장류유래 Exopolysaccharide 생성 유산균의 잠재적 Probiotic 특성 (Potential Probiotic Properties of Exopolysaccharide Producing Lactic Acid Bacteria Isolated from Fermented Soybean Product)

  • 전통 장류에서 유산균 성상을 보이는 균주를 분리하여, TLC와 PCR을 통하여 HoPS를 우수하게 생성하는 균주 HSB15, JSA22, JSA57, JSB22, JSB66 및 JSB89를 선발하였다. 선발된 6균주의 16S rDNA의 염기서열을 분석한 결과 HSB15는 L. alimentarius, JSA22는 L. plantarum, JSA57은 L. pentosus, JSB22는 L. brevis, JSB66는 L. alimentarius, JSB89는 L. parabrevis로 동정되었다. 항균 활성은 6균주 중에서 JSA22가 4개의 병원성균에 대한 항균을 나타내어 항균 활성이 가장 우수하게 나타났고, 인공위액 저항성은 HSB15, JSA22와 JSA57 균주가 70% 이상의 높은 인공위액 저항성으로 가장 우수했으며, 인공담즙 저항성은 HSB15를 제외한 5균주에서 80% 이상의 생존율을 나타내며 담즙액 저항성이 우수하게 나타났다. 또한 probiotic 특성을 지닌 분리 균주의 prebiotics로써 AOS를 이용한 성장율을 관찰한 결과, 모든 분리 균주가 AOS를 선택적으로 이용하며 성장하는 것을 관찰하였고 특히 JSB22가 AOS 이용 성장율이 가장 우수한 것으로 확인되었다. 본 연구를 통해 장류유래 probiotics를 선발하였고 AOS의 새로운 prebiotics로 활용을 검토한 바, synbiotics로 이용할 수 있는 가치가 있다고 판단되었다.

Mitigation of Methane Emission and Energy Recycling in Animal Agricultural Systems

  • Abatement of greenhouse gas emitted from ruminants and promotion of biogas energy from animal effluent were comprehensively examined in each anaerobic fermentation reactor and animal experiments. Moreover, the energy conversion efficiency of biomass energy to power generation were evaluated with a gas engine generator or proton exchange membrane fuel cell (PEMFC). To mitigate safely rumen methanogenesis with nutritional manipulation the suppressing effects of some strains of lactic acid bacteria and yeast, bacteriocin, $\beta$1-4 galactooligosaccharide, plant extracts (Yucca schidigera and Quillaja saponarea), L-cysteine and/or nitrate on rumen methane emission were compared with antibiotics. For in vitro trials, cumulative methane production was evaluated using the continuous fermented gas qualification system inoculated with the strained rumen fluid from rumen fistulated Holstein cows. For in vivo, four sequential ventilated head cages equipped with a fully automated gas analyzing system were used to examine the manipulating effects of $\beta$1-4 galactooligosaccharide, lactic acid bacteria (Leuconostoc mesenteroides subsp. mesenteroides), yeast (Trichosporon serticeum), nisin and Yucca schidigera and/or nitrate on rumen methanogenesis. Furthermore, biogas energy recycled from animal effluent was evaluated with anaerobic bioreactors. Utilization of recycled energy as fuel for a co-generator and fuel cell was tested in the thermophilic biogas plant system. From the results of in vitro and in vivo trials, nitrate was shown to be a strong methane suppressor, although nitrate per se is hazardous. L-cysteine could remove this risk. $\beta$1-4 galactooligosaccharide, Candida kefyr, nisin, Yucca schidigera and Quillaja saponarea are thought to possibly control methanogenesis in the rumen. It is possible to simulate the available energy recycled through animal effluent from feed energy resources by making total energy balance sheets of the process from feed energy to recycled energy.

