Effect of production level and source of fat supplement on performance, nutrient digestibility and blood parameters of heat-stressed Holstein cows

  • The interactive effect of dietary fat supplementation and milk yield level on dairy cows performance under heat stress has not been thoroughly investigated. The purpose of this study was to evaluate the effect of production level, the source of fat supplements and their interaction on dairy cows performance under heat stress. In this study, 64 Holstein multiparous cows were divided into 2 groups and received one of two rations having either calcium salts of fatty acids (Ca-FA) or high-palmitic acid (PA) supplements (2.8% of DM; dry matter). After completing the experiment and based on maturity-equivalent milk, cows were divided into two groups of high-yielding (14,633 kg) and medium-yielding (11,616 kg). Average temperature humidity index (THI) was 71 during the trial period. Apparent digestibility of dry matter (p = 0.04), organic matter (p = 0.05), and neutral detergent fiber (NDF; p = 0.04) for cows fed Ca-FA were greater than cows fed PA. The milk fat content in high-producing cows was 0.3% greater than medium-producing cows (p = 0.03). The milk protein content in cows fed Ca-FA was greater than cows fed PA (p < 0.01). High-producing cows had greater serum cholesterol (p = 0.02) than medium-producing cows. The cows fed PA tended to have a greater BUN than cows fed Ca-FA (p = 0.06). Alanine aminotransferase and aspartate aminotransferase tended to be increased by PA, which indicates that cows in PA treatment may have experienced more adverse effect on the liver function than cows on Ca-FA. Therefore, under heat stress and in 90 d trial, milk production level does not affect the cows' response to PA or Ca-FA. Although cows fed Ca-FA received lower energy than those fed PA, they compensated for this shortage likely with increasing the digestibility and produced a similar amount of milk.

Effects of Salmonella typhymurium Lipopolysaccharide Challenge on the Performance, Immune Responses and Zinc Metabolism of Laying Hens Supplemented with Two Zinc Sources

  • The study was conducted to determine the effect of Salmonella typhymurium lipopolysaccharide (LPS) challenge on egg-laying performance, inflammatory response, zinc metabolism in layer fed diets supplemented with organic or inorganic zinc since 3-wk-old. The three dietary treatments were corn-soybean meal basal diet without supplemental zinc or with supplemental zinc at 60 mg/kg zinc from $ZnSO_4$ or zinc amino acid complex (ZnAA). At the age of 58 wk-old, twelve hens from each dietary treatment were allotted into two sub-groups. On day 1, 3, 5, 7 of the $58^{th}$ week of age, six birds of one sub-group were injected intraperitoneally (i.p.) with 2 ml LPS (1.0 $\ell$/ml) or sterile saline. Neither zinc source ${\times}$ immune challenge interaction nor zinc source effect on egg production performance was observed (p>0.05), LPS-challenge decreased egg production (p<0.04) and increased percentage of cracked eggs (p <0.01). With LPS challenged, the fever response of hens fed ZnAA peaked and subsided earlier than in hens fed $ZnSO_4$ or basal diet. Serum IL-1$\beta$ at 3 h was higher (p<0.01), but lower (p<0.001) at 12 h post-challenge with LPS in hens fed ZnAA than $ZnSO_4$. In salinetreated groups, serum IL 1$\beta$ was higher in hens fed ZnAA than the basal diet at 3 h post-injection (p<0.01). LPS-challenged birds had lower serum zinc and higher zinc sequestered in liver and spleen (p<0.001). In saline-treated birds, there was no difference in zinc concentration of serum, liver and spleen among different dietary treatments (p>0.05). Supplementation of 60 mg/kg zinc from either ZnAA or $ZnSO_4$ significantly (p<0.05) elevated metallothionein (MT) concentration in liver and spleen. MT concentration in liver of birds fed ZnAA diet was higher than in those fed $ZnSO_4$ diet (p<0.05). The magnitude of increase of hepatic and splenic MT due to LPS challenge was higher by supplementation of ZnAA than $ZnSO_4$. The results suggest that zinc amino acid complex enhanceed MT synthesis and zinc sequestered in liver and spleen and increased the sensitivity to immune response due to LPS challenge.

