백년초 유색미의 취반 특성

  • 손바닥 선인장 (Opuntia ficus-indica)은 백년초라고도 불리어지는 다년생 식물로 멕시코가 그 원산지이며, 우리나라에는 약 200년 전에 들어와 제주도 월령마을과 마라도 남쪽해변 산지에 자연상태로 착생되어 제주도 지방 기념물 제 35호로 지정되어 있다. 현재는 북제주군의 월평리를 중심으로 노지에서 재배되고 있으며, 열매는 서양배 모양을 하고 점질물과 함께 다량의 적색계 색소를 함유하고 있다. 열매는 공복에 갈아 마시면 변비치료, 이뇨효과, 장운동 활성화 및 식욕증진에 효과가 있으며, 당뇨, 고혈압, 천식에도 효능이 있다고 하여 민간요법으로 사용되어 왔다. 또한 라디칼 소거, tyrosinase 활성 억제, 항알레르기 활성, 항산화 및 항균활성, 스트레스 항위궤양 효과 등 다양한 기능성이 보고되고 있다. 색소성분은 함질소 anthocyanin의 일종인 betalaines로 적색의 betacyanines과 황색의 betaxanthines 로 구성되어 있으며 75~95%가 betacyanines에 속하는 것으로 알려져 있다. 지금까지 손바닥 선인장에 관한 연구로는 색소의 안정성, 품종별 아미노산 조성, 성분 특성, 추출물을 첨가한 면류의 품질 특성 등으로 상당히 단편적인 연구에 그치고 있다. 본 연구는 최근 식생활의 서구화로 쌀 위주의 식생활에 많은 변화를 가져와 쌀 소비량은 해마다 줄어들고 있는 반면 기능성을 가미한 쌀의 소비가 늘고 있음을 감안하여, 손바닥 선인장 물추출물로 코팅한 유색미의 취반특성을 조사하였다. 그 결과 유색미는 자청색을 띠었으나 그 취반은 연한 황색을 띠었다. 손바닥 선장의 유리아미노산은 $\alpha$-aminoadipic acid가 83.22 mg%, tyrosine이 75.61 mg%로 주 아미노산을 이루었다. 유색미 취반에는 백미 취반에는 검출되지 않는 $\alpha$-aminoadipic acid가 1.66 mg% 함유되었으며, 백미 취반에 비하여 arginine과 leucine은 2배, histidine과 Iysine은 각각 2.3배 및 4.2배, tyrosine은 3.4배가 함유되었다. 손바닥 선인장의 주요 무기질은 Ca, K및 Mg으로 전체 무기질 함량의 약 95%를 차지하였으며 이들 무기질의 함량은 유색미 취반이 백미취반에 비하여 10~45%가 높았다. 밥알의 견고성, 점착성 및 깨짐성은 유색미 취반에서 낮았으나 응집성은 높았다. 단맛은 유색미 취반과 백미 취반 간의 뚜렷한 차이가 없었으나 구수한 맛과 쫄깃한 맛은 유색미 취반에서 높았다. 취반의 종합적인 기호도 및 색상에 대한 기호도는 유의적인 차이가 없었다.

Fresh and Strength Properties of Mortar Produced with Recycled Cactus Stem Powder (자원순환형 선인장 줄기 분말을 혼입한 모르타르의 굳지 않은 특성과 강도)

  • In this study, in order to recycle the stems of opuntia cactus discarded after harvesting the fruit, the cactus stems were powdered and applied as a cement-based admixture. The powder of cactus stem was mixed into the mortar, and its effect on the fresh properties and strength of the mortar was studied. The results were compared with the properties of mortars produced by mixing with a retarder sugar and a viscosity agent methyl-cellulose, which are conventional saccharide-based admixtures. Based on the test results, the cactus stem powder did not clearly show the effect as a retarding agent, whereas the flow and the air content were similar to those of the mortar mixed with methyl-cellulose. This indicated that the cactus stem powder can be used as a viscosity agent. It was found that the strength of the mortar tended to increase when the mixing ratio of the cactus stem powder was lower than 0.3%.