Effect of Prebiotics on Intestinal Microflora and Fermentation Products in Pig In Vitro Model

  • The objective of this study was to evaluate the effect of the different types and levels of prebiotics on intestinal microflora and fermentation products in the in vitro fermentation model. The prebiotcs used in this study were IMO (iso-malto oligosaccharide), CI (partially digested chicory-inulin), RA (raffinose) and CD (cyclodextrin). Experimental diet for growing pigs was predigested by digestive enzymes and this hydrolyzed diet was mixed with buffer solution containing 5% fresh swine feces. Then, the mixture was fermented with or without prebiotics at the concentrations of 0.5 and 1.0% for 24 h. Samples were taken at 24 h, and viable count of micoflora, gas, pH, volatile organic compounds and short-chain fatty acids were determined. The viable count of Enterobacteriaceae was significantly decreased (p<0.001) in all treatments added with prebiotics in comparison to control without prebiotics. However, the increase of lactic acid bacteria was observed in the prebiotics treatment. Gas production increased as the level of prebiotics increased. The pH values in the fermentation fluid decreased in a dose-dependent manner with increasing the concentration of prebiotics. The fermentation with prebiotics resulted in the reduction of malodorous compounds such as ammonia, hydrogen sulfide, indole and skatole. The increase in short-chain fatty acid (SCFA) production was observed in the treatments with prebiotics. In conclusion, the results of this study demonstrated that the fermentation with prebiotics was effective in reducing the formation of malodorous compounds and increasing lactic acid bacteria and SCFA. These effects depended on the concentration of prebiotics. Moreover, further study is needed to determine whether the in vitro efficacy on the reduction of malodorous compounds and increase of SCFA would also be observed in animals.

2'-푸코실락토오스의 자가포식을 통한 멜라닌 감소 연구 (A Study on Melanin Reduction through Autophagy by 2'-Fucosyllactose)

  • 2'-fucosyllactose (2'-FL)는 사람의 모유에 가장 많이 존재하는 올리고당(human milk oligosaccharides, HMOs)으로, 장내 유용 미생물의 성장을 촉진시키고 알레르기, 염증 반응을 완화시키는 것에 도움을 준다. 다양한 긍정적 기능을 가진 2'-FL의 미백 화장품 소재로써의 가능성을 확인하고자, 본 연구를 통해 멜라닌 생성 저해 효능 및 자가포식 유도 가능성을 검토하였다. 인간 유래 멜라닌 생성 세포로 알려진 MNT-1 세포에서 독성 실험을 진행하여 40 g/L 이하에서 세포독성이 없음을 확인하였고, 동일 세포에서 20 g/L 농도로 7 일간 처리하여 멜라닌 생성량을 분석한 결과, 40% 멜라닌 생성 감소를 확인하였다. 멜라닌 생성 관련 인자 TYR 및 TYRP1의 단백질 발현량을 western blot 법을 이용하여 분석한 결과, 2'-FL 처리는 이들을 감소시켰으며, 더불어 자가포식 표지자인 microtubule-associated protein 1 light chain 3 (LC3)의 형태가 LC3-I에서 LC3-𝚷로 변환을 확인할 수 있었다. 공초점 현미경을 통해 2'-FL 처리에 따른 LC3 puncta의 증가가 확인되었다. 따라서, 2'-FL로 활성화된 자가포식이 TYR 및 TYRP1 단백질 발현량을 저해시킴으로서 멜라닌 생성을 감소시키는 것으로 시사된다. 결론적으로 2'-FL은 자가포식을 유도하여 멜라닌 생성이 억제됨이 확인되어 미백 화장품 소재로써의 가능성이 기대된다.

