Study on the Synthsis and Characteristics of Lipophilic Derivatives of β-Sitosterol (β-시토스테롤의 지용성 치환체의 합성 및 특성에 관한 연구)

  • In the paper, lipophilic derivatives of $\beta$-sitosterol, which are known to have a potential to reduce blood cholesterol level, were synthesized by the esterification of $\beta$-sitosterol and fatty acids. When the esterification reactions using stearic acid, oleic acid or linoleic acid as fatty acids were carried out in the presence of an acidic catalyst, the reaction for unsaturated fatty acids such as oleic acid and linoleic acid afforded a significant amount of side products which may be produced by oxidation of unsaturated groups. On the other hand, esterification reactions in the presence of dehydrating agents and a basic catalyst gave pure products regardless of the nature of fatty acids. The solubilities of lipophilic derivatives of $\beta$-sitostero to organic solvents and edible oil were observed to increase as the degree of unsaturation of fatty acids increases.

Nutritional Characteristics of Eels (Auguilla japonica) Fed a Diet of Yuza (Citrus junos Sieb ex Tanaka) (유자 첨가 사료로 사육한 뱀장어의 영양학적 특성)

  • The study investigated the effect of diets supplemented with different levels (0 and 2.5%) of yuza (Citrus junos Sieb ex Tanaka) on the nutritional characteristics of eels (Auguilla japonica). Fish ($9.8{\pm}1.3g$) was fed to apparent satiation twice daily for 8 months. There were no significant differences in proximate composition among the treatment groups, except for the ash and carbohydrate contents (P<0.05). The vitamin C content of eel muscle in the yuza-added group was two-times higher than in non-added groups (P<0.05). Among eight organic acids in eel muscle, lactic acid was predominant, followed by citric acid, oxalic acid, malic acid, and acetic acid. Eels fed a 2.5% yuza diet had the highest lactic acid content in all groups. Six sugars were found in all groups and glucose was the major sugar. Glucose and maltose were the dominant sugars in the yuza-added group. The abundant fatty acids in the yuza-added group were C18:1 n-9, C16:0, and C16:1 n-7, which comprised over 80% of the total fatty acids. The major amino acids in samples were glutamic acid, aspartic acid, lysine, and leucine. There were few differences in the free amino acid compositions among the groups. However, histidine was the predominant amino acid and constituted over 53% of the total free amino acids.

Substrate Effects on Biological Excess Phosphorus Removal (유기물질이 인제거 특성에 미치는 영향)

  • In this research, investigations were made on the effect of type and load of organic substrate on phosphorus release. Reactors of three different sizes were operated, being fed on five kinds of organic substrates. The quantitative analyses were made on phosphorus release and substrate utilization under anaerobic condition. The molar ratios of the uptaken organic substrate to the released phosphorus were 0.5 with acetate, 0.6 with glucose, 0.8 with glucose/acetate, and 1.2 with glucose/acids, respectively. The phosphorus release was inhibited at the higher organic load than the normal at stead state. Both acetate and acids/glucose enhanced phosphorus release- as well as uptake-rate, however, the complete phosphorus removal was achieved after the microbial adaptation to the new environment. In case with acetate, operation was hampered by the poor sludge settleability and phosphorus uptake was not enough although the phosphorus release was active. But with milk/starch, the phosphorus release and uptake was well developed even though phosphorus release was not comparatively high. From this study, it was concluded that organic substrates, such as glucose seemed to be converted fatty acids after fast bio-sorption, followed by concurrent uptake of these acids by excess phosphorus removing bacteria.

Flavor Compounds of Dried Shellfishes 2. Changes of Reducing Sugars, Organic Acids and Fatty Acids Composition in Shellfishes during Drying Process (패류 건제품의 향기성분에 관한 연구 2. 건조가공에 따른 패류의 환원당, 유기산 및 지방산 조성의 변화)

