Attenuated Sympathetic Activity and Its Relation to Obesity in MSG Injected and Sympathectomized Rats

  • In order to characterize the role of sympathetic activity in obesity, we repeatedly assessed sympathetic activity via power spectral analyses of heart rate variability in the same subjects at 7, 11, 25, and 60 weeks, using monosodium glutamate (MSG)-induced obese and control rats. The effects of lower sympathetic activity on obesity were also evaluated. Fat mass in MSG rats was already higher at 7 weeks, but the sympathetic activity did not differ between 7 and 25 weeks. Between 25 and 60 weeks, the increase in fat mass, food efficiency, and body weight gain was higher in MSG rats. The increase in sympathetic activity between 25 and 60 weeks and sympathetic activity at 60 weeks were lower in MSG rats. Fat mass at 60 weeks was inversely correlated with changes in sympathetic activity between 25 and 60 weeks. Reduced plasma epinephrine levels by bilateral adrenal demedullation induced increase of fat mass. In, an attenuated increase of sympathetic activity with age may partly be responsible for aggravated obesity in MSG rats. Additionally, reduced sympathetic activity per se induced obesity in rats. These results suggest that lower sympathetic activity contributes to obesity in rats.

삼황탕(三黃湯)이 고지방식이, MSG 및 난소적출로 유발된 흰쥐의 비만에 미치는 영향 (Effects of Samhwang-tang(Sanhuang-tang) on Obese Rats Induced by High Fat Diet, MSG and Ovariectomy)

  • Objectives : Obesity is a metabolic disease resulted from various causes including nutritional disproportion, hormonal imbalance, and hypothalamic nuclei disorder. Samhwang-tang(Sanhuang-tang, SHT) is composed of 5 herbs (Ephedrae Herba, Scutellariae Radix, Typhae pollen, Acori Rhizoma and Ansu Semen) and is frequently used for obese patients. This study investigated anti-obesity effects of SHT on the rats. Methods : Obesity was induced by high fat diet (HFD), monosodium L-glutamate injection (MSG) and ovariectomy (OVX) in the Sprague-Dawley rats. Water extract of SHT was treated for 2 weeks, then changes of body weight gain and epididymal fat cell size were measured. In OVX animals, c-Fos and neuropeptide Y (NPY) expressions in hypothalamic nuclei were observed with immunohistochemistry. Results : SHT treatment demonstrated significant decrease of body weight gain in obesity induced by HFD and OVX, but not in obesity induced by MSG. SHT treatment demonstrated significant decrease of fat cell size in obesity induced by HFD, but not in obesity induced by MSG. On c-Fos and NPY expressions in hypothalamic nuclei, SHT treatment resulted significant decrease of immuno-reactivity of c-Fos and NPY in obesity induced by OVX. Conclusions : These results suggest that SHT has a weight loss effect against obesity induced by nutritional disproportion of diet intake and hormonal imbalance, especially without central nervous system disorder.

Effects of Hangbisan, an Oriental Medicine, on Body Weight Gain in Diet-Induced Obese (DID) rats

  • This study was designed to determine possible weight loss effects of Hangbisan, an oriental medicine, on Sprague-Dawley (SD) rats. SD rats that were fed a high-fat diet for 6 weeks to induce obesity and subsequently fed with a basic diet containing 10%(w/w) Hangbisan or 10%(w/w) cellulose for 8 weeks. The Hangbisan fed rats demonstrated a significantly (p<0.05) reduced weight gain compared to the cellulose fed rats as well as a reduced level of plasma cholesterol and triglycerides and an increased level of (HDL)-cholesterol. These results suggest that dietary Hangbisan has an anti-obesity effect in the high-fat diet-induced obesity (DIG) rat and therefore a potential use as an anti-obesity agent in the application of oriental medicine compounds.

