Effects of autumn olive berry extract on insulin resistance and non-alcoholic fatty liver in high fructose-fed rat (고과당식이를 급여한 흰쥐에 있어서 토종보리수 추출물의 인슐린 저항성 및 비알콜성 지방간 개선 효과)

  • Purpose: Non-alcoholic fatty liver disease (NAFLD) is characterized by the accumulation of fat in the liver which is not a result of excessive alcohol consumption. Its global prevalence was estimated to be approximately 32% in the years 1994-2019. More than half of obese individuals and patients with diabetes are reported to have NAFLD as a comorbidity. This study aimed to investigate the impact of the autumn olive (Elaeagnus umbellata Thunb.) berry on insulin resistance and steatosis in rats fed a high-fructose diet. Methods: Six-week-old Wistar rats were divided into four groups. The control group received a diet consisting of 65% corn starch, while the fructose and experimental groups were fed a diet comprising 65% fructose (FRU) and an FRU diet containing 0.5% (low-dose autumn olive berry group; LAO) or 1.0% (high-dose autumn olive berry group; HAO) ethanol extract of autumn olive berry, respectively, for 10 weeks. Results: The HAO group exhibited significantly lower blood glucose levels compared to the fructose-fed group. Both the LAO and HAO groups showed a substantial reduction in serum insulin levels and insulin resistance when compared to the fructose-fed group. The consumption of LAO and HAO significantly ameliorated dyslipidemia and reduced the levels of triglycerides in the liver compared to the fructose-fed group. Additionally, the consumption of HAO resulted in lower serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities compared to the fructose group. The hepatic expression of the sterol regulatory element-binding protein-1c (SREBP-1c) and carbohydrate-responsive element-binding protein (ChREBP) was significantly reduced in the LAO and HAO groups compared to the fructose group. Conclusion: Autumn olive berries improved steatosis by ameliorating insulin resistance and down-regulating the lipogenesis proteins in rats fed on high fructose diet.

  • Full-length recombinant VP2 and VP3 proteins of aquabirnavirus isolated from olive flounder were expressed successfully in E. coli expression system. After rats were immunized with these proteins, antisera were used for in vitro and in vivo neutralization test. In in vitro test, VP2 antibody titers were higher than that of VP3. In in vivo assays, fish challenged with aquabirnavirus neutralized with VP2 antibody survived longer than other fish.

  • A biosurfactant producing bacteria strain, D2D2 was selected from diesel-contaminated soil, and identified as Pseudomonas aeroginosa. A glycolipid produced by P. aeroginosa D2D2 was purified by ethyl acetate extraction and adsorption chromatography. The biosurfactant was Identified as glycolipid which has two types of biosurfactants as a results of TLC analysis. The purified glycolipid biosurfactant reduced the surface tension of water to 27 dyne/cm. In time course studies of growth and rhamnolipid production in a minimal salts medium containing 1.5% glucose and 1.5% olive oil, a maximum rhamnolipid yield of $11.45gL^{-1}$ was obtained after 5 days.

  • Experiments were performed on rats to investigate the effect of doses of capsaicin on the immune response. Olive oil and the 0.3 mg, 1.0 mg and 3.0 mg/kg administration of capsaicin in olive oil were injected intraperitoneally every day for 4 weeks. Rats were sensitized and challenged with sheep red blood cells (S-RBC). Immune responses were evaluated by organ weight, HA and HY titer, Arthus reaction, delayed type hypersensitivity (DTH) and Rosette froming cell. Following results have observed. 1) The weight of spleen and thymus were increased by doses of compared with control group, but the body weight of rats was decreased. 2) HA titer, Arthus reaction and DTH were significntly decreased by doses of capsaicin as compared with control group. 3) Rosette forming cell in spleen cells was decreased according to the increase of capsaicin doses. These results suggested that high dose of capsaicin decrease humoral and cellular immune response in rats.

Changes in Steroid Hormones Levels of Olive Flounder, Paralichthys olivaceus Exposed to Phenanthrene

  • Phenanthrene, one of Polycyclic aromatic hydrocarbons with three aromatic rings, is a ubiquitous contaminant in the environment. Phenanthrene has been identified in ambient air, drinking water and sediment. We examined the effect of phenanthrene on steroid hormones level of olive flounder, Paralichthys olivaceus. Plasma testosterone level was increased significantly in fish exposed to phenanthrene($\geq1.0 \mu M$) at 4th week. However, there was no significant changes of estradiol-17$\beta$ concentration in fish exposed to phenanthrene. The physiological variation in phenanthrene exposed fish was a dramatic increase in plasma cortisol level. It is concluded that chronic exposure of phenanthrene can induce increase of plasma testosterone levels and elevate the plasma cortisol level in flounder, Paralichthys olivaceus.
