Production of Phenyl Lactic Acid (PLA) by Lactic Acid Bacteria and its Antifungal Effect

  • Phenyllactic acid (PLA) which is known as antimicrobial compound can be synthesized through the reduction of phenylpyruvic acid (PPA) by lactate dehydrogenase (LDH) of lactic acid bacteria (LAB). LAB producing PLA was isolated from Korea Kimchi and identified to Lactobacillus plantarum SJ21 by 16 rRNA gene sequence analysis. Cell-free supernatant (CFS) from L. plantarum SJ21 was assessed for both the capability to produce the antimicrobial compound PLA and the antifungal activity against four fungal pathogens (Rhizoctonia solani, Aspergillus oryzae, Botrytis cinerea, and Collectotricum aculatum). PLA concentration was investigated to be 3.23mM in CFS when L. plantarum SJ21 was grown in MRS broth containing 5mM PPA for 16 h. PLA production also could be promoted by the supplement of PPA and phenylalanine in MRS broth, but inhibited by the supplement of 4-hydroxyphenylpyruvic acid and tyrosine as precursors. Antifungal activity demonstrated that all fungal pathogens were sensitive to 5% CFS (v/v) of L. plantarum SJ21 with average growth inhibitions ranging from 27.32% to 69.05% (p<0.005), in which R. solani was the most sensitive to 69.05% and followed by B. cinerea, C. aculatum, and A. oryzae. The minimum inhibitory concentration (MIC) for commercial PLA was also investigated to show the same trend in the range from $0.35mg\;mL^{-1}$ (2.11 mM) to $0.7mg\;mL^{-1}$ (4.21 mM) at pH 4.0. The inhibition ability of CFS against the pathogens was not affected by heating or protease treatment. However, pH modification in CFS to 6.5 caused an extreme reduction in their antifungal activity. These results may indicate that antifungal activities in CFS were caused by acidic compounds like PLA or organic acids rather than proteins or peptides molecules.

배양인삼 요구르트의 제조 및 품질 특성 (Preparation and Quality Characteristics of Yogurt Added with Cultured Ginseng)

  • 배양인삼 추출물을 첨가한 요구르트를 제조한 다음 그 품질 특성을 살펴본 결과, 배양인삼의 첨가로 요구르트의 젖산균 증식과 산 생성이 대조군보다 촉진되었고, 발효 24시간 후 생균수는 모든 군에서 $6.0{\sim}8.0×10^8\;CFU/mL$으로 발효시간이 경과할수록 점점 증가하였다. 점도는 대조군에 비하여 증가하였고, 특히 12시간에서 18시간 사이에 점도가 가장 높은 증가를 나타내었다. 총 아미노산 함량의 경우 대조군과 배양인삼 첨가군 모두 17여종의 아미노산이 존재하였으며, 0.5% 첨가군은 1.0% 첨가군에 비해 배양시간이 관계없이 4종의 아미노산을 제외한 대부분의 아미노산 함량이 많이 존재하였다. 유기산 함량은 대조군에 비해 0.5% 첨가군의 경우 24시간 발효시 젖산, 구연산, 개미산, 프로피온산의 생성이 증가되다가, 30시간 이후에는 이들 함량이 다소 감소되었다. 1.0% 첨가군의 경우는 배양시간이 증가할수록 개미산, 젖산, 초산의 함량이 증가되었다. 관능검사에서 배양인삼 0.5%와 1.0%의 첨가 요구르트가 전체적인 기호도에서 우수하였고 다른 항목들에서도 비교적 양호한 값을 보였다. 이와 같은 결과로 보아 $0.5{\sim}1.0%$의 배양인삼을 첨가하여 24시간 발효가 배양인삼 요구르트 제조에 가장 적합한 것으로 생각된다.

Effects of Sown Season and Maturity Stage on In vitro Fermentation and In sacco Degradation Characteristics of New Variety Maize Stover

