본 연구에선 바이오부탄올 생산을 위한 추출발효에 적용하고자 부탄올 추출에 효율적이고 미생물에 독성을 주지 않는 적합한 추출용매를 선정하고자 하였다. 추출 용매를 선정하기 위하여 부틸 부틸레이트, oleyl alcohol, 친환경 용매인 소수성 이온성 액체 2 종류를 이용하여 분배계수를 구한 결과, 부틸 부틸레이트와 oleyl alcohol이 높은 부탄올 분배계수를 나타내었고 부탄올 농도 1-20 g/L와 pH 4-5.5 범위에서 80% 이상의 우수하고 안정적인 추출효율을 나타내었다. 미생물에 독성이 없는 oleyl alcohol를 사용한 회분식 추출발효에서는 추출을 하지 않은 회분식 배양보다 11% 향상된 부탄올 생산을 보였다 (11.2 g/L vs. 12.4 g/L). 이는 배양액 내의 부탄올이 추출되어 배양액내의 부탄올 농도가 낮게 유지됨으로써 부탄올 독성 영향이 감소되었고, 이로 인해 향상된 부탄올 생산이 이루어진 것으로 판단된다. Oleyl alcohol:부틸부티레이트 부피비를 9:1로 혼합한 용매를 사용했을 경우는 미생물 생장에는 저해 영향이 미미했으나 부탄올 생산은 추출발효를 하지 않은 회분식 배양에 비해 60%에 그쳤다. 부틸부티레이트를 20% 이상 미생물 생장에 심각한 저해를 주는 것으로 관찰되어졌다. 본 연구를 통해, 실험에 사용된 4가지 추출 용매 중에서 oleyl alcohol이 미생물에게 독성영향을 끼치지 않으면서 부탄올 추출을 효율적으로 할 수 있음을 알 수 있었고, oleyl alcohol을 바이오 부탄올 연속 추출발효에 적용한다면 향상된 부탄올 생산성을 얻을 수 있을 것으로 기대된다.
Videomicroscopy를 이용하여 비이온성 계면활성제 수용액과 oleyl alcohol, oleic acid와 같은 극성 오일을 접촉시켰을 때 일어나는 동적 거동을 관찰하였다. Vertical-stage의 현미경을 사용하여 비이온성 계면활성제 수용액상 위에 동일한 부피의 오일상을 주입하여 두 상을 접촉시켰을 때 일어나는 계면활성들, 특히 detergency 결과에 중요한 영향을 미치는 중간상 형성, 확산에 의한 과포화로 인하여 일어나는 자발적 유화 현상 등을 관찰하였다.극성 오일로 각각 oleyl alcohol과 oleic acid를 사용하여 1wt% $C_{12}E_5$ 수용액과의 동적 거동을 관찰한 결과는 동일한 경향을 나타내었다. 즉 운점보다 낮은 온도 조건에서는 오일상과 계면활성제 수용액상의 상호작용이 거의 없는 것을 관찰할 수 있었으며, oleyl alcohol을 사용하여 운점보다 높은 온도 조건인 $35^{\circ}C$, $40^{\circ}C$, $51.7^{\circ}C$에서 실험한 lamellar liquid crystal이 중간상으로 형성되었고 확산에 의한 과포화로 인한 자발적 유화 현상을 관찰할 수 있었다. 또한 oleyl alcohol 시스템에 대한 개략적인 상 평형도를 이용하여 각 온도 조건에서 oleyl alcohol과 계면활성제 수용액상이 접촉할 때 일어나는 중간상의 생성 및 자발적 유화 현상 등의 동적 현상들을 diffusion path를 사용하여 이해하였다.
용매추출에 의한 구연산발효를 시도하였다. 발효 배지내 구연산 농도와 추출용매의 독성이 구연산의 생산성을 결정하는 주 인자였다. 특히 이온화되지 않은 구연산은 강력한 발효억제효과를 가져 구연산 농도가 $5g/\ell$ 이상이면 구연산 생산은 거의 중지되었다. 구연산 추출효율을 높이기 위해 3차 아민과 oleyl alcohol의 혼합액을 용매로 사용하였다. 구연산의 추출발효에 척함한 용매조성은 30% 아민 70 % oleyl alcohol로 밝혀졌다. 이 조건에서 추출발효시 구연산농도는 $15.2g/\ell$ 로 동임한 조건에셔 용매 를 첨가하지 않고 발효하여 얻은 구연산농도 $5g/\ell$에 비해 3배 이상 높았다. 아빈 농도가 30%를 념으 면 추출능은 향상되나 용매의 독성효과로 생산성은 오히려 감소하였다. 균체에 대한 용매의 독성효과는 균체를 polyure-thane foam에 고정화함으로써 현저하게 줄일 수 있었다.
