Chemical Composition and Nutritional Value of Algae Meal Produced from Dairy Cow Wastes as a Feedstuff (젖소폐기물에서 생산된 Algae Meal의 화학적조성 및 사료적가치)

  • Algal meal (cell) was produced from the solution of dairy cow wastes by fermentation of ulothrix. sp. and chlorella sp. Raw wastes mainly feces were diluted with ground water to give dry matter concentration of 0.5 w/v of wastes in 20 l amounts of ten plastic containers. Each containers were covered with plastic nets and vinyl films to protect from the insects and rain. Algea cells were harvested every 3 to 5 days and dried by sunlight and artifitial heat. Dried cells were ground by a feed meal, and analyzed and tested for the chemical composition of dry cell, in vitro DM and protein digestibility and the safty of algae. Protein contents in algae meals, ulothrix (29.37%) and chlorella (29.24%) were similar. However, chlorella contained lower Neutral detergent fiber (5.92%) than ulothrix(20,76%), and higher ash (32.86%) and calcium (12.62%) than ulothrix (28.66% and 6.09%) (P<.01). Ulothrix protein had higher for essential amino acids; valine, isoleucine and phenylalanine, than chlorella (P<.05). Algal fats contained high saturated fatty acids, C16:0 and C18:0, for ulothrix and high unsaturated fatty acids, C18:1 and C18:2, for chlorella (P<.01). In vitro digestibility of. ulothrix tended to be higher for DM, but lower for protein than chlorella. The weight gain and survival percentage were higher for pond fishes (loaches, Misgurnus sp. ) fed diet added chlorella meal than diets added ulothrix meal and control diet (P<.05).

Growth Performance of Lambs Fed Diet Supplemented with Rice Bran Oil as Such or as Calcium Soap

  • Forty two Malpura lambs (21 d old) were divided into three groups of 14 each consisting of 8 females and 6 males. Lambs were allowed to suckle their respective dams twice daily up to weaning (13 wks) and offered free choice concentrate and roughage in a cafeteria system. The lambs in control group were fed conventional concentrate mixture, in RBO group concentrate mixture fortified with 4% industrial grade rice bran oil and in Ca-soap rice bran oil (as in RBO group) was supplemented in the form of calcium soap. The concentrate intake decreased($p{\leq}0.05$) in RBO group as a result total dry matter, crude protein and metabolizable energy intake decreased compared to control whereas Ca-soap prepared from the same rice bran oil stimulated the concentrate intake leading to higher total dry matter, crude protein and energy intakes. The digestibility of dry matter ($p{\leq}0.05$), organic matter ($p{\leq}0.05$) and crude protein ($p{\leq}0.05$) was higher in RBO group followed by Ca-soap and control whereas no effect was observed for ether extract digestibility. Higher cholesterol ($p{\leq}0.05$) content was recorded in serum of oil supplemented groups (RBO and Ca-soap) while no effect was recorded for other blood parameters. Rice bran oil as such adversely affected and reduced the body weight gain ($p{\leq}0.001$) of lambs in comparison to control whereas the Ca-soap of rice bran oil improved body weight gain and feed conversion efficiency in lambs. Fat supplementation decreased total volatile fatty acids ($p{\leq}0.05$) and individual volatile fatty acid concentration which increased at 4 h post feeding. Fat supplementation also reduced ($p{\leq}0.05$) total protozoa count. Ca-soap of rice bran oil improved pre slaughter weight ($p{\leq}0.05$) and hot carcass weight ($p{\leq}0.05$). It is concluded from the study that rice bran oil in the form of calcium soap at 40 g/kg of concentrate improved growth, feed conversion efficiency and carcass quality as compared to rice bran oil as such and control groups.

  • Lipases are well known as the enzymes which catalyze the hydrolysis of ester bonds combining aliphatic chains and glycerol on mono-, di- and triglycerides. Their reactions are characterized by be-ing heterogeneous and catalyzing the water-insoluble substrates. This property has been one of the Hurdles which delayed the application of lipases in fats and oils industry, However, with the development of biological reaction system of which organic solvent is introduced in part or whole as the reaction media, enzymatic manipulation of fats and oils is attracting increasing attention from the academic and industrial sectors. Trials in two-phase system and reversed micellar system to produce fatty acids through enzymatic hydrolysis of triglycerides preyed to be efficient in respect to volumetric productivity, fat hydrolysis rate, product separation, etc. In organic solvent system lipases have been found to have the ability to catalyze aminolysis, transesterification, esterification, thiotransesterification and oximolysis that are virtually impossible to catalyze in water. The organic solvent system is being extensively used in interesterifying glycerides to produce a fat with the modified physical and chemical nature.

