Diffusion Weighted Imaging Can Distinguish Benign from Malignant Mediastinal Tumors and Mass Lesions: Comparison with Positron Emission Tomography

  • Background: Diffusion-weighted magnetic resonance imaging (DWI) makes it possible to detect malignant tumors based on the diffusion of water molecules. It is uncertain whether DWI is more useful than positron emission tomography-computed tomography (PET-CT) for distinguishing benign from malignant mediastinal tumors and mass lesions. Materials and Methods: Sixteen malignant mediastinal tumors (thymomas 7, thymic cancers 3, malignant lymphomas 3, malignant germ cell tumors 2, and thymic carcinoid 1) and 12 benign mediastinal tumors or mass lesions were assessed in this study. DWI and PET-CT were performed before biopsy or surgery. Results: The apparent diffusion coefficient (ADC) value ($1.51{\pm}0.46{\times}10^{-3}mm^2/sec$) of malignant mediastinal tumors was significantly lower than that ($2.96{\pm}0.86{\times}10^{-3}mm^2/sec$) of benign mediastinal tumors and mass lesions (P<0.0001). Maximum standardized uptake value (SUVmax) ($11.30{\pm}11.22$) of malignant mediastinal tumors was significantly higher than that ($2.53{\pm}3.92$) of benign mediastinal tumors and mass lesions (P=0.0159). Using the optimal cutoff value (OCV) $2.21{\times}10^{-3}mm^2/sec$ for ADC and 2.93 for SUVmax, the sensitivity (100%) by DWI was not significantly higher than that (93.8%) by PET-CT for malignant mediastinal tumors. The specificity (83.3%) by DWI was not significantly higher than that (66.7%) for benign mediastinal tumors and mass lesions. The accuracy (92.9%) by DWI was not significantly higher than that (82.1%) by PET-CT for mediastinal tumors and mass lesions. Conclusions: There was no significant difference between diagnostic capability of DWI and that of PET-CT for distinguishing mediastinal tumors and mass lesions. DWI is useful in distinguishing benign from malignant mediastinal tumors and mass lesions.

  • 자기지전류(MT) 자료의 3차원 역산에 대해 소개한다. MT 자료의 역산 문제는 기본적으로 악조건이므로 유일한 해가 존재하지 않는다. 이러한 비유일성을 줄이고 정확한 역산해를 구하기 위해서는 역산 시 사전정보를 추가하는 제약조건을 가해야 한다. 물리탐사 분야에서 비선형 역산에 사용되는 가장 일반적인 방법은 일련의 선형화된 역산문제를 푸는 Gauss-Newton법이다. 이 알고리듬은 수렴 시, 모델 공간에서 역산문제에 대한 목적함수를 최소화하는 최적해를 준다. 그러나 이러한 반복적 선형화기법은 3차원 MT 역산의 경우 Jacobian 행렬을 구하기 힘들기 때문에 그 유용성에 한계가 있다. 이러한 어려움은 CG법에 의해 완화할 수 있다. 선형 CG법은 Gauss-Newton 반복의 각 단계를 근사적으로 풀기 위해서 사용된다. 한편 비선형 CG법은 목적함수의 최소화에 직접적으로 적용된다. 이들 CG법은 Jacobian 행렬의 계산 및 대형 선형방정식의 해를 반복 당 세 번의 모델링으로 대치할 수 있어서 3차원 역산에 적합하다.

인공신경망과 SA 알고리즘을 이용한 지능형 생산정 위치 최적화 전산 모델 개발 (Development of Well Placement Optimization Model using Artificial Neural Network and Simulated Annealing)

  • 본 연구에서는 고속의 연산이 가능한 인공신경망 시뮬레이터와 SA 알고리즘을 결합하여 지능형 생산정 위치 최적화 전산 모델을 개발하였다. 기존의 사용하는 상용시뮬레이터의 경우 현장 규모의 저류 전산 시뮬레이션을 수행시 시간이 많이 소모되므로 이를 해결하기 위하여 이 모델에서는 인공신경망을 사용하여 짧은 시간내에 시뮬레이션을 수행할 수 있도록 하였다. 이렇게 얻은 결과를 주관적인 경험에 의거하지 않고 자동으로 최적의 생산정 위치를 선정할 수 있도록 최적화기법인 SA 알고리즘을 적용하였다. 개발된 모델을 사용하여 얻은 결과를 기존 사용 시뮬레이터와 비교하여 예측성능이 양호함을 검증할 수 있었으며, 연산속도 또한 향상됨을 확인하였다. 특히 SA 최적화 알고리즘의 제어변수인 초기온도와 냉각률에 대한 민감도분석을 실시하여 각각에 대한 최적값을 산출하였으며, 이를 통해 개발한 모델의 연산성능을 향상시킬 수 있었다. 마지막으로 개발된 모델을 사용하여 생산정 위치 최적화를 수행한 결과, 생산성이 우수한 지역을 선정하여 최적의 생산정 위치를 도출하였다.

