춘란(Cymbidium goeringii)의 개화정도별 삭과 형성 및 종자 발아에 미치는 NaOCl의 영향 (Effects of NaOCl Treatment on Seed Germination and Capsule Formation of Level of Flowering in Cymbidium goeringii)

  • 본 연구는 춘란 야생 변이종의 육종을 위한 기초 자료를 얻고자 실시하였으며, 야생종의 개화시기를 꽃봉오리, 반개화, 개화, 개화 후 10일, 개화 후 20일의 5단계로 나누어 자가 수분시킨 후 삭과의 형태적 특성과 발아율을 조사한 결과이다. 착과율은 개화 후 20일에 교배한 것에서 100%로 가장 높았고, 삭과의 무게는 반개화 때 교배한 것이 가장 무거웠다. 5단계로 나누어 발아율을 조사한 결과 반개화에서 가장 높게 나타났으며, 교배 후 채종일수를 150일, 165일, 180일의 3단계로 달리하여 발아율을 조사한 결과에서는 교배 후 채종일수를 150일에 파종한 종자에서 5%로 가장 높게 나타났다. NaOCl 처리를 통한 발아율을 조사한 결과에서는 2%로 처리한 것에서 53.3%의 발아율을 보여 가장 좋은 결과를 나타내었다.

  • Orchidaceae species account for one-tenth of all angiosperms including more than 30,000 species having significant ecological, evolutionary, and economic importance. Despite Orchidaceae being one of the largest families among flowering plants, crucial cytogenetic information for studying species diversification, inferring phylogenetic relationships, and designing efficient breeding strategies is lacking, except for 10% or less of orchid species cases involving mostly chromosome number or karyotype analysis. Also, only approximately 1.5% of the identified orchid species from less than a hundred genera have genome size data that provide crucial information for breeders and molecular geneticists. Various molecular cytogenetic techniques, such as fluorescence in situ hybridization (FISH) and genomic in situ hybridization (GISH), have been developed for determining ploidy levels, analyzing karyotypes, and evaluating hybridity, in several ornamental crops including orchids. The estimation of genome size and the determination of nuclear DNA content using flow cytometry have also been employed in some Orchidaceae subfamilies. These different techniques have played an important role in supplementing beneficial knowledge for effective plant breeding programs and other related plant research. This review focused on orchid breeding summarizes the status of current cytogenetic tools in terms of background, advancements, different techniques, significant findings, and research challenges. Principal roles and applications of cytogenetics in orchid breeding as well as different ploidy level determination methods crucial for breeding are also discussed.

BA살포후 주야 온도처리가 동양란 '철골소심'의 개화에 미치는 영향 (Effect of Day and Night Temperatures on Flowering after Spraying Benzyladenine (BA) in Cymbidium ensifolium 'Tekkotsusosin')

  • 본 연구는 동양란 '철골소심'의 개화증진을 위하여 BA처리후 적정한 관리온도를 알아보기 위해 수행되었다. 개화율은 주야간의 기온이 $30/25^{\circ}C$을 유지했을 때 90%, $25/20^{\circ}C$와 대조구에서 50%로 나타났으며 $20/15^{\circ}C$처리에서는 전혀 개화가 되지 않았다. 기형화율은 $25/20^{\circ}C$에서는 나타나지 않았고 $30/25^{\circ}C$에서는 4%, 대조구에서는 9%로 나타났다. $30/25^{\circ}C$에서 화경장, 화경수 등이 증가하였고 화아품질이 향상되었다. BA 살포 후 철골소심의 개화 유도를 위한 한계 기온은 $20^{\circ}C$ 이상인 것으로 나타났다.

Identification of Two Fungal Endophytes Associated with the Endangered Orchid Orchis militaris L.

