자색고구마 Anthocyanin 색소의 추출조건 결정 (Determination of the Conditions for Anthocyanin Extraction from Purple-Flashed Sweet Potato)

  • To establish the optimum conditions for the extraction of anthocyanin pigment from purple-fleshed sweet potato, a suitable extraction solvent with the optimum citric acid concentration for acidification of the solvent, and the optimum extraction time and temperature were determined. Twenty percent ethanol solution acidified with citric acid was found to be a good solvent for the extraction of the pigment from purple-fleshed sweet potato. About 10 hour extraction at room temperature was appropriate for the extraction. pH of the extract was below 3 when more than 0.7% citric acid was added. The higher the concentration of citric acid added was, the higher the total optical density (TOD) of the extract was. However, the increase in TOD of the extract was insignificant when more than 1% of citric acid was added. Therefore, addition of 1% citric acid was determined for acidification of the extracting solvent. Though the initial rate of the pigment extraction increased as the extracting temperature increased, extraction at higher temperatures of 60 or 8$0^{\circ}C$ for an extended time caused a decrease in the extraction yield due to degradation of the pigment. The optimum extraction temperature for the anthocyanin pigment from purple-fleshed sweet potato with the solvent used was determined as 4$0^{\circ}C$.

Extraction of β-glucosidase from Bagasse Fermented by Mixed Culture under Solid State Fermentation

  • Various parameters such as solvent selection, concentration, solid/liquid ratio, soaking time, temperature, stationary, shaking conditions, and repeated extractions were investigated in order to determine the optimum extraction conditions of ${\beta}$-glucosidase from bagasse fermented by mixed culture of Aspergillus niger NRC 7A and Aspergillus oryzae NRRL 447. Among various solvents tested, non ionic detergents gave the best results than the inorganic or organic salt solutions and distilled water. The optimum conditions for extraction of ${\beta}$-glucosidase were 30 min soaking time at $40^{\circ}C$ under shaking condition at 150 rpm, with solid/liquid ratio 1:15 (w/v), which yielded $2882.74{\pm}95.52U/g$ fermented culture (g fc) of enzyme activity. With repeated washes under the above optimum conditions, the results showed that enzyme extracted in the $1^{st}$ and $2^{nd}$ washes represents about 90% of the total activity.

Optimum Conditions for the Extraction of Effective Substances from the Stem of Opuntia fiscus-indica

  • The conditions for the extraction of effective substances from the stem of Opuntia ficus-indica were optimized by a response surface methodology. The total extract yield was maximized under the temperature of $97.77^{\circ}C$, at a time duration of 145.82 min and a water to sample ratio 16.59 mL/g. Moreover, the optimum conditions for the extraction of effective substances were as follows: $84.95^{\circ}C$, 156.50 min and a water/sample ratio of 7.46 mL/g for the phenolics content; and $97.11^{\circ}C$, 139.03 min and a water/sample ratio of 10.91 mL/g for the pectin content. The range of optimum extraction conditions in consideration of the physicochemical properties of the extracts were shown to be as $95-100^{\circ}C$ as the extraction temperature, 120-180 min as extraction time and a water to sample ratio of 5-18 mL/g.

수용성 치자 색소의 추출에 관한 연구 (A Study on the Extraction of Soluble Colorants of the Gardenia jasminoides Ellis)

  • The study was performed to obtain the optimum extraction conditions for crocin from gardenia fructus. Generally crocin is unstable on heat, light, acid and base solution. The extraction efficiency of crocin from gardenia depended upon the extraction time, extraction temperature, pH in the extraction bath and the optimum conditions of crocin extraction were determined as 60 minutes of extraction time, 4$0^{\circ}C$ of extraction temperature, pH 7 of extraction bath. The molar extinction coefficient of crocin was 12,515 and the color yield of purified crocin was about six times higher than that of non-purified crocin. The heat-stability at extraction temperature and lightstability in irradiation with xenon lamp for one hour of the purified crocin were higher than those of non-purified crocin. Intensity of &{\lambda}_{max}&of crocin was decreased by irradiation for one hour but UV-Vis. spectra of crocin was not changed. The colors of purified and non-purified crocin dissolved wit methanol was evaluated by means of CIE L* a* b* system.

