Construction of a Large Synthetic Human Fab Antibody Library on Yeast Cell Surface by Optimized Yeast Mating

  • Yeast surface-displayed antibody libraries provide an efficient and quantitative screening resource for given antigens, but suffer from typically modest library sizes owing to low yeast transformation efficiency. Yeast mating is an attractive method for overcoming the limit of yeast transformation to construct a large, combinatorial antibody library, but the optimal conditions have not been reported. Here, we report a large synthetic human Fab (antigen binding fragment) yeast surface-displayed library generated by stepwise optimization of yeast mating conditions. We first constructed HC (heavy chain) and LC (light chain) libraries, where all of the six CDRs (complementarity-determining regions) of the variable domains were diversified mimicking the human germline antibody repertoires by degenerate codons, onto single frameworks of VH3-23 and $V{\kappa}1$-16 germline sequences, in two haploid cells of opposite mating types. Yeast mating conditions were optimized in the order of cell density, media pH, and cell growth phase, yielding a mating efficiency of ~58% between the two haploid cells carrying HC and LC libraries. We constructed two combinatorial Fab libraries with CDR-H3 of 9 or 11 residues in length with colony diversities of more than $10^9$ by one round of yeast mating between the two haploid HC and LC libraries, with modest diversity sizes of ${\sim}10^7$. The synthetic human Fab yeast-displayed libraries exhibited relative amino acid compositions in each position of the six CDRs that were very similar to those of the designed repertoires, suggesting that they are a promising source for human Fab antibody screening.

Remineralization effect of bamboo salt on incipient subsurface caries enamel (초기우식법랑질 표면 하방에 미치는 죽염의 재광화 효과)

  • Objectives: The aim of this study was to investigate the effects of bamboo salts on remineralization effects on subsurface of artificial carious enamel. Methods: Incipient carious enamel was formed in permanent bovine incisors and then specimens were divided into three groups randomly: 3% bamboo salt (BS), 2% (NaF) and the solution of mixed 3% BS and 2% NaF. For remineralization, specimens of each of the three groups were treated for 24 hrs at $37^{\circ}C$ incubator. After treatment, specimens were analyzed using SEM and VHN. Statistical analysis used was one-way ANOVA. Results: In SEM observation, the BS group showed narrower distances between enamel rods than the cases of incipient subsurface caries enamel. The NaF and BS+NaF groups showed that the enamel rods near the surface were destructed, and innumerable round small particles were deposited near the surface of enamel. The BS+NaF group showed more minerals attachment between enamel rods than the cases of other groups. The differences in subsurface microhardness (${\Delta}VHN$) increased all of three groups in total by $80{\mu}m$. The ${\Delta}VHN$ of the BS+NaF group increased significantly more than NaF and BS groups in depth of $50{\mu}m$, $80{\mu}m$. Conclusions: The 3% bamboo salt with 2% NaF solution was found to increase subsurface hardness of incipient caries enamel. Thus, bamboo salt will be used to contribute to prevention on dental caries.

Scientific Conservation of Seodae of the Chuncheon National Museum (국립춘천박물관 소장 서대(犀帶)의 과학적 보존)

  • Here is an introduction of the conservation and making process of Seodae(Rank Belt with A Rhinoceros' Horn) of the Chuncheon National Museum. The seodae has a structure composed of wrapping fabric around a leather frame and decorated with ttidon(an ornament) made of a rhinoceros' horn and a metal frame. The adhesion between leather was weak and separation was taking place, parts of the horn were missing and the fabric was discoloring. Also the metal frame and the metal wire attaching ttidon were severly corroded. Nondestructive analysis was carried out by using a portable x-ray fluorescence spectroscopy to analyze the components of the metal parts. Conservation was carried out by dismantling parts and carrying out conservation process of each one according to material, then the parts were put back together.

RFID Tag Number Estimation and Query Time Optimization Methods (RFID 태그 개수 추정 방법 및 질의 시간 최소화 방안)

  • An RFID system is an important technology that could replace the traditional bar code system changing the paradigm of manufacturing, distribution, and service industry. An RFID reader can recognize several hundred tags in one second. Tag identification is done by tags' random transmission of their IDs in a frame which is assigned by the reader at each round. To minimize tag identification time, optimal frame size should be selected according to the number of tags. This paper presents new query optimization methods in RFID systems. Query optimization consists of tag number estimation problem and frame length determination problem. We propose a simple yet efficient tag estimation method and calculate optimal frame lengths that minimize overall query time. We conducted rigorous performance studies. Performance results show that the new tag number estimation technique is more accurate than previous methods. We also observe that a simple greedy method is as efficient as the optimal method in minimizing the query time.

  • Ceramic Injection Molding (CIM) is one of wide used processes in industry field and the applications are gradually being expanded to parts of medical and electric devices. In this study, the CIM process were analyzed with FEM and process parameters were studied and analyzed the effect on product quality. The shape of simple flat plate was compared to the shapes with the hole, with the round corner portion or with the side wall portion. If there are holes then the hole around the uneven density distribution and the defects such as weld lines could be occurred. The Large radius of the corners of the product give good formability and fluidity. Not only the shape parameters of product but also the process parameters during CIM are studied. The simulation results showed that the process parameters of temperature, initial fractions and velocity are important design parameters to improve the quality of products.

