  • Obesity-induced disorders contribute to the development of metabolic diseases such as insulin resistance, fatty liver diseases, and type 2 diabetes (T2D). In this study, we evaluated whether the Aloe QDM complex could improve metabolic disorders related to blood glucose levels and insulin resistance. Male C57BL/6 obese mice fed a high-fat diet for 54 days received a supplement of Aloe QDM complex or pioglitazone (PGZ) or metformin (Met) and were compared with unsupplemented controls (high-fat diet; HFD) or mice fed a regular diet (RD). RT-PCR and western blot analysis were used to quantify the expression of obesity-induced inflammation. Dietary Aloe QDM complex lowered body weight, fasting blood glucose, plasma insulin, and leptin levels, and markedly reduced the impairment of glucose tolerance in obese mice. Also, Aloe QDM complex significantly enhanced plasma adiponectin levels and insulin sensitivity via AMPK activity in muscles. At the same time, Aloe QDM decreased the mRNA and protein of $PPAR{\gamma}/LXR{\alpha}$ and scavenger receptors in white adipose tissue (WAT). Dietary Aloe QDM complex reduces obesity-induced glucose tolerance not only by suppressing $PPAR{\gamma}/LXR{\alpha}$ but also by enhancing AMPK activity in the WAT and muscles, both of which are important peripheral tissues affecting insulin resistance. The Aloe QDM complex could be used as a nutritional intervention against T2D.

  • The present study showed WHtR to be significantly better than BMI and WC for prediction of metabolic-related diseases in the middle-aged and older people in Korea, based on Bayesian ordered probit model analysis. The variations of WC, BMI and WHtR were compared according to the number of metabolic-related diseases such as hypertension, dyslipidemia, stroke, myocardial infarction, angina pectoris and diabetes. It was found that the three measures showed the similar variation except a very few extreme cases for age less than 40. For subjects over the age of 40, WC was not significant and WHtR gave more influence in greater variability than BMI on the number of metabolic diseases. Also, the rate of change for WHtR was higher than for BMI as the number of metabolic-related diseases increased. Specifically, the difference of the marginal effect of WHtR between no disease and only one disease was 1.81 times higher than that of BMI. Moreover, it was pointed out that the threshold value of WHtR for obesity should be considered differently by age.

  • The purpose of this study was to estimate the prevalence of osteoarthritis as well as assess the risk factors associated with osteoarthritis in Koreans over 65 years using data from the Fifth Korea National Health and Nutrition Examination Survey (KNHANES V), 2010~2012. Of the participants from KNHANES V, a total of 3,479 subjects were analyzed using SPSS statistics complex samples (Windows ver. 21.0). Osteoarthritis was more frequently found in female (32.5%) or rural (26.8%) groups than male (9.3%) or urban (20.3%) groups. Mean age of the osteoarthritis group was significantly higher than that of the normal group. Mean values of BMI and waist circumference were significantly higher in the osteoarthritis group than in the normal group, whereas height, fasting blood glucose, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, hemoglobin, and hematocrit levels were not. Health-related quality of life (EQ-5D) was significantly higher in the normal group than in the osteoarthritis group. The results of multiple logistic regression showed that obesity and vitamin A/riboflavin intakes were significantly related to the prevalence of osteoarthritis, whereas smoking, alcohol intake, physical activity, hypertension, hypercholesterolemia, hypertriglyceridemia, anemia, and diabetes were not. This study suggests that obesity and nutrient intakes were associated with osteoarthritis, whereas chronic diseases such as hyper-lipidemia, anemia, and diabetes as well as health habits were not. Prospective research of long-term control is needed to establish the effects of those factors on the osteoarthritis.

  • This study was undertaken to examine relations between obesity and birthweight, parental weight and serum biochemical levels(lipids , glucose, insulin and aminotransferase) in elementary school children aged 10-12 . The weight of obese girls was higher than that of obese boys, while WHR was higher in obese boys than in obese girls. The birth weight of the obese children was significantly higher than that of the non-obese children(p<0.01), and was positively correlated with current weight. The concentrations of serum TG, LDL, VLDL, LDL-cholesterol, glucose, insulin and ALT in obese children were much higher than those in non-obese children . However, the serum HDL -cholesterol concentration of the obese children was significantly lower compared with that of the no-obese children. On the other hand, little differences in these levels were found between genders. The concentrations of PL, TC and AST in the groups showed no significant differences. In the obese children, serum concentrations of TG, LDL-cholesterol , glucose and ALT were more closely associated with GMI than with WHR. No significant correlation was found with serum insulin , glucose and TG levels. These data show that childhood obesity may be related to hyperlipidemia , atherosclerosis, hypertension, diabetes mellitus and fatty liver.