  • A survey of the endangered orchid Orchis militaris populations was carried out in north-eastern Italy. The occurrence of fungal root endophytes was investigated by light and electron microscopies and molecular techniques. Two main sites of presence were individuated in the Euganean Hills, differing as to the percentage of flowering individuals and of capsules completing maturity. Fluorescence microscopy revealed an intracellular cortical colonization by hyphal pelotons. Two ITS PCR products co-amplified. Sequencing revealed for the former an identity and a high similarity (99%) with a Tulasnellaceae (Basidiomycota) fungus found within tissues of the same host in independent studies in Hungary and Estonia, suggesting an interesting case of tight specificity throughout the Eurosiberian home range. The second amplicon had 99% similarity with Tetracladium species (Ascomycota) recently demonstrated as potential endophytes. TEM revealed two different hyphal structures. Double fungal colonization appears to occur in Orchis militaris and the possible requirement of a specific fungal partner throws light on the causes of this plant's rarity and threatened status.

Isolation and Characterization of a Doritaenopsis Hybrid GIGANTEA Gene, Which Possibly Involved in Inflorescence Initiation at Low Temperatures

  • In the Doritaenopsis hybrid, like most of the orchid species and hybrids, temperature is crucial for the vegetative-to-reproductive transition, and low temperature is required for bud differentiation. To understand the molecular mechanism of this process, an orchid GIGANTEA (GI) gene, DhGI1, was isolated and characterized by using the rapid amplification of cDNA ends (RACE) PCR technique. Sequence analysis showed that the full-length cDNA is 4,022 bp with a major open reading frame of 3,483 bp, and the amino acid sequence showed high similarity to GI proteins in Zea mays, Oryza sativa, Arabidopsis thaliana and other plants. Semi-quantitative RT-PCR revealed that DhGI1 was expressed throughout development and could be detected in roots, stems, leaves, peduncles and flower buds. The expression level of DhGI1 was higher when the plants were flowering at low temperature (22/$18^{\circ}C$ day/night) than the other growth stages. Further analysis indicated that the accumulation of DhGI1 transcripts was significantly increased at low temperature, and concomitantly, initiation of the peduncle was observed. However, DhGI1 levels were low under high temperature (30/$25^{\circ}C$) conditions, and flower initiation was inhibited. These results indicate that the expression of DhGI1 is regulated by low temperature and that DhGI1 may play an important role in inflorescence initiation in this Doritaenopsis hybrid at low temperatures.

온시디움 'Aloha'의 개화에 미치는 GA3, BA, Zeatin 및 Kinetin의 영향 (Effects of GA3, BA, Zeatin and Kinetin on Flowering of Oncidium 'Aloha')

  • 온시디움 'Aloha'에 $GA_3$를 비롯하여 cytokinin류인 BA, zeatin 및 kinetin을 농도별로 2회(6월1일과 6월 15일) 엽면 살포한 결과 화경수, 소화경수, 소화수 등이 뚜렷하게 증가되었을 뿐만 아니라 개화도 촉진되었다. 특히 BA처리는 꽃대의 수와 소화경수를 가장 많이 증가시켰으며 소화수와 크기는 50mg/L 처리구에서 현저하게 증가되었고 개화소요일수는 무처리구에 비해 100mg/L 처리구에서 26일이나 단축되었다.

Effect of Night Interruption with Mist and Shade Cooling Systems on Subsequent Growth and Flowering of Cymbidium 'Red Fire' and 'Yokihi'

  • Growth and flowering of Cymbidium 'Red Fire' and 'Yokihi' plants were examined in a greenhouse with cooling systems in summer, and with night interruption (NI) lighting in winter as a forcing culture system. The greenhouse was divided into two sections with separate cooling controls during the summer season. One section was cooled by a mist system (mist), while the other section was cooled by a shade screen (shade). During the winter, the greenhouse was redivided into three sections within each cooling system. Plants were grown with NI either at a low light intensity of $3-7{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(LNI) or a high l ight intensity of $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(HNI) u sing h igh-pressure sodium l amps during the 22:00-02:00 HR. The control plants were grown under 9 h short-day condition. NI for 16 weeks and cooling for 9 weeks were employed twice during the 2 years of the experimental period. The air temperature was approximately $2^{\circ}C$ lower in the mist than in the shade and the relative humidity was 80 ${\pm}5%$ in the mist compared to $55{\pm}5%$ in the shade. The daily light integral in the mist section was 48% higher than in the shade section. The time from initial planting to flowering pseudobulb emergence decreased with both LNI and HNI for both cultivars, regardless of the cooling treatments. Under NI conditions, however, between 60% and 1 00% of plants of both cultivars flowered in the mist, whereas no or 20% of 'Red Fire' or 'Yokihi' plants, respectively, flowered in the shade treatment over 2 years. Plants grown under the mist had bigger pseudobulbs than those grown in the shade under both NI treatments. These results show that commercial use of NI in winter and a mist cooling system in summer would decrease crop production time to 2 years and increase profits in Cymbidium forcing culture.