Characterization and Organic Hydrocarbons Degradation Potential of Euryhaline Marine Microorganism, Bacillus sp. EBW4 Isolated from Polychaete (Perinereis aibuhitensis) (갯지렁이(Perinereis aibuhitensis)에서 분리한 광염성 해양 미생물 Bacillus sp. EBW4의 특성 및 유기물 분해능 분석)

  • In this study, euryhaline marine microorganism, Bacillus sp. strain EBW4 isolated from polychaete (Perinereis aibuhitensis) of Suncheon Bay was physiologically, biochemically and genetically characterized. Based on 16S rRNA sequence, EBW14 was found to share 98.25% similarity with Bacillus hemicentroti $JSM076093^T$, 97.96% similarity with Bacillus hwajinponensis SW-$72^T$ and 96.28% similarity with B. algicoa $KMM3737^T$, respectively. The temperature range for the growth of strain EBW4 was $4-40^{\circ}C$, NaCl concentration range 0-17% and pH range pH 5-9, revealing that EBW4 was euryhaline bacterium. Major fatty acids in strain EBW4 were composed of anteiso $C_{15:0}$ (48.2%), iso $C_{16:0}$ (12.1%), anteiso $C_{17:0}$ (11.6%) and iso $C_{14:0}$ (9.4%). EBW4 was found to have DNase, amylase, protease and lipase for the degradation of macromolecules such as DNA, carbohydrates, proteins, lipids, etc. The enzyme activities of alkaline phosphatase, esterase (C4), leucine arylamidase and ${\alpha}$-chymotrypsin were also found in strain EBW4. Analysis of the biodegradation ability of EBW4 for organic hydrocarbons under different salinity conditions using synthetic water waste revealed that EBW4 exhibited the ability to degrade organic hydrocarbons very quickly, suggesting strain EBW4 may be a good candidate for the application to various industries.

  • Among various techniques for hydrogen production from organic wastewater, a dark fermentation is considered to be the most feasible process due to the rapid hydrogen production rate. However, the main drawback of it is the low hydrogen production yield due to intermediate products such as organic acids. To improve the hydrogen production yield, a co-culture system of dark and photo fermentation bacteria was applied to this research. The maximum specific growth rate of R. sphaeroides was determined to be $2.93h^{-1}$ when acetic acid was used as a carbon source. It was quite high compared to that of using a mixture of volatile fatty acids (VFAs). Acetic acid was the most attractive to the cell growth of R. sphaeroides, however, not less efficient in the hydrogen production. In the co-culture system with glucose, hydrogen could be steadily produced without any lag-phase. There were distinguishable inflection points in the accumulation of hydrogen production graph that resulted from the dynamic production of VFAs or consumption of it by the interaction between the dark and photo fermentation bacteria. Lastly, the hydrogen production rate of a repeated fed-batch run was $15.9mL-H_2/L/h$, which was achievable in the sustainable hydrogen production.

Physiochemical and Organoleptic Properties of Feta Cheese Made from Goat Milk (산양유로 제조한 Feta 치즈의 이화학적 및 관능적 특성)

  • We characterized physicochemical properties and examined the organoleptic and textural evaluations of Feta cheese made from goat's milk. Nutritional compositions of goat Feta cheese were fat 23.50%, protein 11.03% with moisture content of 59.54%. Cell numbers of lactic starter cultures in Feta cheese maintained from log 8.46 CFU/g and pH 5.76 during storage at 4℃ for 14 day's aging. The color of Feta cheese was whitish (L. 93.19) at after finishing brine salting, but became a little yellowish(b. 3.52) (a. -0.71). For texture profile analysis of goat Feta cheese, hardness, fracturability springness, and cohesiveness seemed to be week, but adhesiveness gumminess, chewiness, and resilience were enhanced as aging times extended to 14days, resulted in the overall textural properties was to be superior to control cheese(commercial Mozzarella cheese). Organoleptic evaluations were examined based on the intensities and the preferences for flavour, tastes, texture and mouth feeling. saltiness, bitterness and acidity were stronger in the intensities than control cheese, but the preferences were enhanced by aging to be better than control cheese at 14 days and later on, however, the texture changed to be weaker in hardness and unpleasant in mouthfeel. The fatty acid compositions of Feta cheese analysed by Gas chromatography were saturated fatty acid 42.06%, monoenoic acids 29.67%, di-enoic acids 24.24%, tri-enoic acids 1.21%.

Effects of Dietary Organic Sulfur on Performance, Egg Quality and Cell-mediated Immune Response of Laying Hens (유기황의 수준별 급여가 산란계의 생산성, 계란품질 및 세포성 면역능에 미치는 영향)

  • This study aimed to evaluate the dietary effect of organic sulfur (OS) supplementation on performance, egg quality and serum constituents in laying hens. A total of 360 Lohmann brown laying hens at the age of 31 weeks were distributed into four treatments having five replicates of 18 hens each until 54 weeks. The hens were fed four levels (0.0, 0.1, 0.2 and 0.4%) of OS with basal diet. The number of eggs was investigated daily, and egg quality was confirmed every 8 weeks. Sulfur content in eggs, interleukin 2 (IL-2), T help cells (CD4+) and cytotoxicity cells (CD8+) were measured at the termination of the experiment. The result of the study showed that egg production tended to increase with 0.4% OS in diet after 39 weeks of age and, there was a significant effect (P<0.05) from 47 to 54 weeks of age. Egg quality traits of albumen height and haugh unit increased significantly (P<0.05) owing to the addition of OS to the diet. The polyunsaturated fatty acids in yolk were gradually increased while saturated fatty acids were decreased with increasing levels in OS (P<0.05). Total sulfur concentration in the eggs increased significantly (P<0.05) in treatments fed OS. Moreover, albumin, AST and HDL cholesterol levels in serum improved significantly (P<0.05) owing to the addition of OS. The IL-2 concentration and the ratio of CD4+ and CD8+ in blood were generally higher (P<0.05) at 0.4% OS. Therefore, it can be recommended that supplementary OS diet affected the performance, egg quality and stimulated immune response in laying hens.

