Single nucleotide polymorphisms (SNPs) are valuable DNA markers due to their abundance and potential for use in automated high-throughput genotyping. Numerous SNP genotyping assays have been developed. In this report, one of effective and high throughput SNP genotyping assays, which was named the template-directed dye-terminator incorporation with fluorescence polarization detection (FP-TDI) was described. Although the most of this assay succeed, the objective of this work was to determine the reasons for the failures, find ways to improve the assay and reduce the running cost. Ninety $F_2$-derived soybean, Glycine max (L.) Merr., RILs from a cross between 'Pureunkong' and 'Jinpumkong 2' were genotyped at four SNPs. FP measurement was done on $Victot^3$ microplate reader (perkinelmer Inc., Boston, MA, USA). Increasing the number of thermal cycles in the single-base extension step increased the separation of the FP values between the products corresponding to different genotypes. But in some assays, excess of heterozygous genotypes was observed with increase of PCR cycles. We discovered that the excess heterozygous was due to misincorporation of one of the dyeterminators during the primer extension reaction. After pyrophosphatase incubation and thermal cycle control, misincoporation can be effectively prevented. Using long amplicons instead of short amplicons for SNP genotyping and decreasing the amount of dye terminator and Acyclopol Taq polymerase to 1/2 or 1/3 decreased the cost of the assay. With these minor adjustments, the FP-TDI assay can be used more accurately and cost-effectively.
Three dimensional thermal cycle analysis of the plunger is carried out in repeated forming process of the TV glass, which is continued work of two dimensional analysis where an efficient method has been proposed. The plunger undergoes temperature fluctuation during a cycle due to the repeated contact and separation from the glass, which attains a cyclic steady state having same temperature history at every cycle. Straightforward analysis of this problem brings about more than 90 cycles to get reasonable solution. An exponential function fitting method is proposed, which finds exponential function to best approximate temperature values of 3 consecutive cycles, and new cycle is restarted with the fitted value at infinite time. Number of cases are analyzed using the proposed method and compared to the result of straightforward repetition, from which one finds that the method always reaches nearly convergent solution within $9{\sim}12$ cycles, but turns around afterwards without further convergence. Two step use is found most efficient, in which the exponential fitting is carried out fer the first 12 cycles, followed by simple repetition, which shows fast convergence expending only 6 additional cycles to get the accuracy within 2 error. This reduces the computation cycle remarkably from 90 to 18, which is 80% reduction. From the parametric studies, one reveals that the overall thermal behavior of the plunger in terms of cooling parameters and time is similar to that of 2 dimensional analysis.

In order to investigate the initiation and propagation processes of a spherical detonation wave induced by direct initiation, numerical simulations were carried out using two-dimensional compressible Euler equations with an axisymmetric assumption and a one-step reaction model based on Arrhenius kinetics with various levels of ignition energy. By varying the amount of ignition energy, three typical initiation behaviors, which were subcritical, supercritical and critical regimes, were observed. Then, the ignition energy of more than $137.5{\times}10^6$ in non-dimensional value was required for initiating a spherical detonation wave, and the minimum ignition energy(i.e., critical energy) was less than that of the one-dimensional simulation reported by a previous numerical work. When the ignition energy was less than the critical energy, the blast wave generated from an ignition source continued to attenuate due to the separation of the blast wave and a reaction front. Therefore, detonation was not initiated in the subcrtical regime. When the ignition energy was more than the minimum initiation energy, the blast wave developed into a multiheaded detonation wave propagating spherically at CJ velocity, and then a cellular pattern radiated regularly out from the ignition center in the supercritical regime. The influence on ignition energy was observed in the cell width near the ignition center, but the cell width on the fully developed detonation remained constant during the expanding of detonation wave due to the consecutive formation of new triple points, regardless of ignition energy. When the ignition energy was equal to the critical energy, the decoupling of the blast wave and a reaction front appeared, as occurred in the subcrtical regime. After that, the detonation bubble induced by the local explosion behind the blast wave expanded and developed into the multiheaded detonation wave in the critical regime. Although few triple points were observed in the vicinity of the ignition core, the regularly located cellular pattern was generated after the onset of the multiheaded detonation. Then, the average cell width on the fully developed detonation was almost to that in the supercritical regime. These numerical results qualitatively agreed with previous experimental works regarding the initiation and propagation processes.
