Pb의 노출에 따른 넙치, paralichthys olivaceus, 치어의 생존, 성장 및 산소소비율의 변화 (Change of Survival, Growthand Oxygen Consumption Rate in the Juvenile Olive Flounder,Paralichthys olivaceus Exposed to Lead)

  • 넙치의 생존, 성장 및 대사에 미치는 납의 만성적 독성을 파악하기 위하여 6주 동안 납 0, 80, 150, 230, 420 ${\mu}g/L$농도에 노출시켜 실험을 실시하였다. 넙치의 생존율은 납 농도 230 ${\mu}g/L$이상에서 유의한 감소를 나타냈다. 넙치의 성장률과 사료효율은 납 농도가 증가할수록 감소를 하여 150 ${\mu}g/L$이상의 농도에서는 대조구에 비하여 유의한 감소를 나타냈다. 넙치의 대사율은 납 농도 80, 150, 230, 420 ${\mu}g/L$에서 대조구보다 각각 21.4, 23.8, 36.8, 44.9 %가 저하했으며, 80 ${\mu}g$/L이상의 농도에서는 유의한 감소를 나타냈다.

Effects of Lactococcus lactis subsp. lactis I2 with β-Glucooligosaccharides on Growth, Innate Immunity and Streptococcosis Resistance in Olive Flounder (Paralichthys olivaceus)

  • To identify and quantify the effects of a combination of dietary $1{\times}10^8CFU/g$ Lactococcus lactis subsp. lactis I2 ($LI_2$) and 0.1% ${\beta}$-glucooligosaccharides (BGO) on the growth and immunity of olive flounder (Paralichthys olivaceus), a feeding experiment was conducted. Flounder ($14{\pm}0.5g$) were divided into two groups and fed control and synbiotic feeds for 8 weeks. Investigations were carried out on growth and feed utilization, innate immunity, serum biochemical parameters, intestinal lactic acid bacterial (LAB) viability, microvillus length, and changes in the expression levels of genes encoding pro-inflammatory cytokines (tumor necrosis factor $[TNF]-{\alpha}$, interleukin $[IL]-1{\beta}$, and IL-6). Results demonstrated the synbiotic diet had significantly better (p < 0.05) responses in terms of weight gain and specific growth rate, three innate immune parameters (respiratory burst, serum lysozyme, and superoxide dismutase), intestinal LAB viability, and the relative $TNF-{\alpha}$ expression level (p < 0.05). Moreover, after challenge with Streptococcus iniae ($1{\times}10^8CFU/ml$), the synbiotically fed group exhibited significantly higher (p < 0.05) protection against streptococcosis, validating the observed changes in immune parameters and induction of the cytokine-encoding gene. Therefore, according to the results of the present study, synbiotic feed ($LI_2+BGO$) increased growth, modulated innate immune parameters and protected olive flounder against streptococcosis.

  • The viral hemorrhagic septicemia virus (VHSV) has an extensive host range, and infects farmed and wild fish inhabiting both freshwater and marine ecosystems. Enzyme-linked immunosorbent assay (ELISA) is highly useful in diagnosing viral hemorrhagic septicemia. However, ELISA shows high, non-specific background reaction with fish antibodies. In this study, we optimized the antigen and antibody concentrations used for detecting specific antibodies in VHSV-infected olive flounder to reduce non-specific binding, and improve the sensitivity of ELISA. The results suggested that OD (optical Density) values were valid when ELISA was performed with $0.1{\mu}g/well$ of virus, involving blocking with blocking buffer (Roth, Roti-Block), 1:300-1:600 dilution with flounder antisera, and 1:1000 dilution with anti-flounder IgM and HRP-conjugated goat anti-mouse IgG for detecting the VHSV antibody in flounder sera. Furthermore, 11 different VHSV strains isolated in Korea from 2012 to 2016 were used to infect the fish. The results showed no correlation between viral pathogenicity and antibody production. This research is a basic study on the application of antibody detection in the diagnosis of viral hemorrhagic septicemia in the olive flounder.