반응표면분석법에 의한 늙은 호박 추출조건의 모니터링 (Monitoring on Extraction Conditions of Old Pumpkin Using Response Surface Methodology)

  • Response surface methodology was used for monitoring extraction conditions, based on quality properties of old pumpkin extracts. Hunter's color L value of extracts was maximized at 101℃, 2.6 hr and decreased gradually after maximum point. The polynomial equation for Hunter's color L value showed 10% of significance level and 0.8799 of R². Hunter's color a value was minimized at 117℃, 3.9 hr and R² of polynomial equation was 0.9852 within 1% significance level. Hunter's color b value and ΔE value increased as the extracting temperature and time increased. Extraction yield of old pumpkin was maximized at 110℃, 4 hr and increased in proportional to the extracting temperature and time, but decreased after 113℃ and 2 hr. Viscosity of pumpkin extracts were maximized at 120℃, nearly 3 hr. R² of polynomial equations for yield, viscosity and sugar content were 0.9532, 0.9812 and 0.8869, respectively. Optimum ranges of extraction conditions for quality properties of old pumpkin were 102∼109℃, 2.5∼3.5 hr, respectively. Predicted values at the optimum extraction condition agreed with experimental values.

산국(山菊) 에탄올 추출물의 이화학적 특성에 대한 추출조건의 최적화 (Optimization of Extraction Conditions for Physicochemical Properties of Ethanol Extracts from Chrysanthemum boreale)

  • To renew interest in Chrysanthemum boreale as a traditional food material, response surface methodology was used for optimizing extraction conditions and monitoring physicochemical properties of ethanol extracts from Chrysanthemum petal. The phenolic compounds of ethanol extracts were minimized in 87ml/g(solvent per sample), 21%(ethanol concentration), and 15 hours(extraction time). The physicochemical properties of ethanol extracts were maximized in the conditions of 115ml/g, 98% and 16 hours on yellow color intensity, 143ml/g, 75% and 19 hours on carotenoid content, and 148ml/g, 53% and 18 hours on soluble solid content. Optimum ranges of extraction condition for physicochemical properties of Chrysanthemum boreale were 130~150ml/g, 70~85% and 20~28 hours, respectively. Predicted values at the optimum extraction condition were in good agreement with experimental values.

Validation on the Analytical Method of Ginsenosides in Red Ginseng

  • A cross-examination between KT&G Central Research Institute and Guangzhou Institute for Drug Control was carried out in order to select optimum conditions for extraction, separation and determination of ginsenosides in red ginseng and to propose a better method for the quantitative analysis of ginsenosides. The optimum extraction conditions of ginsenosides from red ginseng were as follows: the extraction solvent, $70\%$ methanol; the extraction temperature, $100^{\circ}C;$ the extraction time, 1 hour for once; and the repetition of extraction, twice. The optimum separation conditions of ginsenosides on the SepPak $C_{18}$ cartridge were as follows: the loaded amount, 0.4 g of methanol extract; the washing solvents, distilled water of 25 ml at first and then $30\%$ methanol of 25 ml; the elution solvent, $90\%$ methanol of 5 ml. The optimum HPLC conditions for the determination of ginsenosides were as follows: column, Lichrosorb $NH_2(25{\times}0.4cm,$ 5${\mu}m$, Merck Co.); mobile phase, a mixture of acetonitrile/water/isopropanol (80/5/15) and acetonitrile/water/isopropanol (80/20/15) with gradient system; and the detector, ELSD. On the basis of the optimum conditions a method for the quantitative analysis of ginsenosides were proposed and another cross-examination was carried out for the validation of the selected analytical method conditions. The coefficient of variances (CVs) on the contents of ginsenoside-$Rg_{1}$, -Re and $-Rb_1$ were lower than $3\%$ and the recovery rates of ginsenosides were $89.4\~95.7\%,$ which suggests that the above extraction and separation conditions may be reproducible and reasonable. For the selected HPLC/ELSD conditions, the CVs on the detector responses of ginsenoside-Rg, -Re and $-Rb_1$) were also lower than $3\%$, the regression coefficients for the calibration curves of ginsenosides were higher than 0.99 and two adjacent ginsenoside peaks were well separated, which suggests that the above HPLC/ELSD conditions may be good enough for the determination of ginsenosides.