고지방 식이로 유도된 비만 백서에서 뽕잎 추출물의 항비만 효과 (Anti-obesity Effect of Mulberry Leaves Extraction in Obese Rats High-fat Diet)

  • The aims of this investigation was to examine the anti-obesity effects of oral administration with Mulberry leaves extraction in the high-fat diet induced obesity rats. We investigated the anti-obesity effects of the extract from Mulberry leaves on the progress of high fat diet for 8 weeks. Experimental rats were divided into 5 different experimental groups including an normal group (normal dieat; n=10), control group (high fat diet; n=10), I (Mulberry leaves extraction 100 mg/kg oral administration induced obesity; n=10), II (Mulberry leaves extraction 300 mg/kg oral administration; n=10), and III (Mulberry leaves extraction 500 mg/kg oral administration; n=10). It is to analysis changes in body weight, epididymal fat weight, blood lipid profiles, and fat histological findings. Body weights were significantly increase in control group than normal groups(p<0.05). The level of total cholesterol, LDL-C and TG were significantly lower in experimental groups than control group (p<0.05). In histopathologic finding of fat-tissue around testicle experimental group I and II were more decreased than high fat diet-induced control group in fat cell size. These results suggested that the Mulberry leaves extraction oral administration made the increase of anti-obesity effect in high-fat diet induced obesity rats.

산마늘 기능성 활성물질 탐색 및 자원화 (Effect of Allium victorialis var.platyphyllum Leaves on Triton WR-1339-induced and Poloxamer-407-induced hyperlipidemic Rats and on Diet-Induced Obesity Rats)

  • The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var.platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of IS. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks (p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract of Mt. Odae eco-type showed more potent activity than that of Is. Ullung one. These results suggest that use of the fresh leaves may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.

Anti-Obesity and Hypolipidemic Effects of Dietary Levan in High Fat Diet-Induced Obese Rats

  • We found previously that dietary high fat caused obesity, and levan supplementation to the regular diet reduced adiposity and serum lipids. In the present study, we examined the effects of levan [high-molecular-mass $\beta$-(2,6)-linked fructose polymer] supplement on the development of obesity and lipid metabolism in rats fed with high-fat diet. Thus, to determine whether the dietary levan may have the anti-obesity and hypolipidemic effects, 4-wk-old Sprague Dawley male rats were fed with high-fat diet for 6 wk to induce obesity, and subsequently fed with 0, 1, 5, or 10% levan supplemented high-fat diets (w/w) for another 4 wk. For the comparison, a normal control group was fed with AIN-76A diet. Supplementation with levan resulted in a significant reduction of high-fat-induced body weight gain, white fat (i.e., epididymal, visceral, and peritoneal fat) development, adipocyte hypertrophy, and the development of hyperinsulinemia and hyperlipidemia in a dose-dependent manner. Serum triglyceride and free fatty acid levels were greatly reduced by levan supplementation. Serum total cholesterol level was reduced, whereas the HDL cholesterol level was increased by dietary levan. The expression of uncoupling protein (UCP) was increased by dietary high fat, and was further induced by levan supplementation. The mRNA level of UCP1, 2, and 3 in brown adipose tissue (BAT) and UCP3 in skeletal muscle was upregulated in rats fed with dietary levan. In conclusion, upregulated UCP mRNA expression may contribute to suppression of development of obesity through increased energy expenditure. The present results suggest that levan supplementation to the diet is beneficial in suppressing diet-induced obesity and hyperlipidemia.

고지방 식이를 섭취한 흰쥐의 비만지수 및 식이 아연과 철분 수준에 따른 혈청 미량무기질 함량 변화 (Effects of Dietary Zinc and Iron Levels on Serum Trace Minerals and Obesity Index in High Fat Diet-Induced Obese Rats)

  • The purpose of this study was to evaluate the obesity index and effect of dietary zinc and iron levels on serum trace minerals status in the high fat diet-induced obese rats. Male Sprague-Dawley rats were randomly assigned to control and high fat diet groups. Ten weeks later, the control and high fat diet group were rearranged into six groups by zinc and iron levels. After 16 wk serum zinc, iron, copper and manganese was analyzed. Obesity index was significantly higher in the group fed high fat diet (20% lard) than that of control group (5% corn oil). Body fat content was 12.10$\pm$4.51g/100g BW in high fat diet group and 7.64$\pm$4.18g/100g BW in control group. So, the obese rats were successfully induced by high fat diet. The trace mineral concentration of obese rats in serum were affected by zinc levels. Serum zinc concentration was increased by dietary zinc overload, whereas the iron, copper and manganese were decreased. Specially the manganese concentration was significantly affected by zinc levels. In both groups, serum trace mineral concentration was not changed significantly by the dietary iron levels. There were positive correlations between zinc, iron and manganese concentration according to dietary zinc and iron levels.

