B16 Melanoma 세포에서 Chitosan Oligosaccharide가 Melanin 생성에 미치는 영향 (Effect of Chitosan Oligosaccharide on Melanin Production in B16 Melanoma Cells)

  • To investigate the effect of chitosan oligosaccharide on melanin synthesis, we measured tyrosinase activity and melanin production in B16 melanoma cells. Chitosan oligosacchaide itself did not have any anti-oxidant activity in DPPH radical scavenging, and did not affect the proliferation of B16 melanoma cells. Chitosan oligosaccharide dose-dependently increased melanin production in the absence or presence of MSH. However, chitosan oligosaccharide did not have any influence on the tyrosinase activity and tyrosinase expression in B16 melanoma cells. These results suggest that chitosan oligosaccharide-induced melanin production may be independent on tyrosinase activity in B16 melanoma cells. From the above results. chitosan oligosaccharide dose-dependently appears to increase melanin production in B16 melanoma cells, suggesting that chitosan oligosaccharide may be used as a tanning agent.

  • To investigate the effect of chitosn oligosaccharide on skin care, we measured tyrosinase activity and melanin production in B16 melanoma cells, and elastase and hyaluronidase activity. Chitosan oligosaccharide itself did not have any anti-oxidant activity in DPPH radical scavenging, and did not affect the proliferation of B16 melanoma cells. Chitosan oligosaccharide dose-depednetly increased mealnin production in the absence or presence of MSH. However, chitosan oligosaccharide did not have any influence on the tyrosinase activity and tyrosinase expression in B16 melanoma cells. (omitted)

  • Tyrosinase (monophenol. 3.4-${\beta}$-dihydroxyphenylalanin oxygen oxidoreductase. EC 1.14.18.1 J. which plays a pivotal role in melanogenesis. It is single chain glycoprotein catalyzing the hydroxylation of tyrosine to ${\beta$\mid$$-3.4-dihydroxyphenylalanin (DOPA) and the oxidation of DOPA to DOPA quinone. To investigate whitening effect of chitosan oligosaccharide. we obtained chitosan oligosaccharide [(glucosamine)2-6] by NaNO2 oxidation and measured the effect of chitosan oligosaccharide on tyrosinase activity. Chitosan oligosaccharide dose-dependently inhibited tyrosinase (2 unit) activity and inhibited by 18.8% at dose of 100${\mu}$g/ml. Vitamin C. arbutin and kojic acid that are well known to be inhibitor of melanin production dose-dependently inhibited tyrosinase (2unit) activity. These results suggest that chitosan oligosaccharide may be used as inhibitor of melanin production in melanocyte. which will be further studied.

  • Production of inulo-oligosaccharides from inulin was carried out with the maximum yield of 94% using a purified endoinulinase from Aspergillus ficcum. The optimum reaction temperature and pH were 55-60$^{\circ}C$ and pH 5.5-6.0, respectively. The Michaelis constant and maximum reaction velocity of the endoinuinase for inulin were 13.27 g/L and 0.13 g/L$.$min at 55$^{\circ}C$, pH 5.5, respectively. Inulin source had no significant effect on oligosaccharide yield and product composition, although initial production rate differed according to inulin origins. The reaction pH was a critical factor in inulo-oligosaccharide production because considerable free monosaccharides were released, decreasing oligosaccharide yield at low pH conditions. An empirical relationship describing the reaction performance was developed from kinetic data: the time to reach maximum oligosaccharide yield (tw) as a function of initial concentration of inulin (lo) and enzyme (Eo); i.e., log tM = -1.025 log Eo - 0.011 l0 + 2.655.

천연올리고당 및 Pseudomonas속 길항미생물의 단독 및 혼용처리가 고온기 칼랑코에 생육촉진에 미치는 영향 (Effects of Oligosaccharide and Pseudomonas sp. on the Growth of Potted Kalanchoe During Summer Season)

  • 고온기 분식 칼랑코에 재배에 있어 문제는 고온 스트레서에 의한 생장의 억제로, 본 연구는 지하부 및 지상부의 생장을 촉진을 시킬 수 있다고 알려진 근권 생장촉진미생물(Pseudomonas sp. B와 Pseudomonas sp. D2)을 선발하여 사용하였으며 천연물로서는 alginoligosaccharide와 glucosamine oligosaccharide를 사용하였다. 또한 이들의 AG-용액과 단일 및 복합 처리하여 재배 온도차이를 부여한 경우 칼랑코에의 생장에 미치는 영향을 조사하였다. 상이한 지하부 온도조건은 25$^{\circ}C$, 30$^{\circ}C$ 및 35$^{\circ}C$로 처리하여, 2주후 초장, 엽장,엽폭, 엽면적, 엽중, 지상부 생체중, 근장, 근중을 조사하였다. 25$^{\circ}C$처리구에서는 초장, 엽면적, 엽중, 근중 모두 Pseudomonas sp. B와 glucosamine oligosaccharide를 혼합한 처리구가 가장 높았고, 30$^{\circ}C$의 경우 초장과 엽중은 glusosamine oligosaccharide처리에서, 엽면적과 근중은 Pseudomonas sp. D2와 algin-oligosaccharide를 처리구에서 가장 좋은 생장 효과를 나타내었다. 35$^{\circ}C$의 경우는 초장, 엽면적, 엽중, 근중 모두 Pseudomonas sp. B와 glucosamine oligosaccharide를 혼합한 처리구에서 효과가 컸다. 이상의 결과로 보아 고온기 칼랑코에의 분화 재배시 문제가 되는 생장억제현상은 천연제재와 미생물 제재를 혼합처리 함으로써 크게 개선시킬 수 있었다.

