Development of Optimal Treatment Process Train of Leachate from Industrial Waste Disposal Site

  • Train of chemical and biological processes was investigated to treat leachate from industrial waste landfill. Organics and nitrogen concentrations of landfill leachate studied in this research were high and their BOD/COD ratio was 0.3. Biological process should be combined with chemical process for optimum treatment of leachate. PAC(Polyaluminium chloride) was the best coagulant among three chemicals tested, and the optimum condition of PAC coagulation was pH 6 and 1,250 mg/L of dosage. When SBR was operated for simultaneous removal of organics and nitrogen, removal efficiency of COD and T-N reached over $82\%,\;71\%,$ respectively and time distribution of 2/4/2/1 was most effective for one cycle of anoxic/aerobic/anoxic/aerobic.

  • Precast slab track system(PSTS) is a concrete track laying system where the slab panels are pre-manufactured in factories and assembled and installed on-site. PSTS has been developed for the past 30 years in countries where railway technologies are advanced such as Japan and Germany to improve the various drawbacks of the in-situ concrete slab track. However, the usefulness of PSTS is being continuously approved by many other countries such as China, Taiwan, Austria, Italy, Spain, etc,. Lately, not only Japan and Germany, but also Austria, Italy and China have developed their own PSTS by collaboration between their Governments and private enterprises and are now attempting to expand their businesse soverseas. In accordance to such movement, in 2006, the Korean Railroad Research Institution and Sampyo E&C have developed a Korean PSTS by joint research. PSTS consists of concrete panel, under pouring layer and concrete base layer. Amongst these components, the panel is the main component of PSTS which supports the train load and has a great effect on the track quality, workability and economics. Therefore, a study is to be conducted to select the optimum size for the Panel of the precast slab track system panel by analyzing the various standards & forms, interpretation of finite elements of the selected model and economical analysis.

  • This research is conducted to calculate optimum residents grant regarding types of environmental foundational facilities. In the case of Gangwon province, it was particularly referred in this study, the residents grant of waste treatment facilities was excessively discharged as $1.6{\sim}75.0%$ of construction cost. This kind of excessive residents grant has overweighted the local governments' financial conditions. Accordingly, it is necessary to increase the amount of charge on the national treasury. In the calculation method of optimum residents grant reasonable resident support fund of waste treatment facilities was computed the range of 20%, food waste treatment facilities was 10% ranges and sewage and night soil combination treatment facilities was $3{\sim}5%$ ranges.

  • As for the purpose, we first introduce an random mutation into wild-type gene to expand a mutation space, and then further recombine the mutant genes by staggered extension process PCR. As a result, we obtained the best clones 6-52 that showed a high activity and stability, from a round of error prone and staggered extension process PCR. The purified enzyme showed a similar pH stability to the wild-type enzyme and reveal a slightly high optimum pH at 12. In the optimum temperature, an identical dependency was also showed and a quite high stability in the thermal stability was obtained. Along with this, the enzyme was also stable at a reaction that supplement with a 15 % of ethanol as an additive. The addition of other solvents and surfactants did not improve the reaction and thus resulted in a similar profile to those of wild-type enzyme. The specific activity on the target compound rac-ketoprofen ethyl ester was calculated to be about 85, 000 unit, and the kinetic constants Km and Vmax were determined to be 0.2 mM and 90 mM/mg-protein/min respectively. The deduced amino acid alignment with the wild type enzyme revealed five mutations at L120P, I208V, T249A, D287H and T357A. Based on these observations, the site directed mutagenesis to delineate the mutagenic effect is under progress.

Purification and Characterization of Protein Phosphatase 2C from Rat Liver

  • Protein phosphatase 2C (PP2C) is one of the four major serine/threonine phosphatases which is dependent on $Mg^{2+}$ for its activity. PP2C was purified from rat liver cytosol and its characteristics were investigated. The substrate employed for routine assay was $[^{32}P]casein$ phosphorylated by PKA. The purification process involved DEAE chromatography, ammonium sulfate fractionation, phenyl sepharose chromatography, sephacryl 5-200 gel filtration, and histone agarose chromatography. The SDS-PAGE of PP2C showed one major single protein band at a position corresponding to a molecular mass of 43 kd and the purification fold was 637. The enzyme showed a pH optimum of 8 and $K_M$ value was $1.9\;{\mu}M$. However, when the substrate was changed to $[^{32}P]histone$, the pH optimum was shifted to 7 and $K_M$ value was $2.3\;{\mu}M.\;Mg^{2+}$ was essential to the enzyme activity and okadaic acid did not exert any inhibitory effect on the enzyme. To examine residue in the active site of PP2C effects of some protein-modifying reagents were tested.