A 28-kDa extracellular lipase (pI 8.7) was purified to homogeneity from the culture supernatant of Trichosporon sp. Y- 11 by mmonium sulfate precipitation, DEAE-Sephadex A-50, Bio-Gel P-30, CM- Sephadex C-50, and Bio-Gel P- 10 chromatographies. The purified enzyme exhibited a specific activity of $2,741{\;}{\mu}mol/min/mg$ based on the hydrolysis of triolein, and the optimal hydrolysis activity was dentified at pH 8.0 and $40^{\circ}C$. The enzyme activity was inhibited by $Ag^+$ and enhanced by $Fe^{2+}$, $Fe^{3+}$, $Mg^{2+}$, $Mn^{2+}$, and $Li^{+}$. The enzyme activity exhibited for the hydrolysis of both tributyrin and trilinolein. The ester synthesis of unsaturated fatty acids with various alcohols catalyzed by the purified lipase in a nonaqueous medium or microaqueous system was also investigated. The esterification activity of the lipase increased with an increase of the carbon chain length in the alcohol. The synthesis rate of linoleic acid and oleyl alcohol was the highest with an optimal temperature and pH of $40^{\circ}C$ and 8.0, respectively. The water content and agitation also affected the esterification activity of the lipase.
Retinoids in anti-wrinkle cosmetics are widely used for the treatment of the photoaging skin these days, but very unstable in the presence of water. Multilamellar liquid crystal vesicles consisted of $poly(oxyethylene)_{15}oleyl$ ether $(POE_{15}OE)$ or $poly(oxyethylene)_{15}stearyl$ ether $(POE_{15}SE)$, cetostearyl alcohol (CSA) and cholesterol or oils were prepared to increase the stability of retinyl acetate or retinyl palmitate. $POE_{15}SE-CSA-Water$ systems were more unstable than $POE_{15}SE-CSA-Water$ systems due to the bended structure of oleyl groups. For the modification of vesicles, cholesterol was added. The enthalpy values were decreased reaching to zero and the water permeability of systems was decreased, but the stabilizaton of retinyl acetate was not greatly improved. The stability of retinyl acetate may depend on its location in the structure. Various oils were added for further stabilization and isopropyl myristate was most effective. In practice, retinyl palmitate showed better stability in the same vesicle than retinyl acetate, and the improved vesicles could be used for the anti-wrinkle cosmetics
금속공작유에 의해 발생하는 기름얼룩에 대하여 6종의 유지화합물(rapeseed oil, oleic acid, methyl oleate, oleyl alcohol, lauryl amine, lanolin)에 따른 영향을 조사하였다. 시험방법으로는 MIL-C-22235A의 방법을 사용하였으며 base oil(spindle oil; 1.5wt%의 황함유)에 유지화합물 10wt%와 항산화제 0.5wt%를 첨가시켜 도유용 sample을 만들었다. 시험결과 항산화제 중 B. T가 B. H. T.와 T. C. P보다는 기름얼룩 억제에 효과적이었으며, 산가는 기름얼룩 방지에 나쁜 영향을 미쳤다. 유지화합물의 화학구조에 의해 예견되는 바와 같이 oleic acid는 철이온과 반응이 가능하므로 시편에 심하게 기름얼룩을 발생시켰다.
비이온성 계면활성제 $C_{12}E_5$, 비극성 오일 n-hexadecane, 물을 포함한 계에 oleyl alcohol 혹은 oleic acid 같은 극성 성분을 첨가했을 때의 Phase Inversion Temperature (PIT)를 측정한 결과, 첨가한 극성성분은 친유성 첨가제와 같이 PIT를 급격히 감소시킴을 알 수 있었다. PIT에서의 microemulsion phase의 surfactant film 조성 계산에 의하면 surfactant film에서의 극성 성분/계면활성제 비율이 증가할수록 PIT는 감소하였다. 또한 극성 성분의 첨가는 PIT에서의 microemulsion phase의 부피와 오일, 물의 solubilization parameter에도 큰 영향이 있었다. PIT의 감소에 따라, 예를 들면 극성 성분의 첨가량을 증가시킴에 따라, microemulsion phase의 부피는 급격히 증가하였으며 아울러 오일, 물 모두의 microemulsion phase에 대한 solubilization parameter가 급격히 증가하였다. 본 연구 결과를 비교할 수 있는 세척력 실험결과는 현재 없지만 PIT의 측정은 middle-phase microemulsion phase가 형성되는, 즉 많은 양의 오일을 제거할 수 있는 조건을 예측할 수 있는 유용한 방법임을 알 수 있었다.
To increase the skin permeation rate of flurbiprofen, fatty alcohols were added in propylene glycol vehicle containing 1% flurbiprofen. Their enhancing effect on the skin permeation of flurbiprofen was evaluated using Keshary-Chien diffusion cells fitted with excised rat skins. Lauryl alcohol and oleyl alcohol increased the skin permeation rate of flurbiprofen 11.3 and 8.5 fold, respectively, compared to the control vehicle.
Vortices terminating at free surface have been investigated extensively. however. are focused on surface parallel vortices and little has been known about surface normal vortex or columnar vortex. Visualized experimental results utilizing LlF technique are discussed for the purpose of characterization of columnar vortex interacting with a clean and a contaminated free surfaces and a solid body interface in the present investigation. The results reveal that surface tension changes due to surface contamination although bulk viscosity remains constant and eventually the behavior of a columnar vortex interacting with a contaminated free surface and a solid body interface are totally different from the clean free surface case.