  • 본 실험은 mannan-oligosaccharide(MOS)와 fructo-oligosaccharides(FOS)가 산란계의 생산성과 소장 내 미생물 균총 및 혈액 내 항체 수준에 미치는 영향을 조사하기 위해 실시하였다. 사양 시험은 27주령의 산란계(Hy-Line Brown) 960수를 선별하여 A형 2단 케이지에 대조구 포함 총 6처리구로 구성하여 처리당 4반복, 반복당 40수씩 randomized block design으로 배치하였다. 시험 기간 동안 물과 사료는 자유 섭취하게 하였으며, 일반적인 점등 관리(자연 일조 + 조명; 16 h)를 실시하였다. 실험은 대조구, 항생제구; avilamycin 6 ppm, MOS 0.025%구, MOS 0.05%구, FOS 0.25%구, FOS 0.5%구 등 총 6처리구를 두어 실시하였다. 일계 산란율과 hen-housed 산란율은 처리구간에 유의적 차이가 있었다. 일계 산란율과 hen-housed 산란율은 MOS 0.025% 처리구가 대조구와 비교하여 유의적으로 높았다. 난중, 사료 섭취량에서는 처리구간에 유의한 차이가 없었으나, FOS 0.25%구가 다른 첨가구와 대조구에 비해 낮은 경향을 보였다. 난각 두께에서는 FOS 0.25% 처리구가 MOS 0.025% 첨가구에 비해 유의적으로 높았다. 난각 강도, 난각 색깔 지수, 난황 색깔 지수에서는 처리구간에 유의적 차이가 나타나지 않았다. Haugh unit는 유의한 차이는 나타나지 않았지만 전반적으로 대조구에 비해 모든 첨가구들이 높은 경향을 보였다. 혈청 IgG 농도는 처리구간에 유의적인 차이는 없었지만 모든 첨가구들이 대조구에 비하여 높은 경향을 보여주었으며, 혈청 IgA 농도는 FOS 0.25% 처리구가 대조구에 비해 유의적으로 높았다. Lactobacillus는 첨가구에서 유의하게 증가하고, Cl. perfrigens는 유의하게 감소하였으며, E. coli는 첨가구들에서 감소하는 경향이 있었다. 결론적으로 MOS와 FOS는 산란계에서 산란율을 증가시키는 경향이 있었고 혈중 IgA 및 장내 미생물 균총에도 유의한 영향을 미쳤다.

유전자 재조합 인간의 G-CSF의 생리활성과 EGFP-hG-CSF유전자가 도입된 체세포의 분리 (Biological Activity of Recombinant Human Granulocyte Colony-Stimulating Factor and Isolation of the Somatic Cell Transfected EGFP-hG-CSF Gene)

  • 유전자재조합 hG-CSF의 생리활성을 분석하기 위하여 편상의 암세포로부터 분리되어진 cDNA를 이용하여 hG-CSF 유전자를 분리하여 동물세포(CHO cell lines)를 이용하여 재조합 단백질을 생산하였다. 재조합 단백질의 체내 생리활성을 분석하기 위하여0일과 2일에 피하주사 후 5일에 혈액을 채취하여 백혈구 수를 분석하였다. 투여 전과 비교하여 5일째에 백혈구 수는 현저하게 증가하였다. 또한, pEGFP-mUII-hG-CSF벡터를 소 태아로부터 분리되어진 체세포에 형질전환을 시켜서, EGFP signal을 나타내는 세포를 confocal를 이용하여 분리하여 수립하였다. 따라서, 이러한 결과는 유전자재조합 hG-CSF는 체내에서 강력한 생리활성을 나타내며, 또한 당쇄가 첨가되어지고 이중으로 연결되어진 새로운 돌연변이체를 포함하여 고 활성 재조합체의 생산이 가능할 것으로 보이며, pEGFP-mUII-hG-CSF벡터는 복제 형질전환 가축 생산을 위하여 유용하게 사용되어질 것으로 사료된다.

Effect of Live Yeast and Mannan-oligosaccharides on Performance of Early-lactation Holstein Dairy Cows

  • This study evaluated the effects of live yeast and yeast cell-wall mannan-oligosaccharide supplementation onperformance and nutrient digestibility during early lactation in cows fed a diet based on a mixture of corn silage and alfalfa hay as forage sources. Eight multiparous Holstein dairy cows (average days in milk, 27${\pm}$6) were used in a replicated 4${\times}$4 Latin square design. Diets contained 45% forage and 55% concentrate on a dry matter (DM) basis and treatments were: i) basal diet without additive (Control), ii) basal diet with 32 g/d of mannan-oligosaccharides (MOS), iii) basal diet with $1.2{\times}10^{10}$ colony forming units per day (cfu/d) of live yeast (Saccharomyces cerevisiae CNCM 1-1077; SC), and iv) basal diet with a mixture of MOS (32 g/d) and SC ($1.2{\times}10^{10}$ cfu/d; MOS+SC). Treatments had no effect (p>0.05) on DM intake and yields of milk, 3.5% fat-(FCM) and energy-corrected milk (ECM), and on milk fat percentage, body condition score and blood metabolites. Compared with the Control, only supplementation of SC resulted in numerically higher yields of FCM (41.9 vs. 40.1 kg/d) and ECM (41.8 vs. 40.3 kg/d), and milk fat percentage (3.64 vs. 3.43%). While the MOS diet had no effects on performance compared to the Control, the combination treatment MOS+SC increased milk protein percentage (p<0.05). Also, the MOS supplementation, both alone or in combination with SC, numerically increased milk fat percentage. The SC supplementation increased apparent digestibility of DM and crude protein while the MOS supplementation did not affect digestibility. Concentrations of total volatile fatty acids (VFA) and ruminal pH were similar across treatments. Overall results indicated that supplementation of MOS produced variable and inconsistent effects on rumen metabolism and performance, whereas SC supplementation improved nutrient digestibility and numerically increased FCM and ECM yields, which could not be enhanced by the combined supplementation of MOS+SC. According to our experimental condition, there was no effect of MOS alone or in combination with SC on dairy cow performance.