  • The effects of seedtime and maturity stage on nutritive value of five maize stover varieties, including conventional maize (Kexiangyu 11, CM), fodder maize (Huqing 1, FM), high oil maize (Gaoyou 115, HOM), sweet maize (Kexiangtianyu 1, SM) and waxy maize (Kexiangluoyu 1, WM), were examined based on chemical composition, in vitro gas production and in situ incubation techniques. Maize stover was sampled at d 17 and d 30 after tasseling, and designated as maturity stage 1 and stage 2, respectively. The average dry matter (DM) organic matter (OM), crude protein (CP) and fiber contents were the greatest for HOM, SM and FM, respectively. CM had the highest in vitro organic matter disappearance (IVOMD) and volatile fatty acid (VFA) concentration. The highest ammonia nitrogen ($NH_3$-N) concentration in the incubation solution, and effective degradability of DM ($ED_{DM}$) and neutral detergent fiber ($ED_{NDF}$) were observed in SM. Advanced maturity stage increased (p<0.05) DM content, $ED_{DM}$ and $ED_{NDF}$, but decreased (p<0.05) OM and CP contents, and decreased (p<0.05) b and a+b values, IVOMD and molar proportion of valerate in the incubation solution for maize stover. Maize sown in summer had greater (p<0.05) OM content, but lower DM, CP, neutral detergent fiber (NDF) and acid detergent fiber (ADF) content compared with maize sown in spring. Maize sown in summer had greater (p<0.001) IVOMD, $NH_3$-N concentration in the incubation solution and $ED_{NDF}$, but lower (p<0.01) ratio of acetate to propionate compared to maize sown in spring. The interaction effect of variety${\times}$seedtime was observed running through almost all chemical composition, in vitro gas production parameters and in situ DM and NDF degradability. The overall results suggested that SM had the highest nutrient quality, and also indicated the possibility of selecting maize variety and seedtime for the utilization of maize stover in ruminants.

바이오가스와 균체단백질 생산을 위한 유기질 폐기물의 미생물 전환 연구 (Microbial Conversion of Organic Wastes for Production of Biogas and Algal Biomass)

  • 유기칠 폐기물을 보다 효율적으로 처리하고자, 상 분리 혐기발효와 Chiorella 배양에 의한 생우분의 4 단계-복합처리를 시도하였다. 3배 희석 생우분을 6 일간 산발효시킨 발효액을 4 일의 수리학적 체류시 간으로 1차 에탄발효조에 공급했을 때, 평균 977ml/l C비ture/day의 바이오가스가 생산되었고, 1차 발효 폐액을 역시 4일의 체류시간으로 2차 메탄발효조에 공급했을 때 평균 428ml/l culture/day의 바이오 가스가 생산되었다. 1차 메탄발효의 바이오가스 생 산성은 재래의 단일조식과 비교했을 때 31.4% 증가 되었다. 또한 산발효과정, 1차 및 2차 에탄발효과정 의 유기물 제거율은 각기 71.8%, 42.6% 그리고 2 24.0%였다. 한편, 2차 메탄발효폐액은 Chiarella s sp. PSH3에 의해 10일의 체류시간으로 반연속배양 처리한 결과, 평균 1.8g/l culture/day(2.8$\times$106 cells/ml)의 조체가 생산되었고, 이 과정의 유기물 제거율은 73%였다. 이상의 4단계 처리과정을 통하 여 초발원료인 3배 희석 생우분의 COD가 51, 300ppm에서 최종단계인 Chiarella의 처리에 의해 평균 85ppm으로 감소되어 총유기물 제거율이 99.8 %에 달하였다. 이들의 결과는 유기질 폐기물의 효 율적 처리와 더불어 바이오가스와 균체단백질을 생산하는 복합처리계의 구성이 가능함을 나타낸다.

항균활성 천연물질을 이용한 반추위 메탄저감용 친환경 첨가제 개발 (Development of an Environmental Friend Additive Using Antibacterial Natural Product for Reducing Enteric Rumen Methane Emission)

  • 본 연구는 천연물질에서 유래한 반추위 메탄저감용 친환경 첨가제 개발을 위해 각기 다른 종균을 이용하여 발효한 세신 추출물의 항균활성, 항산화활성 및 in vitro 반추위 발효시험을 체계적으로 실시하였다. 접종된 균주들의 성장효율을 알아보기 위해 실시한 생균수측정 결과 L. curvatus NJ40 균주에서 유의적으로 높은 균주성장을 나타냈다(p<0.05). 항균활성측정 결과는 대조구 대비 L. curvatus NJ40 및 L. plantarum NJ45 균주와 발효된 세신 추출물이 병원균에 대한 항균효과를 나타내는 것으로 조사되었다. In vitro 반추위 발효실험에 세신 및 발효 세신 추출물을 적용한 결과, 휘발성 지방산 생성량 대비 반추위 메탄 저감효과가 나타났다. 특히 반추위 미생물 활력 및 사료이용 효율을 대표할 수 있는 휘발성 지방산 생성효율에 대한 부정적 효과 없이, 오히려 휘발성지방산 생성효율을 향상시키면서 반추위 메탄 저감효과를 가져올 수 있는 것으로 나타났다.