  • The levels of reducing sugars, organic acids and fatty acids, known as precursors of major flavors induced from dried sellfish, were analyzed to investigate their behaviors during drying process. Free reducing sugar contents were markedly decreased in all samples by boiling, Its content in Blue mussel and Short-necked calm samples significantly decreased after drying process. Among the eight different organic acids from blue mussel, Short-necked calm and Pacific oyster samples, succinic acid in blue mussel and short-necked calm samples was measured with $74.9\%\;and\;67.4\%$, respectively. In the sample of Pacific oyster, succinic acid content was found to be $38.4\%$, but pyroglutamic acid content was the highest level with $41.6\%$. Contents of organic acids with exception succinic acid were significantly reduced in all three samples of sun-dried or hot-air dried. The content of succinic acid in the samples of sun-dried and hot-air dried pacific oyster reduced to $53.0\%\;and\;44.2\%$, respectively, but relatively small decrease $(29.0\%\;and\;10.0\%)$ was observed in sun-dried and hot-air dried short-necked calm samples, respectively. Content of polar lipid with the major fatty acids profile of C16:0, C16:1, C20:5 and C22:6 was consisted of $59.1\%,\;66.7\%\;and\;42.4\%$, respectively, in blue mussel, short-necked calm and pacific oyster samples and the content of polyene fatty acids was appeared to be $40.5\%,\;48.6\%\;and\;48.9\%$, respectively. Relatively high peroxide value in all boiled-dried products samples was found to be $41.64\~86.80\;meq/kg$ sample. Carbonyl value in boiled-dried products samples was found to be $15.55\~27.99\;meq/kg$, but its value in broiled products samples was significantly high level of $127.6\~136.5\;meq/kg$.

Effects of Organic Amendments on Soil Microbial Community in Red Pepper Field (시용 유기물의 종류가 고추 재배지 토양 미생물상에 미치는 영향)

  • Diverse organic amendments available in local areas have been used to improve soil quality in red pepper field and so the need for investigating the soil chemical and biological properties changed by the organic amendments application is increasing. Soil microbial diversities were measured by phospholipid fatty acid (PLFA) and Biolog $EcoPlate^{TM}$. Compost was most effective for improving soil chemical properties including pH, EC, total nitrogen, P, K, and Ca, and bark increased soil organic matter significantly (P=0.05). Compost increased the fatty acids indicating actinomycetes and vascular arbuscular fungi, and ratio of cy19:0/18:1w7c and monounsaturated fatty acids/saturated fatty acids in soils in PLFA analysis. Bark increased soil fungal indicators in PLFA analysis (P=0.05). Principal component analysis of Biolog EcoPlate data and PLFA differentiated the compost- and bark-amended soils from other organic matteramended soils especially the soil incorporated with compost. More researches are needed to use bark for improving soil microbial properties because the soil chemical and microbiological properties caused by compost and bark are significantly different.

Development of Gas Chromatography/Mass Spectrometry for the Determination of Essential Fatty Acids in Food Supplemental Oil Products

  • A gas chromatography/mass spectrometric (GC/MS) method was developed as a candidate reference method for the accurate determination of essential fatty acids (linoleic acid, ${\alpha}$- and ${\gamma}$-linolenic acids) in food supplemental oil products. Samples were spiked with three internal standards (stearic acid-$d_{35}$, $^{13}C_{18}$-linoleic acid, and $^{13}C_{18}$-${\alpha}$-linolenic acid). Samples were then subject to saponification, derivatization for methylation, and extraction by organic solvent. For GC/MS measurement, an Agilent HP-88 column, designed for the separation of fatty acid methyl esters, was selected after comparing with other columns as it provided better separation for target analytes. Target analytes and internal standards were detected by selected ion monitoring of molecular ions of their methyl ester forms. The GC/MS method was applied for the measurement of three botanical oils in NIST SRM 3274 (borage oil, evening primrose oil, and flax oil), and measurement results agreed with the certified values. Measurement results for target analytes which have corresponding isotope-labeled analogues as internal standard were calculated based on isotope dilution mass spectrometry (IDMS) approach, and compared with results calculated by using the other two internal standards. Results from the IDMS approach and the typical internal standard approach were in good agreement within their measurement uncertainties. It proves that the developed GC/MS method can provide similar metrological quality with IDMS methods for the measurement of fatty acids in natural oil samples if a proper fatty acid is used as an internal standard.

