탄소원으로 자당을 공급한 합성폐수에 비해 상대적으로 발생수소량은 (7.56 mg $H_2/g$ COD) 적었지만 음식물쓰레기를 기질로 이용한 혐기적 발효공정에서 발생된 가스는 3.47 mg $H_2/g$ COD의 수소생성율을 나타내었다. 자당 합성폐수와 음식물쓰레기의 경우, 각각 8.01, 3.73의 B/A비를 보였으며 수소생성이 많은 경우 주요 유기산 중 Butyric acid가 많이 검출되었다. 동정분석 결과 주요 미생물 군집은 Escherichia 속, Klebsiella 속, Clostridium 속, Bacterium 속, 그리고 Enterobacter 속으로 분석되었다. Clostridium 속은 자당 합성폐수와 음식물 쓰레기 모두에서 관찰되었고 Klebsiella 속은 음식물 쓰레기의 발효과정에서 더욱 활발한 것으로 나타났다. 미생물의 분류학적 관계를 확인한 결과로 60%가 ${\gamma}-proteobacteria$이였으며 Firmicute와 Bacteria가 각각 20%를 차지하였다.
Physiocheical properties and chemical compositions of two plums(Oishiwase, Formosa ) produced in Kimcheon area were investigated . Chemical compositions of two plum cultivars were similar except nitrogen free extract(NFE) . NFE value of Formosa was higher than Oishiwase . Alcohol insoluble substance content, total soluble pectin, total sugar content and total organic acid content of Oishiwase were 2.08%, 14.16%, 5.07% and 949.16mg%, respectively. In case of Formosa cultivar, those were 50%, 15.42%, 5.91% and 985.91mg%, respectively. Fructose content among free sugars of two plum cultivars range from 3.97-3.45% and the other were glucose, sucrose and inositol, 1.83-1.53%, 0.08-0.05% and 0.04-0.03%, respectively. Malic acid content was 35-373mg% and others were levulinic acid, lactic acid, citric acid, succinic acid and fumaric acid in the decreasing order.
G. persimmonis KJ145$^{T}$ 를 사용하여 bacterial cellulose와 식초 동시 생산에 따른 성분변화를 조사하였다. 그 결과 pH는 배양 8일째에 3.22로 감소하는 경향이었으며, 총산은 8일째에 가장 높은 4.66으로 나타났다. Brix는 배양기간 동안 거의 변화가 없었으며, 유리당은 fructose, glucose 및 sucrose로 구성되어 있고 fructose는 배양 종료 후, 10일째에도 거의 감소하지 않고 높게 나타났다. G. Persimmonis KJ145$^{T}$ 균체는 배양 2일부터 급격히 증가하여 4일에 가장 높게 나타났으며, bacterial cellulose는 배양기간이 길수록 증가하는 경향을 나타내었다. 발효액의 유기산은 malic acid, succinic acid, oxalic acid 등이 검출되었으며, acetic acid는 발효 2일째부터 급격히 증가하여 8일째에 가장 높게 생성되었다. 따라서 bacterial cellulose와 초산을 동시에 생산하기 위한 최적배양기간은 8일 정도로 나타났다.
White rot or stem rot caused by Sclerotinia sclerotiorum is one of the most destructive fungal diseases that have become a serious threat to the successful cultivation of oilseed Brassicas. The study was designed with an aim to investigate the association between the pathogenic aggressiveness and pathogenicity determinants of this pathogen specifically in Brassica for the first time. For this, a total of 58 isolates of S. sclerotiorum from different geographical regions were collected and purified. These isolates were inoculated on a Brassica juncea cv. RL-1359 and they exhibited high level of variation in their disease progression. The isolates were grouped and then 24 isolates were selected for the biochemical analysis of pathogenicity determinants. The isolates varied significantly with respect to their total organic acids, oxalic acid production and pectin methyl esterase and polygalacturonase activity. The oxalic acid production corresponded to the disease progression of the isolates; the isolates with higher oxalic acid production were the more aggressive ones and vice-versa. This is, in our knowledge, the first study to establish a correlation between oxalic acid production and pathogenic aggressiveness of S. sclerotiorum on B. juncea. However, the pectinases' enzyme activity did not follow the trend as of disease progression. These suggest an indispensable role of oxalic acid in pathogenicity of the fungus and the potential to be used as biochemical marker for preliminary assessment of pathogenic aggressiveness of various isolates before incorporating them in a breeding program.
