Olfactory stimulation activates multiple signaling cascades in order to mediate activity-driven changes in gene expression that promote neuronal survival. To date, the mechanisms involved in activity-dependent olfactory neuronal survival have yet to be fully elucidated. In the current study, we observed that olfactory sensory stimulation, which caused neuronal activation, promoted activation of the phosphatidylinositol 3'-kinase (PI3K)/Akt pathway and the expression of Bcl-2, which were responsible for olfactory receptor neuron (ORN) survival. We demonstrated that Bcl-2 expression increased after odorant stimulation both in vivo and in vitro. We also showed that odorant stimulation activated Akt, and that Akt activation was completely blocked by incubation with both a PI3K inhibitor (LY294002) and Akt1 small interfering RNA. Moreover, blocking the PI3K/Akt pathway diminished the odorantinduced Bcl-2 expression, as well as the effects on odorant-induced ORN survival. A temporal difference was noted between the activation of Akt1 and the expression of Bcl-2 following odorant stimulation. Blocking the PI3K/Akt pathway did not affect ORN survival in the time range prior to the increase in Bcl-2 expression, implying that these two events, activation of the PI3K pathway and Bcl-2 induction, were tightly connected to promote post-translational ORN survival. Collectively, our results indicated that olfactory activity activated PI3K/Akt, induced Bcl-2, and promoted long term ORN survival as a result.
Generally, odorant molecules are detected by olfactory receptors, which are specialized chemoreceptors expressed in olfactory neurons. Besides odorant molecules, certain volatile molecules can be inhaled through the respiratory tract, often leading to pathophysiological changes in the body. These inhaled molecules mediate cellular signaling through the activation of the Ca2+-permeable transient receptor potential (TRP) channels in peripheral tissues. This review provides a comprehensive overview of TRP channels that are involved in the detection and response to volatile molecules, including hazardous substances, anesthetics, plant-derived compounds, and pheromones. The review aims to shed light on the biological mechanisms underlying the sensing of inhaled volatile molecules. Therefore, this review will contribute to a better understanding of the roles of TRP channels in the response to inhaled molecules, providing insights into their implications for human health and disease.
The histology and anatomy of the olfactory organ in Luciogobius guttatus was investigated using a light microscopy and scanning electron microscopy. The paired olfactory organs in the dorsal part of the snout are situated in between the upper lip and the eyes. They consist of two nostrils, one anterior and the other posterior openings, and a single olfactory cavity. The anterior nostril, an incurrent opening, forms a short tubular structure from the skin. The posterior nostril, an excurrent opening, forms a circular structure opened to the exterior. The distributional pattern of the sensory epithelium is a continuous type. The sensory epithelium with numerous-motile cilia is made up of receptor cells, supporting cells, basal cells, and mucous cells. In contrast, the non-sensory epithelium is comprised of stratified epithelial cells and two types of mucous cells, acidic and neutral cells. The cilia number of the receptor cell is in range of 3 to 4 units. Such results in L. guttatus may reflect its ecological habit and microhabitat in the tidal zone with a periodic tide.
흰쥐에 항정신병약물인 haloperidol과 sulpiride를 장기간 투여한 뒤 줄무늬체와 후결절조직에서의 DA 수용체 의 변동에 대해 조사하였다. 이와 함께 apomorphine 투여에 따른 상동행동점수도 평가하였다. 흰쥐에 haloperidol(0.5mg/kg/day) 이나 sulpiride(40mg/kg/day)를 4주간 투여하고, 3일간의 약물 배설기간을 거친 후에 apomorphine(0.5mg/kg)을 투여하여 상동행동점수를 평가하였다. Haloperidol을 투여한 군에서는 대조군이나 sulpiride를 투여한 군에 비하여 상동행동에 대한 접수가 높게 나타났고, sulpiride를 투여한 군에서는 대조군과 유사한 정도의 상동행동을 나타내었다. 한편 줄무늬체 조직의 [$^3H$]spiperone의 결합에 대한 포화실험에서는 대조군에 비해 haloperidol이나 sulpiride를 투여한 군에서 최대결합치가 유의하게 증가되었다. 해리상수는 sulpiride투여시 줄무늬체에서 유의하게 감소되어 있었다. Sulpiride를 투여했을때 haloperidol 투여 시와 마찬가지의 DA수용체의 증식이 일어나지만, sulpiride 투여군에서만 상통행동 접수가 낮은 것으로 볼 때, 뇌의 DA계에서 sulpiride가 haloperidol과는 다르게 작용한다는 것을 간접적으로 시사하고 있다. 후결절에서는 sulpiride와 haloperidol을 투여한 양군에서 모두 최대결합치가 현저하게 증가되어 있었다. 또한 해라상수도 양군의 후결절에서 유의하게 증가되었다. 더욱이, 대조군은 후결절에서의 해리상수가 37.5pM, 줄무늬체에서는 86.2pM로 나타나, 후결절에서 줄무늬체에서보다 친화도가 두배 이상 높게 측정되었다. 위의 결과로 미루어 볼 때 sulpiride는 후결절에서 DA수용체의 증식을 유도할 뿐만 아니라 높은 친화력을 가진 몇몇 DA수용체 아형과도 상호작용을 한다는 것을 나타낸다. 또한 대조군에서 줄무늬체와 후결절에서의 친화도가 다른 점은, [$^3H$]spiperone에 반응한 DA수용체 아형은 줄무늬체와 후결절에서 각기 다르다는 점을 시사하고 있다.