넙치(Paralichthys olivaceus) 사료 내 phytosterol의 이용 가능성 평가 (Dietary Supplementation of Phytosterol for Olive Flounder Paralichthys olivaceus)

  • We evaluated the effects of dietary supplementation of phytosterol on the growth, feed utilization, immunity, digestibility, wound healing ability and disease resistance of olive flounder Paralichthys olivaceus. We conducted two consecutive feeding trials at different growth stages of the fish: EXP-1 (68.9 g) and EXP-2 (16.5 g). The experimental diets were supplemented with graded levels of phytosterol (1% ecdysteroid extracted from Achyranthis radix) at 0, 0.025, 0.05, 0.1, 0.2 and 0.4% (designated as Con, PHY0.025, PHY0.05, PHY0.1, PHY0.2 and PHY0.4, respectively). Dietary phytosterol did not significantly affect growth and cholesterol concentration. Feed utilization was higher in fish fed phytosterol-supplemented diets than in fish fed the control diet. Dietary phytosterol increased innate immunity and digestibility of protein and dry matter. Wound healing ability was also increased by the phytosterol supplementation. The survival against Edwardsiella tarda challenge was higher in fish fed low-phytosterol diets than in fish fed the control diet. The optimum dietary level of phytosterol seems to be approximately 0.05%. The results in this study indicate that the phytosterol could be used as a functional supplement in diets to improve feed utilization, immunity, digestibility and wound healing ability of olive flounder.

Effect of vegetable oils with different fatty acid composition on high-fat diet-induced obesity and colon inflammation

  • BACKGROUND/OBJECTIVES: Different fatty acids exert different health benefits. This study investigated the potential protective effects of perilla, olive, and safflower oils on high-fat diet-induced obesity and colon inflammation. MATERIALS/METHODS: Five-week old, C57BL/6J mice were assigned to 5 groups: low-fat diet (LFD), high-fat diet (HFD) and high-fat diet supplemented with-perilla oil (HPO), olive oil (HOO), and safflower oil (HSO). After 16 weeks of the experimental period, the mice were sacrificed, and blood and tissues were collected. The serum was analyzed for obesity- and inflammation-related biomarkers. Gene expression of the biomarkers in the liver, adipose tissue, and colon tissue was analyzed. Micro-computed tomography (CT) analysis was performed one week before sacrifice. RESULTS: Treatment with all the three oils significantly improved obesity-induced increases in body weight, liver weight, and epididymal fat weight as well as serum triglyceride and leptin levels. Treatment with perilla oil (PO) and safflower oil (SO) increased adiponectin levels. The micro-CT analysis revealed that PO and SO reduced abdominal fat volume considerably. The mRNA expression of lipogenic genes was reduced in all the three oilsupplemented groups and PO upregulated lipid oxidation in the liver. Supplementation of oils improved macroscopic score, increased colon length, and decreased serum endotoxin and proinflammatory cytokine levels in the colon. The abundance of Bifidobacteria was increased and that of Enterobacteriaceae was reduced in the PO-supplemented group. All three oils reduced proinflammatory cytokine levels, as indicated by the mRNA expression. In addition, PO increased the expression of tight junction proteins. CONCLUSIONS: Taken together, our data indicate that the three oils exert similar anti-obesity effects. Interestingly, compared with olive oil and SO, PO provides better protection against high-fat diet-induced colon inflammation, suggesting that PO consumption helps manage inflammation-related diseases and provides omega-3 fatty acids needed by the body.

반수체 넙치(Paralichthys olivaceus)의 발생학적 특성 분석 (Embryonic Development of Haploid Olive Flounder Paralichthys olivaceus)

  • We investigated the characteristics of embryonic and abnormal organ development in haploid olive flounder, Paralichthys olivaceus, by comparing egg development and histological sections in haploid and diploid individuals. After the mid-blastula transition, abnormal development was observed in haploid fish, including delayed epiboly and malformation of the germ ring and embryonic body. In haploid flounder, Kupffer’s vesicles are irregularly shaped and of variable size compared to diploids. The embryonic body of haploids was shorter and broader than that of diploids and the tail length and size were variable. Most haploid embryos failed to hatch and the few larvae that did, did not survive for more than 6 h. The histological analysis of haploid larvae revealed deformed development in diverse organs, including the eye, otic vesicles, notochord, and neural tube. These results may be related to an abnormality in the axial system of haploid larvae. This study confirmed that the abnormalities of haploid olive flounder were similar to the reported characteristics of haploid syndrome. The abnormalities are caused by delayed epiboly and involution and deformity of Kupffer’s vesicle during egg development.

  • We investigated the effects of feeding rate on the growth performance of growing and sub-adult olive flounder Paralichthys olivaceus, and the optimum feeding rate at high water temperature (25-31℃). In experiment I, two replicated groups of fish (113 g) were fed a commercial diet (CD) at rates of 0, 0.7, 1.2, 1.7, 2.2, and 2.4% (satiation) body weight (BW) per day for 4 weeks. In experiment II, two replicated groups of fish (313 g) were fed a CD feeding rates of 0, 0.3, 0.5, 0.7, 0.9, and 1.0% (satiation) BW per day for 4 weeks. In experiment I, the weight gain (WG) and specific growth rate (SGR) of fish fed at 0.7 and 0.9% BW per day was significantly higher than that of unfed fish (0%) and fish fed at 0.3, 0.5, and 1.0% (satiation) BW per day. In experiment II, the WG and SGR increased significantly as feeding rate increased from 0 to 2.2% BW per day, but decreased in the satiation group (2.4%). Broken-line regression analysis of WG showed that the optimum feeding rate of growing and sub-adult olive flounder were 1.98% and 0.55% BW per day, respectively, at the high water temperature.