Effect of Opuntia Ficus-indicts variety Saboten Makino and Satsuma Mandarin Dyeing on Physical Characteristics and Colors of Protein Fabrics (선인전 및 감귤염색에 의한 단백질 직물의 물성 및 색상에 관한 연구)

  • The physical characteristics of silk and wool fabrics dyed with OFS-FR and SM-FR were examined and color difference and colorfastness of these fabrics were analyzed. The results are as follows; 1. The thickness, density and weight of both dyed fabrics were increased than those of undyed fabrics independently of dyeing materials and mordants. 2. Air permeability of all the silk samples increased than that of undyed silk, on the other hand, air permeability of most of wool samples decreased than that of undyed wool. 3. Thermal insulation values of silk samples were not consistent with dyeing conditions, but those of wool samples increased than those of undyed fabrics regardless of dyeing materials and mordants. 4. Color difference was clear between OFS-FR and SM-FR dyed fabrics. The colors dyed with OFS-FR-Al and OFS-FR-chitos an showed violet, those dyed with SM-FR displayed yellowish, in addition to in case of wool, dyed with OFS-FR-Cu the color was greenish yellow, in case of silk dyed with SM-FR-Cu that was yellowish red. 5. Sense of silk fabrics by sensory-test was soft and smooth. Feeling of colors in silk fabrics was wick, warm and slightly brilliant. In case wool, sense of dyed fabrics was slightly soft and slightly rough. Feeling of colors in wool fabrics was warm but not elegant. Therefore the result of sensory-test in silk might be better than that of in wool. 6. Colorfastness to dry cleaning and perspirations was good, but that to washing and sunlight was not fine.

Characterization and sequence analysis of half of genome RNA of a new Tobamovirus (Cactus mild mottle virus) from cultivated cactus plants in Korea

  • A new isolate of rod-shaped virus was identified from grafted cactus, Gymnocalycium mihanovichii grafted onto Hylocereus trigonus, in Korea. The virus proved to be a new Tobamovirus and called previously as Tobamovirus-Ca for which we suggest the name Cactus mild mottle virus(CMMoV), because it produced systemic mild mosaic symptoms on its original host. CMMoV is distantly related to known species of the genus Tobamovirus on the basis of host range, serological and sequence analyses. Western blot analysis showed that CMMoV is serologically unrelated to Summons' Opuntia virus which is the only known species of the genus found in cactus plants. The 3'-terminal 2,910 nucleotides have been sequenced for the virus. The coat protein (CP) and movement protein (MP) genes encode 161 and 306 amino acids residues, respectively. The nucleotide and amino acid sequences of the CP were 39.6 % to 49.2 % and 26.4 % to 40.3 % identical to other tobamoviruses, respectively. The MP and 3' noncoding region shared 16.3 % to 23.3 % and 44.6 % to 63.4 % identities, respectively, with the members of the genus. Phylogenetic tree analysis of the CP gene revealed that CMMoV clusters with members of subgroup I of Tobamovirus. CMMoV particles contained genomic RNA along with two subgenomic RNAs, and this characteristics is common in the members of the subgroup II. This is the first information of sequence and comparative analysis of a Tobamovirus that infects cactus.

Effect of Ginseng and Herbal Plant Mixtures on Anti-Obesity in Obese SD Rat Induced by High Fat Diet (홍삼 및 천연물 혼합소재의 비만 유도 흰쥐에 대한 항비만 효과에 관한 연구)