A Study on the Pb-contaminated Soil Remediation by Organic Acid Washing (유기산을 이용한 납 오염토양의 복원에 관한 연구)

  • A study on the removal of Pb ion from Pb-contaminated soil was carried out using ex-site extraction process. Tartaric acid (TA) and iminodiacetic acid sodium salt(IDA) as a washing agent were evaluated as a function of concentration reaction time mixing ratio of washing agent and recycling of washing agent. TA showed a better extraction performance than IDA. The optimum washing condition of TA and IDA were in the ratio of 1:15 and 1:20 between soil and acid solution during 1 hr reaction. The total concentrations of Pb ion by TA and IDA at three repeated extraction were 368.8 ppm and 267.5 ppm respectively. The recovery of Pb ion from washing solution was achieved by adding calcium hydroxide and sodium sulfide form the precipitation of lead hydroxide and lead sulfide and optimum amounts of sodium sulfide and calcium hydroxide were 7 g/$\ell$ for the TA washing solution and 4 g/$\ell$, 5g/$\ell$ for the IDA washing solution respectively. The efficiency of recycle for TA and IDA washing solution were 78.8% , 95.1%, and 89.2%, 96.6% at third extractions under $Na_2S$ and $Ca(OH)_2$, respectively.

  • From traditional ceremony to science technology, origami was used as technique for artistic creativity and functionality based on formative characteristics. The purpose of this study is to analyze the expressive characteristics of functionality in fashion design using origami technique. The method of this study was to analyze recent documentaries, fashion books, fashion magazines, internet web site and so forth. The results were as follows: In various fields of design, the category of the expressive characteristics was classified as efficient correspondence of space, compositive convergence of function, variable potentiality of form and human ergonomic optimum. In recent origami fashion based on functionality, the efficient correspondence of space was expressed constructive or decorative pocket design, using on clothing surface and varied design by lifestyle. The compositive convergence of function was expressed innovative connection of fashion items or non-fashion items and deconstructed classical clothing pattern. The variable potentiality of form was expressed control of form by irregular fold, dynamic metamorphosis by organic fold and creation of varied clothing silhouette. The human ergonomic optimum was expressed metamorphosis mechanism from two dimension to three dimension, suitable construction to physical ability and limit and organic form based on articular folding. The functional origami fashion means reflection of nomadic lifestyle, maximum of usability, pursuit of playful sensibility, creation of new body aesthetic and pursuit of technology and humanism. And the result of these study may be used for creative inspiration in fashion design because of unique characteristics.

Characterization of ${\beta}-Galactosidase$ from a Bacillus sp. with High Catalytic Efficiency for Transgalactosylation

  • A ${\beta}$-galactosidase with high transgalactosylic activity was purified from a Bacillus species, registered as KFCC10855. The enzyme preparation showed a single protein band corresponding to a molecular mass of 150 kDa on SDS-PAGE and gave a single peak with the estimated molecular mass of 250 kDa on Sephacryl S-300 gel filtration, suggesting that the enzyme is a homodimeric protein. The amino acid and sugar analyses revealed that the enzyme is a glycoprotein, containing 19.2 weight percent of sugar moieties, and is much more abundant in hydrophilic amino acid residues than in hydrophobic residues, the mole ratio being about 2:1. The pI and optimum pH were determined to be 5.0 and 6.0, respectively. Having a temperature optimum at $70^{\circ}C$ for the hydrolysis of lactose, the enzyme showed good thermal stability. The activity of the enzyme preparation was markedly increased by the presence of exogenous Mg (II) and was decreased by the addition of EDTA. Among the metal ions examined, the most severely inhibitory effect was seen with Ag (I) and Hg (II). Further, results of protein modification by various chemical reagents implied that 1 cysteine, 1 histidine, and 2 methionine residues occur in certain critical sites of the enzyme, most likely including the active site. Enzyme kinetic parameters, measured for both hydrolysis and transgalactosylation of lactose, indicated that the enzyme has an excellent catalytic efficiency for formation of the transgalactosylic products in reaction mixtures containing high concentrations of the substrate.

Optimum Maintenance and Retrofit Planning for Reliable Seismic Performance of the Bridges (내진성능확보를 위한 교량의 최적유지보수계획법)

  • In the maintenance and retrofit planning of a bridge system, the optimal strategy for inspection and repair are suggested by minimizing the expected total life-cycle cost, which includes the initial cost, the costs of inspection, repair, and failure. Degradation of seismic performance is modeled by using a damage function. And failure probability is computed according to the degree of damage detection by random vibration theory and the event tree analysis. As an example to illustrate the proposed approach, a 10-span continuous bridge structure is used. The numerical results show that the optimum number of the inspection and the repair are increased, as the seismic intensity is increased and the soil condition of a site becomes more flexible.

  • In teleoperation, it is important for an operator to feel as if he really were in a distant place. To realize this objective, the various information from a remote site must be presented to the operator. Even though tactile information is very important to efficiently execute a task, it is not yet sufficiently provided for the operator. In this paper, we propose the new mechanism that can provide the more dexterous tactile information to the operator This device utilizing the electromagnetic force is designed to be compact and light enough to be attached to the fingerpad, and designed to be controlled continuously. The magnetic circuit is derived by the probable flux paths method in order to take forces at any given dimension. An optimization technique is also proposed to maximize the tactile force that humans can perceive under the same conditions. The objective function is formulated as maximizing displacements indented on the fingerpad, considering the mechanism of human tactile perception. The optimization formulation is subject to the geometric and rising temperature constraints in the coil. It is demonstrated that, by optimization, the tactile force increases by 24%, compared with that obtained from the initial design.