익모초 에탄올 추출물의 항산화 및 항염증 활성 (Anti-oxidant and Anti-inflammatory Effects of Ethanol Extracts from Leonurus japonicus Houtt. on LPS-induced RAW 264.7 Cells)

  • 본 실험에서는 익모초 지상부 에탄올 추출물의 항산화 및 항염증 효과를 확인하여 건강 기능성 소재로서의 가능성을 평가하고자 하였다. DPPH 및 ABTS radical 소거 활성, 환원력, 총 폴리페놀 및 플라보노이드 함량을 통해 익모초의 항산화능을 측정한 결과, 400 ㎍/mL, 1500 ㎍/mL의 농도에서 57.8%, 62.3%의 DPPH 및 ABTS radical 소거활성을 보였고 환원력 또한 농도 의존적으로 증가하는 경향을 보였다. 총 폴리페놀 함량 및 플라보노이드 함량은 1 mg/mL의 농도에서 각각 51.40 ± 0.47 mg of gallic acid equivalents/g, 73.28 ± 0.10 mg of rutin equivalents/g로 나타났으며, 익모초 추출물은 세포 내 ROS의 생성 억제에 있어서 유의적인 효과를 보였다. 익모초의 항염증 효과를 측정한 결과, LPS로 자극해 활성화된 RAW 264.7 cell에서 익모초 추출물(0~400 ㎍/mL)의 세포 독성은 없었으며 LPS 처리로 유도된 세포의 형태학적 변화도 농도의존적으로 완화되는 경향을 보였다. NO 발생량은 LPS 처리군과 비교해 익모초 추출물 처리 시 농도 의존적으로 감소하였고, 400 ㎍/mL에서는 90.3%로 NO의 생성이 억제되었다. 염증성 cytokine (TNF-α, IL1-β)의 생성도 유의적으로 감소하였고 NO를 생성하는 염증성 단백질 iNOS의 발현 또한 억제되었으며 이와 같은 염증성 단백질의 전사를 조절하는 NF-κB (NF-κB, IκB-α) 및 MAPK (ERK, p38)의 인산화 및 활성화 또한 익모초 추출물 처리로 인해 억제됨을 확인하였다. 따라서 익모초 추출물은 NF-κB signaling pathway 및 ERK/p38 MAPK cascade pathway의 조절을 통해 염증성 단백질 및 염증인자의 발현을 감소시킨다고 볼 수 있다. 이러한 결과를 토대로 익모초 지상부 에탄올 추출물은 천연 기능성 소재로서 활용될 가능성이 있으며, 본 연구 결과는 익모초의 고부가가치 향상을 위한 기초자료로 도움이 될 것으로 생각된다.

Oil supplementation improved growth and diet digestibility in goats and sheep fed fattening diet

  • Objective: This study evaluated the growth, digestibility and rumen fermentation between goats and sheep fed a fattening diet fortified with linseed oil. Methods: Twelve 3 to 4 months old male goats and sheep were randomly allocated into two dietary treatment groups in a $2(species){\times}2$ (oil levels) factorial experiment. The treatments were: i) goats fed basal diet, ii) goats fed oil-supplemented diet, iii) sheep fed basal diet, and iv) sheep fed oil-supplemented diet. Each treatment group consisted of six animals. Animals in the basal diet group were fed with 30% alfalfa hay and 70% concentrates at a rate equivalent to 4% of their body weight. For the oil treatment group, linseed oil was added at 4% level (w:w) to the concentrate portion of the basal diet. Growth performance of the animals was determined fortnightly. Digestibility study was conducted during the final week of the feeding trial before the animals were slaughtered to obtain rumen fluid for rumen fermentation characteristics study. Results: Sheep had higher (p<0.01) average daily weight gain (ADG) and better feed conversion ratio (FCR) than goats. Oil supplementation did not affect rumen fermentation in both species and improved ADG by about 29% and FCR by about 18% in both goats and sheep. The above enhancement is consistent with the higher dry matter and energy digestibility (p<0.05), as well as organic matter and neutral detergent fiber digestibility (p<0.01) in animals fed oil- supplemented diet. Sheep had higher total volatile fatty acid production and acetic acid proportion compared to goat. Conclusion: The findings of this study suggested that sheep performed better than goats when fed a fattening diet and oil supplementation at the inclusion rate of 4% provides a viable option to significantly enhance growth performance and FCR in fattening sheep and goats.