  • Engine control/performance model for helicopter simulator if one of the most important models which affect flight performance and handling quality. It is typical to develop the model based on the raw data and models from the engine designers/manufacturers. The approaches in this study were to develop the basic model bated on the available resources and to tune and verify it based on the ground/flight test results. The maintenance manuals of TB3-117 which is installed in KA-327 were reviewed and the components to be simulated for the engine control model were categorized and modeled. Piece-wise linear modeling method was used for the engine performance model. The engine performance data in the engine maintenance manuals were incorporated into the engine steady state performance tablet, which were incorporated with the transfer functions for the dynamic performance. Engine control/performance model was compared and tuned with the round/flight test results. It was verified that the fidelity of the model was within the tolerances in FAA AC120-63.

Design of Optimized ARIA Crypto-Processor Using Composite Field S-Box (합성체 S-Box 기반 최적의 ARIA 암호프로세서 설계)

  • Conventional ARIA algorithm which is used LUT based-S-Box is fast the processing speed. However, the algorithm is hard to applied to small portable devices. This paper proposes the hardware design of optimized ARIA crypto-processor based on the modified composite field S-Box in order to decrease its hardware area. The Key scheduling in ARIA algorithm, both diffusion and substitution layers are repeatedly used in each round function. In this approach, an advanced key scheduling method is also presented of which two functions are merged into only one function for reducing hardware overhead in scheduling process. The designed ARIA crypto-processor is described in Verilog-HDL, and then a logic synthesis is also performed by using Xilinx ISE 14.7 tool with target the Xilnx FPGA XC3S1500 device. In order to verify the function of the crypto-processor, both logic and timing simulation are also performed by using simulator called ModelSim 10.4a.

Mineral Uptake and Soluble Carbohydrates of Tomato Plants as Affected by Air Temperatures and Mineral Treatment Levels

  • Both low and high temperatures affect plant growth and development at whole plant level, tissue and even cell level through a variety of metabolic changes. Temperature stress is one of frequently occurring problems in greenhouse crops in summer and winter seasons due to the wide-spread year-round cultivation. In the present study, we investigated the extent of the inhibition of growth, macro-element uptake and soluble carbohydrate production, and the effect of extra-supply of minerals as a means of the recovery from temperature damage. Tomato plants were grown five different growth temperatures (15/8, 20/13, 28/21, 33/23 and $36/26^{\circ}C$), and extra-supply of minerals was composed of 1.5- and 2.0-fold stronger than the standard nutrition (1/2 strength of Hoagland's solution). Temperature stress significantly adversely affected tomato growth and mineral uptake, whereas soluble carbohydrate accumulation represented temperature-dependent response, more accumulation at low temperature and more consumption at high temperature. The soluble sugars in leaves and stems were mostly declined with the supply of extra-minerals at low and optimal temperatures, whereas remained unchanged at high temperature. The starch levels also remained unchanged or slightly decreased.

Effect of the Crucible Cover on the Distillation of Cadmium

  • The distillation of liquid cathode is necessary to separate cadmium from the actinide elements in the pyroprocessing since the actinide deposits are dissolved or precipitated in a liquid cathode. It is very important to avoid a splattering of cadmium during evaporation due to the high vapor pressure. Several methods have been proposed to lower the splattering of cadmium during distillation. One of the important methods is an installation of crucible cover on the distillation crucible. A multi-layer porous round cover was proposed to avoid a cadmium splattering in our previous study. In this study, the effect of crucible cover on the cadmium distillation was examined to develop a splatter shield. Various surrogates were used for the actinides in the cadmium. The surrogates such as bismuth, zirconia, and tungsten don't evaporate at the operational temperature of the Cd distiller due to their low vapor pressures. The distillation experiments were carried out in a crucible equipped with cover and in a crucible without cover. About 40 grams of Cd was distilled at a reduced pressure for two hours at various temperatures. The mixture of the cadmium and the surrogate was heated at $470{\sim}620^{\circ}C$. Most of the bismuth remained in the crucible equipped with cover after distillation under $580^{\circ}C$ for two hours, whereas small amount of bismuth decreased in the crucible without cover above $580^{\circ}C$. The liquid bismuth escaped with liquid cadmium drop from the crucible without cover. It seems that the crucible cover played a role to prevent the splash of the liquid cadmium drop. The effect of the cover was not clear for the tungsten or zirconia surrogate since the surrogates remained as a solid powder at the experimental temperature. From the results of this work, it can be concluded that the crucible cover can be used to minimize the deposit loss by prevention of escape of liquid drop from the crucible during distillation of liquid cathode.

Convergence Study on Flow due to the Configuration of Bobsleigh (봅슬레이의 형상에 따른 유동에 관한 융합 연구)

  • The front bumper of bobsleigh is mounted to alleviate the impact, but the air resistance to the bobsleigh depends on the body shape positioned in front. This study was conducted the flow analyses about three kinds of bobsleigh configurations. Models B and C with the sharp type of the front can reduce the air resistance than model A with the round type of the front. And the type that the back of the bumper narrows can generate a flow smoother than the one widening. It is thought that the results of this study can be devoted at ensuring the body design to reduce the flow resistance most at bobsleigh. As the design data with the durability of bobsleigh obtained on the basis of this study result are utilized, the esthetic sense can be shown by being grafted onto the machine or structure at real life.