  • An 8-week feeding experiment was conducted to evaluate three types of squid Sepia esculenta liver powder (SLP) as a dietary protein source replacing fish meal (FM) in juvenile olive flounder Paralichthys olivaceus. To replace FM, six experimental diets were formulated with three types (A, B, C) of SLP at the 5 and 10%: SLP-A5, SLP-A10, SLP-B5, SLP-B10, SLP-C5, and SLP-C10. One control diet contained 100% FM as the main protein source and another was a commercial diet (Com). Fish with an average body weight of $22.8{\pm}0.4$ g ($mean{\pm}SD$) were allocated randomly to aquaria in groups of 20 fish and fed the experimental diets in triplicate to satiation. The weight gain (WG) and specific growth rate (SGR) of fish fed the SLP-C10 diet were lower than those of fish fed the FM and SLP-B5 diets. No significant difference was observed in the WG and SGR among fish fed the diets other than SLP-C10. The feed efficiency (FE) and protein efficiency ratio (PER) of fish fed each SLP diet did not differ from those fed the control diet. However, fish fed SLP-C5 and SLP-C10 had a lower FE and PER than the fish fed commercial, SLP-A5 and SLP-B5 diets. Each SLP diet except for SLP-C10 could replace up to 10% of FM for juvenile olive flounder. The results of this experiment provide information that will assist in formulating an inexpensive practical diet containing SLP for juvenile olive flounder.

양식넙치에 경구투여한 Ciprofloxacin의 근육조직 잔류량을 이용한 휴약 기간 설정 연구 (Withdrawal Times of Ciprofloxacin in Oliver Flounder (Paralichthys olivaceus) after Oral Administration)

  • Ciprofloxacin is the most commonly used fluoroquinolones for treating bacterial disease in olive flounder (Paralichthys olivaceus) farming, but its withdrawal time for industrial-scale farming has not been established. The depletion of ciprofloxacin was investigated in the olive flounder under field conditions. Fish were kept in an inland fish farm and fed a commercial diet containing 5 mg/kg of ciprofloxacin for 13 days. Seven fishes per sampling time were examined during and after the treatment. Ciprofloxacin and its major metabolite, enrofloxacin, were analyzed using HPLC with a fluorescence detector. The concentrations of ciprofloxacin and enrofloxacin in muscle increased during the medication period, and then decreased rapidly. The ciprofloxacin and enrofloxacin concentration in the olive flounder peaked on days 11 and 13, respectively, with maximum concentrations in muscle of 0.58 and 0.73 mg/kg. Residual ciprofloxacin and enrofloxacin were eliminated rapidly; 6 days after treatment, the respective levels in muscle were 0.04 and 0.10 mg/kg and neither was detected 15 days post treatment. The level of ciprofloxacin accumulation at the beginning of oral administration was variable according to the farming conditions, but the overall exhaustion time was similar. In conclusion, an adequate withdrawal period for enrofloxacin is 15 days in the case of oral ciprofloxacin administration.

천연 및 인공 넙치종묘의 성장 단계에 따른 지질조성 및 비타민 E 함량 비교 (Comparison of Lipids and Vitamin E Content with Growth Stages Between Cultured and Wild Olive Flounder Larvae (Paralichthys olivaceus))

  • 천연 및 인공 넙치 종묘의 성장 단계에 따른 체지질 조성 변화를 비교 검토한 결과는 다음과 같다. 1) 어체의 지질 및 비타민 E 함량은 천연 종묘 보다 인공 종묘에서 높은 값을 나타내었다. 2) 어체 성장에 따른 지질, 지방산(18: 2n-6, 22: 6n-3) 및 비타민 E 함량 변화는 부화 후 전장 5-10mm 전후의 배합 사료 공급기에서 급격한 증감 현상을 보였다. 3) 어체 지질, 지방산 조성 및 비타민 E 함량은 먹이 조성에 직접적인 영향을 받았다. 4) 천연 종묘는 인공 종묘에 비해 자연 상태에서 지질 함량이 낮고 고농도의 DHA를 함유하고 있는 천연 먹이를 섭취한 것으로 추정되었다.
