Implementing of Ozonated Olive Oil Manufacturing Device to Ensure Biocompatibility (생체 적합성이 확보를 위한 오존화 올리브오일 제조 장치 구현)

  • As research results showed that ozonated olive oil has excellent therapeutic effects on skin diseases, attempts were made to develop cosmetics manufacturing technology using ozone. Ozone is harmful to the human body, so a separate facility must be prepared to manufacture ozonated olive oil as a cosmetic raw material. The manufacturing device developed in this study was designed as an optimized process for manufacturing cosmetics while ensuring the safety of workers involved in production. To verify the ozonated olive oil produced from the implemented device, a toxicity test was conducted on animals. After applying ozonated oil (high concentration) to the rabbit's back for 24 hours, mortality, general symptoms and symptoms were measured. A skin irritation evaluation was performed. As a result of the experiment, as a result of evaluating the test substance treatment area after a certain period of time, it was confirmed that no skin irritation was observed in all animals, confirming the safety of the ozonated oil production process and the safety of the product.

Development of antibodies after combination Streptococcus iniae and Streptococcus parauberis of commercial vaccine injection in olive flounder (Paralichthys olivaceus) in farm (양식 넙치에서 연쇄구균 혼합백신 접종 후 항체가 변화 조사)

  • Streptococcosis, caused by Streptococcus iniae and Streptococcus parauberis is an important bacterial disease that affects in olive flounder in Korea. In Korea, multivalent bacterial vaccines are used to prevent streptococcal diseases in aquaculture. In this study, commercial vaccines containing formalin-inactivated bacterial cells of S. iniae and S. parauberis were administered at six fish farms and one unvaccinated fish farm were designated for investigation (Wando; 4 sites and Jeju; 3 sites). Blood was collected from vaccinated and unvaccinated olive flounders, and titers of antibodies against S. iniae and S. parauberis in serum were analyzed using ELISA. After a one shot vaccination in the farms at Jeju (farm A) and Wando (farm D), the proportion of individuals with specific antibodies against S. parauberis OD values of 0.4 or higher was 60% and 53.5%, respectively. But after booster vaccination, the proportion of individuals with serum OD values of 0.4 or higher was higher substantially increased to 96.6% (farm A) and 100% (farm D). The levels of S. parauberis specific antibodies of olive flounder were increased after vaccination in three fish farms (farm D, E, and F), but not S. iniae specific antibodies.

Physicochemical Properties of Olive Flounder Paralichthys olivaceus, Red Seabream Pagrus major and Jacopever Sebastes schlegeli (넙치(Paralichthys olivaceus), 참돔(Pagrus major) 및 조피볼락(Sebastes schlegeli)의 이화학적 성분 특성)

  • Olive flounder (Paralichthys olivaceus) including red seabream (Pagrus major) and jacopever (Sebastes schlegeli) were known to be suitable characteristics for preparing a processed food because of white meat fish containing low fat in muscle. In this study, the physicochemical properties of olive flounder (46-50 cm of length, 950-1,050 g of weight), red seabream (30-33 cm of length, 1,250-1,350 g of weight) and jacopever (20-23 cm of length, 550-650 g of weight) were studied to obtain a basic data for the development of a new processed food. Head, scale, bone and viscera of each samples were removed, and then grinding with homogenizer before experiment. Moisture content of red seabream is lower than that of olive flounder and jacopever, therefore, crude protein and crude lipid are higher content. Red seabream showed higher redness, yellowess and color difference value than those of olive flounder and jacopever except lightness. The major total amino acid in olive flounder, seabream and jacopever were glutamic acid, lysine and aspartic acid in order. Especially, the highest content of free amino acid was lysine for olive flounder and jacopever and alanine for red seabream. The highest content of mineral in olive flounder, seabream and jacopever was potassium (K) and then phosphorous (P) and sodium (Na) in order. The highest amount of fatty acid in olive flounder, red seabream and jacopever was polyenoic acid in which especially contained 6.8, 7.1 and 6.7% for EPA, and 15.2, 15.2 and 17.1% for DHA, respectively. From the result of sensory evaluation, there was no significant difference in color and odor, but not in taste and texture.

Comparison of Pood Chemical Properties in Olive Flounder (Paralichthys olivaceus), Spotted Flounder (Verasper variegatus) and Their Hybrid Muscle (넙치, 범가자미 및 잡종 (넙치 male $\times$ 범가자미 female) 근육의 식품화학적 특성 비교)

  • The proximate composition, mineral content, fatty acid composition, ATP related compounds, amino acid composition, color and texture were investigated with dorsal and ventral muscle from olive flounder, spotted flounder and hybrid (olive flounder m $\times$spotted flounder f). Spotted flounder and hybrid were higher in moisture content, and lower in crude protein content than those of olive flounder, Potassium content in hybrid was higher than that in olive flounder and spotted flounder. Hybrid was lower in calcium, iron, manganese content, and higher in magnesium content than olive flounder and spotted flounder. The contents of saturated fatty acid, unsaturated fatty acid and docosahexaenoic acid (DHA) in hybrid were intermediate level of spotted flounder and olive flounder. Fatty acid composition of dorsal muscle was slightly similar to ventral muscle. Adenosine triphosphate (ATP) and its related compounds contents and amino acid content in hybrid were intermediate level of spotted flounder and olive flounder, and these compounds of dorsal muscle were slightly similar to those of counterpart. The major amino acids such as aspartic acid, glutamic acid, leucine and Iysine were very similar to all the samples. Total amino acid and essential amino acid contents in dorsal muscle were slightly higher than those in ventral muscle. Free amino acid content and composition in hybrid were similar to spotted flounder, and free amino acid content in dorsal muscle was higher than in ventral muscle, The lightness of hybrid and spotted flounder was stronger than that of olive flounder, and was stronger in dorsal muscle. The breaking strength of hybrid was slightly lower than that of spotted flounder, and was stronger in dorsal muscle.