Diffusion-Weighted Imaging for the Left Hepatic Lobe has Higher Diagnostic Accuracy for Malignant Focal Liver Lesions

  • Background: This study was conducted to investigate whether apparent diffusion coefficient (ADC) measurements by dividing the liver into left and right hepatic lobes may be utilized to improve the accuracy of differential diagnosis of benign and malignant focal liver lesions. Materials and Methods: A total of 269 consecutive patients with 429 focal liver lesions were examined by 3-T magnetic resonance imaging that included diffusion-weighted imaging. For 58 patients with focal liver lesions of the same etiology in left and right hepatic lobes, ADCs of normal liver parenchyma and focal liver lesions were calculated and compared using the paired t-test. For all 269 patients, ADC cutoffs for focal liver lesions and diagnostic accuracy in the left hepatic lobe, right hepatic lobe and whole liver were evaluated by receiver operating characteristic curve analysis. Results: For the group of 58 patients, mean ADCs of normal liver parenchyma and focal liver lesions in the left hepatic lobe were significantly higher than those in the right hepatic lobe. For differentiating malignant lesions from benign lesions in all patients, the sensitivity and specificity were 92.6% and 92.0% in the left hepatic lobe, 94.4% and 94.4% in the right hepatic lobe, and 90.4% and 94.7% in the whole liver, respectively. The area under the curve of the right hepatic lobe, but not the left hepatic lobe, was higher than that of the whole liver. Conclusions: ADCs of normal liver parenchyma and focal liver lesions in the left hepatic lobe were significantly higher than those in the right hepatic lobe. Optimal ADC cutoff for focal liver lesions in the right hepatic lobe, but not in the left hepatic lobe, had higher diagnostic accuracy compared with that in the whole liver.

Association of the A-G Polymorphism in Porcine Adiponectin Gene with Fat Deposition and Carcass Traits

  • The adiponectin gene is known to be involved in the regulation of energy homeostasis involving food intake, carbohydrate and lipid metabolism. Human adiponectin gene polymorphisms have been recently reported to be associated with obesity, insulin sensitivity and the risk of type 2 diabetes. The present study was carried out to investigate the porcine adiponectin gene as a candidate gene for fat deposition and carcass traits. A mutation of A178G of the porcine adiponectin gene that resulted in substitution of the amino acid Isoleucine to Valine was identified. AcyI PCR-RFLP was used to detect the polymorphism of the genotypes in five different pig populations (Large White, Landrace, Duroc, Chinese breeds Meishan and Qingping). The A allele frequency was significantly higher among subjects from Chinsese lard type breeds, while the G allele was the only one present in those from Western lean type breeds. To determine if there was an association of the polymorphism with phenotypic variation, the mutation was tested in 267 pigs of the "Large $White{\times}Meishan$" F2 resource population. The results of association analyses showed significant associations of the genotypes with fat deposition and carcass traits. Allele G was significantly associated with increase in loin eye height, loin eye area and lean meat percentage and bone percentage, and decrease in fat mean percentage, ratio of lean to fat, shoulder fat thickness, 6-7 rib fat thickness, thorax-waist fat thickness and buttock fat thickness. The substitution of A178G (Ile60Val) happened to be located at amino acid 60 in the collagenous domain of porcine adiponectin which might affect the association into higher-order structures, and accordingly affect the posttranslational modifications and optimal biological activity of the multimeric forms. The identified functional polymorphism provides new evidence of adiponectin as an important candidate gene affecting fat deposition and carcass traits in pigs.