  • Anti-obesity effects of ginseng and herbal plant mixtures were investigated to develop natural materials for anti-obesity. After inducing obesity with high fat diet for 8 weeks in male SD rats, ginseng and herbal plant mixtures DM1 (ginseng, puer tea, opuntia) and DM2 (ginseng, puer tea) were administrated orally to rats for another 8 weeks. During administration, food efficacy ratio and body weight of rat were measured twice weekly. After administration, body weight, body fat contents, and serum lipid level were estimated for anti-obesity effect and hematological analysis blood level of ALP and ASP was checked for safety. Body weight in rats fed high fat diet was significantly increased. Body weight in obese rats induced by high fat diet was significantly decreased by DM1 and DM2 feedings. The amount of body fat (epididymal, perirenal and visceral fat, brown adipose tissue) was significantly reduced by DM1 and DM2 treatments. The amount of TG, the concentration of leptin in blood plasma, and the concentration of insulin in blood plasma were significantly diminished by DM1 and DM2. Lipid accumulation on liver was reduced in DM2. There were no side effects among all groups according to blood analysis, hematological findings, and body weight. The findings of this study suggest that DM1 and DM2 may be effective materials for anti-obesity through reducing plasma triglyceride and body fats, and also decreasing body weight without side effects.

Study on the storage stability of the white internal organs using natural materials (천연소재를 활용한 백내장의 저장성 증진 연구)

  • The purpose of this study was to investigate the ability of natural antioxidants as stabilizers for meat by-products to prevent lipid oxidation. The white internal organs were evaluated using different treatments: no antioxidant (control), ascorbic acid (T1), Artemisiacapillaris Thunb. (T2), Opuntia (T3), Schisandra chinensis (T4), and Saururuschinensis (Lour.) Baill (T5). Antioxidant activities were analyzed by measuring DPPH contents, superoxide anion radical levels, nitrate scavenging activities, and total polyphenol contents. T1 and T2 showed higher antioxidant activities and total polyphenol contents (p<0.05). Additionally, changes in physicochemical properties (pH, color, volatile basic nitrogen [VBN], and thiobarbituric acid reactive substances [TBARS]) and microbiological aspects in white internal organs processed with antioxidants were investigated. As storage time increased, the CIE $a^*$ and $b^*$ values of the white internal organs processed with natural antioxidants were decreased (p<0.05), and CIE $L^*$ values were low, particularly for the T3 sample relative to that in the control. Moreover, the pH, VBN, and TBARS values of samples T2-T5 were increased after 7 days of storage, but showed low values compared with those of the control (p<0.05). Moreover, compared with the control group, the treatments showed antimicrobial effects. Our results indicated that these natural antioxidants could be used as lipid oxidation stabilizers of meat by-products during storage and that Artemisiacapillaris Thunb. and Opuntia may have applications as natural antioxidants in the meat by-product industry.

Cooking Characteristics of Rice Coated with Prickly Pear Water Extracts (손바닥 선인장 물추출물로 가공한 유색미의 취반 특성)

  • Cooking characteristics of rice uncoated and coated with prickly pear water extracts (4% v/w for rice) were investigated. Prickly pear contained 83.22 mg% of $\alpha$-aminoadipic acid and 75.61 mg% of tyrosine as major free amino acids. After cooking, the coated rice contained 1.66 mg% of $\alpha$-aminoadipic acid which was not found in the uncoated rice. The free amino acid composition of the coated rice revealed significantly higher contents (about 2.0 to 4.2 times) of arginine, histidine, leucine, lysine and tyrosine compared with those of the uncoated rice. The major minerals in the prickly pear were Ca, K and Mg accounting for 95% of the total minerals present. After cooking, the mineral contents in the coated rice were higher by 10~45% than those in the uncoated rice. The coated rice showed lower hardness, gumminess and brittleness, and higher cohesiveness than the uncoated rice. In sensory evaluation, there were no differences in sweet taste, and overall and color acceptability between the uncoated and coated rice. However, tile coated rice showed higher scores for savory and sticky taste than the uncoated rice.