수중과 육상의 이중적인 생활을 하는 남방짱뚱어 Scartelaos gigas (amphibious mudskipper)의 후각기관을 해부학과 조직학적으로 연구하였다. 그 결과 후각기관은 좌우 한 쌍이 존재하며 각 기관은 한 개의 가느다란 긴관 (an elongated canal)과 사비강과 누비강의 두 개의 비강 (ethmoidal and lacrimal nasal sacs)으로 구성되어 있었다. 각 관의 내벽에 발달한 감각 상피는 3~4개의 섬모를 가지는 감각 수용기 세포 (receptor cell), 지지세포 (supporting cell), 기저세포 (basal cell) 그리고 점액세포 (mucus cell)들로 구성되는 다세포성 구조로 이루어져 있었다. 반면에 비감각 상피는 감각기를 갖지 않는 주름진 표면만을 가지고 있었다. 또한 두 개의 비강은 상피 표면에 점액 방울 (mucin droplet)을 가지고 있었다. 따라서 남방짱뚱어의 이러한 후각기관의 특징은 생태적 습성과 서식처에 관련된 반수중 생활 (semi-aquatic life)에 적응한 특징으로 사료된다.
Xenopus를 사용하여 후각원판이 정상적인 상태에서 어떠한 과정을 거쳐 발달하는지 연구하였다. 특히 뉴런의 형태적 분화, 초기 발달과 성숙 양상, 축색과 원시전뇌의 접촉 등에 초점을 맞추었다. 후각원판은 stage 23에 외배엽이 두터워진 형태로 처음 나타나는데, 쌍을 이루며 각각은 표피측에 비신경층(NNL)과 안쪽의 신경층(NL)의 두 층으로 되어 있다. stage 26 후에 원판 세포는 NNL세포 틈을 비집고 상피쪽으로 이동하기 시작하며, stage 28이 되면 선단 돌기가 표피 끝에 도달한다. stage 29/30에는 NL의 기부에서 기부 돌기(미래의 축색)가 나타나 stage 32무렵 종뇌에 도달한다. 시냅스는 stage 37/38에 처음 나타난다. 일부 원판 세포들이 후각 뉴런으로 분화하는 동안 많은 원판 세포들은 기저세포로서 후각상피에 그대로 남아 있다. 연구 결과는 뉴런 외배엽의 NL에서 기원하고 지지세포는 NNL층에서 기원함을 보여주었다. 또한 시냅스 형성 전에 뉴런의 분화가 완성됨으로써 후각뉴런의 분화는 뇌의 발달과 독립적으로 일어나며 뇌의 영향을 받지 않는다는 사실을 알 수 있었다.
한국산 대륙송사리 Oryzias sinensis의 후각기관을 실체현미경과 광학현미경을 이용하여 해부 및 조직 화학적 특징들을 조사하였다. 전체적 구조에서 후각기관은 두부의 주둥이 위에 한 쌍으로 존재하였으며 다소 떨어져있는 전비공과 후비공, 한 개의 비강, 한 개의 비낭으로 구성되었다. 비강내 상피는 감각상피와 비감각상피층으로 구분되었다. 감각상피는 후감각세포, 지지세포, 기저세포 그리고 공포들로 구성되었다. 비감각상피는 층상상피 세포, 산성과 중성 점액세포들로 구성되었다. 비낭의 상피층은 층상상피세포와 배상세포들로 구성되었다. 결과적으로 이러한 대륙송사리의 후각기관의 해부 및 조직화학적 특징들은 농경지에서의 정체되고 오염되어있는 수환경을 반영하는 것으로 사료된다.
본 연구에서는 드렁허리 Monopterus albus 후각기관의 형태 및 조직학적 특성을 실체, 광학 및 주사전자현미경하 관찰을 통하여 조사하였다. 후각기관의 외부 형태는 피부와 평행한 폐쇄형 전비공과 후비공을 나타냈다. 내부 구조는 파이프형 비강과 뒤쪽의 위아래의 두 개 비낭으로 구성되었다. 비강의 내벽은 위중층상피층의 감각상피와 중층편평상피층의 비감각상피, 미확인기관으로 이루어졌다. 감각상피는 후감각뉴런, 지지세포, 기저세포, 림프구로 구성되며, 비감각상피는 중층상피세포와 산성의 점액다당류를 보유하는 점액세포를 보유했다. 특히, 감각상피층 내 모세혈관과 풍부한 진피에서의 혈관화는 논, 농수로, 연못, 늪과 같은 진흙 하상의 정체된 수환경에 적응 된 후각기관 내 부가적인 호흡기능의 증거이며 이는 이전에 원구류 이상의 어류 분류군에서 확인되지 않은 새로운 결과로 확인된다.

Olfactory receptors (OR) are primarily responsible for the detection of odorant molecules. We previously demonstrated that OR7D4, an OR for androstenone, is expressed in the vomeronasal organ and olfactory epithelium tissue of stallions. Recently, the expression of OR1I1 in the human testes was reported and the possible roles of OR1I1 in the testicular cells were suggested. The objectives of this study were 1) to explore the expression of OR7D4 and OR1I1 in stallion testes, and 2) to define the specific localization of OR7D4 and OR1I1 in the testicular tissues. Stallion testicular tissue samples were used for this study. Western blot was performed to confirm the cross-reactivity of OR7D4 and OR1I1 antibody with stallion testicular tissue samples. OR7D4 and OR1I1 gene expressions were investigated using reverse transcription-polymerase chain reaction (RT-PCR) in stallion testes. Immunofluorescence was performed to investigate the expression of OR7D4 and OR1I1 in stallion testicular tissues. The protein bands for OR7D4 and OR1I1 from the testes were observed at approximately 38 kDa and 43 kDa, respectively. The mRNA of OR7D4 and OR1I1 were detected in stallion testes. Immunolabeling of OR7D4 and OR1I1 in the cytoplasm of both spermatogonia and Leydig cells was observed. In conclusion, androstenone and another odorant chemical, which is recognized by OR1I1, may play an important role in stallion testes.