Effect of feeding of blend of essential oils on methane production, growth, and nutrient utilization in growing buffaloes

  • Objective: An experiment was conducted to study the effect of a blend of essential oils (BEO) on enteric methane emission and growth performance of buffaloes (Bubalus bubalis). Methods: Twenty one growing male buffaloes (average body weight of $279{\pm}9.3kg$) were divided in to three groups. The animals of all the three groups were fed on a ration consisting of wheat straw and concentrate mixture targeting 500 g daily live weight gain. The three dietary groups were; Group 1, control without additive; Group 2 and 3, supplemented with BEO at 0.15 and 0.30 mL/kg of dry matter intake (DMI), respectively. Results: During six months feeding trial, the intake and digestibility of dry matter and nutrients (organic matter, crude protein, ether extract, neutral detergent fibre, and acid detergent fibre) were similar in all the groups. The average body weight gain was tended to improve (p = 0.084) in Group 2 and Group 3 as compared to control animals. Feeding of BEO did not affect feed conversion efficiency of the animals. The calves of all the three groups were in positive nitrogen balance with no difference in nitrogen metabolism. During respiration chamber studies the methane production (L/kg DMI and L/kg digestible dry matter intake was significantly (p<0.001) lower in Group 2 and Group 3 as compared to control animals. Conclusion: The results indicated that the BEO tested in the present study have shown potential to reduce enteric methane production without compromising the nutrient utilization and animal performance and could be further explored for its use as feed additive to mitigate enteric methane production in livestock.

Effects of compound organic acid calcium on growth performance, hepatic antioxidation and intestinal barrier of male broilers under heat stress

  • Objective: The aim of this study was to evaluate the effects of compound organic acid calcium (COAC) on growth performance, hepatic antioxidant status and intestinal barrier of male broilers under high ambient temperature (32.7℃). Methods: Nine hundred healthy one-d-old Cobb-500 male broiler chicks were randomly assigned into three groups with six replicates of 50 birds each. A basal diet supplemented with 0% (control), 0.4% and 0.8% COAC, respectively were fed to birds for 6 weeks. All treatments were under high ambient indoor temperature of 32.7℃, and had a constant calcium and available phosphorus ratio. Results: The results showed that, compared with control, the average daily gain of broilers in 0.4% and 0.8% was significantly increased and the ratio of feed to gain in in 0.4% and 0.8% was significantly decreased at 1 to 21, 22 to 42 and 1 to 42 days of age (p<0.05). Compared with control, 0.8% COAC slightly decreased (p = 0.093) the content of malondialdehyde in liver at 42 days of age while 0.4% COAC significantly decreased (p<0.05) the activity of alkaline phosphatase. Furthermore, 0.4% COAC significantly enhanced the intestinal barrier function via increasing jejunal and ileal ocln transcription, promoting jejunal mucin 2 transcription at 42 days of age (p<0.05), and decreasing jejunal toll-like receptor 2 (TLR-2) and ileal TLR-15, inducible nitric oxide synthase compared with control group (p<0.05). Whereas, no significant differences on the transcription of interleukin-1β in jejunum and ileum were observed among three treatments (p>0.05). Overall, heat stress caused by high natural environment temperature may induce the damage to hepatic antioxidation and intestinal barrier. Conclusion: Dietary inclusion of COAC can improve the tolerance of broilers to thermal environment through the modification of antioxidative parameters in liver and the mRNA expression of genes in intestinal barrier, resulting in an optimal inclusion level of 0.4%.

Effect of Microwave Treatment on Chemical Composition and In sacco Digestibility of Wheat Straw in Yak Cow

  • Wheat straw was treated with microwave for 4 min and 8 min at a power of 750 W and frequency of 2,450 MHz. Chemical compositions of untreated, 4 min treated and 8 min treated straws were analyzed and in sacco degradabilities of all these straws in yak rumens were measured. Microwave treatment didn't significantly (p>0.05) affect the chemical composition of the straw. In sacco dry matter (DM) degradability of the straw after 18 h incubation in rumen was significantly (p<0.01) improved by microwave treatment. In sacco crude protein (CP) degradability of the straw was not (p>0.05) affected by microwave treatment. In sacco organic matter (OM) degradability of the straw was increased (p<0.01) by around 20% for both the 4 min and 8 min microwave treatment, that of acid detergent fibre (ADF) was increased (p<0.01) by 61.6% and 62.8%, and that of ash free ADF was enhanced by 72.1% and 69.6% for the 4 min and 8 min microwave treatment respectively. No significant difference was observed between the 4 min and 8 min microwave treatment on the degradability of DM, OM, CP, ADF and ash-free ADF of the straw.

