physicochemical properties of the soils in the absence of rice plants, whereas $21{\sim}50%$ of the originally applied $^{14}C$ was percolated in the presence of rice plants, suggesting that the amounts of $^{14}C$ leached decreased remarkably in rice-cultivating soils. Bentazon leached faster in soil with higher pH and with lower organic matter content in the presence of growing rice plants. The amounts of $^{14}CO_{2}$ evolved from the soil columns were less than 0.2% of the originally applied $^{14}C$. Smaller amounts of $^{14}C$ were translocated into shoots via roots in soils with higher organic matter content. $^{14}C$ activities distributed into the aqueous phase of the leachate collected from the soil columns increased with leaching period and by rice cultivation, whereas the physico-chemical properties of soils did not exhibit any effect. The amounts of soil-bound residues increased remarkably by cultivating rice plants.

The Contents of Nitrogen, Phosphorus, Silicon Nutrient and Algal Growth Potential (AGP) in the Sediment of Taechong Reservoir (대청호 저토의 N, P및 Si 영양염 함량과 조류생장잠재력)

  • The chemistry of porewater and exchangeable nutrients of sediment was determined to define potential influence of sediment nutrients on the water quality and the phytoplankton growth in Taechong Reservoir in March and May of 2000. The sediment-water interface showed almost anoxic state, < 0.5 mg $O_2/l$. Conductivity of the porewater was higher 1.9${\sim}$2.6 fold than that of sediment, and its variation was greater in the shallow water. Eh ranged from -12mV to -148 mV and bulk density from $1.17\;g/cm^3$ to $1.30\;g/cm^3$ and they did not differ among stations. The water content in the sediment ranged from 58% to 72% and organics from 8% to 13%, and they were higher toward the lower part of reservoir. Soil texture was in the order of sand>clay>silt and sandy sediment accounted for 97% of the total sediment. The total bacterial numbers and diatom abundance were high in the downstream of the reservoir. In porewater and exchangeable nutrients of sediment, nitrogen and phosphorus were mostly consist of the particulate form. Inorganic nitrogen was mostly composed of $NH_4$. Nitrogen of porewater was mostly an inorganic form while exchangeable nutrients were composed an organic form. However, phosphorus was composed of dissolved organic fraction in the porewater while inorganic fraction in the exchangeable nutrients. Silicon content of sediment was much exchangeable nutrients with 63%, and it was higher than in the porewater. In summary, the sediments of Taechong Reservoir were mostly composed of organics and assessed to be a eutrophic state.

Changes in Properties of Kimchi Prepared with Different Kinds and Levels of Salted and Fermented Seafoods during Fermentation (젓갈의 종류 및 첨가수준에 따른 배추 김치의 발효기간 중 특성변화)

  • This study was conducted to examine the effects of the kinds and the levels of salted and fermented seafoods, shrimp and juice of anchovy, on the properties of kimchi during fermentation. There were no significant differences in pH and contents of total acid, reducing sugar, and organic acid examined except lactic acid contents among the kimchi samples at the same fermentation period. Until four days of fermentation, kimchi containing higher level of salted and fermented anchovy juice tended to have greater number of total microorganisms and of Leuconostoc genus microorganisms than the other kimchi samples. Sensory characteristics were not significantly different among the samples at the same fermentation period. significant decrease in pH and in reducing sugar content, and increase in total acid content were noticed in all the kimchi samples during fermentation. This was more marked in kimchi samples containg salted and fermented seafoods. Contents of malic and succinic acids decreased while those of actic and acetic acids increased with longer fermentation. There were significant changes in the numbers of microorganisms during fermentation. Carbonic mouthfeel, salty taste, sour taste and staled flay or of all the kimchi samples increased while firmness and crispness decreased with extended fermentation.

Changes in Organic Acid Composition of Dried Lavers during Storage (마른김의 유기산조성(有機酸組成)과 저장중의 변화)

  • Dried lavers, Porphyra yezoensis Ueda, were stored at $20^{\circ}C$ for 10, 25 and 45 days under a dry and humid conditions respectively, and changes in the contents of organic acids were studied by GLC and supplementary use of GC-MS. 1) Out of eight acids determined, i. e., pyruvic, lactic, oxalic, malonic, fumaric, succinic, malic, and citric acid, relatively abundant ones were oxalic, succinic, malic, and citric acid. 2) Oxalic, malic, and succinic acid did not show any notable changes in their contents under a dry condition, however, under a humid condition the contents of the former tow acids decreased rapidly showing contrast to that of succinic acid, of which content increased markedly. The content of citric acid rapidly· decreased independent of the storage conditions. 3) Among some unidentified components, two of them were presumed to be aromatic acids from their mass spectra. The contents of the two acids decreased slowly under a dry condition, however, rapidly decreased under a humid condition and after 45 days storage almost all of them disappeared. 4) The contents of total volatile acids were increased under the both storage conditions, however, the rate of increment was larger under a humid than under a dry condition.