Penicillium oxalicum strain CBPS-3F-Tsa, an efficient phosphate solubilizing fungus, was evaluated for its production of organic acid in vitro and effect of inoculation on the growth promotion of Maize under greenhouse conditions. The fungus solubilized 129.1, 118.8, and 54.1 mg P/1 of tricalcium phosphate [$Ca_{3}(PO_{4})_{2}$], aluminum phosphate ($A1PO_{4}$),and ferric phosphate ($FePO_{4}$), respectively, after 72 h of incubation. Malic acid, gluconic acid, and oxalic acid were detected in the flasks supplemented with various phosphate sources [240, 146, 145 mM/1 $A1PO_{4},\;FePO_{4},\;and\;Ca_{3}(PO_{4})_{2}$, respectively] together with a large amount of malic acid followed by the other two. The effects of inoculation of P. oxalicum CBPS-3F-Tsa on maize plants were studied under pot culture conditions. P. oxalicum CBPS-3F-Tsa was inoculated to maize plants alone or together with inorganic phosphates in the form of fused phosphates (FP) and rock phosphates (RP). Inoculation of P. oxalicum CBPS-3F-Tsa increased the plant growth and N and P accumulation in plants, compared with control plants, and also had positive effects when applied with RP. The results of this study show that the fungus P. oxalicum strain CBPS-3F-Tsa could solubilize different insoluble phosphates by producing organic acids, particularly malic acid, and also improved the efficiency of RP applied to maize plants.
두유박의 단백질 소재화를 위하여 셀룰레이즈(cellulase), 단백질 분해효소, 코지(koji), 효모 등의 복합처리에 의한 가용화 방법이 고형분 및 단백질 회수율, 아미노산, 유기산 조성에 미치는 영향을 고찰하였다. 셀룰레이즈와 단백질 분해효소를 복합 사용하여 두유박을 가수분해하는 것이 높은 고형분 회수율(80.2%) 및 단백질회수율(83.5%)을 얻을 수 있었고, 코지와 효모로 처리할 경우 유기산의 함량을 증대시킬 수 있었으며 양조간장 특유의 맛과 향을 내는데 효과적이었다. 특히 셀룰레이즈로 처리한 가수분해물에서 유기산 함량이 높았다. 유리 아미노산 함량은 코지보다 단백질 분해효소로 분해한 경우에서 높았으며 셀룰레이즈 처리는 유리 아미노산 함량에 큰 영향을 미치지 않았다. 두유박 가수분해물에 대한 관능검사 결과 코지로 가수분해한 시료가 관능적으로 우수하였으나 전체적인 기호도는 대조구로 사용된 양조간장에 비해 낮았다.
본 연구를 통하여 유기산 생산을 위한 본 연구를 통하여 유기산 생산을 위한 Propionibacterium acidipropionici의 발효 특성을 규명하여 보았다. P. acidipropionici ATCC 25562, 4875, 및 4965 등 세 균주를 선별하여 발효 실험에 의한 최적 성장조건을 규명한 결과, peptone $1.5\%$ (w/v), yeast extract $0.75\%$ (w/v)에서 유기산 생산성이 최대로 나타났다. 또한 주어진 영양 조건에서 온도, pH 등 환경을 제어하며 회분식 발효를 실시한 결과 세 균주 모두 pH 6.0에서 유기산 수율과 생산성이 가장 높았으며, 그 중에서도 P acidipropionci ATCC 4965 균주가 유기산 생산성이 0.29 g total acids/L/h로 가장 우수한 것으로 판명 되었다. 세 균주의 화학양론적인 유기산 생산비를 비교해본 결과 1.5몰의 글루코오스로부터 $1.60{\sim}2.34$몰의 프로피온산과 $0.74{\sim}1.05$몰의 초산을 생산하는 것으로 나타났다.
