Effect of Mineral-induced Alkaline Reduced Water on Sprague-Dawley Rats Fed on High-fat Diet

  • Mineral-induced alkaline-reduced water (MRW) is generated by the chemical reaction of water with alkaline earth metals and characterized by high pH and low oxidation-reduction potential. As ROS are believed to have a role in the pathogenesis of obesity, we attempted to determine the effect of MRW on obesity in Sprague-Dawley (SD) rats fed on a high-fat diet. The body weight of the MRW group was significantly lower than that of the control group in most periods of the examination (P<0.05). Serum level of triglycerides (p<0.05) and fat deposition in the livers of the MRW group were found to have been significantly reduced. This suggests that MRW down-regulates lipid metabolism, thereby suppressing obesity. Possible mechanisms of MRW related to reactive oxygen species were also discussed. Our results suggest that MRW is effective in the alleviation of obesity in SD rats fed on high-fat diet.

  • In Oriental medicine, there has been a theory that the deficiency of the Qi(氣) and the Phlegm(濕)-Damp(痰) bring Obesity. And so a clinically representative herb-medicine of the obesity treatments is Chegameyiin-tang . We observed the effects of Chegameyiin-tang on the fat tissues and what the function of Chegameyiin-tang is. These experimental studies were designed to investigate the effects of Chegameyiin-tang on the weight and lipid metabolism of obesity rats induced by high fat diet. And what is changed in the blood and how the leptin and uncoupled protein is affected. The measurement has been performed on (1) the weight of obese rats fed high fat diet, (2) the average size and number of epididymal fat cells, (3) the total cholesterol, triglyceride, glucose. and free fatty acids in the blood. and (4) the leptin and uncoupled protein in the blood are observed. The results are as follows; 1. In the sample group, the weight decrease occured significantly throughout the whole research period than that of control group. 2. In the sample group, epididymal fat weight showed significantly decrease in the 8th and 14th weeks than that of control group.3. In the sample group, epididymal fat cell size was decreased significantly in the 8th and 14th weeks than that of control group. 4. In the sample group, total cholesterol. triglyceride and glucose increased rather than control group in 8 weeks, those decreased significantly in 14 weeks. 5. In the sample group, free fatty acids and insulin increased rather than control group in 8 weeks. those showed some decrease in 14 weeks . 6. In the sample group, leptin decreased significantly than control group in 8, 14 weeks. Uncoupled protein showed some decrease in 8 weeks. that decreased significantly in 14 weeks.

Effect of the Combination of Total Saponin of Red Ginseng and Coisis Semen for the Prevention and Treatment of Obesity (홍삼(紅蔘) 총사포닌과 의이인(薏苡仁) 혼합물이 비만(肥滿) 치료 및 억제에 미치는 영향)

  • Objectives: This study was on the pharmaceutical components in purified ginseng total saponin (GTS), coisis semen (CS), the combination of these drugs, and the major component of coicis coixol for the prevention and treatment of obesity. Methods: In this study, to evaluate the effect on the suppression of obesity, high fat diet-induced obese rats were treating with the drugs, the effects on the balance of energy and diet activity were examined, and the change of weight, the change of the intake of diet, body fat rate, etc. were assessed. Results: The results demonstrate that in high fat diet-induced obese white rats, the combination treatment of ginseng total saponin and coicis was effective in suppression of weight gain, reduction of intake of food, and reduction of body fat. Conclusions: The results suggest that a combination treatment with major components of red ginseng total saponin and coicis may be used therapeutically for the suppression and treatment of obesity.

  • This study was performed to investigate the anti-obesitic effect of mulberry root-bark on male and female Zucker FA/FA or FA/fa rats. Obesity in the rats was induced by feeding high-lipid diet contained 3% corn oil and 1% cholesterol. Experimental groups in male and female rats were assigned to normal diet group (normal control), high-lipid diet group(positive control) and 3% mulberry root-bark powder in high-lipid diet group (MRC). The mulberry treated-group showed decreases of body weight, FER (food efficiency ratio) value and lipid peroxidation in the liver and increase of high density lipoprotein cholesterol (HDL-C) value, compared with positive control groups. Our findings suggest that mulberry root-bark has a potential role in preventing or improving obesity from the following points of view, body weight, serum lipids and antioxidant enzyme activities.