Application of Joint Detection of AFP, CA19-9, CA125 and CEA in Identification and Diagnosis of Cholangiocarcinoma

  • Objective: To explore the application of joint detection of serum AFP, CA19-9, CA125 and CEA in identification and diagnosis of cholangiocarcinoma (CC). Materials and Methods: The levels of serum AFP, CA19-9, CA125 and CEA of both 30 patients with CC and 30 patients with hepatocellular carcinoma (HCC) were assessed. Receiver operating characteristic (ROC) curves were used to evaluate the diagnostic effects of single and joint detection of those 4 kinds of tumor markers for CC. Results: The levels of serum CA19-9, CA125 and CEA in CC patients were higher than that in HCC patients,whereas that of serum AFP was significantly lower s. The area under ROC curve of single detection of serum AFP, CA19-9, CA125 and CEA were 0.05, 0.86, 0.84 and 0.83, with the optimal cutoff values of 15.4 ng/ml, 125.1 U/ml, 95.7 U/ml and 25.9 ng/ml, correspondingly, and the percentage correct single diagnosis was <79%. With joint detection, the diagnostic effect of combined AFP, CA19-9, CA125 and CEA was the highest, with an area under the ROC curve of 0.94 (95%CI 0.88~0.99). Conclusions: Single detection of serum CA19-9, CA125 and EA is not meaningful. The sensitivity, specificity, the rate of correct diagnosis and the area under ROC curve of joint detection of AFP, CA19-9, CA125 and CEA are highest, indicating that the joint detection of these 4 tumor markers is of great importance in the diagnosis of CC.

차량용 레인센서에서 생성된 센서시그널을 이용한 강우량 측정 (Measurement of Rainfall using Sensor Signal Generated from Vehicle Rain Sensor)

  • 본 연구에서는 차량용 레인센서를 이용하여 고해상도의 강우관측을 위한 관계식을 개발하였다. 차량용 레인센서는 8개의 채널로 이루어져 있으며, 각 채널은 강우발생시 차량의 전면유리창에 내리는 우적량을 감지하여 센서시그널을 생성하는데, 강우량이 높을수록 센서시그널은 낮은 값으로 형성된다. 레인센서에서 생성되는 센서시그널의 이러한 특징을 이용하여 관계식을 개발하였다. 특정강우를 발생시키기 위하여 인공강우 발생장치를 제작하였으며, 인공강우발생장치에서 분사되는 강우량의 변화에 따른 센서시그널의 변화 값을 분석하였다. 이 중 민감도 분석을 통해 다양한 강우량을 잘 반영하는 최적의 센서 채널을 선정하였다. 선정된 채널을 이용하여 5분 단위 센서시그널를 생성하였고 생성된 5분 단위 센서시그널의 대표 값을 평균, 25분위, 50분위, 75분위로 설정하여 관계식을 구축하였다. 구축된 관계식을 이용하여 실강우 데이터에 적용하여 강우량 값을 환산하였고, 환산된 강우량은 지상강우관측소에서 관측된 강우량 값과의 비교를 통해 관계식의 신뢰도를 검증하였다. 검증결과 데이터의 이상치가 발견되어 관계식의 신뢰도는 다소 떨어졌지만, 해당 잔차 범위의 실험 데이터가 부족한 것으로 판단되었다. 개발된 관계식을 실강우에 적용시켜 강우량 값을 환산한 하였고, 신뢰도 검증을 위해 동시간에 관측된 지상강우 관측 장비에서 생성된 강우량 값과 비교를 하였고 관측 결과 레인센서는 0.5mm 이하의 미세한 강우량까지 측정하였고 평균 관측 오차는 0.36mm로 나타났다.

염화비닐의 요중 대사물질인 thiodiglycolic acid의 분석을 위한 전처리 조건 (Pretreatment method of urinary thiodiglycolic acid as metabolite of vinyl chloride)