Effect of Nitrogen Fertilization and Stage of Maturity of Mottgrass (Pennisetum purpureum) on its Chemical Composition, Dry Matter Intake, Ruminal Characteristics and Digestibility in Buffalo Bulls

  • Four ruminally cannulated buffalo bulls were fed mottgrass diets using a $4{\times}4$ Latin square design. Treatments were arranged factorially and consisted of mottgrass fertilized with 0 for Kg nitrogen (N) per acre and harvested at 40 and 60 days of age. Nitrogen fertilization improved the concentrations of neutral detergent fiber (NDF), and acid detergent fiber (ADF) in early-cut mottgrass (ECM) but, the acid detergent lignin (ADL) contents were higher in the late-cut mottgrass (LCM). The crude protein (CP) contents of the mottgrass decreased with advancing maturity, but N fertilization increased CP at both maturities. The intake of dry matter (DM), organic matter (OM), CP, NDF and ADF were higher by buffalo bulls fed ECM than those fed LCM. The ruminal pH increased in first 6 hours post feeding in animals fed N fertilized mottgrass and may be due to higher concentration of ruminal ammonia. The digestibilities of DM, OM, CP, NDF and ADF were higher by buffalo bulls fed ECM than those fed LCM. However, the application of N fertilizer did not affect the digestibilities of these nutrients.

Comparative analysis of silage fermentation and in vitro digestibility of tropical grass prepared with Acremonium and Tricoderma species producing cellulases

  • Objective: To find out ways of improving fermentation quality of silage, the comparative analysis of fermentation characteristics and in vitro digestibility of tropical grasses silage applied with cellulases produced from Acremonium or Tricoderma species were studied in Thailand. Methods: Fresh and wilted Guinea grass and Napier grass silages were prepared with cellulases from Acremonium (AC) or Trichoderma (TC) at 0.0025%, 0.005%, and 0.01% on a fresh matter (FM), and their fermentation quality, chemical composition and in vitro digestibility were analyzed. Results: All silages of fresh Napier grass were good quality with lower pH, butyric acid, and ammonia nitrogen, but higher lactic acid content than wilted Napier grass and Guinea grass silage. Silages treated with AC 0.01% had the best result in terms of fermentation quality. They also had higher in vitro dry matter digestibility and in vitro organic matter digestibility at 6 and 48 h after incubation than other silages. Silages treated with lower levels at 0.005% or 0.0025% of AC and all levels of TC did not improve silage fermentation. Conclusion: The AC could improve silage fermentation and in vitro degradation of Guinea grass and Napier grass silages, and the suitable addition ration is 0.01% (73.5 U) of FM for tropical silage preparation.

BNR 효율개선을 위한 산 발효조 최적운전 조건에 관한 연구 (A Study on the Optimum Operating Condition of Acid Fermenter for the BNR Performance Improvement)

  • 국내 하수처리장에 적용되는 BNR(Biological Nutrients Removal) 공정의 효율저하 원인은 유입원수의 낮은 C/N비와 탄소 (Carbon source) 부족이 가장 큰 것으로 지적되어 왔다. 탄소원의 효율적인 공급을 위해 1차 슬러지 산 발효조(Acid fermenter)를 이용하여 SCFAs(Short-Chain Fatty Acids) 생성을 유도할 수 있다. 본 연구에서는 side-stream의 산 발효조와 조합된 $A_2/O$ 프로세스를 운전하여 영양염류 제거에 미치는 영향을 연구하였다. 그 결과 산 발효조의 최대 SCFAs생성은 중온상의 SRT 4~5일에서 약 3.000mg/L(as COD), 대략 0.10~0.16 mg SCFA(as COD)/mg 1차 슬러지(as COD)로 나타났다. 또한 생성된 SCFAs의 성상비는 Acetic, Propionic, (iso)Butyric 및 (iso)Valeric이 1, 0.7, 0.5 그리고 0.6 순으로 나타났다. BNR공정의 side-stream 산 발효조 적용에 의한 영양염류 제거효율은 크게 증가하는 것으로 나타났다. T-N은 인공하수(R1)의 경우 35% 정도의 낮은 처리효율을 나타냈으나 인공 SCFA 주입(R2) 및 발효산물 투입(R3)의 경우 제거효율 74%, 76%로 크게 향상되는 것으로 나타났다. 또한 Phosphate 는 R1의 경우 45% 의 처리효율을 나타냈으나 R2, R3의 경우 73~74% 정도로 양호한 처리효율을 나타냈다. 이는 발효산물 및 SCFA 주입에 의해 혐기조에서 인의 방출 및 호기조에서 과잉섭취가 잘 이루어졌기 때문으로 R3의 경우 인의 방출율은 $0.34g\;PO_4-P/g$ COD로 기존 연구와 비교해서 유사한 것으로 나타났다.