Changes in Sugar Level, Acidity, Viscosity, and Color of Lactic Acid Bacteria- Fermented Waxy Rice Paste Containing Colored Agro-food Products (유색 식물을 이용한 약초부각용 발효찹쌀풀의 당, 산도, 점도 및 색도 변화)

  • To develop new high-quality Yakchobugak, features of Lactococcus lactis-fermented waxy rice paste after addition of some colored powdered agro-food products were investigated. Total and reducing sugars of waxy rice paste fermented by lactic acid bacteria were higher than those of control raw waxy rice paste. Total acidity gradually increased as powder concentration rose, being 1.02-1.56% and 0.96-1.87% in samples fermented with Cucurbita maxima and Capsicum annuum powders, respectively; these values were 3-4 times those in rice fermented with other powders. Fermented waxy rice paste viscosities were lower than those of non-fermented samples. The viscosities of samples fermented with Curcuma longa and Opuntia ficus powders were in the range $100-160{\times}10^4$ centipoise($mPa{\cdot}s$), and those of pastes fermented with Robus coreanus and Camellia sinensis extracts were under $40{\times}10^4mPa{\cdot}s$. Hunter color lightness(L) values decreased and yellowness(b) values rose after fermentation. Waxy rice paste fermented with Robus coreanus showed uniform particle size distribution, and many pores, by scanning electron micrography.

The Antimicrobial Activity of Medicinal Plants Extracts against Helicobacter Pylori (약용식물 추출물의 Helicobacter pylori에 대한 항균활성)

  • Antimicrobial activities were assayed through the hot-water extracts from 41 species of medicinal plants against Helicobacter pylori which is known as the ulcerogenic phathogen. Opuntia ficus-indica, Houttuynia cordata, Sinomenium acutum, and Coptis japonica showed the MIC at the concentrations less than 100 ppm, Pulsatilla koreana, Forsythia koreana, Rheum undulatum, and Perilla frutescens less than 200 ppm, Belamcanda chinensis, Arctium lappa, Cassia tora, Citrus tachibana, Siegesbeckia orientalis, and Caesalpinia sappan less than 300 ppm by the 2-fold dilution method. In disc method only three of them were confirmed to have antimicrobial activities which were increased in the order Perilla frutescens, Coptis japonica, Caesalpinia sappan. Three extracts were partitioned with chloroform, ethyl acetate and butanol in sequence and examined for the activity to inhibit H. pylori. The major ativities were observed in ethyl acetate fraction of Caesalpinia sappan, butanol fraction of Perilla frutescens, butanol and chloroform fraction of Coptis japonica. The partitioned fractions were found to have increased antimicrobial activities in all extracts. The experiments in which the extracts were added into urea R broth containing the crude urease derived from H. pylori resulted in the increase of pH and optical density at 560 nm to 8.15 and 1.7 respectively. Urease activity of H. pylori was inhibited over 80% by Caesalpinia sappan, Perilla frutescens, and Coptis japonica, of which Caesalpinia sappan suppressed up to 95%.

Chemical compositions and antioxidant activities of Cheonnyuncho (Opuntia humifusa) stems and fruit (천년초의 성분특성과 항산화 활성)

  • The purpose of this study was to investigate the chemical composition of the freeze-dried stems and fruit of the cactus Cheonnyuncho. The analysis showed that powdered stems have the highest fat content (1.91%) and the powdered fruits have he highest protein content (2.62%). The K content of the fruits higher than that of the stems, while the Ca, Mg, Na and P contents of the stems were higher than those of the fruits. Both the stems and fruits powders contained high levels of the amino acids glutamic acid and aspartic acid. The free sugars such as sucrose, fructose, and glucose were detected in both the stems and fruits. The 75% ethanol (EtOH) extract showed a relatively high antioxidative activity compared to those of the water and 75% methanol (MeOH) extracts. Furthermore, the 75% EtOH extract of the stem powder exhibited a total polyphenol content of 3.60 g/100 g, and a total flavonoid content of 2.00 g/100 g. The antioxidant activities of the stem and fruit powder extracts, measured in DPPH radical scavenging experiments, were higher than that of the control group.
