Objectives This study was conducted to determine whether nano-sized carbon black exposure results in greater damage in high fat diet-induced overweight rats than normal weight ones and to identify the possible causes of any differences. Methods Two groups of Sprague-Dawley rats allocated by body weight (normal and overweight) were exposed to aerosolized nano-sized carbon black for 6 hours a day, 5 days per week over a 4-week period. Differential cell counts, lactate dehydrogenase (LDH) activities and albumin concentrations were measured in bronchoalveolar lavage (BAL) fluid, and histopathological findings in the lungs were evaluated. Tumor necrosis factor-alpha (TNF-${\alpha}$) and interleukin (IL)-6 were measured in BAL fluid and supernatants of lipopolysaccharide(LPS)-stimulated lymphocyte culture. Results Rats exposed to high concentrations of nano-sized carbon black showed significantly increased (p <0.05) polymorphonuclear leukocyte number and LDH activity in the BAL fluid from both overweight and normal rats. Mild histopathological changes were observed in normal rats irrespective of carbon black concentrations. However, severe histological scores were found in overweight rats ($1.75{\pm}0.46$, $2.25{\pm}0.46$, and $2.88{\pm}0.35$ after low, medium, and high concentration exposures). Proinflammatory cytokine levels of TNF-${\alpha}$ and IL-6 were significantly higher in the supernatant of LPS-stimulated lymphocytes of overweight rats, whereas there was no significant difference in the BAL fluid between normal and overweight rats. Conclusions Inflammation and damage to lungs exposed to nano-sized carbon black was more severe in high fat diet-induced overweight rats compared to normal rats.
This study investigated the effects of Wolbiga-banha-tang (WBBT) on high fat diet-induced rats. We divided the rats into 4 groups: normal (fed a general-fat diet), control (fed a high-fat diet and no treatment), $WBBT{\times}1$ (fed a high-fat diet and 250 mg/kg of Wolbiga-banha-tang extracts), and $WBBT{\times}2$ (fed a high-fat diet and 500 mg/kg of Wolbiga-banha-tang extracts). We measured the body weight, total cholesterol, HDL cholesterol, LDL cholesterol, triglycerides, free fatty acids, total lipids, AST, ALT, and CBC in the serum of the obese rats for 7 weeks. There was a significant decrease in the control group, $WBBT{\times}1$, and $WBBT{\times}2$ in triglycerides and free fatty acids. The results suggest that WBBT is effective for lowering triglycerides and free fatty acids. Therefore, WBBT may be applied to the treatment of lipid metabolism in obesity and further clinical studies are needed.
This study evaluated the effect of biocellulose and the diet formulation on reducing body weight gain of obese rats induced by high-fat diet for 10 weeks. Thirty male Sprague-Dawley rats were randomly assigned to high-fat diet group (OB-CON), high-fat diet group containing 5% biocellulose (OB-BIO), and high-fat diet group containing 5% dietary formulation (OB-DF). After feeding each diet for additional 10 weeks, body weight gains of OB-BIO and OB-DF groups were lower by 3.3 and 4.8%, respectively as compared with that of OB-CON group. Although not significant, measured values of the perirenal and visceral fat pads of OB-BIO and OB-DF groups were lower than those of the OB-CON group. The weight of interscapular brown adipose tissue did not show significant difference in all groups. The size of adipocyte in rats was lower in both OB-BIO and OB-DF groups. Thus, biocelluose and the diet formulation showed the anti-obesity effect.
The protective effects of BNs-3 and BNs-7, extracts of citrous orange peel, on the obesity induced by ad libitum feeding a cholesterol-containing diet to rats were investigated. The animals were fed on the diet including cholesterol (0.5%) with or without the citrous orange peel extracts BNs-3 (5%) and/or BNs-7 (0.1%) for 8 weeks. The ad libitum feeding a diet containing cholesterol to rats from 6 weeks of age increased the body weight gain compared with that of rats fed on a normal diet. Such an increase in body weights was markedly attenuated by the addition of BNs-3 or BNs-7 to the diet. Especially, a combinational feeding on BNs-3 and BNs-7 significantly reduced the body weight gain below that of normal diet-fed animals. Interestingly, the weights of abdominal adipose tis-sues surrounding epididymides were greatly reduced by the citrous orange peel extracts, in parallel with the decrease in body weights. In addition, blood concentrations of lipids including cholesterol were also lowered by the combinational treatment with BNs-3 and BNs-7. Taken together, it is suggested that the obesity and overweight produced by unrestricted overfeeding on diet with cholesterol may be partially due to the accumulation of abdominal adipose tissues, around the epididymides in rats, and that citrous orange peel extracts might exert antiobese activities by reducing the adipose tis-sues as well as blood lipid concentrations.
This study investigated the inhibitory effects of sea mustard on high-fat diet-induced obesity and hyperlipidemia in Sprague-Dawley rats. Sea mustard (Undaria pinnatifida) powder, sea mustard ethanol extract, and sea mustard ethanol-extracted residue were tested. The ethanol extracted residue had the most beneficial anti-hyperlipidemic activity. Alginate in the sea mustard was considered to be the key component. The ethanol-extracted residue of sea mustard also had antioxidant activity, which may be effective in preventing hyperlipidemia by increasing the enzymatic activity of superoxide dismutase, which can remove active oxygen from the bloodstream.