Characterization of Vibrio harveyi, the Causal Agent of Vibriosis in Cultured Marine Fishes in Korea

  • An epizootic causing mortality among cultured marine finfishes occurred in 1999 in the province of Kyoungsang, Korea. The disease was characterized by the presence of enterocele, abdominal swelling, and gastroenteritis. The causative bacteria were isolated from olive flounder (Paralichthys olivaceus), black rockfish (Sebastes schlegeli), turbot (Scophthalmus maximus) and the rearing water. These bacteria showed swarming activity on agar plates and yellowish or greenish colonies on thiosulfate-citrate-bile salts-sucrose (TCBS) agar plates, but no luminescence. The pathogen was identified as Vibrio harveyi based on morphological and biochemical characteristics and the sequence of l6S rDNA. The lethal doses (LD$_{50}$) of olive flounder and black rockfish were estimated to be $1.24\times10^6-1.36\times10^8$ and $3.24\times10^5-5.8\times10^7$ CFU/fish respectively following intraperitoneal injection.

Environmental stress-related gene expression and blood physiological responses in olive flounder (Paralichthys olivaceus) exposed to osmotic and thermal stress

  • We isolated warm temperature acclimation-related protein 65-kDa (Wap65) cDNA from the liver of olive flounder and investigated the mRNA expression of Wap65 and HSP70 in olive flounder exposed to osmotic (17.5, 8.75, and 4 psu) and thermal stress (25 and $30^{\circ}C$). The mRNA expression of Wap65 and HSP70 was increased by thermal stress. The mRNA expression of HSP70 was also increased by osmotic stress, whereas no significant change in Wap65 expression was detected. These results indicate that Wap65 mRNA expression occurs specifically in response to increases in water temperature, but not in response to osmotic stress. Plasma cortisol levels were also increased by osmotic and thermal stress. We also utilized the stress hormone cortisol to examine whether Wap65 expression is thermal-stress-specific. Cortisol treatment increased HSP70 mRNA expression in vitro, but had no significant effect on Wap65 mRNA expression. Thus, thermal stress, but not osmotic stress, induces Wap65 expression.

  • Different phytochemicals obtained from various natural plant sources are used as reduction agents for preparing gold, copper, silver and platinum nanoparticles. In this work a green method of reducing graphene oxide (rGO) by an inexpensive, effective and scalable method using olive leaf aqueous extract as the reducing agent, was used to produce rGO. Both GO and rGO were prepared and investigated by ultraviolet and visible spectroscopy, Fourier-transform infrared, scanning electron microscopy, atomic force microscopy, thermogravimetric analysis, cyclic voltammetry, X-ray photoelectron spectra, electrochemical impedance spectroscopy and powder X-ray diffraction.

  • Morphological structure and sporogenesis of myxosporean parasite, Parvicapsula anisocaudata from cultured olive flounder, Paralichthys olivaceus were examined by light and transmission electron microscopies. Numerous round early stages, spindle-shaped disporic or monosporic pseudoplasmodia, asymmetrical thin-walled mature spores were found inthe lumen of renal tubule and urinary bladder. Long pseudopodia or short projections from cytoplasm of pseudoplasmodia make the parasite attach firmly to host tissues. Spore development was mono- or disporous with no pansporoblast formation. Sporoplasmic cell was partially surrounded by two capsulogenic cells, and capsulogenic cells were enveloped by two flattened valvogenic cells. Capsulogenesis and valvogenesis followed general patterns seen in that of other myxosporean.

Tribological Properties of Aqueous Solutions Composed of Aminated Olive Oil Derivative (수용성 아민화 올리브유 유도체의 트라이볼로지적 특성고찰)

  • Aminated oilve oil derivative as the new organic disperse phases of the water soluble metal working fluid has been synthesized and tribological properties of the aqueous solutions composed of aminated olive oil derivative investigated using Four Ball Wear Tester and Falex EP Tester. The formulated aqueous solutions showed higher antiwear and extreme pressure properties and also lower friction coefficient. On the basis of the the results, water soluble aminated oilve oil derivative showed excellent tribological properties due to the polarizability of oleic acid derivative composed of majority part in oilve oil.

Hepatic Response in Cytochrome P45O and De-alkylase Activity of Olive Flounder (Paralichthys olivaceus) Exposed to Water-borne Phenanthrene

  • Olive flounder (Paralichthys olivaceus) was exposed to water-borne phenanthrene for 4 weeks. After the exposure for 2-weeks, hepatic cytochrome P450 contents were significantly elevated. Induction of hepatic ethoxy resorufin-O-deethylase (PROD) activity was significantly increased in flounders treated with 1.0 and 2.0 $\mu$M phenanthrene, compared to untreated group and 0.5 $\mu$M treated group. However, there were no significant changes in pentoxyresorufin-O-deethylase (PROD) activity in hepatic microsome of all the phenanthrene-treated groups, compared to the untreated group. Phenanthrene has the potential to induce cytochrome P450 and EROD enzyme of the olive flounder.