The purpose of this study was to classify the types of tweezers used across time and to examine the social culture of periods through the use of tweezers. Chapter II summarized the terms for tweezers and looked at the use of tweezers in two cases related to the social culture and politics of a period. The first is the use of tweezers for beauty purposes. Men used tweezers as a kind of self-management to maintain their social status and power, and thus they helped develop a wide range of tweezers cultures. People with gray hair are usually old. The perception that we should step down from politics when we are aged has become strong. Therefore, politicians pulled out gray hairs with tweezers to maintain only black hair and show youth, which indicates that their social influence is still strong. The second is the use of tweezers for first aid. Chapter III classified tweezers by type during the Goryeo and Joseon periods. The tweezers are largely divided into basic and composite types. The basic type of tweezers is a form that has been continued since the Three Kingdoms Period and consists of tongs and handles, and a fixed type of tweezers with a fixed ring appeared. Composite tweezers are made by grafting earpick or multipurpose knives. Composite tweezers are all-in-one tweezers with an earpick and an all-in-one knife. Tweezers are usually all-in-one. Among the composite tweezers, rivet separation and ring separation are characterized by separation, unlike the all-in-one tweezers. The method of connecting is divided into rivets and connection rings. The all-in-one tweezers appear only during the Goryeo Dynasty and are characterized by the lifestyle that provides a glimpse of the tastes of contemporary users. The manufacturing takes shape after making a thin metal plate. Decorative techniques are carved on soft metal tweezers, such as silver and bronze with a line, point, and a pressed angle. These tweezers are presumed to have been used by the royal family or aristocrats. However, most tweezers are made of strong bronze or iron. Therefore, the majority of simple X-shaped patterns are sampled or without patterns. The biggest reason why there are such diverse types of tweezers is that the culture of tweezers was widespread regardless of the times. In addition, the basic type of tweezers has been used since the Three Kingdoms Period and has been modified and used together as necessary because the shape of tweezers is a practical daily tool. Study of metal crafts have been limited to royal objects and Buddhist crafts. We hope that research on everyday tools such as tweezers will continue to serve as an opportunity to examine the social and cultural aspects of the times in various ways.

Trip generation is the first step in the conventional four-step model and has great effects on overall demand forecasting, so accuracy really matters at this stage. A linear regression model is widely used as a current trip generation model for such plans as urban transportation and SOC facilities, assuming that the relationship between each socio-economic index and trip generation stays linear. But when rapid urban development or an urban planning structure has changed, socio-economic index data for trip estimation may be lacking to bring many errors in estimated trip. Hence, instead of assuming that a socio-economic index widely used for a general purpose, this study aims to develop a new trip generation model by type based on the market separation for the variables to reflect the characteristics of various zones. The study considered the various characteristics (land use, socio-economic) of zones to enhance the forecasting accuracy of a trip generation model, the first-step in forecasting transportation demands. For a market separation methodology to improve forecasting accuracy, data mining (CART) on the basis of trip generation was used along with a regression analysis. Findings of the study indicated as follows : First, the analysis of zone characteristics using the CART analysis showed that trip production was under the influence of socio-economic factors (men-women relative proportion, age group (22 to 29)), while trip attraction was affected by land use factors (the relative proportion of business facilities) and the socio-economic factor (the relative proportion of third industry workers). Second, model development by type showed as a result that trip generation coefficients revealed 0.977 to 0.987 (trip/person) for "production" 0.692 to 3.256 (trip/person) for "attraction", which brought the necessity for type classifications. Third, a measured verification was conducted, where "production" and "attraction" showed a higher suitability than the existing model. The trip generation model by type developed in this study, therefore, turned out to be superior to the existing one.
Distillation columns are one of the main equipment used for the separation of chemical components in petrochemical process design. However, in spite of the efficient operation in wide range, and the advantage of data collection for equipment verification, the distillation columns are inherently known for high energy consumption and capital cost. Hence, the trade-off analysis needs to be done between investment cost and operation cost to develop the most economical distillation columns. This study was conducted using Aspen Plus, a popular process simulation program, in the pursuit of broad application by as many process engineers as possible. In this paper, design variables for optimization of distillation columns were defined to improve emphatically the design quality with reducing erratic practice of many engineers. In addition, by eliminating unnecessary reviewing step and establishing systematic and efficient procedures, the amount of time for design and human resources were minimized. Aspen Process Economic Analyzers (APEA) program was introduced in order to calculate the investment cost reliably, and the efficient systematic procedure for utilization of APEA was established.