찹쌀의 종류와 전처리를 달리한 약주의 특성 및 휘발성 향기 성분 (Fermentation Characteristics and Volatile Compounds in Yakju Made with Various brewing Conditions; Glutinous Rice and Pre-treatment)

  • 한국의 전통 알코올 음료인 약주를 찹쌀가루, 찹쌀 전분 그리고 누룩을 사용하여 제조하였다. 본 연구에서는 찹쌀의 종류와 전처리의 차이에 따른 발효 특성을 조사하였다. 효모는 Y90-9를 사용하였고 발효제로는 시판되는 개량누룩(SP1800)을 사용하였으며 덧술에 엿기름을 첨가하여 저온에서 발효하였다. 발효기간 중 pH와 당도는 찹쌀가루를 이용한 약주가 찹쌀전분을 이용한 경우보다 높았으며 전처리로 호화를 거친 약주가 높게 나타났고 발효초기 0.3 이상이었던 산도는 발효 종료 후 전처리에 관계없이 비슷한 수준을 나타내였으며, 알코올 함량은 전처리로 호화를 거친 찹쌀가루 약주에서 가장 높게 나타났다. 아미노산 함량은 찹쌀가루를 사용한 경우가 더 높게 나타났고 자외부 흡수와 착색도 또한 찹쌀 가루를 사용한 약주도 같은 결과를 나타내었다. 환원당 함량은 3.5-3.9 mg/mL의 수준이였다. 술에서 맛과 산성도 조정 역할을 하는 유리당의 조성은 glucose > maltose > fructose 순으로 나타났으며 유기산은 검출된 6개 중 succinic acid가 가장 높게 나타났다. 검출된 21개의 휘발성 향기 성분 중 alcohol은 3 종, ester는 17종, 그리고 aldehyde는 1종이 동정되었으며 그 중 부드러운 향을 나타내는 ethyl palmitate가 높게 나타났다. 이상의 결과들로 미루어 호화한 찹쌀가루와 개량누룩을 사용하여 제조한 약주가 알코올의 수율이 높고 풍부한 향미를 갖는 것으로 나타났다.

The Effect of Different Sources of Urease Enzyme on the Nutritive Value of Wheat Straw Treated with Urea as a Source of Ammonia

  • Wheat straw samples (3-4 cm) were sprayed with solutions of urea (U) alone or with a dry addition of garden soil (GS), midden soil (MS), soya bean meal (SM) or jack bean meal (JM) as crude urease sources and with a pure urease (UR) enzyme. Each of the urease sources was included at two levels: 30 and 60 g/kg except pure urease, which was added at a level of 2.5 & 5.0 g/kg treated straw dry matter. Untreated straw without urease source was used as a control. After treatment, samples were sealed in polythene bags and stored for 2, 7, 14, 21 and 35 days at $19{^{\circ}C}$. The urease sources, their levels and treatment time produced significant effects on ammonia production (p<0.01). The addition of urease offered more flexibility in hydrolyzing urea in the shortest possible time. Incorporation of soya bean and jack bean meal was effective in reducing the modified acid detergent fiber (MADF) content of straw and the same time increasing organic matter (OM) digestibility. Overall effect, addition of soya bean to urea at a ratio of 1:1 appeared to be the most satisfactory urease source for the treatment of urea and wheat straw.