  • The analysis of thiodiglycolic acid in urine has been used as an index of biological exposure to vinyl chloride. Unfortunately thiodiglycolic acid has a strong hydrophilic character, because it has two carboxylic groups, so that it can only be extracted with organic solvent with a great difficulty. Underivatized thiodiglycolic acid tends to tail because of non-specific interaction with the inert support. Therefore, esterification is the obvious first choice for derivatization of thiodiglycolic acid, particularly for gas chromatography. In this study, the focus of interest is to compare two method of esterifications (methylation and silylation). Methylation is to make the methyl ester of thiodiglycolic acid by reaction with diazomethane. Silylation is to make the trimethylsilyl ester of thiodiglycolic acid by reaction with N-trimethylsily-ldiethylamine. The results and conclusions are as the following: 1. The detection limit (sensitivity) of methylated thiodiglycolic acid was $5.00{\mu}g/m{\ell}$ and silylated thiodiglycolic acid was $3.07{\mu}g/m{\ell}$ by gas chromatography with flame ionization detector. 2. The optimal liquid-liquid extraction of thiodiglycolic acid was as following: To each of the tubes, $15m{\ell}$ of urine, concentrated sulfuric acid (pH 1 - 2) and 5 gsodium sulfate were added. The samples was extracted three times with $5m{\ell}$ ethylacetate each time. 3. The methylated thiodiglycolic acid was more stable than silylated thiodiglycolic acid in extractional solvent which contained humidity. 4. The precision (pooled coefficient of variation for 4 days) of the analysis was 0.07324 in methylated thiodiglycolic acid with external standard calibration, and 0.07033 in methylated thiodiglycolic acid with internal standard calibration. 5. The precision (pooled coefficient of variation for 4 days) of the analysis was 0.10914 in silylated thiodiglycolic acid with external standard calibration, and 0.13602 in silylated thiodiglycolic acid with internal standard calibration. From the above results, the analysis of methylated thiodiglycolic acid was more sensitive (limit of detection) than silylated thiodiglycolic acid by gas chromatography. However, the methylated thiodiglycolic acid was stable in the humidity and was separated sharply on chromatogram. Also, analysis of methylated thiodiglycolic acid was more precise (pooled coefficient of variation for 4 days) than silylated thiodiglycolic acid. In conclusion, it is established that the analysis of methylated thiodiglycolic acid is appropriate for biological monitoring of exposure to vinyl chloride.

NPTII 및 HPT 유전자가 삽입된 현사시의 항생제에 대한 저항성 분석 (Comparative Analysis of Resistance to Antibiotics in Populus alba$\times$P. glandulosa Transformed by nptII or hpt Gene)

  • 본 실험은 nptII 및 hpt 유전자가 삽입된 현사시나무를 이용하여 각 항생제 저항성 유전자로 형질전환된 세포의 효율적인 선발 조건을 규명하기 위하여 수행되었다. 액아가 포함된 줄기절편과 잎절편 조직의 생장, 발근 유도, 캘러스 유도로 항생제에 대한 감수성에 대한 효과를 검정하였다. 형질전환되지 않은 대조식물체의 잎절편을 이용한 경우 50 mg/L의 kanamycin이나 2 mg/L hygromycin으로 캘러스 유도 및 생장을 억제할 수 있었으나 형질전환된 식물은 100 mg/L kanamycin, 50 mg/L hygromycin에서도 왕성한 생장을 나타냈다. 절간조직의 개아의 경우 100 mg/L kanamycin, 5 mg/L hygromycin으로 줄기신장을 완전히 억제 가능하였으며, 뿌리유도는 50 mg/L kanamycin, 5 mg/L hygromycin에서 선발이 가능하였다. 형질전환체는 모두 이보다 높은 농도에서 왕성한 생장을 보였다. 형질전환이 되지 않은 세포의 생장을 억제하는 데는 hygromycin이 kanamycin보다 더 효율이 좋은 것으로 나타났다. 따라서 hpt유전자가 npt II 유전자보다 훨씬 강력한 선발표지임이 확인되었으며 엽절편의 캘러스 유도와 생장과 절간조직의 발근유도 모두 항생제에 예민하게 작용하여 형질전환체의 식별에 효과적으로 사용할 수 있음을 알 수 있었다.

  • SYMMV는 콩에서 빈번하게 발생하는 바이러스이며, 이 바이러스의 발생률은 계속해서 증가하고 있다. 본 연구에서는 콩에 발생하는 SYMMV를 신속하게 검출하기 위해서 RT-LAMP를 적용하였다. RT-LAMP 방법은 등온에서 단시간에 유전자 증폭이 가능하고, 전기영동 없이도 형광물질을 이용해 바이러스를 검출할 수 있는 이점이 있다. 프라이머는 SYMMV coat protein gene의 염기서열을 기반으로 4개의 프라이머를 설계하였다. 실험결과 SYMMV RT-LAMP는 $65^{\circ}C$에서 50분간 증폭시켰을 때 최적의 효율을 보였다. 또한, RT-LAMP와 RT-PCR과의 민감도를 비교한 결과 RT-LAMP 방법이 10-100배 정도 더 우수한 민감도를 가지는 것으로 밝혀졌다. 본 실험 결과를 토대로 기존의 진단법과 비교하여 높은 민감도와 짧은 소요시간에 이점이 있는 RT-LAMP는 SYMMV의 현장 및 연구실에서의 진단에 적용될 수 있을 것이라 생각된다.