Laboratory experiments were carried out to investigate the performance of anaerobic sequencing batch reactor(ASBR) for digestion of a municipal sludge. Each cycle of the ASBR comprised feeding, two-or three-day reaction, one-day thickening, and withdrawal. The reactors were operated at an HRT of 10days and 5days with an equivalent organic loading rate of 0.8-1.54 gVS/l/d, 1.81-3.56 gVS/l/d at 35$\circ$C, respectively. Solids accumulation was remarkable in the ASBR during start-up period, and directly affected by settleable solids in the feed sludge. Floatation thickening occured in the ASBRs, and Solids profiles at the end of thickening step dramatically changed at solid-liquid interface. Slight difference in solids concentrations was observed within thickened sludge bed. Efficiencies through floatation thickening were comparable to that of additional thickening of the completely mixed control reactor. Average solids concentrations in the ASBRs were 2.2-2.6 times higher than that in the control throughout the total operation period. The dehydrogenase activity had a strong correlation with the solids concentration. Organics removals based on clarified effluent of the ASBRs were consistently above 86%. Remarkable increase in equivalent gas production of 27-52% was observed at the ASBRs compared with the control though the control and ASBRs showed similiar effluent quality. Thus, digestion of a municipal sludge was possible using the ASBR in spite of high concentration of solids in the sludge.
Aluminum can to can recycling was divided into two stpes. The first step was composed of the processes such as collection of used beverage cans (UBC), shredding, magnetic separation, De-laquiring, melting and casting. The second one was remelting and casting, heat treating, hot and cold rolling, annealing, and can making. In this study, the effect of alloying elements on the microstructure and texture of the secondary ingots made by Al UBC was investigated. In aluminum can to can recycling, the second phase particles appeared in the solidification stage must be controlled by heat treatment. The optimum heat treatment condition was $615^{\circ}C$ for 5hrs. the texture in hot rolled sheet was depressed with increasing Mn content, on the other hand, Si and Fe elements promoted the texture development. The textures of can-body sheet should be controlled in the hot rolling and annealing stage because can was formed from cold rolled sheet without heat treatment.
To address organic dye wastewater, economic and effective adsorbents are required. Here, magnetic biochar from alkali-activated rice straw (AMBC) was successfully synthesized using one-step magnetization and carbonization method. The alkaline activation caused the large specific surface area, high pore volume and abundant oxygen-containing groups of the AMBC, and the magnetization gave the AMBC a certain degree of electropositivity and fast equilibrium characteristics. These characteristics collectively contributed to a relative high adsorption capacity of 53.66 mg g-1 for this adsorbent towards rhodamine B (RhB). In brief, RhB can spontaneously adsorb onto the heterogeneous surface of the AMBC and reach the equilibrium in 60 min. Although the initial pH, ionic strength and other substances of the solution affected the adsorption performance of the AMBC, it could be easily regenerated and reused with considerable adsorption content. Based on the results, H-bonds, π-π stacking and electrostatic interactions were speculated as the primary mechanisms for RhB adsorption onto the AMBC, which was also demonstrated by the FTIR analysis. With the advantageous features of low cost, easy separation, considerable adsorption capacity and favorable stability and reusability, the AMBC would be a potential adsorbent for removing organic dyes from wastewater.

In this paper, a flight test method is described for the system identification of the unmanned aerial vehicle equipped with an automatic flight control system. Multistep inputs are applied for both longitudinal mode and lateral/directional excitation. Optimal time step for excitation is sought to provide the broad input bandwidth. A programmed mode flight test method provides high-quality flight data for system identification using the flight control computer with the longitudinal and lateral/directional autopilot which enables the separation of each motion during the flight test. In addition, exact actuating input that is almost equivalent to the designed one guarantees the highest input frequency attainable. Several repetitive flight tests were implemented in the calm air in order to extract the consistent system model for the air vehicle. The enhanced airborne data acquisition system endowed the high-quality flight data for the system identification. The flight data were effectively used to the system identification of the unmanned aerial vehicle.