High Food Efficiency Ratio of Prepubertal Growth Period Leads to a Long-Term Susceptibility for Obesity and Insulin Resistance in Obesity-Prone and Obesity-Resistant Sprague Dawley Rats

  • Excessive body weight gain during the growth period of early life may predispose individuals towards obesity and metabolic disorder in later life. We investigated the possibility of using the food efficiency ratio as an early indicator for predicting susceptibility to diet-induced obesity and insulin resistance. Four-week-old, prepubertal, male Sprague Dawley rats were divided into obesity-prone and obesity-resistant groups based on food efficiency ratio values after five days on a high-fat diet. Metabolic parameters measured after 2, 6, and 10 weeks, and specific phenotypes were compared with each group. Obesity-prone rats had higher increases in body weight and fat mass compared to obesity-resistant rats over the study period. Obesity-prone rats became glucose intolerant early in this study and remained so throughout the experimental period, with increases in fat weight and leptin levels occurring first, followed by increases in insulin level. Gluconeogenesis and insulin resistance significantly increased in obesity-prone groups in which activities of glucose-6-phosphatase and phosphoenolpyruvate carboxykinase were increased and glucokinase activity decreased. Higher food efficiency ratio at an early age was closely correlated with body fat accumulation, hyperleptinemia, and hyperinsulinemia of middle and elderly age. We suggest a high food efficiency ratio in prepubertal subjects may be a useful predictor of future obesity and insulin resistance.

고지방식이로 유발된 비만 백서에서 가미감비제습탕이 비만 유발에 미치는 영향 및 기전 연구 (Anti-obesity Effects and Mechanism of Original and Modified Gambejaeseup-tang in Female Rats with Diet-induced Obesity)

  • Gambejaeseup-tang (GBJST) have recently been used as an anti-obesity herbal medicine but their effect and mechanism of action have not been studied. We modified ingredients of GBJST based on the previous experiments about exploring herbs to suppress triglyceride accumulation in 3T3-L1 adipocytes. We investigated the effects of modified GBJST on energy, glucose and lipid homeostasis using female rats with diet-induced obesity and their action mechanism was also determined. Rats fed a high-fat diet (HFD) were divided into 3 groups: rats in each group received 0.2 or 2 g water extracts of modified GBJST (L-GBJST or H-GBJST) or 2 g cellulose per kg body weight (a negative control) on a daily basis. A further group was fed a low-fat diet (LFD) as a positive control. We found that modified GBJST dose-dependently decreased body weight and mesenteric and retroperitoneal fat more than the control. This decrease was due to the reduction in energy intake and the increase of energy expenditure. HFD increased fat oxidation more than LFD and modified GBJST further increased fat oxidation as a major energy source more than the control in a dose-dependent manner. In addition, H-GBJST improved glucose tolerance without changing serum insulin levels during an oral glucose tolerance test. H-GBJST also suppressed the increase of serum total and LDL cholesterol and triglyceride levels by HFD. In conclusion, modified GBJST have a good anti-obesity effect by decreasing energy intake and increasing energy expenditure mainly as fat in female rats with diet-induced obesity. It also improves glucose tolerance and lipid metabolism.

  • 고지방 식이 유도 비만 흰쥐에 Monascus purpureus균주로 발효시킨 참당귀 추출물을 식이에 첨가하였을 때 나타나는 체중 및 내장지방의 감소, 혈중 및 조직 내 중성지질 및 과산화지질 수준에 미치는 영향을 평가하였다. FAG섭취로 인해 체중증가 및 내장지방 축적 비율을 효과적으로 낮추었으며, 간 기능 지표로 활용되는 혈중 AST, ALT, LDH 및 ALP 활성 변화는 정상 수치와 비슷한 수준으로 감소되었다. FAG를 섭취함으로서 혈중 중성지질 농도가 정상 수준으로 감소하였으며, 혈중 leptin 호르몬 함량 또한 정상 수치와 비슷한 수준으로 증가하였다. 고지방 식이 섭취로 인한 각 조직 과 혈청 내 과산화지질(TBARS)을 측정한 결과 고지방 식이만 섭취한 C군에 비해 각 실험군에서 유의적으로 감소하였으며, 특히 FAG10군에서 가장 낮은 함량을 나타내었다. Hematoxylin과 eosin 염색 및 Oil red O 염색을 통해 FAG를 섭취함에 따라 간장 내 지질침착도의 감소를 확인였으며, 부고환 주변 지방 조직의 hematoxylin과 eosin 염색으로 지방세포의 크기가 감소되었음을 확인하였다.