Chemical Components in Different Parts of Korean Sword Bean(Canavalia gladiata) (한국산 작두콩의 부위별 화학성분)

  • The chemical components in different parts such as seed, cotyledon, hull, pod, leaf, stem and root of Korean sword bean(Canavalia gladiata) were determined in this study. The contents of total sugar and protein were relatively higher in cotyledon than other parts. The highest mineral component was K, Ca in root and leaf, respectively. In case of organic acids, relatively higher content of oxalic acid(2,556 mg%), citric acid(573 mg%) was found in leaf and pod, respectively. Among free sugars, the contents of sucrose(3.80%), fructose(2.17%) were the highest contents in hull and stem, respectively. The highest component total amino acids in cotyledon and leaf, pod, stem, and root was glutamic acid(592 mg%), glutamic acid(429 mg%), proline(497 mg%), lysine(328 mg%) and arginine(714 mg%), respectively. Among fatty acids in hull and pod, palmitic acid(32.75%, 21.93%) was high in saturated fatty acid fraction, while linoleic acid(39.15%, 43.03%) was high in polyunsaturated fatty acid fraction.

Characterization of Itraconazole Semisolid Dosage Forms Prepared by Hot Melt Technique

  • The objective of this study was to formulate itraconazole semisolid dosage forms and characterize their physicochemical properties. Itraconazole and excipients such as polysorbate 80, fatty acids, fatty alcohols, oils and organic acids were melted at $160^{\circ}C$. The fused solution was then cooled immediately at $-10^{\circ}C$ to make wax-like semisolid preparations. Their physicochemical attributes were first characterized using differential scanning calorimetry, Fourier transform infrared spectroscopy and nuclear magnetic resonance spectrometry. The solubility of itraconazole in semisolid preparations and their dispersability in the simulated gastric fluid were also determined. Our semisolid preparations did not show any distinct endothermic peak of a crystalline form of itraconazole around $160-163^{\circ}C$. This suggested that it was changed into amorphous one, when it was formulated into semisolid preparations. In addition, the distinctive functional peaks and chemical shifts of itraconazole were well retained after processing into semisolid preparations. It could be inferred from the data that itraconazole was stable during incorporation into semisolid preparations by the hot melt technique. In particular, itraconazole semisolid preparations composed of polysorbate 80, fatty acids and organic acids showed good solubility and dissolution when dispersed in an aqueous medium. It was anticipated that the semisolid dosage forms would be industrially applicable to improving the bioavailability of poorly water-soluble drugs.

Acid Fermentation Characteristics of Waste Activated Sludge using Acids and Ultrasonication (산용해 및 초음파를 이용한 하수 슬러지의 산발효 특성)

  • The Carbon source to enhance the denitrification is essential matter in the advanced sewage treatment. For the high level of nutrient removal, external carbons such as ethanol, methanol, volatile fatty acids and so on should be needed. In this study, the methods to increase the sludge solubilization and acidification rate were compared with waste activated sludges and food waste leachate. Ultrasonication and acids were used for the pretreatment of organic particles in sludges. As a results, the optimal temperature and HRT were $60^{\circ}C$ and 5 days, respectively. HAc, HPr, HBr, and other VFAs for acid fermentations reduced up to 22, 16, 14, and 48% with HRT reduction. For the increase of solubilization, 28% of solids destruction rate was shown at 0.3 watts/mL.

Properties of Photoisomerization of Organic-Ultrathin Films (아조벤젠기를 갖는 장쇄지방산LB막의 광이성화 특성)

  • The absorption spectra of synthesis of azobenzene containing long chain fatty acids were investigated by UV spectrophotometer in chloroform solution. Also, the pressure-area isotherms of the molecules on the water-air interface were obtained and the LB films were fabricated onto a quartz slides and quartz crystals by the conventional Langmuir-Blodggett(LB) method. The UV absorption spectra of Langmuir-Blodggett(LB) films on quartz slides have been measured. From these measurements, following conclusions were obtained. Azobenzene containing long chain fatty acids show a photoisomerization by irradiation of UV light and visible light alternatively. At the pressure-area isotherms, the value of surface pressure increment were decreased when the number of $C_n$ increased. Also the LB films show a photoisomerization characteristics. So the LB film of azobenzene containing long chain fatty acid has possibility to being applied to functional molecular device such as photomemory and light switching.