Studies on the Processing and Utilization of Seaweeds - Studies on the Processing of Sea Mustard Jam- (해조류의 가공 및 이용에 관한 연구 -미역쨈의 제조에 관한 연구-)

  • In order to utilize sea mustard, Undaria pinnatifida, effectively. sea mustard jam was prepared by use of physical properties of polysaccharides, mainly alginic acid and then its chemical composition, nutritive qualifies and the stability of pigments were examined. Suitable processing condition for sea mustard jam was as fellows : as the first stage, fresh sea mustard was scalded for 20 sec at $85^{\circ}C$ and dried sea mustard was dipped for 20 min in cold water for rehydration, then both sea mustards were treated by draining and chopping. Next, after adding 4 fold of $0.5\%\;K_2HPO_4$ solution to weight of chopped sea mustards, the mixed solutions were agitated for 15 min at $95^{\circ}C$, and gelated sea mustards were filterated by pressing. Flow characteristics of those sea mustard jam were regarded mixed type having pseudoplastic type and yield stress. Judging from sensory evaluation, adding $0.375\%$ of saccharin, $2\%$ of sorbitol, $0.25\%$ of citric acid and $0.5\%$ of powder of roasted soybean to sea mustard jam were suitable for enhancing taste and flavor of product. Chemical composition of products were scarcely changed throughtout processing. Jam processed by fresh sea mustard was better than dried one in contents of chlorophyll and total carotenoid pigments. In fatty acid composition, polyenoic acids of $C_{18:3},\;C_{20:4},\;C_{20:5},\;C_{22:6}$ were held high contents as known to have lowering function of cholesterol contents. It was presumed that dominant contents in free amino acids such as lysine, alanine, glutamic acid and organic acids such as citric acid, oxalic acid, $\alpha-ketoglutaric$ acid, lactic acid and succinic acid held important role for the flavor of sea mustard jam.

Properties of Cheongkukjang Prepared with Admixed Medicinal Herb Powder (생약초(표고버섯, 더덕, 어성초)를 첨가한 청국장의 특성)

  • Cheongkukjang was prepared by addition of medicinal herb powder (from Lentinus edodes, Codonopsis lanceolata BENTH et HOOK, or Houttuynia cordata THUNB) to improve Cheongkukjang quality and functional properties. Redness and yellowness (measured using Hunter’s color values) increased with increasing amounts of medicinal herb powder, whereas lightness (the third Hunter's color value) and pH decreased. Crude protein and lipid levels of Cheongkukjang decreased with increasing amounts of added medicinal herb powder, whereas powder addition did not affect either the moisture or crude ash content of Cheongkukjang. The major organic acids of Cheongkukjang were lactic acid, acetic acid, succinic acid, and citric acid, and acid levels increased as the proportion of medicinal herb powder in the Cheongkukjang preparation increased. The major amino acids of Cheongkukjang were glutamic acid, aspartic acid, leucine, arginine, and lysine and the principal fatty acids were linoleic acid, oleic acid, palmitic acid, and linolenic acid. Addition of medicinal herb powder to Cheongkukjang increased both crude saponin and quercetin contents. Sensory scores of Cheongkukjang containing 2% (w/v) medicinal herb powder were optimal in terms of both quality characteristics and sensory evaluation.

Effect of Transplanting Times on the Content of Aromatic Substances in Tobacco Cultivar, Hyangcho (이식시기(移植時期)가 향초(香草)(Nicotiana tabacum L.)의 향끽미(香喫味) 물질(物質) 발현(發現)에 미치는 영향(影響))

  • This study was conducted to establish the most appropriate transplanting time which can attribute to increase of aromatic substances in tobacco variety, Hyangcho, maintaining its original aroma. The highest content of total alkaloid, total nicotine and total nitrogenous compounds were observed in the late transplanting time such as July 5, showing 4.16%, 3.83% and 3.68%, respectively. However, the petroleum ether extract was highest in the early transplanted one such as April 5, showing 5.77% and total sugar content, 12% in April 20. The content of petroleum ether extract decreased as the transplanting time delayed. The early transplanting of Hyangcho on April 20 under vinyl mulching showed the increase of fatty acid content, but the decrease of non-volatile organic acids such as malic and citric acids than those of the conventional transplanting time on March 5. The earlier transplanting time also increased the content of volatile acids such as 2,3-methylbutanoic acid and 3-methyl-pentanoic acid, which seem to be related to tobacco aroma.