Aureobasidium pullulaans를 이용한 Maltose로부터 Isomalto-oligosaccharides의 생산 (Isomalto-oligosaccharide Production from Maltose by Intact Cells of Aureobasidium pullulaans)

  • A Aureobasidium pullulans 균체를 이용하여 malt-ose로부터 isomalto-oligosaccharides를 생산하였다. 저농도의 maltose를 기질로 이용할 경우에는 가수분해활성이 transglucosylation활성보다 매우 강하여 isomalto-oligosaccharides 합성수율이 매우 낮았으나 400g/ P 이상의 고농도기질에서는 isomalto-oligosaccharides 합성이 유리하였다. isomaltomalto-oligosaccharides 생산의 반응 최적조건은 pH 4.5, 온도 $65^{\circ}C$, 균체농도는 기질 그램당 10unit이었다. 이때 생산된 isomaltooligosa-ccharides의 수율은 최고 48.35%였다. 반응초기에 maltose로부터 panose가 선택적으로 생산되었고, maltose가 더 이상 기질로 이용되지 못하는 반응시점에서 panose를 기질로 이용하여 isomaltotriose가 생산되었다.

  • 대두유의 oligosaccharide를 분해할 수 있는 S. uvarum을 이용하여 L. acidophilus와 혼합배양을 행하여 당대사의 상호작용을 검토한 결과 대두유에 glucose, fructose, galactose 등의 단당류를 첨가하여 L. acidophilus 단독배양만으로도 높은 산생성을 보여주었으며 혼합배양의 경우도 단당류를 잘 이용하였다. Sucrose가 첨가된 배지에서의 L. acidophilus의 단독배양에서는 산생성이 낮았으나 혼합배양을 했을 때는 다른 단당류만큼 증가되었다. 배양시간에 다른 soywhey의 당변화를 검토한 결과 L. acidophilus 배양에서는 당변화가 거의 없었으나 S. uvarum은 sucrose, stachyose, raffinose와 같은 oligosaccharide를 분해하고 있어 혼합배양중 oligosaccharide로부터 분해된 단당류를 L. acidophilus가 젖산 발효에 이용하는 것으로 생각되었다.

키토산 올리고당 첨가량에 따른 콩다식의 품질 특성 (A Study on the Quality Characteristics of Soybean Dasik by Addition of Chitosan-Oligosaccharide)

  • Along with development of modem medicine, the average life span has been increased but diseases of lifestyle such as cancer, obesity atheroscerosis, cardiac disorder become social issues. This study was designed to examine the availability of the functional food by adding chitosan-oligosaccharide to Korean traditional food, Soybean Dasik. For the study, 0, 30, 50 and $70\%$ of chitosan-oligosaccharide power that contains $23\%$ of chitosan-oligosaccharide were added to soybean Dasik in proportion to the total powder weight These were compared with the Soybean Dasik itself in relation to the physicochemical characteristics, sensory evaluation, and mechanical characteristics. The results of the research are as follows. Physicochemical characteristics showed that chitosan-oligosaccharide soybean Dasik had less ash, K, and Mn. As for the result of sensory evaluation, Dasik added $0\%$ chitosan-oligosaccharide was evaluated very good in its color. Sweetness and softness was good on $0\%\;30\%\;and\;50\%$ ones, while moistness and overall quality on $30\%\;and\;50\%$ ones. As the test results of mechanical characteristics, the chitosan-oligosaccharide soybean Dasik was good for hardness, cohesiveness, gumminess, and brittleness, especially on $0\%\;30\%\;and\;50\%$ ones. Springiness was good in all groups. In color, L value of chitosan-oligosaccharide soybean Dasik decreased but a and b values increased as the substitution percentage increased Therefore, the right amount of addition fit for the production properties was $30-50\%$ chitosan-oligosaccharide of total powder weight.

Inhibition of Proinflammatory Cytokine-induced Invasiveness of HT-29 Cells by Chitosan Oligosaccharide

  • The effect of chitosan oligosaccharide (COS, 1 kDa${\gamma}$, 10 ng/ml IL-$1{\alpha}$, and 25 ng/ml TNF-${\alpha}$) in HT-29 cells. Inducible nitric oxide synthase (iNOS) expression induced by these cytokines was inhibited by COS. COS pretreatment inhibited the invasiveness of cytokines-treated HT-29 cells through Matrigel-coated membrane in a dose-dependent manner. COS also inhibited cytokines-induced matrix metalloproteinase (MMP)-2 activity. This study shows that proinflammatory cytokines induce NO production, iNOS expression, and invasiveness of human colorectal adenocarcinoma HT-29 cells. COS pretreatment inhibited cytokines-mediated NO production, iNOS expression, and invasiveness of HT-29 cells. These results provide sufficient information for the further development of COS as an antitumor metastatic agent for the treatment of colon cancer.

고정화 세포에 의한 Fructo-oligosaccharides 생산 반응메카니즘에서 물질전달 영향에 대한 수학적 모델 (Modeling the effect of mass transfer on the kinetics of fructo-oligosaccharide production by immobilized cells)

  • Calcium Alginate gel에 고정화된 Aureobasidium pullulans cell을 이용한 fructo-이igosaccharides 생산 반응메카니즘에 있어서 포도당, sucrose 및 fructo-oligosaccharides의 유효확산계수를 구하고, 반응메카니즘에 대한 수학적 모벨을 제안하였다. $770g/\ell$, $860g/\ell$ 의 고농도 당용액에서 이들의 유효확산계수는 $1.2-7.6\times10^{-7}\textrm{cm}^2$/s 범위로 매우 낮은 값을 보였고, 이 결과를 모댈식에서 적용하여 기질농도, bead 크기 및 첨가비율 등을 변화시키면서 실험적으로 검증해 본 결과 실험치와 모델식이 대체로 잘 일하였다.