Effects of Dietary Synbiotics from Anaerobic Microflora on Growth Performance, Noxious Gas Emission and Fecal Pathogenic Bacteria Population in Weaning Pigs

  • Synbiotics is the term used for a mixture of probiotics (live microbial feed additives that beneficially affects the host animal) and prebiotics (non-digestible food ingredients that beneficially affect the organism). This study investigated the effect of probiotics from anaerobic microflora with prebiotics on growth performance, nutrient digestibility, noxious gas emission and fecal microbial population in weaning pigs. 150 pigs with an initial BW of 6.80${\pm}$0.32 kg (20 d of age) were randomly assigned to 5 dietary treatments as follows: i) US, basal diet+0.15% antibiotics (0.05% oxytetracycline 200 and 0.10% tiamulin 38 g), ii) BS, basal diet+0.2% synbiotics (probiotics from bacteria), iii) YS, basal diet+0.2% synbiotics (probiotics from yeast), iv) MS, basal diet+0.2% synbiotics (probiotics from mold), v) CS, basal diet+0.2% synbiotics (from compounds of bacteria, yeast and mold). The probiotics were contained in $10^{9}$ cfu/ml, $10^{5}$ cfu/ml and $10^{3}$ tfu/ml of bacteria, yeast and molds, respectively. The same prebiotics (mannan oligosaccharide, lactose, sodium acetate and ammonium citrate) was used for all the synbiotics. Pigs were housed individually for a 16-day experimental period. Growth performance showed no significant difference between antibiotic treatments and synbiotics-added treatments. The BS treatment showed higher (p<0.05) dry matter (DM) and nitrogen digestibility while ether extract and crude fiber digestibility were not affected by the dietary treatment. Also, the BS treatment decreased (p<0.05) fecal ammonia and amine gas emissions. Hydrogen sulfide concentration was also decreased (p<0.05) in BS, YS and MS treatments compared to other treatments. Moreover, all the synbioticsadded treatments increased fecal acetic acid concentration while the CS treatment had lower propionic acid concentration than the US treatment (p<0.05) gas emissions but decreased in fecal propionate gas emissions. Total fecal bacteria and Escherichia coli populations did not differ significantly among the treatments, while the Shigella counts were decreased (p<0.05) in synbiotics-included treatment. Fecal bacteria population was higher in the YS treatment than other treatments (p<0.05). The BS treatment had higher yeast concentration than YS, MS and CS treatments, while US treatment had higher mold concentrations than MS treatment (p<0.05). Therefore, the results of the present study suggest that synbiotics are as effective as antibiotics on growth performance, nutrient digestibility and fecal microflora composition in weaning pigs. Additionally, synbiotics from anaerobic microflora can decrease fecal noxious gas emission and synbiotics can substitute for antibiotics in weaning pigs.

유청 단백질에서 유도되는 생리활성 펩타이드에 관한 연구 (Isolation of Mitogenic Glycophosphopeptides from Cheese Whey Protein Concentrate)