A study was conducted to investigate the effect of supplementing earthworm meal (EWM) on the performance of spent laying hens after induced molting, and fatty acid composition in egg yolks. A total of 90 laying hens at 73 weeks of age were fed the experimental diets containing 0.0% (Control) and 0.2% of EWM for 4 weeks. Eggs were collected and weighted every day and Egg production and feed conversion were recorded every week during the experimental period. However fatty acid composition of egg yolk were measured at last week of experimental period. An amount of feed intake increased by supplemental with EMW except for Erst week experimental period (P<0.05). When 76 and 77 week of age, egg production and daily egg mass were improved by the supplemented with EWM (P<0.01), but average egg weight was not different when fed a EWM, therefore feed/egg mass was significantly decreased when fed a EWM. The ratio of egg yolk n-6/n-3 fatty acids contents was significantly improved fed a EMW (P<0.05). It is concluded that supplementing 0.2% of earthworm meal in the spent laying hens after induced molting was fast in recover of physical strength, therefore improves laying performance.
효모를 이용하여 높은 생산성으로 고농도의 erythritol을 생산하기 위하여 ‘세포 성장단계’와 ‘erythritol 생성단계’로 나누어 2 단계 유가식 배양을 수행하였다. 세포 성장 단계에서는 pH-stat 유가식 배양을 수행하여 3.62 g/L-H의 생산성으로 76 g/L의 균체 농도를 얻을 수 있었다. pH-stat 유가식 배양을 수행하여 고농도의 균체 농도를 얻은 후에 멸균되지 않은 분말형태의 glucose를 발효조에 400 g/L가 되도록 공급하여 세포에 osmotic stress를 가해준 결과 41%의 수율로 187 g/L의 erythritol을 생산할 수 있었으며, 이때의 생산성은 2.79 g/L-h로서 기존에 보고된 어느 결과보다도 높은 값을 얻을 수 있었다. Production phase에서 erythritol 생산에 대한 삼투압의 영향을 알아 본 결과 glucose의 농도가 증가할 수록 생성되는 erythritol 농도가 증가하였으며 세포 농도에 따른 erythritol 생산 수율은 큰 차이는 없었으나 세포 농도가 증가할수록 이에 비례하여 erythritol 생산성이 크게 증가하였다. Erythritol 생산에서 용존 산소량에 관계없이 butyric acid가 생성되었으며 40%의 DO 미만에서는 citric acid가 생성되고, 40% 이상의 DO에서는 gluconic acid가 생성되어 용존 산소량에 의하여 metabolic shift가 유도되었다. 고 농도의 glucose 첨가 후 용존 산소량을 20%로 조절하면서 pH를 3.0으로 낮춘 결과 citrate synthetase의 활성이 억제되어 citric acid가 생성되지 않았으며 gluconic acid 농도는 $KH_2PO_4$에 의하여 증가하고 $MgSO_4{\cdot}7H_2O$ 또는 $(NH_4)_2SO_4$에 의하여 감소되었다. Butyric acid 농도는 $KH_2PO_4$ 또는 $(NH_4)_2SO_4$에 의하여 감소되었으며 synergic effect에 의하여 억제되었다
본 연구에서는 Clostridium ljungdahlii 배양에서 유기질소공급물인 tryptone과 sodium tungstate의 배양액 내 농도가 균주 성장과 아세트산, 에탄올 생산에 대한 영향을 확인하기 위한 실험을 수행하였다. 대조군 실험의 배지 조건(0 g/L tryptone)보다 tryptone 2.5 g/L를 투입한 경우 균주 성장이 144.6% 증가하였고, 에탄올 및 아세트산 생산은 각각 8.6%와 36.7% 향상되었다. 대조군 실험에 사용된 배지의 조건(0.01 µM Na2WO4·2H2O)보다 100배인 1 µM Na2WO4·2H2O를 사용한 실험군에서, 균주 성장과 C2 대사산물의 총생산량에는 큰 차이가 없지만, 에탄올 생산 증가하고 아세트산 생산은 감소하여 에탄올/아세트산 생산비가 0.56로 대조군의 0.24에서 크게 증가하였다.