소형 다화 분지성 호접란 'SM 333' 육성 (Breeding of Phalaenopsis 'SM 333' with Mini Multiple Flower Formation)

  • 신품종 Phalaenopsis 'SM 333'은 2002년 상미원에서 Phalaenopsis 'Odoriko'와 Phalaenopsis 'Be Tris'를 모부본으로 인공교배하여 육성한 $F_1$ 개체 중에서 선발한 핑크 소형 다화 분지성이다. 2003년부터 2004년까지 2년에 걸쳐 실생 300개체를 양성하여 이들 중에서 영양생장과 개화특성이 우수한 개체 02-03-33을 선발하였다. 2004년과 2005년에 1차, 2차 특성검정을 통하여 품종의 안정성과 균일성을 확인하고 'SM 333'으로 명명하였다. 'SM 333'은 화색이 화사하고 선명한 핑크(RP69D)이며, 화형은 앉아피기로 안정되어 있다. 꽃의 길이와 폭은 각각 5.0, 5.8cm로 소형이다. 복총상화서로 볼륨이 있으며 소형 캐주얼분화에 적합하다. 잎의 길이와 폭은 각각 20.8, 6.5cm이며 엽형은 수평이다. 기내증식율이 높고 변이가 거의 없으며 영양생장 우수하여 재배관리가 용이하다. 2009년 12월 1일 품종등록(등록번호 제2916호)하여 종자산업법에 의해 보호받고 있다.

  • A New Cymbidium variety "Yellow Evening" was developed from a cross between "Oji" and "Valley Flower Cherry Life" followed by seedling and line selections at the National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA) in 2004. After investigation of the characteristics and selection for 9 years (1996-2004), it has been selected specifically pot orchid flower. "Yellow Evening" has medium plant height and semi-erect peduncle. It has medium flowering habit and brownish yellow flowers (RHS, Y11B: petal and sepal) with grayed purple GP183A) spot on lip, when fully opened. Each peduncle has 15.3 flowers with diameter of about 7.4 cm. It has adequate peduncle height, with some curved and spreading petal and sepals. Under optical culture conditions, it started to bloom early-winter (November).

고온기 폐광 자연냉풍 처리가 호접란의 화아분화와 개화에 미치는 영향 (Effect of Treatment of Natural Cool Air from an Abandoned Mine on Flower Bud Formation and Flowering of Doritaenopsis 'Happy Valentine')

  • 호접란 촉성재배를 위한 하계 고온기 폐광 자연냉풍처리가 개화에 미치는 영향을 밝히고자 본 실험을 실시하여 다음과 같은 결과를 얻었다. 보령지방 폐광의 자연냉풍온도는 $12{\sim}14^{\circ}C$이였으며 이 온도를 주온 $22^{\circ}C$, 야온 $20^{\circ}C$로 조절하여 6월부터 8월까지 처리하였다. 이 온도는 호접란 화성유도 기본 온도범위이었으며, 이 때 외부 평균기온은 $28.4{\sim}32.8^{\circ}C$로 나타났다. 화경 발생은 20일저온경과에서 3% 발생되었으며, 45일 경과에서 65% 이상 발생되었고, 90일 경과에서 100% 발생하였다. 화경장은 30일 경과에서 48.7cm로 짧았으며, 60일 경과에서 62.4cm로 길어져 처리기간이 길수록 화경장이 신장되었다. 화수장, 소화수도 같은 경향이었고, 화경 마디수는 6~7개로 저온 경과일수와는 큰 영향이 없는 것으로 나타났다. 개화는 45일 경과에서 9월 15일에 가장 빨리 개화되었고 저온 경과일수가 길수록 늦어지는 경향을 나타내었다. 고온기 폐광 자연냉풍처리는 무처리에 비하여 화경 발생시기는 3개월, 개화기는 4개월 정도 촉진되었다.