  • 1. CWPC중의 새로운 생리활성물질의 검색 Mouse 임파세포의 증식효과를 지표로 하는 면역기능을 검토하여 CWPC중의 면역 부활작용을 갖는 새로운 성분의 검색을 실시하였다. CWPC를 여러 가지 분획법으로 분획하여 mouse 임파세포의 증식효과를 지표로 면역 활성성분을 검색하였다. 그 결과 gel filtration, 음이온교환법을 사용하여 분획한 당을 다량 포함한 부분에 강한 면역 부활담당세포에 대하여 증식활성을 나타내는 물질을 발견하였다. 이 물질은 SDS-PAGE상에서 분자량이 약 16kDa에 위치하여 Ca, p및 당쇄를 포함한 물질이며, 이것을 GPP로 하였다. GPP에는 우유케이신의 trypsin분해물이며 Ca와 무기인을 풍부하게 포함하는 ${\beta}$-CPP와 유사한 phosphoserin 영역을 갖는 성분과 갖지 않는 성분의 2종류가 존재하며, 각각의 면역 부활활성이 인정되었다. 각 성분의 아미노산 분석, 당 분석의 결과에서 지금까지 보고된 우유중의 면역담당세포에 대한 증식활성을 갖는 물질과는 상이한 성분인 것으로 밝혀졌다. 더우기 이 활성물질(GPP)은 PP cell에서도 동등한 활성이 있는 것으로 판단되었다. 이러한 결과를 종합하여 보면 CWPC중에는 지금까지 알려지지 않았던 새로운 면역 부활물질이 존재하며, 그 성분에는 CPP와 유사한 phophoserine영역이 존재하는 성분이 포함되어 있고, N-글리코실 결합의 당쇄가 존재하는 것으로 시사되었다. 이 성분은 전신면역의 지표인 비장세포에 대해서만이 아니고, 장관면역계에 중요한 역할을 담당하는 PP Cell에서도 활성이 있는 것으로 보아 전신 및 국부적인 면역기능의 부활성분으로서 응용의 가능성이 시사되었다.

Some Prophylactic Options to Mitigate Methane Emi ssion from Animal Agriculture in Japan

  • The abatement of methane emission from ruminants is an important global issue due to its contribution to greenhouse gas with carbon dioxide. Methane is generated in the rumen by methanogens (archaea) that utilize metabolic hydrogen ($H_2$) to reduce carbon dioxide, and is a significant electron sink in the rumen ecosystem. Therefore, the competition for hydrogen used for methanogenesis with alternative reductions of rumen microbes should be an effective option to reduce rumen methanogenesis. Some methanogens parasitically survive on the surface of ciliate protozoa, so that defaunation or decrease in protozoa number might contribute to abate methanogenesis. The most important issue for mitigation of rumen methanogenesis with manipulators is to secure safety for animals and their products and the environment. In this respect, prophylactic effects of probiotics, prebiotics and miscellaneous compounds to mitigate rumen methanogenesis have been developed instead of antibiotics, ionophores such as monensin, and lasalocid in Japan. Nitrate suppresses rumen methanogenesis by its reducing reaction in the rumen. However, excess intake of nitrate causes intoxication due to nitrite accumulation, which induces methemoglobinemia. The nitrite accumulation is attributed to a relatively higher rate of nitrate reduction to nitrite than nitrite to ammonia via nitroxyl and hydroxylamine. The in vitro and in vivo trials have been conducted to clarify the prophylactic effects of L-cysteine, some strains of lactic acid bacteria and yeast and/or ${\beta}$1-4 galactooligosaccharide on nitrate-nitrite intoxication and methanogenesis. The administration of nitrate with ${\beta}$1-4 galacto-oligosaccharide, Candida kefyr, and Lactococcus lactis subsp. lactis were suggested to possibly control rumen methanogenesis and prevent nitrite formation in the rumen. For prebiotics, nisin which is a bacteriocin produced by Lactococcus lactis subsp. lactis has been demonstrated to abate rumen methanogenesis in the same manner as monensin. A protein resistant anti-microbe (PRA) has been isolated from Lactobacillus plantarum as a manipulator to mitigate rumen methanogenesis. Recently, hydrogen peroxide was identified as a part of the manipulating effect of PRA on rumen methanogenesis. The suppressing effects of secondary metabolites from plants such as saponin and tannin on rumen methanogenesis have been examined. Especially, yucca schidigera extract, sarsaponin (steroidal glycosides), can suppress rumen methanogenesis thereby improving protein utilization efficiency. The cashew nutshell liquid (CNSL), or cashew shell oil, which is a natural resin found in the honeycomb structure of the cashew nutshell has been found to mitigate rumen methanogenesis. In an attempt to seek manipulators in the section on methane belching from ruminants, the arrangement of an inventory of mitigation technologies available for the Clean Development Mechanism (CDM) and Joint Implementation (JI) in the Kyoto mechanism has been advancing to target ruminant livestock in Asian and Pacific regions.