추출조건에 따른 삼백초 추출물의 항산화 활성 및 성분의 변화 (Change of Antioxidant Activity and Antioxidant Compounds in Saururus chinensis by Extraction Conditions)

  • 삼백초 지상부의 추출조건을 설정하고자 추출용매 및 시간에 따른 이화학적 특성을 살펴보았다. 추출용매 조건은 40% 에탄올을 사용하여 추출할 때로 나타났으며, 이때, 총수율, 전자공여능, 총페놀함량, 총플라보노이드 함량은 각각 13.50%, 83.50%, 2.60 mg/ml 및 2.09 mg/ml으로 나타났다. 최적의 추출 시간은 4시간으로 나타났으며, 이때, 총수율, 전자공여능,총폐놀함량, 총플라보노이드 함량은 각각 12.79%, 71.13%, 2.83 mg/ml 및 2.16 mg/ml으로 나타났다. 삼백초 지상부를 40% 에탄을 추출물로 4시간 추출하였을때, quercetin 함량은 360.13 mg/100 g으로 나타났으며, quercetin 함량은 1379.54 mg/100 g으로 나타났다. 삼백초 지상부를 이용하여 추출조건을 설정한 결과 40% 에탄올을 이용하여 4시간 추출하는 것이 가장 적합한 것으로 나타났다.

반응표면 분석법을 이용한 치아씨 항산화 활성 추출의 최적화 조건 (Optimization for Chia Seed Antioxidative Activity of Solvent Extraction Using the Response Surface Methodology)

  • 본 연구에서는 치아씨의 항산화 활성을 증가시키기 위하여 추출용매 종류와 용매농도, 추출시간과 추출온도를 달리하여 총 폴리페놀 함량과 DPPH 라디칼 소거능을 측정한 후 반응표면분석을 이용하여 최적조건을 수립하였다. 치아씨 추출조건은 에탄올과 메탄올용매 모두에서 추출용매 60%, 추출시간 130분과 추출온도 $20^{\circ}C$에서 총 폴리페놀 871.00mg%($R^2=0.9507$), 557.70mg%($R^2=0.9784$)로 가장 함량이 많았고, DPPH 라디칼 소거능은 72.14%($R^2=0.9675$), 52.79%($R^2=0.9524$)의 항산화력을 보였다. 용매종류에서는 에탄올 추출용매가 메탄올 추출용매보다 같은 조건에서는 월등히 높은 항산화력을 보여 에탄올이 치아씨 추출용매로 더 우수한 용매로 선정되었다. 또한 반응표면분석 결과, 용매추출 조건에서는 에탄올 용매에서는 용매추출 시간이나 온도보다는 용매농도에 가장 큰 영향을 받았고, 메탄올 추출조건에서는 용매추출 시간에서 가장 큰 영향력을 주었다. 반응표면분석 후 용매 농도, 시간과 온도의 최적 추출조건의 범위는 에탄올 추출조건은 에탄올농도 63%, 추출시간 100분과 추출온도 $18^{\circ}C$가 최적조건이고, 메탄올 추출조건에서는 추출농도 65%, 추출시간 120분과 추출온도 $16^{\circ}C$를 보였다.

마이크로웨이브 추출조건에 따른 곰취 추출물의 총 폴리페놀 함량 및 항산화작용의 변화 (Changes of Total Polyphenol Content and Antioxidant Activity of Ligularia fischeri Extracts with Different Microwave-Assisted Extraction Conditions)

  • 마이크로웨이브 추출방법과 환류 냉각 추출방법을 비교한 결과, 물과 에탄올의 혼합용매로 추출한 경우 마이크로웨이브 추출 방법에 의하여 추출시간을 단축시키면서 환류 냉각 추출 방법에서와 같은 수준의 가용성 고형분 및 총 폴리페놀 함량을 갖는 곰취 추출물을 얻을 수 있었다. 마이크로웨이브 추출시 최적 마이크로웨이브 에너지는 120∼150 W 였고 추출시간은 4∼8분이 적당하였다. 추출에 사용한 용매들 가운데 에탄올, 메탄올 보다 물 그리고 물과 에탄올 또는 메탄올 혼합용매를 사용한 추출물의 가용성 고형분, 총 폴리페놀 함량 및 항산화 효과가 높은 것으로 나타났다.