Anti-obesity Effect of Fermented Whey Beverage using Lactic Acid Bacteria in Diet-induced Obese Rats

  • High-protein fermented whey beverage (FWB) was manufactured using whey protein concentrate (WPC) and Lactobacillus plantarum DK211 isolated from kimchi. This study was designed to evaluate the anti-obesity activity of FWB in male rats fed a high-fat diet. Male Sprague-Dawley rats were randomly assigned to three groups (n=8 per group). The three groups differed in their diet; one group received a normal diet (ND), another, a high-fat diet (HD), and the third, a HD plus fermented whey beverage (HDFWB), for 4 wk. Supplementation with FWB (the HDFWB group) prevented weight gain and body fat accumulation. The food intake in the HDWFB group was significantly lower (p<0.05) than that of the HD group. The HDWFB group also showed a significant decrease in organ weights (p<0.05), except for the weight of the testis. There was a significant decrease in total cholesterol, LDL-cholesterol, and triglycerides in the HDFWB group compared with the HD group (p<0.05), but there was no significant difference in serum HDL-cholesterol levels among the experimental groups. Rats ingesting FWB (the HDFWB group) also showed a significant decrease in blood glucose levels, and plasma levels of insulin, leptin, and ghrelin compared to HD group (p<0.05). These results indicate that FWB has beneficial effects on dietary control, weight control, and reduction in fat composition and serum lipid level; consequently, it may provide antiobesity and hypolipidemic activity against high fat diet-induced obesity in rats.

The Dietary Effect of Patty Made with Added Glucomannan in High Fat Diet-induced Obese Rats (글루코만난을 첨가하여 제조한 패티가 고지방 식이로 유도된 비만 흰쥐의 체중 감소 및 영양상태에 미치는 영향)

  • This study was carried out to investigate the effect of body weight reduction of patty containing glucomannan in high fat diet-induced obese rats. Male Spraque-Dawley rats were randomly assigned to control and high fat diets groups for 4 weeks. After 4 weeks, the control and high fat diet groups were rearranged into 5 groups by different levels of patty containing glucomannan (60%). After 5 weeks, serum and whole blood levels were analyzed. Obesity index was significantly lower in the group fed patty containing glucomannan than that of high fat diet groups. The status of white blood cells in hematological examination was significantly higher in rats fed high fat diet and not significantly different from those fed patty. Serum albumin levels were not significantly different, while serum glucose levels were significantly different among groups. Serum Triglyceride and total cholesterol levels were the highest in rats fed high fat diets, while the lowest in rats fed patty containing glucomannan.

High Hydrostatic Pressure Extract of Red Ginseng Attenuates Inflammation in Rats with High-fat Diet Induced Obesity

  • Chronic low-grade inflammation is associated with obesity. This study investigated effect of high hydrostatic pressure extract of red ginseng (HRG) on inflammation in rats with high-fat (HF) diet induced obesity. Male, Sprague-Dawley rats (80~110 g) were randomly divided into two groups, and fed a 45% HF diet (HF) and a 45% HF diet containing 1.5% HRG (HF+HRG) for 14 weeks. At the end of the experiment, the serum leptin level was reduced by the HRG supplementation. The mRNA expression of genes related to adipogenesis including peroxisome proliferator-activated receptor-gamma and adipocyte protein 2 was down-regulated in the white adipose tissue (WAT). The mRNA levels of major inflammatory cytokines such as tumor necrosis factor-${\alpha}$, monocyte chemoattractant protein 1, and interleukin-6 were remarkably down-regulated by the HRG in WAT. These results suggest that HRG might be beneficial in ameliorating the inflammation-associated health complications by suppressing adipogenic and pro-inflammatory gene expression.