The Effects of Additives in Napier Grass Silages on Chemical Composition, Feed Intake, Nutrient Digestibility and Rumen Fermentation

  • The effect of silage additives on ensiling characteristics and nutritive value of Napier grass (Pennisetum purpureum) silages was studied. Napier grass silages were made with no additive, fermented juice of epiphytic lactic acid bacteria (FJLB), molasses or cassava meal. The ensiling characteristics were determined by ensiling Napier grass silages in airtight plastic pouches for 2, 4, 7, 14, 21 and 45 d. The effect of Napier grass silages treated with these additives on voluntary feed intake, digestibility, rumen fermentation and microbial rumen fermentation was determined in 4 fistulated cows using $4{\times}4$ Latin square design. The pH value of the treated silages rapidly decreased, and reached to the lowest value within 7 d of the start of fermentation, as compared to the control. Lactic acid content of silages treated with FJLB was stable at 14 d of fermentation and constant until 45 d of ensiling. At 45 d of ensiling, neutral detergent fiber (NDF) and acid detergent fiber (ADF) of silage treated with cassava meal were significantly lower (p<0.05) than the others. In the feeding trial, the intake of silage increased (p<0.05) in the cow fed with the treated silage. Among the treatments, dry matter intake was the lowest in the silage treated with cassava meal. The organic matter, crude protein and NDF digestibility of the silage treated with molasses was higher than the silage without additive and the silage treated with FJLB. The rumen parameters: ruminal pH, ammonia-nitrogen ($NH_3$-N), volatile fatty acid (VFA), blood urea nitrogen (BUN) and bacterial populations were not significantly different among the treatments. In conclusion, these studies confirmed that the applying of molasses improved fermentative quality, feed intake and digestibility of Napier grass.

상향류식 혐기성반응조와 산발효조의 병합처리시 유기물질과 질소거동에 관한 연구 (A Study on the Behaviour of Organics and Nitrogen Using Upflow Anaerobic Reactor When Acid Fermenter is Added)

  • This study was aimed to behaviour of organics and nitrogen on the upflow anaerobic reactor when a acid fermenter is added. Up flow anaerobic reactor (UAR) reaction will result which operates, COD removal efficiencies of reactor with nitrate loading rate 0.11, 0.66g/L/d were over 77%, but one with 1.0g/L/d was 73.5%. Especially, on NLR 0.11g/L/d, COD removal was 77% and nitrate removal efficiency was 93% simultaneously. The other side upflow anaerobic reactor and acid fermenter (UAR+AF) reaction will result witch operates, COD removal efficiencies of reactor with nitrate loading rate 0.11, 0.66g/L/d were over 85%, but one with 1.0g/L/d was 80%. Especially, on NLR 0.11g/L/d, COD removal was 85% and nitrate removal efficiency was 98% simultaneously. Also, without in reaction condition increase of influent nitrate concentration resulted in the linear decrease of nitrate removal efficiency and nitrate removal efficiency at influent nitrate-nitrogen 800mg/L was 50%. Alkalinity was increased theoretically by denitrification at low nitrate-N concentration, however, it was not increased theoretically at high nitrate-No 40% nitrate-N of UAR was denitrified until 70% height of reactor and 90% nitrate-N of UAR+AF was denitrified until 30% height of reactor Upflow anaerobic reactor was to occur accumulate acid, which TVA/Alkalinity is 0.3$\sim$0.47. 발효된 물김치인 동치미에서 분리한 혼합 젖산균의 생리활성 증진에 대한 연구 (Studies on the Enhanced Physiological Activities of Mixed Lactic Acid Bacteria Isolated from Fermented Watery Kimchi, Dongchimi)

  • The aim of this study was to investigate the efficacy of enhanced physiological activities in cultures isolated from Korean fermented watery Kimchi, Dongchimi, of single lactic acid bacteria (LAB), and when these three are mixed LAB as probiotics. Using the BIOLOG system and 16S rRNA sequencing, the isolates were characterized, and identified and assigned to Leuconostoc mesenteroides DK-3, Leuconostoc dextranicum DK-6, and Lactobacillus curvatus DK-13, respectively. Growth rate and pH changes, production of organic acids as metabolites, and physiological activities of the single and mixed LAB cultures, were monitored and compared. In mixed LAB cultures after 72 h of incubation, the maximum concentrations of lactic acid and acetic acid were approximately 340.5 mM and 191.9 mM, respectively, and pH changed from 7.00 to 3.62. Mixed LAB cultures were able to eliminate 96.3% of nitrite. Activities of antioxidant and ${\beta}$-galactosidase were 60.3% and 16.8 units/mg, respectively. Significant antibacterial activity of the concentrated supernatants was demonstrated against several food-poisoning bacteria. Physiological activities obtained from the mixed LAB cultures have been shown to be considerably higher than those of single LAB cultures. In conclusion, these studies demonstrate that compared to the single cultures, all physiological activities in mixed LAB cultures are significantly enhanced.