The roots of Rosa rugosa have been used to treat diabetes mellitus in the folkloric society of Korea. To demonstrate the active component for the rat obesity induced by high fat diet for 6 weeks, the phytochemical fractionation and the pharmacological activity test were performed on this crude drug. It was shown that the methanolic extract and its EtOAc fraction inhibited the weight increase of the rat body, abdominal fat pad and hyperlipidemia at 200 mg/kg dose. Further, the triterpenoids, euscaphic acid and tormentic acid, isolated from R. rugosa roots were active at 30 mg/kg in the same assay. The two components shifted serum total-, HDL, and LDL-cholesterol levels toward the values of the unteated group, suggesting that the active compounds has hypolipidemic effects. The rats fad euscaphic acid and tormentic acid also reduced thiobarbituric acid-reactive substance (TBARS) and hydroxyl radical in the rat blood and increased superoxide dismutase activity compared to the control. TBARS values and carbonyl contest of the hepatic protein were reduced by treatment with the two triterpenoids. Antioxidative enzyme (SOD, glutathione peroxidase, and catalase) activities in hepatic were increased by treatment of rats with the triterpenoids, which suggests that triterpenoids inhibited the reduction of hepatic antioxidative activity caused by high fat diet. Taken together, these results support that euscaphic acid and tormentic acid improve a high fat diet-induced hyperlipidemia via the activation of antioxidative mechanism.

The study aimed to investigate the anti-obesity effects of 1% Rhus verniciflua vinegar (RV) supplementation in high-fat-diet (60% fat)-induced obese rats. A total of 50 4-wk-old male Sprague-Dawley rats were fed normal chow diet or maintained on high-fat diet (HFD) for 12 weeks to induce obesity and were then randomized into five groups as follows: normal diet+ultra-pure water (CON), HFD+ultra-pure water (OB-DW), HFD+1% acetic acid (OBAA), HFD+1% RV (OB-RV), and HFD+0.1% caffeine (OB-CF). AA was used as a control for RV, and caffeine was used as a positive control due to its weight reducing effect. After 2 months, body weight, organ and adipose tissue weights, serum lipids, hepatic lipids, adipocyte size, and cell number per spot level were analyzed. As a result, food efficiency ratio, abdominal adipose tissue weight, serum levels of total cholesterol, triacylglycerol, free fatty acids, coronary artery index, and fecal lipid were significantly reduced in the RV treatment group. In this study, we found that dietary RV improved obesity by increasing lipid excretion and reducing lipogenesis. These results suggest that RV has potential as a functional anti-obesity food.

This study was conducted to investigate the effects of ethanol extract from Lycii folium (L. folium) leaves on obesity index, serum lipids, homocysteine, leptin, ghrelin, and glucose in obese rats. Sprague-Dawley rats were administrated high-fat diets to induce obesity. Then, obese rats were divided into three diet groups: a basal-diet obese group (BO group), high-fat diet obese group (FO group), and high-fat diet plus L. folium extract obese group (FLEO group). Three groups were each fed experimental diet for 8 weeks. There were no significant differences in body weight and FER among the groups. However, obesity index such as Ro?hrer index, Lee index, and T.M. index of FLEO group was significantly decreased as compared to FO group. While serum triglyceride of BO group was significantly decreased as compared to FO group, there were no significant differences in serum lipids, homocysteine, leptin, ghrelin and glucose between FLEO group and FO group. In conclusion, these results indicated that ethanol extract from L. folium leaves might be beneficial with anti-obese effect by reduction of obesity indices in obese rats.
This study was performed to investigate the effect of acorn supplementation on the lipid profile and redox antioxidant enzyme activities in obese rat. Obesity in the rats was induced by feeding diet contained 10% lard and 0.5% cholesterol for 4 week. After 4 weeks, rats were divided into the following 5 groups; high fat diet (Control), high fat diet plus 10% Acorn powder (APlO%), high fat diet plus 20% Acorn powder (AP20%), high fat diet plus 0.2% Acorn extract (AE0.2%), high fat diet plus 0.5% Acorn extract (AE0.5%). Total food intake and food efficiency ratio (FER) was not significantly different by acorn powder and extract supplementation. But, body weight was decreased by 20% acorn powder. Acorn powder and extract supplementation for 4 weeks tend to decrease total cholesterol and triglyceride level on the serum and hepatic tissue. There was no significant difference in hepatic glutathione (GSH) content among all the groups. The hepatic GST activity in acorn supplemented groups was lower than that of control. Glutathione peroxidase and catalase activities were higher in acorn supplemented groups than that of control. Hepatic TBARS levels of experimental groups were also significantly lower than that of control group. Our finding suggest that acorn powders and extract might have potential role for improving lipid profiles and antioxidant enzyme activities in obese rats.