  • This present study was carried out to investigate the body weight-regulatory effects of Chekamhanghyuluiyiin-tang plus Zeae Stigma in high fat diet-induced obese rats. Control group rats were fed with high fat diet and administered normal saline for 8weeks. Experimental groups rats were fed with high fat diet and administered extract of 2 kind of Chekamhanghyuluiyiin-tang plus Zeae Stigma each other for 8 weeks. And observed that, body weight of rats and total cholesterol, triglyceride, free fatty acid, total lipid, phospholipid, high density lipoprotein(HDL)-cholesterol, low density lipoprotein(LDL)-cholesterol in serum of rats, and glucose, insulin of rats. The results were as follows; There were significant decrease of serum free ratty acid level in 4CH/sub 5/ group. There were significant decrease of serum glucose level in 4CH/sub 10/ group. According to above mentioned results, Chekamhanghyuluiyiin-tang plus Zeae Stigma was expected to be applied to the prevention or treatment of obesity and its complications.

The Effects of Daecheongryoung-tang on Anti-weight Gain and Anti-hyperlipidemic in Obese Sprague Dawley Rat Induced by High Fat Diet (대청룡탕이 비만 유도 흰쥐의 체중 감량과 혈중지질농도 개선에 미치는 영향)

  • Objectives: The aim of this study is to investigate the effects of Daecheongryoung-tang (DCR) therapy on body weight, serum total cholesterol, high density lipoprotein (HDL) cholesterol, low density lipoprotein (LDL) cholesterol, triglyceride, free fatty acid, total lipid, phospholipid level and complete blood cell count of obese rats. Methods: 34 rats are divided into 4 groups, the rats in the normal group are 7 and the rats in the other group are 9 per group; Normal group (general fat diet and no medication), Control group (high-fat diet and no medication), DCR_L group (high-fat diet and DCR 250 mg medication) and DCR_H group (high-fat diet and DCR 500 mg medication). DCR is administrated for 6 weeks. Results: There is significant statistical difference between Control group and DCR-H group for the body weight, the total cholesterol, LDL cholesterol, triglyceride, free fatty acid level. Also, there is significant statistical difference among Control group, DCR_L group and DCR_H group for body weight, triglyceride, free fatty acid and phospholipid level. Conclusions: These results suggest that medication of DCR_L and DCR_H is effective for the treatment of obesity.

Study on Effect of GyeongshinhaeGihwan 1 in Body Weight and Food Intake for High fat Diet Induced Obese Male Rats (고지방식이 수컷 비만백서에서 경신해지환(輕身解脂丸)이 체중 및 사료섭취량에 미치는 영향)

  • To investigate the effect of the frequently used anti-obese medicine GyeongshinhaeGihwan 1 (GGT1), in food intake, body weight and food efficiency ratio for high fat diet induced obese male rats. Also, to value the diffences between GyeogshinhaeGihwan 1 and FDA approved Sibutramine in anti-obesity effect. High fat diet induced obese male rats were classified into four groups - positive control group, negative control group, GyeongshinhaeGihwan 1 group and Sibutramine group - and their food intake and body weight were observed for eight weeks. Anti-obesity effect was estimated with food efficiency ratio which is calculated by weight inclose divided by food intake. The result shown in Fig. 2 suggests that the GyeongshinhaeGihwan 1 group is more effective on food intake control than the Sibutramine group. Average weight variation shows an increase in both positive/negative control group and medication group. Also, the result in Fig. 3 indicates that average food efficiency ratio decreases contrary to the average weight variation. In addition, repeatedly estimated variance analysis on average food efficiency ratio of the GyeonushinhaeGihwan 1 group shows (1) the result corollary to the time of observation of food efficiency ratio was effective under 0.05 variance (P-value 0.000). The differences between each groups were not shown under 0.05 variance. Compared to the control group, medication groups were visually more effective on food intake control. Although both groups had a tendency of weight increasing, food efficiency ratio considering food intake and weight variation rate showed a decrease. Especially, the medication group variated less than the control group corollary to the point of time, proving the individuals react less sensitive to the medicine. Moreover, there were no differences in the anti-obesity effect between GyeongshinhaeGihwan 1 group and Sibutramine group studied by repeatedly estimating variance analysis(P-value: 0.610). When considering Sibutramine as an anti-obesity medicine approved by FDA, the point of being classifed in the same group proves the effect of GyeongshinhaeGihwan 1 as an anti-obesity medicine.