  • Purpose: Metabolic syndrome (also known as insulin resistance syndrome) represents a constellation of hypertriglyceridemia, hypertension, impaired glucose tolerance, and obesity. Presently, the influence of various factors on metabolic syndrome was assessed in patients of a university hospital comprehensive medical examination center. Methods: Age, sex, blood pressure, height, weight, triglyceride level, high-density lipoprotein cholesterol, and glucose levels were measured in 67 people (37 males and 30 females). These factors were correlated with tobacco use, alcohol consumption, and exercise habits. Metabolic syndrome and abdominal obesity were assessed according to NCEP-ATP III criteria and the Asia-Pacific guidelines (male obesity defined as a waist circumference exceeding 90 cm), respectively. Data was analyzed using t-test, 2-test, and logistic regression. Results: Respective percentages were: tobacco use (14.9% of the 67 people), no tobacco use (85.1%), alcohol consumption (62.7%), no alcohol consumption (37.3%), regular exercise (25.4%), no regular exercise (74.6%). Logistic regression analysis revealed a gender-related odds ratio of 2.3 for metabolic syndrome and no exercise. Conclusions: Weight reduction and physical exercise may decrease the prevalence of metabolic syndrome. Early identification of metabolic syndrome and risk factor modification is prudent in cases of obesity, diabetes, hyperlipidemia, and hypertension.

  • Obesity is caused by unbalance of energy intake and expenditure, which results in extra accumulation of adipose tissue. Obesity is directly related to metabolic diseases such as diabetes, hyperlipidemia, fatty liver and so on. To investigate the anti-obesity effects of Scutellariae Radix, 70% EtOH extract and water extract of it were tested by in vitro and in vivo studies of fat accumulation. 3T3-L1 preadipocyte cell line was used in a in vitro study of fat accumulation. After 3T3-L1 cells were induced to differentiate into adipocytes, S. radix extract were added and fat accumulation was measured by oil red O staining. In vivo study showed that weight and epididymal/ retro-peritoneal adipose tissues were significantly reduced in mice fed Scutellariae Radix extract compared with control group. Especially, mice fed Scutellariae Radix extract showed reduced serum triglyceride and glucose levels. When adipose tissues were analyzed by microscope, mean adipocyte size was significantly reduced in Scutellariae Radix extract-fed mice. Therefore, this study showed inhibitory effects of Scutellariae Radix on in vitro and in vivo fat accumulation.

  • Objectives: The aim of this study was to determine the affecting factors of metabolic syndrome (MetS) and it's components in Korean adults in their 30s and 40s. Methods: The present study assessed 1,215 subjects who participated in the Korea National Health and Nutrition Examination Survey in 2014. We analyzed the prevalence rate of MetS and it's components and their affecting factors. Results: The prevalence rate of MetS was 16%. Family history of diabetes mellitus and, low or high saturated fat intake were associated with MetS in males. Low income, low education, excessive sleep, excessive carbohydrate and low saturated fat intake were associated with MetS in females. Triglycerides showed the highest prevalence in males and were related to white collar socioeconomic status, smoking, and obesity. Triglycerides and high-density lipoprotein cholesterol were high in females, and triglycerides were affected by excessive sleep, family history, low saturated fat intake, and obesity. High-density lipoprotein cholesterol was affected by obesity. Conclusions: Because sex differences exist in MetS, it is necessary to intervene through diverse approaches. In particular, a continuous management and preventive intervention for obesity is needed.

  • Obesity increases the risk of mortality and morbidity because it results in hypertension, heart disease, and type 2 diabetes. Therefore, there is an urgent need for pharmacotherapeutic drugs to treat obesity. We performed a screening assay using natural products with anti-adipogenic properties in 3T3-L1 cells and determined that tschimganidine, a terpenoid from the Umbelliferae family, inhibited adipogenesis. To evaluate the anti-obesity effects of tschimganidine in vivo. Mice were fed either a normal chow diet (NFD) or a high-fat chow diet (HFD) with or without tschimganidine for 12 weeks. Treatment with tschimganidine decreased lipid accumulation and adipogenesis, accompanied by reduced expression of adipogenesis and lipid accumulation-related factors. Tschimganidine significantly increased the phosphorylation of AMP-activated protein kinase (AMPK) and decreased that of AKT. Depletion of AMPK relieved the reduction in lipid accumulation resulting from tschimganidine treatment. Moreover, tschimganidine administration drastically reduced the weight and size of both gonadal white adipose tissue (WAT) and blood glucose levels in high-fat diet-induced obese mice. We suggest that tschimganidine is a potent anti-obesity agent, which impedes adipogenesis and improves glucose homeostasis. Tschimganidine can then be evaluated for clinical application as a therapeutic agent.

  • Obesity-induced inflammation plays a crucial role in obesity-related pathologies such as type II diabetes and atherosclerosis. Adipose tissue macrophages and the cell-derived proinflammatory chemokines are key components in augmenting inflammatory responses in obesity. Anthocyanins such as cyanidin and $cyanidin-3-O-{\beta}-D-glucoside$ (C3G) are known to elicit anti-inflammatory activities by suppressing the production of proinflammatory mediators such as tumor necrosis factor alpha and nitric oxide in LPS-stimulated macrophages. In the present study, we investigated whether cyanidin and C3G have the potential to suppress the inflammatory responses of adipose cells. Cyanidin and C3G not only suppressed the migration of RAW 264.7 macrophages induced by mesenteric adipose tissue-conditioned medium, but also inhibited the activation of the cells to produce inflammatory chemokines such as monocyte chemoattractant protein-1 (MCP-1) and macrophage inflammatory protein-related protein-2 (MRP-2) in a dose-dependent manner. Cyanidin and C3G also inhibited the release of MCP-1 and MRP-2 from adipocytes and/or macrophages. These findings suggest that cyanidin and C3G may suppress the inflammatory responses of adipose tissue in obesity.
